Comparison of three inoculum sources for acetate production and microbial succession in H2/CO2-fed anaerobic system.

Introducing a synthetic H2/CO2 gas mixture into anaerobic systems is a promising strategy for C1 gas valorization, renewable energy storage, and sustainable chemical synthesis. However, systematic comparisons of inoculum sources under identical operational conditions remain limited. In this study, three inoculum sources, fermentation sludge (S1), lake sediment (S2), and brewery anaerobic sludge (S3), were compared for acetate production from CO2 and H2, with particular attention to microbial community succession and reactor stability. S3 showed the highest CO2 consumption rate (331 ± 4 mg CO2/

H2 Consumption by Various Acetogenic Bacteria Follows First-Order Kinetics up to H2 Saturation.

Acetogenic bacteria play an important role in various biotechnological processes, because of their chemolithoautotrophic metabolism converting carbon dioxide with molecular hydrogen (H2) as electron donor into acetate. As the main factor limiting acetogenesis is often H2, insights into the H2 consumption kinetics of acetogens are required to assess their potential in biotechnological processes. In this study, initial H2 consumption rates at a range of different initial H2 concentrations were measured for three different acetogens. Interestingly, for all three strains, H2 consumption was found

CO-driven electron and carbon flux fuels synergistic microbial reductive dechlorination.

BACKGROUND: Carbon monoxide (CO), hypothetically linked to prebiotic biosynthesis and possibly the origin of the life, emerges as a substantive growth substrate for numerous microorganisms. In anoxic environments, the coupling of CO oxidation with hydrogen (H2) production is an essential source of electrons, which can subsequently be utilized by hydrogenotrophic bacteria (e.g., organohalide-respring bacteria). While Dehalococcoides strains assume pivotal roles in the natural turnover of halogenated organics and the bioremediation of chlorinated ethenes, relying on external H2 as their electron

Unraveling the source of corrosive microorganisms from fracturing water to flowback water in shale gas field: evidence from gene sequencing and corrosion tests.

As an insidious and often underestimated phenomenon, microbially influenced corrosion (MIC) poses a significant threat to the integrity and longevity of oil and gas pipelines. However, the complex corrosive microorganisms, that might induce MIC in underground pipelines, might be introduced by the fracturing water during the production period, or they may also exist in the native corrosive microbial community underground. In this study, microbial community analysis was conducted to unravel the source of corrosive microbes in oil and gas pipelines. Meanwhile, the corrosion rate caused by the fra

Simultaneous removal of sediment and water contaminants in a microbial electrochemical system with embedded active electrode by in-situ utilization of electrons.

In the water environment such as lakes, there is a phenomenon that the sediment and overlying water are polluted at the same time. In this study, A microbial electrochemical system with an embedded active electrode was developed for simultaneous removal of polycyclic aromatic hydrocarbons in sediment and antibiotics in overlying water by in-situ utilization of electrons. In the closed-circuit group, the pyrene concentration in sediment decreased from 9.94 to 2.08 mg/L in 96 d, and the sulfamethoxazole concentration in water decreased from 5.12 to 1.12 mg/L in 168 h. These values were 18.71 % a

Start-Up of Chitosan-Assisted Anaerobic Sludge Bed Reactors Treating Light Oxygenated Solvents under Intermittent Operation.

Quality of the granular sludge developed during the start-up of anaerobic up-flow sludge bed reactors is of crucial importance to ensure the process feasibility of treating industrial wastewater such as those containing solvents. In this study, the microbial granule formation from suspended-growth biomass was investigated in two chitosan-assisted reactors. These reactors operated mimicking industrial sites working with night closures treating a mixture of ethanol, ethyl acetate, and 1-ethoxy-2-propanol. Each reactor operated under different hydrodynamic regimes typical from UASB (R1: <0.15 m h

Electron-acceptor loadings affect chloroform dechlorination in a hydrogen-based membrane biofilm reactor.

Chloroform (CF) can undergo reductive dechlorination to dichloromethane, chloromethane, and methane. However, competition for hydrogen (H2 ), the electron-donor substrate, may cause poor dechlorination when multiple electron acceptors are present. Common acceptors in anaerobic environments are nitrate (NO3- ), sulfate (SO42- ), and bicarbonate (HCO3- ). We evaluated CF dechlorination in the presence of HCO3- at 1.56 e- Eq/m2 -day, then NO3- at 0.04-0.15 e- Eq/m2 -day, and finally NO3- (0.04 e- Eq/m2 -day) along with SO42- at 0.33 e- Eq/m2 -day in an H2 -based membrane biofilm reactor (MBfR). Molecular and carbon isotopic characterization of an anaerobic stable enrichment culture containing Dehalobacterium sp. during dichloromethane fermentation.

