Gating current experiments on frog nodes of Ranvier treated with Centruroides sculpturatus toxins or aconitine.

(1) Gating currents were recorded from frog nodes of Ranvier treated either with toxins III or IV from the venom of the scorpion Centruroides sculpturatus or with the alkaloid toxin aconitine. (2) Toxins III or IV from Centruroides sculpturatus (which drastically reduce the sodium permeability PNa and slightly shift its voltage dependence in the depolarizing direction) caused a small depolarizing shift of the relation between charge (Qon) and membrane potential (E) without affecting the maximum charge Qon max. (3) On nodes treated with toxins III or IV from Centruroides sculpturatus, a depolar

Effects of aconitine on the respiratory activity of brainstem-spinal cord preparations isolated from newborn rats.

Aconitine is a sodium channel opener, but its effects on the respiratory center are not well understood. We investigated the dose-dependent effects of aconitine on central respiratory activity in brainstem-spinal cord preparations isolated from newborn rats. Bath application of 0.5-5 μM aconitine caused an increase in respiratory rhythm and decrease in the inspiratory burst amplitude of the fourth cervical ventral root (C4). Separate application of aconitine revealed that medullary neurons were responsible for the respiratory rhythm increase, and neurons in both the medulla and spinal cord wer

Pseudo-ginsengenin DQ ameliorated aconitine-induced arrhythmias by influencing Ca2+ and K+ currents in ventricular myocytes.

Pseudo-ginsengenin DQ (PDQ) is the product of the oxidative cyclization of protopanaxadiol. PDQ exhibits various bioactivities, including reversal of multidrug resistance in cancer, renal protective effects against acute nephrotoxicity and attenuating myocardial ischemia injury induced by isoproterenol or ligation of coronary arterials, but its effect on arrhythmias has not been clear until now. Because of the complicated effects of ginseng on the cardiovascular system, it is necessary to investigate whether PDQ affects arrhythmias, which are always concomitant with other cardiac diseases. Sarcoplasmic reticulum Ca2+ release channel ryanodine receptor (RyR2) plays a crucial role in aconitine-induced arrhythmias.

The present study established a model of RyR(2) knockdown cardiomyocytes and elucidated the role of RyR(2) in aconitine-induced arrhythmia. Cardiomyocytes were obtained from hearts of neonatal Sprague-Dawley rats. siRNAs were used to down-regulate RyR(2) expression. Reduction of RyR(2) expression was documented by RT-PCR, western blot, and immunofluorescence. Ca(2+) signals were investigated by measuring the relative intracellular Ca(2+) concentration, spontaneous Ca(2+) oscillations, caffeine-induced Ca(2+) release, and L-type Ca(2+) currents. In normal cardiomyocytes, steady and periodic spo

Case report: Accidental aconitine poisoning caused by the inappropriate use of a type of Chinese patent medicine.

BACKGROUND: Compound schizonepeta fumigation lotion is a type of Chinese patent medicine for external use. It has the effect of dispelling wind, eliminating dampness, reducing swelling, and relieving pain. Clinically, it is used for anal fumigation and treatment of external hemorrhoids, anal fissures, and other diseases. Aconitum species are widely used in the field of traditional Chinese medicine. However, improper use and overdose can easily cause acute poisoning, leading to malignant arrhythmias, cardiogenic shock, and even death. METHODS: This study retrospectively analyzed the clinical d

Effect of quercetin on aconitine-induced renal interstitial fibrosis and its relationship with PI3K/Akt pathway.

BACKGROUND: Renal interstitial fibrosis (RIF) is a critical pathological outcome of chronic kidney disease (CKD). Aconitine has been shown to induce RIF, with the phosphatidylinositol 3-kinase/protein kinase B (PI3K/Akt) signaling pathway potentially playing a key role in this process. Quercetin, a natural flavonoid compound, exhibits anti-fibrotic effects that may be associated with this pathway. However, there is no clear evidence yet regarding its role in aconitine-induced RIF. OBJECTIVES: This study aimed to investigate the intervention effect of quercetin on aconitine-induced RIF and the

HBB contributes to individualized aconitine-induced cardiotoxicity in mice via interfering with ABHD5/AMPK/HDAC4 axis.

The root of Aconitum carmichaelii Debx. (Fuzi) is an herbal medicine used in China that exerts significant efficacy in rescuing patients from severe diseases. A key toxic compound in Fuzi, aconitine (AC), could trigger unpredictable cardiotoxicities with high-individualization, thus hinders safe application of Fuzi. In this study we investigated the individual differences of AC-induced cardiotoxicities, the biomarkers and underlying mechanisms. Diversity Outbred (DO) mice were used as a genetically heterogeneous model for mimicking individualization clinically. The mice were orally administere

Aconitine-induced Ca2+ overload causes arrhythmia and triggers apoptosis through p38 MAPK signaling pathway in rats.

Aconitine is a major bioactive diterpenoid alkaloid with high content derived from herbal aconitum plants. Emerging evidence indicates that voltage-dependent Na(+) channels have pivotal roles in the cardiotoxicity of aconitine. However, no reports are available on the role of Ca(2+) in aconitine poisoning. In this study, we explored the importance of pathological Ca(2+) signaling in aconitine poisoning in vitro and in vivo. We found that Ca(2+) overload lead to accelerated beating rhythm in adult rat ventricular myocytes and caused arrhythmia in conscious freely moving rats. To investigate eff

Discovery of novel 2-azabicyclo[3.2.1]octane derivatives against non-small cell lung cancer from aconitine simplification.

