Ca2+ signaling of pancreatic acinar cells in malignant hyperthermia susceptibility.

BACKGROUND: Malignant hyperthermia susceptibility (MHS) and acute pancreatitis (AP) share a common cellular pathomechanism that is Ca2+-overload of the muscle fiber and the pancreatic acinar cell (PAC). In the muscle, gain-of-function mutations of the ryanodine receptor (RyR1) make the Ca2+-release mechanism hypersensitive to certain ligands, including Ca2+, volatile anaesthetics and succinylcholine, creating a medical emergency when the patient is exposed to these drugs. As RyR1 was shown to contribute to Ca2+-overload in PAC, we presumed that pancreata of MHS individuals are more prone to AP

Western diet-induced ultrastructural changes in mouse pancreatic acinar cells.

Mouse models of diet-induced type 2 diabetes mellitus provide powerful tools for studying the structural and physiological changes that are related to the disease progression. In this study, diabetic-like glucose dysregulation was induced in mice by feeding them a western diet, and light and transmission electron microscopy were used to study the ultrastructural changes in the pancreatic acinar cells. Acinar necrosis and vacuolization of the cytoplasm were the most prominent features. Furthermore, we observed intracellular and extracellular accumulation of lipid compounds in the form of lipid

Inducing differentiation of human dental pulp cells toward acinar-lineage.

OBJECTIVE: To induce acinar-differentiation from human dental pulp cells for potential application in aiding treatment of dry-eye syndromes. METHOD: Human dental pulp cells were co-cultured with human submandibular gland acinar cells using a transwell construction for 2 weeks. The two populations of cells were physically separated while chemical and biochemical components can be exchanged. Fibroblasts were included as a negative control. Expression of amylase, cytokeratin 8 and vimentin were examined by immune-staining. Amylase activity was measured using an AMS Assay Kit. RESULT: Cobbleston

Characterization of cysteine string protein in rat parotid acinar cells.

Cysteine string proteins (CSPs) are secretory vesicle chaperone proteins that contain: (i) a heavily palmitoylated cysteine string (comprised of 14 cysteine residues, responsible for the localization of CSP to secretory vesicle membranes), (ii) an N-terminal J-domain (DnaJ domain of Hsc70, 70kDa heat-shock cognate protein family of co-chaperones), and (iii) a linker domain (important in mediating CSP effects on secretion). In this study, we investigated the localization of CSP1 in rat parotid acinar cells and evaluated the role of CSP1 in parotid secretion. RT-PCR and western blotting revealed

Alcohols enhance caerulein-induced zymogen activation in pancreatic acinar cells.

Activation of zymogens within the pancreatic acinar cell is an early feature of acute pancreatitis. Supraphysiological concentrations of cholecystokinin (CCK) cause zymogen activation and pancreatitis. The effects of the CCK analog, caerulein, and alcohol on trypsin and chymotrypsin activation in isolated pancreatic acini were examined. Caerulein increased markers of zymogen activation in a time- and concentration-dependent manner. Notably, trypsin activity reached a peak value within 30 min, then diminished with time, whereas chymotrypsin activity increased with time. Ethanol (35 mM) sensitiz

Muscarinic signalling in submandibular salivary acinar cells of ageing rats.

Muscarinic cholinergic stimulation of submandibular acinar cells results in the activation of Ca(2+)-dependent ion-transport pathways responsible for the secretion of primary saliva. Decreased saliva production is common among elderly people and may compromise oral health with implications for systemic health, nutrition, and quality of life. The density and affinity of muscarinic receptors in the submandibular gland of rats and the Ca2+ responses to stimulation of these receptors in the acinar cells were examined. An increase in the number of receptors and increases in the affinities of the re

Dispersed rat parotid acinar cells. II. Characterization of adrenergic receptors.

The in vitro characterization of adrenergic receptors in isolated rat parotid acinar cells was accomplished through investigations of the transmembrane influxes of K and of the secretion of amylase in response to interactions of the cells with selected agonists and antagonists. Interaction of epinephrine (EPI) at concentrations of 10(-3)-10(-9) M with the alpha-adrenergic receptors resulted in rapid efflux of K from the cells. This effect was inhibited by phentolamine but not by propranolol or atropine. The process of secretion of amylase by these cells involved the activation of the beta-adre

HDL inhibits pancreatic acinar cell NLRP3 inflammasome activation and protect against acinar cell pyroptosis in acute pancreatitis.

BACKGROUND AND PURPOSE: Recent clinical studies have shown that serum high-density lipoprotein (HDL) levels are correlated with acute pancreatitis (AP) severity. We aimed to investigate the role of HDL in pancreatic necrosis in AP. EXPERIMENTAL APPROACH: ApoA-I is the main constitution and function component of HDL. The roles of healthy human-derived HDL and apoA-I mimic peptide D4F were demonstrated in AP models in vivo and in vitro. Constitutive Apoa1 genetic inhibition on AP severity, especially pancreatic necrosis was assessed in both caerulein and sodium taurocholate induced mouse AP mod

Aberrant acinar cell CA 19-9 expression and peri-insular acinar cell alterations in an adult human pancreas.

