Valproic Acid Induces Decreased Expression of H19 Promoting Cell Apoptosis in A549 Cells.

It has been suggested that the imprinted gene, H19, plays a crucial role in the development of cancer. In the present study, we attempted to treat the abnormal expression and methylation status of H19 in A549 cells using valproic acid (VPA), ascorbic acid (Vc), and 5-aza-Cytidine (5-Aza). The results suggested that VPA administration could alter the expression pattern of H19, while the hypomethylation status of H19 DMR was unchanged. Furthermore, overexpression of HDAC1 and DNMT1 was associated with decreased expression of H19 in VPA-treated cells. Western blot results showed that the expressi

DYRK1B overexpression promotes senescence-like transcriptional and secretory programs in A549 lung cancer cells.

The dual-specificity tyrosine-regulated kinase DYRK1B is frequently expressed at elevated levels in solid tumors and has been implicated in cancer cell survival, therapy resistance, and cell cycle arrest. While DYRK1B is best known for maintaining quiescence, its broader impact on cancer cell phenotypic plasticity remains incompletely understood. Using a doxycycline-inducible A549 lung adenocarcinoma model, we show that sustained overexpression of catalytically active DYRK1B induces senescence-related phenotypic changes. DYRK1B overexpression led to increased senescence-associated β-galactosid

Streptococcus pneumoniae and Influenza A Virus Co-Infection Induces Altered Polyubiquitination in A549 Cells.

Epithelial cells are an important line of defense within the lung. Disruption of the epithelial barrier by pathogens enables the systemic dissemination of bacteria or viruses within the host leading to severe diseases with fatal outcomes. Thus, the lung epithelium can be damaged by seasonal and pandemic influenza A viruses. Influenza A virus infection induced dysregulation of the immune system is beneficial for the dissemination of bacteria to the lower respiratory tract, causing bacterial and viral co-infection. Host cells regulate protein homeostasis and the response to different perturbance

Vitamin D Inhibits Lipopolysaccharide (LPS)-Induced Inflammation in A549 Cells by Downregulating Inflammatory Cytokines.

BACKGROUND Studies have shown that lung inflammation affects lung function, with life-threatening results. Vitamin D may play an important role in inhibiting inflammatory cytokines. Vitamin D deficiency is related to several lung problems, including respiratory distress syndrome, alveolar inflammation, epithelial damage, and hypoxia. Few studies have evaluated the benefits of vitamin D in preventing inflammation in alveolar cells. MATERIAL AND METHODS We developed a cell inflammation model induced by lipopolysaccharide (LPS) treatment. The effects of vitamin D on LPS-induced inflammation in A5

New copper complexes inducing bimodal death through apoptosis and autophagy in A549 cancer cells.

Two copper complexes, Cu1 (CuL1Cl2, L1 = 2-(6,7-dimethoxyisoquinolin-1-yl) aniline) and Cu2 (CuL2Cl2, L2 = 2-(6-methoxyisoquinolin-1-yl) aniline), were synthesized and characterized. These complexes exhibited high cytotoxic activity toward different cancer cell lines, including the A549 lung cancer cell line, and low cytotoxicity toward normal human cells. Mechanistic studies have shown that these complexes induce bimodal death of cancer cells through apoptosis and autophagy, including the activation of apoptotic and autophagic cell signaling pathways. In addition, Cu1 and Cu2 interacted with

Zika virus subversion of chaperone GRP78/BiP expression in A549 cells during UPR activation.

Flaviviruses replicate in membranous factories associated with the endoplasmic reticulum (ER). Significant levels of flavivirus polyprotein integration contribute to ER stress and the host cell may exhibit an Unfolded Protein Response (UPR) to this protein accumulation, stimulating appropriate cellular responses such as adaptation, autophagy or cell death. These different stress responses support other antiviral strategies initiated by infected cells and can help to overcome viral infection. In epithelial A549 cells, a model currently used to study the flavivirus infection cycle and the host c

Feroniellin A-induced autophagy causes apoptosis in multidrug-resistant human A549 lung cancer cells.

Источник пометил публикацию как отозванную

During the screening of natural chemicals that can reverse multidrug resistance in human A549 lung cancer cells resistant to etoposide (A549RT-eto), we discovered that Feroniellin A (FERO), a novel furanocoumarin, shows toxicity toward A549RT-eto cells in a dose- and time-dependent manner. FERO reduced the expression of NF-κB, leading to downregulation of P-glycoprotein (P-gp), encoded by MDR1, which eventually sensitized A549RT-eto cells to apoptosis. FERO specifically diminished transcription and promoter activity of MDR1 but did not inhibit the expression of other multidrug resistance genes

Psoralen reverses docetaxel-induced multidrug resistance in A549/D16 human lung cancer cells lines.

Chemotherapy is the recommended treatment for advanced-stage cancers. However, the emergence of multidrug resistance (MDR), the ability of cancer cells to become simultaneously resistant to different drugs, limits the efficacy of chemotherapy. Previous studies have shown that herbal medicine or natural food may be feasible for various cancers as potent chemopreventive drug. This study aims to explore the capablility of reversing the multidrug resistance of docetaxel (DOC)-resistant A549 cells (A549/D16) of psoralen and the underlying mechanisms. In this study, results showed that the cell viab

Effects of Mycoplasma pneumoniae infection on sphingolipid metabolism in human lung carcinoma A549 cells.

