[The effect of melatonin and mexamine on the human thyroid under in-vitro conditions].

In in vitro study of the human euthyroid and thyrotoxic thyroid gland melatonin (N-acetyl-5-methoxytryptamine) and, to a lesser measure mexamine (5-methoxytryptamine) had a dose-dependent inhibiting effect on thyroxine secretion. Moreover, melatonin weakened the TSH stimulating effect in relation to the secretory process in the thymus while mexamine did not. Despite the similarity in the quality of the effect of both methoxyindoles on the release of thyroxine, the mechanism of its realization differs: the action of melatonin is mediated by the adenylate-cyclase-cAMP system, but in the action o

Melatonin deacetylase activity in the pineal gland and brain of the lizards Anolis carolinensis and Sceloporus jarrovi.

Melatonin modulates a variety of rhythmic processes in vertebrates, and is synthesized in both the retina and pineal gland. We have shown previously that retinal melatonin is deacetylated generating 5-methoxytryptamine, which is then deaminated by monoamine oxidase, producing 5-methoxyindoleacetic acid and 5-methoxytryptophol. This process occurs within the eyes of a variety of vertebrates including the iguanid lizard Anolis carolinensis. To determine whether melatonin deacetylase activity also occurs in the pineal organ or in other parts of the lizard brain, pineals and brains of Anolis carol

Examination of pineal indoles and 6-methoxy-2-benzoxazolinone for antioxidant and antimicrobial effects.

Oxidative modification of low density lipoprotein (LDL) induced by free radicals is implicated in the development of atherosclerosis. The aim of the present study was to examine the ability of various pineal indoles in inhibiting LDL oxidation which is accompanied by an increase in mobility in agarose gel electrophoresis and by an augmented generation of thiobarbituric acid-reactive substance induced by Cu2+. It was found that the order of potencies in inhibiting malondialdehyde formation was 5-hydroxytryptamine (serotonin)>5-hydroxytryptophol and 5-hydroxyindole-3-acetic acid when tested at 4

Melatonin deacetylation: retinal vertebrate class distribution and Xenopus laevis tissue distribution.

Deacetylation is a rapid clearance mechanism for ocular melatonin. We have studied the distribution of retinal melatonin deacetylase activity among vertebrate classes. Exogenous radiolabeled melatonin is metabolized by ocular tissue prepared from the amphibian Xenopus laevis, the reptile Anolis carolinensis, the teleost fish Carassius auratus, and the bird Gallus domesticus. In contrast, we were unable to detect ocular melatonin breakdown in rat or pig. In each species exhibiting ocular melatonin breakdown, melatonin is first deacetylated to 5-methoxytryptamine, which is deaminated, producing

Indoleamines and 5-methoxyindoles in trout pineal organ in vivo: daily changes and influence of photoperiod.

This study describes the diel rhythms in several indoleamines, melatonin, and related 5-methoxyindoles in the pineal organ of rainbow trout in vivo. In addition, the effect of different photoperiod conditions was evaluated. Melatonin levels displayed clear daily rhythms in the pineal organ of rainbow trout kept experimentally under long (LD 16:08), neutral (LD 12:12), and short (LD 08:16) photoperiods. Duration of melatonin signal was dependent on the night length of prevailing photoperiod, while peak amplitude was higher when lengthening the photoperiod. Significant daily rhythms in 5-HT cont

Identification of Gut Microbiome Signatures Associated with Serotonin Pathway in Tryptophan Metabolism of Patients Undergoing Hemodialysis.

Serotonin, a tryptophan metabolite, exerts a significant influence on both brain and gut functionality. While previous research has elucidated the intricate dynamics of the gut-brain axis, the interplay between serotonin pathway metabolites and gut microbiota in individuals undergoing hemodialysis remains largely unexplored. Therefore, this study aimed to investigate gut microbiota composition corresponding to serotonin pathway metabolite levels among patients with hemodialysis. A total of 85 patients undergoing hemodialysis were selected. Their gut microbiota was analyzed using shotgun metage

[Epiphysis cerebri-thyroid interrelations. Distorting effect of partial thyroidectomy on the antithyroid action of epiphyseal methoxyindoles in animal experiments].

The effect of pineal methoxyindoles (MI) on hypothalamus-hypophysis-thyroid gland system was studied in intact and partially thyroidectomized male Wistar rats in conditions of a short light day (winter). Melatonin administration for 10 days suppressed 131I uptake by the thyroid gland and decreased the levels of T3, T4 and thyrotropic hormone (TTH) in the blood serum of intact animals, with TTH reaction to thyroid hormone (TH) retained. 5-methoxytryptamine administration was less effective. Partial thyroidectomy distorted the direction of MI effect: melatonin and to a lesser extent 5-methoxytry

Biosynthetic Origin of the Methoxy Group in Quinine and Related Alkaloids.

Quinine is a historically important natural product containing a methoxy group that has been assumed to be incorporated at a late pathway stage. Here we show that the methoxy group in quinine and related alkaloids is introduced onto the starting substrate tryptamine. Feeding studies definitively show that 5-methoxytryptamine is utilized as a quinine biosynthetic intermediate in planta. We discover the biosynthetic genes that encode the responsible oxidase and methyltransferase, and we use these genes to reconstitute the early steps of the alkaloid biosynthetic pathway in Nicotiana benthamiana

Inhibitory action of melatonin and structurally related compounds on testosterone production by mouse Leydig cells in vitro.

