Effect of Cyclodextrin Complex Formation on Solubility Changes of Each Drug Due to Intermolecular Interactions between Acidic NSAIDs and Basic H2 Blockers.

One of the solubilization of poorly water-soluble drugs is the use of cyclodextrin (CD)-based inclusion complexes. On the other hand, few studies have investigated how CD functions on the solubility of drugs in the presence of multiple drugs that interact with each other. In this study, we used indomethacin (IND) and diclofenac (DIC) as acidic drugs, famotidine (FAM) and cimetidine (CIM) as basic drugs, and imidazole (IMZ), histidine (HIS), and arginine (ARG) as compounds structurally similar to basic drugs. We attempted to clarify the effect of β-CD on the solubility change of each drug in th

Cyclodextrin inclusion complex enhances solubility and antimicrobial activity of chlortetracycline hydrochloride.

Chlortetracycline hydrochloride (CTC) is a broad-spectrum tetracycline antibiotic used to prevent or resist different bacterial infections. However, due to its poor aqueous stability and physicochemical instability, its clinical development is limited. To improve such properties, CTC complexation with β-cyclodextrin (β-CD) and 2-hydroxypropanol-β-cyclodextrin (HP-β-CD) was performed using different preparation techniques (Freeze drying and ball milling). Inclusion complexes were comprehensively characterized using different analytical techniques and showed different properties distinct to pure

Cyclodextrin-Mediated Enantiomeric Separation of Idelalisib: A Validated Capillary Electrophoresis and NMR Study.

Idelalisib (IDE) is a marketed chiral anticancer drug administered as the S-enantiomer, requiring sensitive monitoring of the R-enantiomer to ensure enantiomeric purity. However, no dedicated capillary electrophoresis (CE) method has been reported for trace-level quantification of R-IDE. In this study, a cyclodextrin-mediated CE method was developed for reliable detection of the R-enantiomer at the 0.1% level (LOD 2 µg/mL; LOQ 5 µg/mL). Systematic screening identified hydroxypropyl-β-cyclodextrin (HP-β-CD) with an intermediate degree of substitution (DS~6.8) as the optimal chiral selector, pro

Cyclodextrin complexes of sulfonamide carbonic anhydrase inhibitors as long-lasting topically acting antiglaucoma agents.

Complexes of several 1,3,4-thiadiazole-2-sulfonamide derivatives possessing strong carbonic anhydrase (CA) inhibitory properties with beta-cyclodextrin and hydroxypropyl-beta-cyclodextrin were obtained and characterized. Although the investigated CA inhibitors possessed very powerful inhibitory properties against the two CA isozymes involved in aqueous humor production within the eye, i.e., CA II and CA IV, these compounds were topically ineffective as intraocular pressure (IOP) lowering agents in normotensive/hypertensive rabbits, due to their very low water solubility. On the contrary, the c

α-Cyclodextrin decreases cholera toxin binding to GM1-gangliosides.

Cholera toxin (CT), the principal virulence factor secreted by Vibrio cholerae, is an A-B5 type exotoxin that binds to host cell GM1-gangliosides and is responsible for cholera diarrhoea. We tested the hypothesis that the cyclic hexasaccharide α-cyclodextrin (α-CD), but not the cyclic heptasaccharides methyl-β-cyclodextrin (MD-β-CD) and hydroxypropyl-β-cyclodextrin (HP-β-CD) inhibit binding of CT to GM1-gangliosides. We report that α-CD decreases CT binding to GM1-ganglioside-coated microtitre plate wells and on the surface of fixed HeLa cells in a concentration-dependent manner, suggesting th

Spectroscopic Study of R- and S-Ketoprofen Inclusion Complexes With 2-Hydroxypropyl-β-cyclodextrin.

Ketoprofen, also known as (RS)-2-(3-benzoylphenyl)-propionic acid, has the molecular formula C16H14O3 and is classified as a non-steroidal anti-inflammatory medication. R-ketoprofen has stronger analgesic effects than S-K, and as a result, there is a growing interest in enantio-recognition research, which may be accomplished through supramolecular interactions, particularly host-guest reactions. A 1:1 molar ratio was used to produce the combination of separate RK and SK and a 0.01 M stock solution of HP-β-CD to get the final concentration of 6 × 10-4 M. Ethanol was used to make stock solutions

Dynamic high-resolution computer simulation of isotachophoretic enantiomer separation and zone stability.

The development of electrophoretic computer models and their use for simulation of electrophoretic processes has increased significantly during the last few years. Recently, GENTRANS and SIMUL5 were extended with algorithms that describe chemical equilibria between solutes and a buffer additive in a fast 1:1 interaction process, an approach that enables simulation of the electrophoretic separation of enantiomers. For acidic cationic systems with sodium and H3 0(+) as leading and terminating components, respectively, acetic acid as counter component, charged weak bases as samples, and a neutral

Dissolution, cyclodextrin-enhanced solubilization, and mass removal of an ideal multicomponent organic liquid.

Laboratory experiments and mathematical modeling were conducted to examine the influence of a hydroxypropyl-beta-cyclodextrin (HPCD) solution on the dissolution of single- and three-component organic liquids. The results of batch experiments showed that HPCD-enhanced solubilization of the organic-liquid mixtures was ideal (describable using Raoult's Law), and that solubilization-enhancement factors were independent of mixture composition. Addition of the HPCD solution to columns containing residual saturations of the organic liquid enhanced the dissolution and removal of all three compounds in

Preparation and validation of cyclodextrin-based excipients for radioiodinated hypericin applied in a targeted cancer radiotherapy.

