Development of molecular markers for marker-assisted breeding and quality evaluation of Aconitum carmichaelii cultivars.

BACKGROUND: Aconitum carmichaelii (AC), a traditional Chinese medicinal herb, provides substantial economic benefits to the pharmaceutical industry and rural development. Its broad genetic diversity has led to multiple cultivars with distinct biological traits. However, accurately identifying these cultivars is challenging due to the morphological similarities, particularly at the seedling stage. Misidentifying cultivars or selecting inappropriate cultivation regions may lead to crop failure. Moreover, the chemical profiles of different cultivars remain insufficiently characterized. Therefore,

The global profiling of alkaloids in Aconitum stapfianum and analysis of detoxification material basis against Fuzi.

Aconitum alkaloids are versatile in chemical structures and are well known for their bioactivity and toxicity. Cases of analogs with closely similar structures or positional isomers are widespread in herbs of the Aconitum genus. It is still challenging to rapidly identify unknown compounds via mass spectrometry, especially positional isomers. Herein, to profile the alkaloids of Aconitum stapfianum that possess bioactivity against intoxication by the lateral root of Aconitum carmichaelii (Fuzi), a strategy was developed by carefully determining the fragmentation pathways of authentic standards.

The local anesthetic activity of Aconitum alkaloids can be explained by their structural properties: a QSAR analysis.

Alkaloids isolated from Aconitum roots exhibit anesthetic effects at peripheral nerves. We performed the present quantitative structure-activity relationship (QSAR) analysis in order to understand the mechanism of action as local anesthetics of 11 Aconitum alkaloids. The alkaloids with the highest anesthetic activity had an aroyl/aroyloxy group at R14 position while the weaker anesthetic alkaloids had the aroyloxy group at R4. The stable compounds exhibited a higher local anesthetic activity than the unstable compounds. In relation to the reactivity indexes of atoms on the aromatic ring, C2' w

C‑20 Diterpenoid Alkaloids with PD-L1 Inhibitory Activity from the Roots of Aconitum coreanum.

Fifteen previously unreported diterpenoid alkaloids, named aconicorines A-O (1-15), and 13 known diterpenoid alkaloids (16-28) were isolated from the roots of Aconitum coreanum. Their structures and configurations were determined by comprehensive spectroscopic analysis including HRMS, NMR, and single-crystal X-ray diffraction. Structurally, all new compounds are classic C20-hetisine-type diterpenoid alkaloids with various oxidation sites. Among them, aconicorines A-D (1-4) are unique C20-diterpenoid alkaloids containing a rare sulfonate group. Bioassay results showed that 1, 7, 13-17, and 20 d

A review of traditional and current methods used to potentially reduce toxicity of Aconitum roots in Traditional Chinese Medicine.

CONTEXT: Aconitum species are well-known for their medicinal value and high lethal toxicity in many Asian countries, notably China, India and Japan. The tubers are only used after processing in Traditional Chinese Medicine (TCM). They can be used safely and effectively with the methods of decoction, rational compatibility, and correct processing based on traditional experiences and new technologies. However, high toxicological risks still remain due to improper preparation and usage in China and other countries. Therefore, there is a need to clarify the methods of processing and compatibility
