Effects of acrylamide on locomotion and central monoamine function in the rat.

Male rats receiving acrylamide (ACR) in their drinking water (100 ppm) for a six-week period displayed increased psychomotor stimulation to d-amphetamine (d-A; 1.0 mg/kg SC) under several conditions of handling and drug administration. Following behavioral tests a subset of the animals was sacrificed at 15, 50, 80 and 120 minutes following d-A and the brains removed and dissected for determinations of regional brain levels of several monoamine neurotransmitters and metabolites. ACR rats had elevated levels of 5-hydroxyindoleacetic acid (5-HIAA) in the striatum, septal area, and thalamus. The e

Reducing acrylamide in bread with plant additives: a systematic review.

Reducing acrylamide (AA) formation in thermally processed foods, particularly bread, is a major food safety concern. Plant-based additives, including powders and extracts, offer a promising natural strategy for AA mitigation due to their bioactive compounds, such as polyphenols and flavonoids, which can scavenge reactive intermediates, modulate Maillard reactions, and inhibit lipid oxidation. This study systematically evaluated the effects of various plant powders and extracts, including green tea, rosemary, turmeric, buckwheat, and traditional spices, on AA formation in bread. Based on report

Comprehensive analysis of metabolic changes in rats exposed to acrylamide.

Acrylamide (ACR) is a widely used environmentally hazardous compound that is known to be neurotoxic, genotoxic, carcinogenic, and reproductive toxicity. It is widely present in soil, water, agents used in chemical industries, and food. It can be distributed to all organs and tissues, and can cause damage to various human systems and those of other animals. Previous metabolomics studies have mainly focused on metabolites in serum and urine, but have lacked comprehensive analysis of major organs and tissues. In the current study, a gas chromatography-massspectrometry method was used to investiga

Influence of Composition on the Patterns of Electrokinetic Potential of Thermosensitive N-(Isopropyl)Acrylamide Derivatives with Poly(Ethylene Glycol) Dimethacrylate and N-(2-Hydroxyethyl)Acrylamide.

The synthesis of poly(N-isopropyl acrylamide) (pNIPA)-based polymers via the surfactant-free precipitation polymerization (SFPP) method produced thermosensitive nanospheres with a range of distinctive physicochemical properties. Nano- and microparticles were generated using various initiators, significantly influencing particle characteristics, including the hydrodynamic diameter (DH), which varied from 87.7 nm to 1618.1 nm. Initiators, such as potassium persulfate and 2,2'-azobis(2-methylpropionamidine) dihydrochloride, conferred anionic and cationic functionalities, respectively, impacting t

Ginger as a nutraceutical shield: Counteracting acrylamide-induced liver injury.

Acrylamide, a contaminant formed during high-temperature cooking of common foods, is increasingly recognized as a silent and underestimated contributor to liver injury. In this editorial, we comment on the study by Nour El Deen et al, demonstrating that a chemically standardized ginger (Zingiber officinale) extract (≥ 20% 6-gingerol) mitigates acrylamide-induced hepatotoxicity in rats through antioxidant, anti-inflammatory, and cytoprotective mechanisms. By combining biochemical, histopathological, and molecular evidence, the authors establish a coherent experimental basis for future translati

One-Dimensional Acrylamide Gel Electrophoresis for Analysis of Plant Samples.

Polyacrylamide gel electrophoresis (PAGE) is a widely used technique for separating proteins from complex plant samples. Prior to the analysis, proteins must be extracted from plant tissues, which are rather complex than other types of biological material. Different protocols have been applied depending on the protein source, such as seeds, pollen, leaves, roots, and flowers. Total protein amounts must also be determined before conducting gel electrophoresis. The most common methodologies include PAGE under native or denaturing conditions. Both procedures are used consequently for protein iden

Carnosic acid attenuates acrylamide-induced retinal toxicity in zebrafish embryos.

Acrylamide (ACR) is a water-soluble chemical used widely in industry, which can be formed in tobacco smoke and in starchy foods cooked at high temperatures. ACR is considered to be a neurotoxin, genotoxin and carcinotoxin. Previous studies reported that ACR-exposed workers and experimental animals exhibited visual function defects, although the underlying mechanisms have not been elucidated. In this study, we found that zebrafish embryos exposed to 1 mM and 2 mM ACR showed significantly increased reactive oxygen species (ROS), decreased expression of the antioxidant genes Sod1, Sod2, Catalase,

Subacute Acrylamide Intoxication with Severe Visual Disturbance: A Case Report.

A 35-year-old man was admitted due to somnolence and progressive sensory-motor polyneuropathy, followed by severe visual impairment in both eyes after direct skin exposure to an acrylamide monomer solution. The results of an ophthalmological examination including central critical flicker fusion frequency and the decreased amplitude of action potentials observed in the visual evoked potential studies suggested that acrylamide intoxication resulted in neuronal degeneration in the optic pathways. Additional attention should be directed to the potential effect of acrylamide on the human visual sys

Current issues in dietary acrylamide: formation, mitigation and risk assessment.