BACKGROUND: Iodine-131 labeled hypericin (131I-Hyp) has been utilized as a necrosis-avid theragnostic tracer in a dual targeting pan-anticancer strategy called OncoCiDia. Widespread use of previously-tested solvent dimethyl sulfoxide (DMSO) is limited by safety concerns. To tackle this, the present study was designed to explore a clinically feasible excipient for the formulation of the hydrophobic 131I-Hyp for intravenous administration. METHOD: Solubility of Hyp in serial solutions of already-approved hydroxypropyl-β-cyclodextrin (HP-β-CD) was evaluated by UVspectrophotometry and 50% HP-β-CD

Prediction of PAH biodegradation in field contaminated soils using a cyclodextrin extraction technique.

Biodegradation has been identified as a major loss process for organic contaminants in soils and, as a result, microbial strategies have been developed for the remediation of contaminated land. Prediction of the biodegradable fraction would be important for determining bioremediation end-points in the clean-up of contaminated land. The aim of this study was to investigate the ability of a cyclodextrin extraction to predict the extent to which polycyclic aromatic hydrocarbons (PAHs) would be degraded microbiologically in field contaminated soils; further testing the robustness and reproducibili

Prediction of polycyclic aromatic hydrocarbon biodegradation in contaminated soils using an aqueous hydroxypropyl-beta-cyclodextrin extraction technique.

This study investigated the use of an aqueous hydroxypropyl-beta-cyclodextrin (HPCD) shake extraction to predict the degree of microbial degradation of polycyclic aromatic hydrocarbons (PAHs) in soils. Three different aged PAH-contaminated soils were studied: A soil from a former coke works (CW) and two artificially contaminated soils (AC1 and AC2). First, the catabolic activity of the indigenous soil microflora was assessed with 14C-respirometry, using a range of 14C-labeled aromatic compounds. Extensive mineralization of several compounds occurred in the CW and the AC2 soils, suggesting that

Hydroxypropylcyclodextrins in parenteral use. II: Effects on transport and disposition of lipids in rabbit and humans.

Hydroxypropyl ethers of cyclodextrins, after parenteral administration, come into contact with lipids in tissues and in circulation and form water-soluble inclusion complexes with these lipids. A single intravenous administration of hydroxypropyl-beta-cyclodextrin to a hereditary hyperlipidemic Watanabe rabbit slightly and temporarily decreased the level of total cholesterol in serum. Single injections of hydroxypropyl-alpha-cyclodextrin and of the corresponding gamma-homologue, both of which are less potent solubilizers of cholesterol, had lesser effects. Enhancing the dissolution of PCE with the mixed cyclodextrin-ethanol solution for aquifer remediation.

Groundwater contaminated by dense non-aqueous phase liquids (DNAPLs) such as chlorinated hydrocarbons is difficult to remediate. The combination of flushing agents has a synergistic solubilization effect on the removal of DNAPLs. However, the solubilization mechanisms of DNAPLs under the mixed flushing agent treatment remains poorly understood. In this work, the dissolution process of tetrachloroethylene (PCE) in Hydroxypropyl-β-cyclodextrin (HPCD)-ethanol mixed solution system was observed through one-dimensional (1D) capillary experiments. The mixed HPCD-ethanol solution has a synergistic so

Effects of cyclodextrin in two patients with Niemann-Pick Type C disease.

Niemann-Pick Type C disease (NPC) is an autosomal recessive lysosomal storage disorder characterized by progressive neurological deterioration. Currently, there are no effective treatments for NPC, although miglustat has shown some effectiveness in stabilizing neurological status in juvenile-onset NPC patients. Recent studies have demonstrated the efficacy of hydroxypropyl-β-cyclodextrin (HPB-CD) in NPC mice. Herein, we describe the effects of HPB-CD in two patients with NPC. The two patients received HPB-CD infusions twice (Patient 2) or thrice (Patient 1) weekly, starting with a dose of 80 m

Molecular Mind Games: The Medicinal Action of Cyclodextrins in Neurodegenerative Diseases.

Cyclodextrins are often used as molecular carriers for small active ingredients in medicine. Recently, the intrinsic medicinal activity of some of these compounds has been under investigation, mainly related to their ability to interfere with cholesterol and, therefore, prevent and treat cholesterol-related diseases such as cardiovascular disease and neuronal diseases arising from altered cholesterol and lipid metabolism. One of the most promising compounds within the cyclodextrin family is 2-hydroxypropyl-β-cyclodextrin (HPβCD), owing to its superior biocompatibility profile. This work presen

Supramolecular interaction of ketoprofen with native and hydroxypropyl beta-cyclodextrin: Improved solubility and anti-inflammatory.

Ketoprofen (KTP), an active pharmaceutical ingredient with poor solubility in water and other aqueous solvents, has gained attention for its improved solubility and quicker therapeutic effects when combined with cyclodextrin (CD). This study thoroughly examines how varying concentrations of CD affect KTP, leading to notable increases in absorbance and fluorescence intensity. The binding constant for the 1:1 complex is determined from these changes. Ultrasonication enabled the successful formation of inclusion complexes (ICs) of KTP with β-cyclodextrin (βCD) and hydroxypropyl-β-cyclodextrin (Hβ

Evaluation of pharmaceutical excipients as cosolvents in 4-methyl umbelliferone glucuronidation in human liver microsomes: applications for compounds with low solubility.

Standard incubation procedures for carrying out microsomal assays involve the use of less than 1% w/v organic solvents to minimize the potential inhibitory effects of organic solvents on metabolic activity. This presents a practical limitation for poorly soluble xenobiotics, which cannot be incubated at concentrations high enough to obtain a V(max), and therefore subsequent values for K(m) and Cl(int) cannot be calculated. Our goal was to study the application of a variety of pharmaceutical excipients to aid the solubilization of compounds in vitro in glucuronidation incubations, without affec
