Selective inhibition of replicative and repair DNA synthesis in mouse colon following administration of 1,2-dimethylhydrazine.

Early alterations in normal semiconservative and repair DNA synthesis were determined in gastrointestinal tissues of HalCR mice following administration of the colon carcinogen 1,2-dimethylhydrazine (DMH). Following DMH injections of 60 and 200 mg/kg, normal DNA synthesis was rapidly inhibited in all tissues. The greatest depressions were observed in the descending colon, followed closely by the ascending colon. DNA repair was estimated by measuring unscheduled DNA synthesis. No repair was observable in the descending or ascending colon. The esophagus, forestomach, jejunum, and ileum demonstra

Inhibition of DNA synthesis by 1,2-dimethylhydrazine and methylazoxymethanol acetate in rabbit colon mucosa in organ culture.

When maintained in organ culture, colon mucosa from male New Zealand White rabbits showed a near-normal mucosal morphology and linear incorporation of [3H]thymidine into mucosal DNA up to 36 hours of incubation. Explants from the descending colon had a higher DNA synthetic activity than did other segments of the large bowel. Inhibition of DNA synthesis in colon explants by 1,2-dimethylhydrazine (DMH) and methylazoxymethanol (MAM) acetate was dose-dependent. When DNA synthesis was determined after an 18-hour incubation, MAM acetate inhibited DNA synthesis at concentrations of 50, 100, 150, and

Protective effect of Callistemon citrinus on oxidative stress in rats with 1,2-dimethylhydrazine-induced colon cancer.

Callistemon citrinus has terpenes effective in inducing antioxidant enzymes, an important mechanism involved in cancer chemoprevention. This study investigated the chemopreventive efficacy of herbal preparation of C. citrinus leaves against the oxidative stress produced during the colorectal cancer (CRC) in male Wistar rats. The amelioration of toxicity in a model of CRC induced with 1,2-dimethylhydrazine (DMH) was determined by assessing antioxidant enzymes, phase II enzymes activities and lipid peroxidation (LPO) products after 22 weeks of treatment. C. citrinus was administered at a daily o

Influence of dietary resveratrol on early and late molecular markers of 1,2-dimethylhydrazine-induced colon carcinogenesis.

OBJECTIVE: Colon cancer is an exceptionally aggressive disease, and the use of natural or synthetic substances to prevent or decrease cancer risk without adverse effects remains a major challenge. In this study, the mechanistic basis for the chemopreventive effect of resveratrol (Res) on 1,2-dimethylhydrazine-induced colon carcinogenesis in an rat model was evaluated. METHODS: Rats were randomized into six groups. Group 1 were control rats, group 2 were control rats that received Res (8mg/kg of body weight orally every day), and rats in groups 3-6 were treated once per week with 1,2-dimethylh

Gut-associated lymphoid tissue and 1,2-dimethylhydrazine intestinal tumors in the rat: an histological and immunoenzymatic study.

The association between chemically-induced intestinal carcinoma and gut lymphoid patches was studied in 20 male Sprague-Dawley rats 7 months after the first of 16 weekly injections of 1,2 dimethylhydrazine (DMH). The lymphoid patches of DMH-treated rats and of 14 untreated control animals were systematically studied histologically on sections of "swiss-rolled" whole intestine. It was found that 78% of the small-intestine carcinomas and 73% of the colorectal carcinomas were associated with intestinal lymphoid patches. Furthermore, misplaced and often atypical glandular crypts were often found i

Effect of dietary selenium deficiency on incidence and size of 1,2-dimethylhydrazine-induced colon tumors in rats.