The possible effect of melatonin, 5-methoxytryptamine, 5-methoxytryptophol, 6-chloromelatonin and 2-iodomelatonin on testosterone production by Leydig cells in vitro was investigated. The ability of individual indoles to inhibit testosterone production was found to depend on the concentration used. The relative inhibitory potency of the compounds tested was: 6-chloromelatonin greater than 2-iodomelatonin greater than melatonin greater than 5-methoxytryptamine greater than 5-methoxytryptophol. The results revealed that natural indoles which are synthesized in the pineal gland and their halogeni

Secretion of methoxyindoles from trout pineal organs in vitro: indication for a paracrine melatonin feedback.

Synthesis and release of the pineal hormone melatonin is in all vertebrates primarily regulated by the light/dark cycle. In pineal organs of teleost fish, like in other non-mammalian vertebrates, melatonin formation is regulated by a direct photoreception of the pineal organ. We performed measurements in explanted, perifused pineal organs of the rainbow trout, Oncorhynchus mykiss, to examine whether melatonin can influence its own production. For this purpose we have continuously perifused isolated pineal organs under light- and dark-adapted conditions and measured the release of melatonin and

Effect of pinealectomy and 5-methoxyindoles on Harderian gland activity of the Indian Jungle bush quail Perdicula asiatica.

The present study reveals that the annual Harderian gland weight cycle of P. asiatica has a direct relationship with the testicular cycle and an inverse relationship with the pineal gland cycle. Pinealectomy resulted in a significant decrease of the Harderian gland weight during the recurdescence (March-May) and active (April-June) phases. Subcutaneous implantation of melatonin (aMT) and 5-methoxytryptamine (MT) brought about a significant reduction during the recrudescence and active phases whereas subcutaneous injections of aMT, MT and 5-methoxytryptophol (ML) reduced Harderian gland weight

"Synaptic" ribbons and spherules lacking in the pineal gland of the European hamster appear after ganglionectomy.

"Synaptic" ribbons and spherules are typical organelles of the mammalian pinealocytes, that present quantitative day-night variations parallel to pineal melatonin levels. The European hamster is a wild photoperiodic mammal whose pineal melatonin concentrations do not vary during the 24 hr light-dark cycle. The aim of this study was thus to see if a rhythmic variation in synaptic structures was nevertheless present in this species. It was found that, in contrast to all of the photoperiodic species studied to date, the European hamster pinealocyte contains no ribbons, and virtually no spherules.

Histological studies on the effects of pineal 5-methoxyindoles on the reproductive organs of the male golden hamster.

The effects of late-afternoon injections of melatonin (MEL), 5-methoxytryptamine (MTN), 5-methoxytryptophol (MTOL), and 5-methoxyindole-3-acetic acid (MIAA) on testicular and seminal vesicular histology in the golden hamster were examined. MEL, MTN, and MTOL injections caused a reduction in the diameters of seminiferous tubules and an inhibition of spermatogenesis. Testicular regression ranged from a decrease in the abundance of late spermatids and mature spermatozoa in some animals to an almost complete loss of spermatogenesis in others. Sertoli cells were more resistant to the treatment than

A foxy intoxication.

Foxy is the colloquial name for the hallucinogen 5-ethoxy-diisopropyltryptamine (5-MeO-DIPT). A non-fatality involving a 23-year-old Caucasian man who ingested a capsule containing 5-MeO-DIPT is described. He presented to the Emergency Department, not with visual nor auditory hallucinations but with sensory hallucinations, that of formication and paranoia. He was observed and given supportive care for 4 h, then discharged without any known sequelae. Blood and urine were collected for laboratory analyses. Foxy and its metabolites were identified in urine by gas chromatography-mass spectrometry.

[Epiphyseal-thyroid interrelations: effect of calcitonin on indole metabolism in health and in the presence of an excess of thyroid hormones].

A new regularity was established in a study of epiphyseal thyroid interrelationships: the opposite effect of the thyroid hormones (thyroxin and calcitonin) on the formation of methoxyindoles (melatonin, 5-methoxytryptamine) in the epiphysis. Thyroxin was shown to stimulate and calcitonin to inhibit O-methylation of serotonin in the pineal gland, and combined administration of both hormones levelled their action on one another. The inhibitory effect of calcitonin was more marked if a high basal level of melatonin was stimulated by preliminary administration of thyroxin. The authors assume that

[Morphofunctional changes in the pineal gland during adaptation to hypothermia].

The influence of hypothermal stress (+4 degrees during 3 h) on the ways of serotonin metabolism in pineal gland and its structure has been studied in dynamics on adult male Wistar rats. It has been revealed that melatonin-producing epiphyseal function suffers from phase changes in dynamics of adaptation--significant rising during 15 min. after beginning of the experiment, rehabilitation up to normal--in 30 min, and fast suppressing--in 3 hrs. Suppressing of the functional pineal activity is not due to switched serotonin metabolism with melatonin and new indoles release, but to a partial pineal