Aconitine, a diterpenoid alkaloid, exhibits promising bioactivities, but its clinical application is limited by severe toxicity. Through a structural simplification and derivatization strategy, we designed and synthesized a novel series of 2-azabicyclo[3.2.1]octane derivatives. This effort culminated in the identification of compound 8q, which demonstrated potent and selective anti-tumor efficacy against ALK (anaplastic lymphoma kinase) -positive cancer cells (NCI-H2228 and Karpas-299), significantly outperforming ceritinib in enzymatic and cellular assays. Mechanism studies revealed that 8q e

Successful rescue of aconitine poisoning with extended cardiopulmonary resuscitation combined with hemoperfusion: A case report and literature review.

RATIONALE: Aconitine, a highly toxic alkaloid derived from Aconitum plants, exhibits a narrow therapeutic window, with a poisonous dose as low as 0.2 mg and lethal dose of 2 to 5 mg. Currently, no specific antidote exists for aconitine poisoning, which frequently triggers refractory electrical storms and cardiopulmonary arrest, culminating in high mortality. Elderly patients face particularly dire prognoses due to diminished physiological reserves. This study evaluated the efficacy of extended cardiopulmonary resuscitation (CPR) combined with hemoperfusion (HP) as a rescue strategy for aconiti

Inhibition of the INa/K and the activation of peak INa contribute to the arrhythmogenic effects of aconitine and mesaconitine in guinea pigs.

Aconitine (ACO), a main active ingredient of Aconitum, is well-known for its cardiotoxicity. However, the mechanisms of toxic action of ACO remain unclear. In the current study, we investigated the cardiac effects of ACO and mesaconitine (MACO), a structurally related analog of ACO identified in Aconitum with undocumented cardiotoxicity in guinea pigs. We showed that intravenous administration of ACO or MACO (25 μg/kg) to guinea pigs caused various types of arrhythmias in electrocardiogram (ECG) recording, including ventricular premature beats (VPB), atrioventricular blockade (AVB), ventricula

Multimodal Management of Refractory Ventricular Tachycardia Due to Aconitine Poisoning: A Case Report in a Resource-Limited Setting.

Aconitine, a potent plant alkaloid used in traditional Asian medicine, possesses significant toxicity risks. This case report presents a 42-year-old male with alcohol use disorder who developed life-threatening ventricular tachycardia after ingesting a homemade herbal liqueur. Initially thought to contain Cassia alata (candle bush), further investigation revealed that the tincture was instead prepared from Aconitum root (Wolfsbane) based on advice from an online social media video. He subsequently developed life-threatening ventricular tachycardia and cardiac arrest refractory to initial inter

Stir Fry with a Side of Extracorporeal Membrane Oxygen: Management of Cardiogenic Shock Secondary to Unintentional Aconitine Ingestion.

Plant exposures leading to systemic or topical toxicity are common presentations seen in the emergency department. While often nonfatal, certain highly toxic plants result in cardiovascular or respiratory failure requiring invasive management. We describe a 65-y-old patient who presented with a refractory ventricular dysrhythmia secondary to an unintentional ingestion of an aconitine-containing plant after incorrect identification. Despite aggressive treatment with vasopressors, intravenous fluids, antiarrhythmics, as well as electrolyte correction and multiple attempted synchronized cardiover

Aconitine promotes injured peripheral nerve recovery through restraining the activation of inflammasome-mediated cell pyroptosis and pathological inflammation.

Peripheral nerve injury (PNI) represents a common neurological condition with significant social and economic implications. Aconitine, a diterpenoid alkaloid derived from Aconitum species, exhibits potent anti-cancer, anti-viral, anti-inflammatory, analgesic, and immunomodulatory activities against malignancies, rheumatic disorders, arthralgia, and select endocrine pathologies. However, the neuroprotective potential of aconitine in PNI repair remains unclear. Here, we revealed that aconitine treatment at the optimal dose significantly improved SFI values, electrophysiological conduction, axon

The effects of B0, B20, and B100 soy biodiesel exhaust on aconitine-induced cardiac arrhythmia in spontaneously hypertensive rats.

CONTEXT: Diesel exhaust (DE) has been shown to increase the risk of cardiac arrhythmias. Although biodiesel has been proposed as a "safer" alternative to diesel, it is still uncertain whether it actually poses less threat. OBJECTIVE: We hypothesized that exposure to pure or 20% soy biodiesel exhaust (BDE) would cause less sensitivity to aconitine-induced arrhythmia than DE in rats. METHODS: Spontaneously hypertensive (SH) rats implanted with radiotelemeters were exposed once or for 5 d (4 h) to either 50 mg/m(3) (low), 150 mg/m(3) (medium), or 500 mg/m(3) (high) of DE (B0), 20% (B20) or 100%

A single exposure to particulate or gaseous air pollution increases the risk of aconitine-induced cardiac arrhythmia in hypertensive rats.

Epidemiological studies demonstrate an association between arrhythmias and air pollution. Aconitine-induced cardiac arrhythmia is widely used experimentally to examine factors that alter the risk of arrhythmogenesis. In this study, Wistar-Kyoto (WKY) and spontaneously hypertensive (SH) rats acutely exposed to synthetic residual oil fly ash (s-ROFA) particles (450 mug/m(3)) were "challenged" with aconitine to examine whether a single exposure could predispose to arrhythmogenesis. Separately, SH rats were exposed to varied particulate matter (PM) concentrations (0.45, 1.0, or 3.5 mg/m(3) s-ROFA)