Torula yeast diets deficient (less than or equal to 0.02 ppm) in selenium (Se) and a control diet supplemented with sodium selenite (0.1 ppm Se), were fed to 52 male Sprague-Dawley rats per diet for 23 wk following weaning. 1,2-dimethylhydrazine (DMH) was administered (20 mg/kg body weight) in 20 weekly i.p. injections after 3 wk of feeding. After 23 wk, 67% of the rats fed the Se-deficient diet had intestinal adenocarcinomas versus 63% on the 0.1 ppm Se diets. Diet also had no effect on the number or size of tumors per tumor-bearing animal or on the location of the tumors within the colon. Th

Aberrant and nonrandom methylation of chromosomal DNA-binding proteins of colonic epithelial cells by 1,2-dimethylhydrazine.

The alkylating carcinogen 1,2-dimethylhydrazine (DMH) induces colonic tumors with high incidence. The sites of in vivo modification of target macromolecules were studied using [methyl-3H]DMH. Carcinogen binding to subcellular fractions of colonic epithelial cells was analyzed at time intervals ranging from 10 min to 36 hr, with particular emphasis on the alkylation of nuclear constituents. DMH modifies not only nucleic acids but also histones and other DNA-binding proteins in the target cells. Separation of the 3H-methylated amino acids showed aberrant methylation patterns after exposure to [3

Chemical interactions with herpes simplex type 2 virus: enhancement of transformation by hydrazine and 1,2-dimethylhydrazine.

Quantitative assays for the morphological transformation of 3T3 Swiss mouse cells by herpes simplex type 2 virus (HSV-2) were employed to examine the effect on cell transformation of chemical carcinogens and suspected carcinogens. Exposure of the cells to the chemical compound, followed by virus infection, resulted in enhancement of transformation when compared to that observed with chemical or virus alone. Enhancement occurred in tests utilizing either UV light-inactivated HSV-2 (strain 333) or a temperature-sensitive (ts) mutant of HSV-2 (A8(293)]. A series of seven ts-mutants were tested an

Cell population kinetics of 1,2-dimethylhydrazine-induced colonic neoplasms and their adjacent colonic mucosa in the mouse.

The parameters of cell population kinetics of symmetrical 1,2-dimethylhydrazine-induced colonic neoplasms and their adjacent colonic mucosa in the mouse were analyzed using the fraction labeled-mitoses curve method and compared with those of three groups of epithelial cells in the crypt of the descending colon of normal mouse. The analysis of three groups of epithelial cells in the crypt of normal mouse indicates that differentiation of epithelial cells was associated not only with a smaller proliferative pool of cells but also with a shortening of the duration of G2 phase and a prolongation o

Inhibition of 1,2-dimethylhydrazine-induced neoplasia of the large intestine in female CF1 mice by carbon disulfide.

Disulfiram, diethyldithiocarbamate, and bis-(ethylxanthogen) inhibit symmetrical 1,2-dimethylhydrazine dihydrochloride (DMH)-induced neoplasia of the large bowel in female CF1 mice. These three compounds contain a carbon disulfide (CS2) moiety in their structure. In the present study, CS2 itself was found to inhibit DMH-induced neoplasia of the large bowel in female CF1 mice. Thus a relatively simple compound is now available for investigation of the mechanism of inhibition of DMH-induced neoplasia by a group of sulfur-containing inhibitors having common structural features.

L-theanine's therapeutic effects on colorectal cancer: Mechanistic insights from a 1,2-dimethylhydrazine-induced rat model.

This study aimed to elucidate the novel effects and underlying mechanisms of L-theanine on colorectal cancer (CRC) in a rat model. Specifically, it focused on how L-theanine uniquely impacts cell proliferation, apoptosis, epithelial-mesenchymal transition (EMT), oxidative stress, inflammation, intestinal microbiota, and short-chain fatty acids in rat CRC compared to existing studies. In the 1,2-dimethylhydrazine (DMH) group, serum levels of CRP, AFP, CEA, and CA199 were elevated compared to controls, but L-theanine treatment reduced these levels. IHC analysis showed a significant decrease in K

Protective effect of p-coumaric acid against 1,2 dimethylhydrazine induced colonic preneoplastic lesions in experimental rats.

