Determinants of personal demand for an AIDS vaccine in Uganda: contingent valuation survey.

OBJECTIVE: To assess the factors affecting demand for an HIV/AIDS vaccine among adults in their prime earning and childbearing years and the impact of vaccination on risk behaviour in a high-prevalence, low-income country. METHODS: A contingent valuation survey of 1677 adults aged 18-60 years was conducted in 12 districts in Uganda. Respondents were asked about a hypothetical vaccine to prevent HIV infection. Households were randomly assigned survey questionnaires with one of two levels of vaccine efficacy (50% or 95%) and one of five prices. The influence of demographic characteristics, vacc

Control of a mucosal challenge and prevention of AIDS by a multiprotein DNA/MVA vaccine.

Heterologous prime/boost regimens have the potential for raising high levels of immune responses. Here we report that DNA priming followed by a recombinant modified vaccinia Ankara (rMVA) booster controlled a highly pathogenic immunodeficiency virus challenge in a rhesus macaque model. Both the DNA and rMVA components of the vaccine expressed multiple immunodeficiency virus proteins. Two DNA inoculations at 0 and 8 weeks and a single rMVA booster at 24 weeks effectively controlled an intrarectal challenge administered 7 months after the booster. These findings provide hope that a relatively si

Co-design and user testing of a Japanese encephalitis vaccine decision aid (JEVaDA).

BACKGROUND: Japanese encephalitis (JE) is a rare but potentially serious infection in travellers. While effective vaccines are available, uptake remains low. Vaccine decision aids are evidence-based tools designed to help users make informed vaccination decisions. This study details the development of a novel web-based Japanese encephalitis vaccine decision aid (JEVaDA) for travellers, following globally recognised standards. METHODS: Collaborating with community members, healthcare providers and experts, we followed a multi-step approach, involving a scoping review, a survey of user needs, c

Computer-Aided Design of an Epitope-Based Vaccine against Epstein-Barr Virus.

Epstein-Barr virus is a very common human virus that infects 90% of human adults. EBV replicates in epithelial and B cells and causes infectious mononucleosis. EBV infection is also linked to various cancers, including Burkitt's lymphoma and nasopharyngeal carcinomas, and autoimmune diseases such as multiple sclerosis. Currently, there are no effective drugs or vaccines to treat or prevent EBV infection. Herein, we applied a computer-aided strategy to design a prophylactic epitope vaccine ensemble from experimentally defined T and B cell epitopes. Such strategy relies on identifying conserved

Polio, hepatitis B and AIDS: an integrative theory on a possible vaccine induced pandemic.

The hypothesis that simian virus 40 (SV40) infected polio vaccines may be linked to the evolution of acquired immunodeficiency disorder (AIDS), and certain cancers, has been advanced. Most recently, investigators discussed the likelihood of gene-reshuffling following SV40 infection as a precursor to acquired immune dysfunction. Findings of recent SV40 infections in four children born after 1982 suggest infections were transmitted vertically along gene lines. Earlier observations proved activation of a retrovirus gene by a hepatitis B virus (HBV) protein. This paper proposes a new integrative t

The dilemma of AIDS vaccine and therapy. Possible clues from comparative pathogenesis with measles.

AIDS viruses, because of their unique properties, are extraordinary. Past successes achieved with vaccines against ordinary viruses do not provide the guidelines needed to develop successful vaccines against HIV. Neither vaccines nor drugs can be relied upon to provide an answer to AIDS. AIDS is a disease of immune dysfunction and destruction, and an alternative to prevention of infection or cure might lie with elimination of the clinical consequences of infection. This might find a basis in precise definition of its pathogenesis. The enormity of possible pathogenetic changes in HIV infection

Therapeutic Transcutaneous Immunization with a Band-Aid Vaccine Resolves Experimental Otitis Media.

Transcutaneous immunization (TCI) is a noninvasive strategy to induce protective immune responses. We describe TCI with a band-aid vaccine placed on the postauricular skin to exploit the unique organization of the stratum corneum and to promote the development of immune responses to resolve active experimental otitis media due to nontypeable Haemophilus influenzae (NTHI). This therapeutic immunization strategy induced significantly earlier resolution of middle ear fluid and rapid eradication of both planktonic and mucosal biofilm-resident NTHI within 7 days after receipt of the first immunizin

Protection against late-onset AIDS in macaques prophylactically immunized with a live simian HIV vaccine was dependent on persistence of the vaccine virus.

