[Necrotizing scleritis after acanthamoeba keratitis].

Nodular scleritis and necrotizing scleritis are rare complications of acanthamoeba keratitis. This article presents the case of a 61-year-old female patient who had suffered from persistent keratitis in the right eye for more than 4 months. The patient was initially treated with propamidine isethionate and polyhexamethylene biguanide eye drops. A penetrating limbo-keratoplasty was performed. Examination of the corneal explant showed acanthamoeba cysts. In the following 5 months the sclera showed recurrent abscesses. A total of two thermal cauterizations and three amniotic membrane grafts were

[Delayed course of Acanthamoeba keratitis].

A 35-year-old male presented with corneal ulceration on the left eye with a history of treatment over several months. At the first visit in our department we saw an elliptically shaped ulcerative stromal keratitis with circular peripheral neovascularization. There was organized hypopyon with hyphemia. The best corrected visual acuity (BCVA) was light perception. The patient had used contact lenses for many years. Under the suspicion of herpetic keratitis due to a positive "dendrite" the patient had undergone antiviral therapy for 6 months in a different department. Our diagnosis was Acanthamoe

Photochemotherapeutic strategies against Acanthamoeba keratitis.

Here, we determined the potential of photochemotherapy, namely the application of photodynamic compounds followed by exposure to a suitable source of UV-visible radiation against corneal pathogen, Acanthamoeba. Organometallic macromolecule, tin porphyrin [Sn(IV)porphyrin] was synthesized and purity confirmed using nuclear magnetic resonance spectroscopy. The Sn(IV)porphyrin was tested against a keratitis isolate of Acanthamoeba castellanii belonging to the T4 genotype using growth and viability assays. The effects of Sn(IV)porphyrin on A. castellanii binding to and cytopathogenicity of human c

Subepithelial infiltrates in Acanthamoeba keratitis.

Six patients had Acanthamoeba keratitis that manifested an unusual type of subepithelial corneal infiltrate late in the course of the disease. These infiltrates occurred in the anterior corneal stroma, often in a location removed from the site of infection. In four of these patients the infiltrates occurred with no other clinical signs of inflammation. Two patients had mild foreign-body sensation that resolved despite the persistence of the subepithelial infiltrates. The infiltrates resolved with topical corticosteroid therapy in three patients; however, in two of these three patients the infi

Naegleria and Acanthamoeba infections: review.

Infections caused by small, free-living amebas are still unfamiliar to many clinicians, pathologists, and laboratorians. As of 31 July 1989, more than 140 cases of primary amebic meningoencephalitis caused by Naegleria fowleri and more than 40 cases of granulomatous amebic encephalitis caused by Acanthamoeba species (including two cases in patients with AIDS) and possibly by other free-living amebas had occurred worldwide. The recent increase in acanthamoeba keratitis (more than 200 cases), especially in contact lens wearers, has generated new interest in this group of amebas. Effective treatm

Acanthamoeba keratitis due to Acanthamoeba genotype T4 in a non-contact-lens wearer in Turkey.

An otherwise healthy 22-year-old man presented with Acanthamoeba keratitis (AK) in the right eye. He was not a contact lens wearer and had no history of corneal trauma. The Acanthamoeba strain isolated from a corneal scraping was identified as morphological group II and genotype T4. Three more Acanthamoeba strains isolated from sites of possible human contact with acanthamoebae in the same geographical region, including a lens storage case, tap water and soil, were subjected to morphological and molecular biological identification. Whereas the strain from tap water also exhibited genotype T4,

Effects of electroporation on Acanthamoeba Polyphaga.

BACKGROUND: Species of the genus Acanthamoeba spp. are ubiquitous and can cause Acanthamoeba keratitis (AK), a serious corneal infection. Due to the toxicity and ineffectiveness of currently available prolonged therapies, we investigated electroceutical treatment aimed at facilitating the permeation of molecules through the membrane of cysts and trophozoites, which allows for faster elimination of the parasite. METHODS: Cysts and trophozoites of Acanthamoeba polyphaga (ATCC® 30461TM) were exposed in vitro to an electric field with intensities of 2,000 volts and 2,500 volts. Viability after el

Incidence of Acanthamoeba Keratitis in Switzerland.