Oxidative stress and gut microbial enzymes are intricately linked to the onset of colon carcinogenesis. Phytochemicals that modulate these two factors hold promise for the development of such agents as anticancer drugs. The present study evaluates the chemopreventive potential of p-coumaric acid (p-CA) - a phenolic acid in rats challenged with the colon specific procarcinogen DMH (1,2 di-methyl hydrazine). Rats were randomized into six groups (n=7/group). Group 1 (control); Group 2 (p-CA 200mg/kg b.w.); Group 3 (DMH 40mg/kg b.w.); Groups 4 (DMH+p-CA 50mg/kg b.w.) and Group 5 (DMH+p-CA 100mg/kg

Food deprivation increases apoptotic cell counts induced by 1,2-dimethylhydrazine in rat descending colonic and rectal crypts.

The frequency of apoptosis after treatment with 1,2-dimethylhydrazine (DMH) was counted in the descending colonic and rectal crypts of food-deprived and fed rats. Food-deprived or fed rats were subcutaneously injected with DMH (100 mg/kg body weight). Six hours after the injection, apoptotic cells were observed in crypt regions by light microscopy. The incidence of DMH-induced apoptosis in food-deprived rats was significantly higher than in fed rats. The incidence appeared to be higher in descending colon than in rectum. PAS staining revealed that DMH treatment lowered mucin secretion in crypt

Early stages of 1,2-dimethylhydrazine-induced colon carcinogenesis suppress immune-regulated ion transport of mouse distal colon.

Genetic events, such as gene deletion, mutation, and amplification, are involved in the development and progression of colonic neoplasia. The importance of interactions between immune cells and neoplastic cells in influencing the pathogenesis of colon cancer is suggested by the clinical efficacy of levamisole, an immunostimulant, in the treatment of colon cancer and by the association of local lymphocytic infiltration with improved prognosis. Thus, the immune cell-neoplastic cell interaction can be viewed as being analogous to various host-parasite relationships that involve the immune system

Metabolism of the carcinogen 1,2-dimethylhydrazine by isolated human colon microsomes and human colon tumor cells in culture.

Human colon microsomes catalyze the metabolism of the model colon carcinogen 1,2-dimethylhydrazine. Activity appears to be distributed in a gradient towards the lower end of the colon. Highest activities were observed for microsomes prepared from the descending segment of the colon with the transverse segment exhibiting lower activities, while the ascending segment showed the lowest rate of metabolism. Dimethylhydrazine metabolism in each segment is inhibited significantly by inhibitors of the cytochrome P-450-dependent mixed function oxidase system. Microsomes prepared from a human colon tumo

Effect of metabolic inhibitors, methylxanthines, antioxidants, alkali metals, and corn oil on 1,2-dimethylhydrazine carcinogenicity in rats.

The effect of the oral administration of 10 compounds on 1,2-dimethylhydrazine (DMH) carcinogenesis was investigated in 180 male Wistar rats and 510 male BD6 rats. DMH, administered s.c. once per week for 20 consecutive weeks (20 mg/kg body wt/dose), produced intestinal (mainly colon) tumors of various histological type in 100% of both rat strains and, in addition, caused Zymbal gland carcinomas in 79.7% of Wistar rats. Pretreatment with disulfiram (DSF, 500 mg/kg), a known inhibitor of DMH metabolism, totally prevented intestinal and Zymbal gland tumors in Wistar rats. When DSF treatment star

Different dose-response relationships in the induction of different types of colonic tumors in Wistar rats by 1,2-dimethylhydrazine.

Four groups of 21 male Wistar rats were given weekly subcutaneous injections of 1,2-dimethylhydrazine at a dose of 20 mg/kg body weight for 2, 4, 6 and 8 weeks, respectively, and 7 animals of each group were killed in weeks 24, 32 and 40. This treatment induced a clear dose-related increase in the total number of colonic carcinomas. The number of tubular adenocarcinomas, which were mostly located in the descending colon, was clearly dose-related, whereas the number of signet ring cell carcinomas, which developed in the ascending colon, was apparently dose-independent.