UNLABELLED: Biodegradation of dichloromethane (DCM) under reducing conditions is of major concern due to its widespread detection in contaminated groundwaters. Here, we report an anaerobic enrichment culture derived from a membrane bioreactor operating in an industrial wastewater treatment plant, capable of fermenting DCM and the brominated analogue dibromomethane (DBM). Comparative analysis of bacterial 16S rDNA-DGGE profiles from fresh liquid medium inoculated with single colonies picked from serial dilution-to-extinction agar vials showed that cultures degrading DCM contained a predominant

Genomic potential and physiological characteristics of C1 metabolism in novel acetogenic bacteria.

Acetogenic bacteria can utilize C1 compounds, such as carbon monoxide (CO), formate, and methanol, via the Wood-Ljungdahl pathway (WLP) to produce biofuels and biochemicals. Two novel acetogenic bacteria of the family Eubacteriaceae ES2 and ES3 were isolated from Eulsukdo, a delta island in South Korea. We conducted whole genome sequencing of the ES strains and comparative genome analysis on the core clusters of WLP with Acetobacterium woodii DSM1030T and Eubacterium limosum ATCC8486T. The methyl-branch cluster included a formate transporter and duplicates or triplicates copies of the fhs gene

Isolation of acetogenic bacteria that induce biocorrosion by utilizing metallic iron as the sole electron donor.

Corrosion of iron occurring under anoxic conditions, which is termed microbiologically influenced corrosion (MIC) or biocorrosion, is mostly caused by microbial activities. Microbial activity that enhances corrosion via uptake of electrons from metallic iron [Fe(0)] has been regarded as one of the major causative factors. In addition to sulfate-reducing bacteria and methanogenic archaea in marine environments, acetogenic bacteria in freshwater environments have recently been suggested to cause MIC under anoxic conditions. However, no microorganisms that perform acetogenesis-dependent MIC have

Microbial community succession during lactate amendment and electron acceptor limitation reveals a predominance of metal-reducing Pelosinus spp.

The determination of the success of in situ bioremediation strategies is complex. By using controlled laboratory conditions, the influence of individual variables, such as U(VI), Cr(VI), and electron donors and acceptors on community structure, dynamics, and the metal-reducing potential can be studied. Triplicate anaerobic, continuous-flow reactors were inoculated with Cr(VI)-contaminated groundwater from the Hanford, WA, 100-H area, amended with lactate, and incubated for 95 days to obtain stable, enriched communities. The reactors were kept anaerobic with N(2) gas (9 ml/min) flushing the hea

Active methylotrophic methanogenesis by a microbial consortium enriched from a terrestrial meteorite impact crater.

Microbial methane generation (methanogenesis) is an important metabolic process in the terrestrial deep biosphere and is an analog to early Earth as it is proposed to be one of the most ancient metabolisms on Earth. Signs of methanogenesis in meteorite impact craters are of particular interest in this respect as these settings are proposed hot spots for deep microbial colonization of the upper crust. Yet, reports of active deep rock-hosted methanogenesis are scarce, particularly for methylotrophic methanogenesis, while reports from terrestrial meteorite impact craters are completely lacking. H

Wastewater-based human gut microbiome surveillance links microbial signatures to population health in Ohio communities.

Wastewater-based epidemiology (WBE) has been widely used to track infectious diseases, yet its potential for population-level gut microbiome surveillance and broader public health assessment remains limited. Here, we present a multi-city study in Ohio, USA, demonstrating how sewage-derived human gut microbiome profiles can be leveraged to infer community health patterns. In this retrospective cross-sectional study, influent wastewater samples were collected from eight Ohio cities (May - June 2022). Community-level gut bacterial profiles were characterized and integrated with publicly available

Mildly acidic pH boosts up CO2 conversion to isobutyrate in H2 driven gas fermentation system.