We herein report on a 23-year-old female patient who suffered from Crohn's disease and anorexia nervosa and died after long-term malnutrition and a perforated colitis. At autopsy, her pancreas displayed two peculiar findings. First, there was a constant and aberrant expression of CA 19-9 in the acinar cells. The expression of the carbohydrate antigen CA 19-9 is normally confined to duct-like epithelia, including centroacinar cells, while islet and acinar cells have repeatedly been reported to be immunohistochemically negative. Thus, to the best of our knowledge, our case is the first to show a

Spink1 promotes the survival of Kras-mutant pancreatic acinar cells.

BACKGROUND: Pancreatic cancer remains one of the most intractable malignancies due to its strong therapeutic resistance and the difficulty of early detection. Previous studies have suggested that pancreatic inflammation can promote carcinogenesis. In contrast, the regulatory mechanisms controlling trypsin activity are closely associated with susceptibility to pancreatitis. Genetic variants that lead to sustained trypsin activity-thereby promoting protease activation-have been identified in hereditary pancreatitis. Serine protease inhibitor Kazal type 1 (SPINK1) functions as an endogenous inhib

Apical Ca2+-activated potassium channels in mouse parotid acinar cells.

Ca(2+) activation of Cl and K channels is a key event underlying stimulated fluid secretion from parotid salivary glands. Cl channels are exclusively present on the apical plasma membrane (PM), whereas the localization of K channels has not been established. Mathematical models have suggested that localization of some K channels to the apical PM is optimum for fluid secretion. A combination of whole cell electrophysiology and temporally resolved digital imaging with local manipulation of intracellular [Ca(2+)] was used to investigate if Ca(2+)-activated K channels are present in the apical PM

Sialoglycoconjugate expression in acinar cells of rat developing submandibular gland.

Direct and indirect staining procedures were developed to characterize sialoglycoconjugates in developing rat submandibular gland. Lectin histochemistry, with and without prior sialidase digestion, combined with differential oxidation and deacetylation procedures was performed in situ. This allowed the expression of sialic acids to be followed during acinar cell development. It was found that terminal periodate-labile sialic acids linked to beta-galactose occurred early. In contrast, the terminal disaccharide sialic acid-N-acetylgalactosamine was only detectable at the adult stage and so was c

Acyl-coenzyme A causes Ca2+ release in pancreatic acinar cells.

The regulation of cytosolic Ca2+ is important for a variety of cell functions. One non-inositol 1,4,5-trisphosphate (IP3) compound that may regulate Ca2+ is palmitoyl-coenzyme A (CoA), a fatty acid-CoA that is reported to cause Ca2+ release from intracellular stores of oocytes, myocytes, and hepatocytes. To study the role of palmitoyl-CoA in the pancreatic acinar cell, rat pancreatic acini were isolated by collagenase digestion, permeablized with streptolysin O, and the release of Ca2+ from internal stores was measured with fura-2. Palmitoyl-CoA released Ca2+ from internal stores (EC50 = 14 mi

Na+, K+, and Cl- transport in resting pancreatic acinar cells.

To understand the role of Na+, K+, and Cl- transporters in fluid and electrolyte secretion by pancreatic acinar cells, we studied the relationship between them in resting and stimulated cells. Measurements of [Cl-]i in resting cells showed that in HCO3(-)-buffered medium [Cl-]i and Cl- fluxes are dominated by the Cl-/HCO3- exchanger. In the absence of HCO3-, [Cl-]i is regulated by NaCl and NaK2Cl cotransport systems. Measurements of [Na+]i showed that the Na(+)-coupled Cl- transporters contributed to the regulation of [Na+]i, but the major Na+ influx pathway in resting pancreatic acinar cells

Calcium metabolism and amylase release in rat parotid acinar cells.

1. A method is described for the isolation of rat parotid acinar cells by controlled digestion of the gland with trypsin followed by collagenase. As judged by Trypan Blue exclusion, electron microscopy, water, electrolyte and ATP concentrations and release of amylase and lactate dehydrogenase, the cells are morphologically and functionally intact. 2. A method was developed for perifusion of acinar cells by embedding them in Sephadex G-10. Release of amylase was stimulated by adrenaline (0.1-10muM), isoproternol (1 or 10 MUM), phenylephrine (1 muM), carbamoylcholine (0.1 or 1 muM), dibutyryl cy

Dispersed rat parotid acinar cells. III. Characterization of cholinergic receptors.

The in vitro characterization of cholinergic receptors in dispersed rat parotid acinar cells was accomplished through investigations of the net transmembrane fluxes of K in response to exposure of the cells to selected cholinergic agonists and antagonists. Interaction of acetylcholine bromide (ACh) and carbamylcholine (carbachol) with the cholinergic receptors resulted in rapid net efflux of K from the cells. This cellular response was demonstrable in concentrations of carbachol as low as 10(-8) M. With gradual increase in the concentrations of the agonist an increase in the K efflux was obser

Primary acinar cell carcinoma of the ampulla of Vater.

Acinar cell carcinoma of the pancreatobiliary system is a relatively rare malignant neoplasm arising usually in the pancreatic parenchyma. We experienced a 68-year-old woman who presented with obstructive jaundice due to an ampullary mass 1.0 cm in diameter, detected by abdominal computed tomography and endoscopic examination. The patient underwent a curative surgical operation, and histopathological examination revealed that the tumor was confined to the ampulla of Vater with no continuity to the pancreatic parenchyma. The tumor cells showed acinar or tubular arrangement with eosinophilic to