Acrylamide (AA) is known as a neurotoxin in humans and it is classified as a probable human carcinogen by the International Agency of Research on Cancer. AA is produced as by-product of the Maillard reaction in starchy foods processed at high temperatures (>120 °C). This review includes the investigation of AA precursors, mechanisms of AA formation and AA mitigation technologies in potato, cereal and coffee products. Additionally, most relevant issues of AA risk assessment are discussed. New technologies tested from laboratory to industrial scale face, as a major challenge, the reduction of AA

Evidence-based dose-response assessment for thyroid tumorigenesis from acrylamide.

Acrylamide is commonly found in various foods. Cancer studies in rats have reported increases in tumors of the thyroid, mammary tissues, tunica vaginalis of the testis, and sometimes tumors at other sites. We review relevant studies on acrylamide's DNA toxicity, tumor formation and the manner of its tumor formation. We find, as do others, that glycidamide (a metabolite of acrylamide) causes point mutations, but acrylamide does not. We also find that thyroid tumors are most consistently sensitive in rats, being evoked in each of four long-term experiments. We evaluate the common manners of this

Acrylamide: its metabolism, developmental and reproductive effects, genotoxicity, and carcinogenicity.

Monomeric acrylamide is an important industrial chemical primarily used in the production of polymers and copolymers. It is also used for producing grouts and soil stabilizers. Acrylamide's neurotoxic properties have been well documented. This review will focus on pertinent information concerning other, non-neurotoxic, effects observed after exposure to acrylamide, including: its genotoxic, carcinogenic, reproductive, and developmental effects. It will also cover its absorption, metabolism, and distribution. The data show that acrylamide is capable of inducing genotoxic, carcinogenic, developm

Peripheral neuropathy with sympathetic overactivity from industrial contact with acrylamide.

This paper describes the experimental and clinical findings of acrylamide intoxication in a human being. It is believed that this is the first such case to be recorded in the medical literature.Acrylamide is widely used as a "chemical grout". It is pumped into dirt, clay and stone walls of excavations in a liquid state together with a catalyst, and it then polymerizes to make a watertight shield. This chemical is neurotoxic in its non-polymerized form and can be absorbed through the intact skin, mucous membranes and lungs. In spite of warnings with regard to its handling, this worker became car

ATRA-mediated RAR-α activation attenuates acrylamide-induced testicular toxicity.

Acrylamide (ACR) is an environmental reproductive toxicant with unclear testicular toxicity mechanisms. Retinoids are a group of vitamin A-related compounds that function by activating retinoid receptors. We aimed in this study to further explore All-trans retinoic acid's (ATRA) protective response against an acrylamide-induced testicular insult model and the underlying possible mechanisms. Fifty male rats were allocated into control, DMSO, ACR (40 mg/kg bwt, i.p. daily for 14 days), ATRA (7.5 mg/kg bwt, i.p. daily), and ACR + ATRA groups. Body and testes weights, sperm parameters, testosteron

Deletion of IL-1β exacerbates acrylamide-induced neurotoxicity in mice.

Acrylamide is a neurotoxicant in human and experimental animals. Interleukin-1β (IL-1β) is a proinflammatory cytokine known as a critical component of brain reaction to any insult or neurodegenerative pathologies, though its role in electrophile-induced neurotoxicity remains elusive. The aim of this study was to investigate the role of IL-1β in acrylamide-induced neurotoxicity in mice. Ten-week-old male wild-type and IL-1β knock-out mice were allocated into 3 groups each and exposed to acrylamide at 0, 12.5, 25 mg/kg body weight by oral gavage for 28 days. Compared with wild-type mice, the res

Acrylamide based molecularly imprinted polymer for detection of m-nitrophenol.

Molecular imprinting has already proven to be one of the most promising artificial receptors that can lead to specific recognition at molecular level. In this work, molecular imprinting polymers (MIPs) were prepared from acrylamide (ACM) as functional monomer and m-nitrophenol as template respectively. Bis-acrylamide was used as cross-linker. Synthesis was carried out first by ultrasonication of the reaction mixture containing monomer, cross-linker and template in chloroform. Subsequently, the polymerization was conducted at high temperature catalyzed by initiator. Interaction between function

Toxicological effects of acrylamide on rat testicular gene expression profile.

Toxicological effects of acrylamide on differential gene expression profile of rat testis were evaluated. Acrylamide induced morphological sperm defects, and decreased sperm concentration in cauda epididymis. Serum testosterone level and Leydig cell viability were also decreased dose-dependently, which resulted in decreased spermatogenesis. Acrylamide-induced histopathological lesions, such as formation of multinucleated giant cells and vacuolation, and numerous apoptotic cells were observed in seminiferous tubules. cDNA microarray analysis revealed that genes related to testicular-functions,