As a greenhouse gas, massive carbon dioxide (CO2) has been generated due to organic matter degradation in wastewater treatment processes. Microbial gas fermentation offers a promising approach to capture CO2 and generate various valuable chemicals. However, limited studies have achieved branched or medium-chain fatty acids production via gas fermentation. This study reported the production of isobutyrate and hexanoate by feeding H2 and CO2 into membrane biofilm reactors (MBfRs). The gas fermentation product in the reactor with neutral pH (pH of 7) was dominated by acetate (accounting for 90 %

Electron acceptors determine the BTEX degradation capacity of anaerobic microbiota via regulating the microbial community.

Anaerobic degradation is the major pathway for microbial degradation of benzene, toluene, ethylbenzene, and xylenes (BTEX) under electron acceptor lacking conditions. However, how exogenous electron acceptors modulate BTEX degradation through shaping the microbial community structure remains poorly understood. Here, we investigated the effect of various exogenous electron acceptors on BTEX degradation as well as methane production in anaerobic microbiota, which were enriched from the same contaminated soil. It was found that the BTEX degradation capacities of the anaerobic microbiota gradually

Metagenomic insights into the effects of Chive seed flavonoid on intestinal fermentation, morphology, and microbiota composition in sheep.

Chive seed flavonoid (CSF) exhibits antioxidant, digestive, and immunomodulatory properties, yet their effects on ruminant intestinal development and microbial ecosystems remain underexplored. This study systematically evaluated CSF supplementation (0.25% of concentrate) on intestinal morphology, volatile fatty acids (VFAs) dynamics, and microbiota composition in 16 weaned Hu lambs (3 months; 19.57 ± 1.56 kg initial weight) using histomorphometry, VFA quantification, and full-length 16S rRNA sequencing. Animals were randomly allocated to a control (basal diet) or CSF-supplemented group (T), fo

Sulfate-Reducing Bacteria Isolated from an Oil Field in Kazakhstan and a Description of Pseudodesulfovibrio karagichevae sp. nov.

Sulfidogenic bacteria cause numerous issues in the oil industry since they produce sulfide, corroding steel equipment, reducing oil quality, and worsening the environmental conditions in oil fields. The purpose of this work was to isolate and taxonomically identify the sulfidogenic bacteria responsible for the corrosion of steel equipment at the Karazhanbas oil field (Kazakhstan). In this study, we characterized five sulfidogenic strains of the genera Pseudodesulfovibrio, Oleidesulfovibrio, and Acetobacterium isolated from the formation water of the Karazhanbas oil field (Kazakhstan). Hydrogeological compartmentalization outweighs hydrochemistry in structuring microbial communities across contrasting subsurface habitats.

Deep groundwater ecosystems are energy-limited microbial habitats in which hydrochemical conditions, hydrogeological structure, and habitat isolation jointly influence community assembly. However, the relative importance of environmental filtering and hydrogeological compartmentalization in structuring subsurface microbial communities remains poorly understood. We characterized hydrochemistry, bacterial communities, and archaeal signatures across two contrasting groundwater systems in Québec, Canada: a shallow permeable groundwater system at pointe-du-lac (PDL) and deep fractured formation wat

Uncovering potential autotrophs in coal seams: characterization, enrichment, and genome-resolved metabolisms of a subsurface coal seam microbiome.

UNLABELLED: Biological transformation of inorganic carbon into organic compounds is an extremely important biosynthetic process, and many organisms catalyze this reaction autotrophically, using CO2 as the sole carbon source. Natural autotrophic pathways are diverse, as are the organisms that employ them, driven in part by the varied ecological niches they inhabit. One understudied ecological niche, coal seams, contains autotrophic acetogens that catalyze an intermediate step in the conversion of coal to methane. Here, we characterize the geochemistry and microbiology of coal seam produced wate

Influence of vitamin B12 and cocultures on the growth of Dehalococcoides isolates in defined medium.

Bacteria belonging to the genus Dehalococcoides play a key role in the complete detoxification of chloroethenes as these organisms are the only microbes known to be capable of dechlorination beyond dichloroethenes to vinyl chloride (VC) and ethene. However, Dehalococcoides strains usually grow slowly with a doubling time of 1 to 2 days and have complex nutritional requirements. Here we describe the growth of Dehalococcoides ethenogenes 195 in a defined mineral salts medium, improved growth of strain 195 when the medium was amended with high concentrations of vitamin B(12), and a strategy for m