This is a 5-year follow-up study on 12 macaques that were immunized orally with two live SHIV vaccines, six with V1 and six with V2. All 12 macaques became persistently infected after transient replication of the vaccine viruses; all were challenged vaginally 6 mo later with homologous pathogenic SHIV(KU-1). Two of the V1 group developed full-blown AIDS without evidence of vaccine virus DNA in tissues. The data on the 10 vaccinated survivors showed that all 10 became infected with SHIV(KU-1) and that DNA of both vaccine and SHIV(KU-1) viruses were present 6 mo postchallenge, with minimal repli

Dendritic cell-based vaccines, a promising prospect for ending AIDS and eradication of HIV worldwide: A narrative review on the latest updates.

Significant challenges are presented as a result of the ongoing HIV/AIDS pandemic, particularly given the ongoing burden of new infections and the lack of a proven treatment. Among other approaches, vaccinations based on Dendritic Cells (DCs) have become a viable tactic in the fight against HIV. These vaccines take advantage of DCs' special capacity to deliver antigens, which makes them crucial mediators between innate and adaptive immune responses. DC-based vaccines seek to offer a prolonged and effective protection against the virus by stimulating immunological responses against HIV-infected

Immunoinformatics-Aided Design of a Multiepitope Vaccine Against Eimeria Tenella-Associated Chicken Coccidiosis.

Avian coccidiosis, caused by Eimeria protozoa, presents a significant threat to poultry, with Eimeria tenella being particularly harmful due to its impact on the chicken cecum. Growing resistance to current treatments necessitates alternative therapeutic approaches. Consequently, this study employed an immunoinformatics approach to design a multiepitope vaccine targeting E tenella. Key proteins, including the sporulated oocyst TA4 antigen, alkylglycerone-phosphate synthase, and apical membrane antigen-1, were analysed for epitope prediction. Further comprehensive downstream analysis identified

Immunoinformatics-aided design of a potential multi-epitope peptide vaccine against Leishmania infantum.

Visceral leishmaniasis (VL) or kala-azar, the most severe form of the disease, is endemic in more than eighty countries across the world. To date, there is no approved vaccine against VL in the market. Recent advances in reverse vaccinology could be promising approach in designing the efficient vaccine for VL treatment. In this study, an efficient multi-epitope vaccine against Leishmania infantum, the causative agent of VL, was designed using various computational vaccinology methods. Potential immunodominant epitopes were selected from four antigenic proteins, including histone H1, sterol 24-

[Studies on virulence of HIV and development of non-virulent live AIDS vaccine using monkeys].

A great effort for developing AIDS vaccine has been carried out in the world, designed by various new ideas based on basic research information obtained in recent virology and immunology. Withall it, to obtain effective AIDS vaccine is considered skeptical. One of the reasons of its difficulty is a lack of experimental animals susceptible to HIV-1. In our laboratory, we have succeeded in developing chimeric SIV having 3' half of HIV-1 genome including env (SHIV), which is infectious to macaque monkeys. One of SHIVs has been proved nonpathogenic in monkeys from various aspects and it afforded p

Increasing prebunking effectiveness to aid disinformation-susceptible publics in overcoming vaccine-related disinformation.

In the dynamic intersection of health communication and disinformation, this study investigates tailored intervention strategies for disinformation-susceptible public segments classified by the motivation-knowledge-attitude framework. Using a two-phase survey and experimental design with American adults, participants were segmented into four public types (disinformation-immune, vulnerable, receptive, and amplifying) and exposed to one of three inoculation message conditions: autonomy supportive, autonomy controlling, or control. Data were analyzed using ANCOVA with Bonferroni-adjusted post-hoc

Co-designing a shared digital decision aid for HPV vaccination in French general practice.

BACKGROUND: Human papillomavirus (HPV) is responsible for the most common sexually transmitted infection in the world and persistent infection of oncogenic types is associated with an increased risk of cancers. This infection could be prevented by vaccination. However, in a time of vaccine hesitancy, the decision to get vaccinated or to have a child vaccinated is the result of a complex process and decision aids are tools that may be of help in this context. OBJECTIVE: The aim of this article is to present the development of a shared decision aid for HPV vaccination in France, destined to be

Real-time prediction model of cVDPV2 outbreaks to aid outbreak response vaccination strategies.

BACKGROUND: Circulating vaccine-derived poliovirus outbreaks are spreading more widely than anticipated, which has generated a crisis for the global polio eradication initiative. Effectively responding with vaccination activities requires a rapid risk assessment. This assessment is made difficult by the low case-to-infection ratio of type 2 poliovirus, variable transmissibility, changing population immunity, surveillance delays, and limited vaccine supply from the global stockpile. The geographical extent of responses have been highly variable between countries. METHODS: We develop a statisti