The role of sphingolipids in bacterial pathogenesis has been gradually recognized. In an effort to identify the possible involvement of sphingolipids during Mycoplasma pneumoniae (M. pneumoniae) infection, we first adopted a lipidomic approach to achieve the profiles of major sphingolipid species of M. pneumoniae as well as human lung carcinoma A549 cells, and further evaluated the effects of M. pneumoniae infection on sphingolipid metabolism in A549 cells. It was shown that M. pneumoniae and A549 cells share many common sphingolipid species, however, M. pneumoniae possesses certain specific m

ATM activation accompanies histone H2AX phosphorylation in A549 cells upon exposure to tobacco smoke.

BACKGROUND: In response to DNA damage or structural alterations of chromatin, histone H2AX may be phosphorylated on Ser139 by phosphoinositide 3-kinase related protein kinases (PIKKs) such as ataxia telangiectasia mutated (ATM), ATM-and Rad-3 related (ATR) kinase, or by DNA dependent protein kinase (DNA-PKcs). When DNA damage primarily involves formation of DNA double-strand breaks (DSBs), H2AX is preferentially phosphorylated by ATM rather than by the other PIKKs. We have recently reported that brief exposure of human pulmonary adenocarcinoma A549 cells or normal human bronchial epithelial ce

Phosphorylation of p53 protein in A549 human pulmonary epithelial cells exposed to asbestos fibers.

We examined effects of asbestos exposure on the phosphorylation of p53 protein in human pulmonary epithelial type II cells (A549), which express wild-type p53. In cells exposed to two different types of asbestos, chrysotile (approximately 1-6% iron content) and crocidolite (approximately 27% iron content) fibers, at the doses of 1, 5, and 10 microg/cm2 for 24 hr, the levels of p53 phosphorylated at Ser15 and p53 protein were correlated with the dose. On a per-weight basis, chrysotile was more potent in inducing Ser15 phosphorylation and accumulation of p53 protein than was crocidolite. After e

[Action of calmodulin antagonist on the proliferation and cytoskeleton of A549 lung cancer cells].

Trifluoperazine (TFP) could inhibit the proliferation of A549 lung cancer cells and human embryo lung (HEL) cells. The inhibitory effects were dose dependent. The inhibitory effects were stronger in A549 lung cancer cells than in HEL cells at the same TFP dose. Cytoskeleton in A549 lung cancer cells treated with TFP was increased as observed electron microscopically. The role of calmodulin in the proliferation and the changes in cytoskeleton of A549 lung cancer cells is discussed.

Coralyne enhances radiosensitivity of A549 cells by suppressing radiation-induced pro-survival transcriptional programs.

Radioresistance remains a major cause of radiotherapy failure in non-small cell lung cancer (NSCLC), driven in part by rapid activation of pro-survival transcriptional programs following ionising radiation (IR). Targeting this adaptive response represents a promising strategy for radiosensitisation. Here, we show that coralyne, a synthetic protoberberine isoquinoline alkaloid, enhances A549 cell radiosensitivity by suppressing radiation-induced transcriptional reprogramming. Coralyne interacts with DNA and is associated with alterations in radiation-responsive transcriptional programs. Clonoge

Time-resolved proteomic and phosphoproteomic profiling of Angiotensin-(1-7) signaling in A549 cells.

Angiotensin-(1-7) [Ang-(1-7)] is a heptapeptide of the renin-angiotensin system (RAS) with antitumoral effects reported in various tumoral cell lines, including the human lung adenocarcinoma A549 lineage. While previous studies have shown that Ang-(1-7) modulates MAPK and PI3K-AKT signaling, the precise molecular mechanisms involved remain incompletely understood. To investigate the signaling events of Ang-(1-7) in lung cancer-derived cells, we employed an integrated proteomic and phosphoproteomic approach in A549 cells. We analyzed early (minutes) and late (hours) molecular responses to Ang-(

Yellow Fever Virus Induces Golgi Stress and CREB3L1 Nuclear Translocation in Human A549 Cells.

Yellow fever is an endemic disease in Africa and South America caused by the homonymous flavivirus (YFV). The disease can manifest as a febrile syndrome and, in the most severe cases, liver failure and death. Although it can be prevented by a vaccine, there is no specific treatment. Recent studies have reported a Golgi stress response following infection with other flaviviruses such as DENV or ZIKV. Here, we investigated the effects of YFV infection on the phenotype of the Golgi apparatus and the consequences of drugs that affect Golgi function in YFV replication using the human lung cancer ce

MUC5B regulates the airway inflammation induced by atmospheric PM2.5 in rats and A549 cells.

Atmospheric PM2.5 can induce airway inflammation and mucin secretion. MUC5B is required for airway defense. However, the research on the role of MUC5B in airway inflammation induced by atmospheric PM2.5 remains limited. This study was designed to explore the role of MUC5B in airway inflammation induced by atmospheric PM2.5. In vivo, Wistar rats were exposed to 0, 1.5, 7.5, 37.5 mg/ kg PM2.5 saline suspension via intratracheal instillation. HE staining and AB-PAS staining were used to observe the airway inflammation and goblet cell hyperplasia. In vitro, normal A549 cells and MUC5B-knockdown A5