Despite rising global reports of Acanthamoeba keratitis (AK), the incidence of AK in Switzerland remains unknown. This investigator-initiated, retrospective, multicenter study assessed the nationwide incidence of PCR- and/or culture-positive Acanthamoeba results from January 2010 to December 2023. Data were collected from all tertiary care and large ophthalmological facilities in Switzerland, fully anonymized, and aggregated by month and year. We considered all corneal scraping results, whereby the detection method was specific to local standards. We identified 271 PCR- or culture-positive Aca

Biological characteristics and pathogenicity of Acanthamoeba.

Acanthamoeba is an opportunistic protozoa, which exists widely in nature and is mainly distributed in soil and water. Acanthamoeba usually exists in two forms, trophozoites and cysts. The trophozoite stage is one of growth and reproduction while the cyst stage is characterized by cellular quiescence, commonly resulting in human infection, and the lack of effective monotherapy after initial infection leads to chronic disease. Acanthamoeba can infect several human body tissues such as the skin, cornea, conjunctiva, respiratory tract, and reproductive tract, especially when the tissue barriers ar

Acanthamoeba as a temporal vehicle of Cryptosporidium.

The capacity of Acanthamoeba to predate Cryptosporidium oocysts was demonstrated. A maximum of six oocysts per Acanthamoeba trophozoite were detected, and a slow elimination of the internalized oocysts to the surrounding culture medium was observed. Free-living amoebae may act as carriers of Cryptosporidium oocysts and, thus, may play an important role in the transmission of cryptosporidiosis.

Phagocytosis and pinocytosis in Acanthamoeba castellanii.

Endocytotic activity of Acanthamoeba trophozoites attenuates once the cells enter stationary phase in liquid culture. Phagocytosis, monitored by the ingestion of polystyrene latex beads, essentially ceases and the uptake of [3H]inulin, known to be mediated by pinocytosis, is reduced by about half. The reduced pinocytotic activity of stationary-phase cells remains sensitive to respiratory inhibitors. Preincubation of stationary-phase cells in fresh growth medium for 1-5 h before the initiation of endocytosis has no effect on phagocytosis and only marginally increases pinocytosis. This impairmen

Enhancing Acanthamoeba diagnostics: rapid detection of viable Acanthamoeba trophozoites and cysts using viability PCR assay.

Acanthamoeba keratitis (AK) is a sight-threatening corneal infection that is challenging to diagnose and treat due to the resistance of Acanthamoeba to standard antimicrobial agents. Current detection methods have limitations. This study aimed to develop and validate a sensitive viability PCR (v-PCR) assay using a photoreactive dye to distinguish viable from non-viable Acanthamoeba for rapid identification of viable Acanthamoeba trophozoites and cysts. Propidium monoazide (PMAxx) was used as a photoreactive dye. Mixtures containing decreasing percentages of viable Acanthamoeba, including refer

The increasing importance of Acanthamoeba infections.

Free-living amebae belonging to the genus Acanthamoeba are the causative agents of granulomatous amebic encephalitis, a chronic progressive disease of the central nervous system, and of amebic keratitis, a chronic eye infection. Granulomatous amebic encephalitis occurs more frequently in immunocompromised patients while keratitis occurs in healthy individuals. The recent increased incidence in Acanthamoeba infections is due in part to infection in patients with acquired immune deficiency syndrome, while that for keratitis is due to the increased use of contact lenses. Understanding the mechani

Acanthamoeba keratitis: report of a case.

Acanthamoeba keratitis, rarely seen in Taiwan, has aroused much attention in western countries in recent years because of the increasing number of reported cases and its usually disastrous outcome. Most of the reported cases were closely associated with contact lens wearing. Others were related to a minor trauma or contacting contaminated water. We report a case of recalcitrant corneal ulcer following trauma, which is the first reported case of Acanthamoeba polyphaga keratitis proven by a special culture method and pathological examination in Taiwan. We reviewed the nature and characteristics
