Herein, we report that NaBH4 facilitates an unusual reductive cyclization of N-(2-iodoaryl)acrylamide derivatives to afford oxindoles without the need for external initiators. While other hydride donors failed to achieve this transformation, NaBH4 enabled it with a broad scope. Control experiments and DFT studies support a unimolecular radical-nucleophilic substitution (SRN1) pathway.

We describe a visible-light photoredox-catalyzed alkylation/arylative cyclization of N-acrylamides─from 2-arylindoles, 2-arylbenzimidazoles, or N-substituted anilines─with ketone-derived dihydroquinazolinones, accessing indolo- and benzimidazolo[2,1-a]isoquinolines or 2-oxindoles. The consecutive incorporation of alkyl- and aryl-carbogenic motifs across a C=C bond via formal cleavage of ketone α-C-C and arene C-H bonds leads to the formation of five- and six-membered rings, with an all-carbon quaternary stereocenter. This dicarbofunctionalization elaborates aromatization-driven radical C-C fun

Acrylamide related substances (AAs), including N-methylacrylamide and N-hydroxymethylacrylamide, are highly polar, low-molecular-weight organic compounds that have raised significant food safety concerns due to their prevalence in thermally processed foods. AAs exhibit potential neurotoxicity and AA itself also demonstrates reproductive toxicity and possible carcinogenic effects. The International Agency for Research on Cancer has classified AA as a Group 2 A probable human carcinogen, underscoring the health risks associated with long-term dietary exposure to AAs. Consequently, efficient and

BACKGROUND: Acrylamide (ACR) is an important chemical raw material, and its toxic effects have been confirmed in vitro and in vivo. However, only a few studies have investigated ACR-induced liver injury. As a naturally occurring flavonoid widely distributed across the plant kingdom, rutin (Rut) possesses notable pharmacological properties. This study aimed to demonstrate its therapeutic potential in mitigating ACR-induced hepatic injury in rats. Accordingly, an intervention model was established to explore the mechanistic basis of the protective effects of Rut against ACR-induced liver injury.

Data has revealed bread to be one of the sources of acrylamide in human diet. Significant efforts have been dedicated to find efficient methods for reducing acrylamide formation in bread and this review offers a detailed examination of the formulation and technological strategies for this purpose. Key formulation approaches include using organic acids, amino acids competing with asparagine, replacing reducing sugars with non-reducing alternatives, application of L-asparaginase, addition of divalent cations which affect pH and Maillard reaction pathways, addition of antioxidant compounds and hy

Acrylamide (ACR) is formed during food processing by Maillard reaction between sugars and proteins at high temperatures. It is also used in many industries, from water waste treatment to manufacture of paper, fabrics, dyes and cosmetics. Unfortunately, cumulative exposure to acrylamide, either from diet or at the workplace, may result in neurotoxicity. Such adverse effects arise from covalent adducts formed between acrylamide and cysteine residues of several neuronal proteins via a Michael addition reaction. The molecular determinants of acrylamide reactivity and its impact on protein function

The aim of this study is to explore the potent neuroprotective effect of calpeptin (CP) on neuron damage induced by acrylamide (ACR) and its mechanism. Behavioural indicators such as hind limb splay, rota-rod performance, and gait analysis were assessed weekly to evaluate neurobehavioural changes after ACR and/or CP administration. The histopathological alterations and the changes of μ-calpain, m-calpain, microtubule-associated protein 2 (MAP2), and α-tubulin and β-tubulin protein levels in spinal cord were determined. Results showed that after administration of 30 mg/kg ACR, decreased body we

This research aimed at analysing the protective effect of quercetin against the nephrotoxicity of acrylamide (AA) using metabonomics technology. Rats were randomly divided into six groups, including the control group, AA-treated group (5 mg per kg bw per d), two dosages of quercetin-treated groups (10 and 50 mg per kg bw per d) and two dosages of quercetin plus AA-treated groups. Quercetin and AA were administered to rats daily via gavage and drinking water, respectively. After a 16 week treatment, the kidneys were collected for metabonomics analysis. A total of 14 metabolites were identified

The degradation of acrylamide (AA) during UV/chlorine advanced oxidation process (AOP) was investigated in this study. The degradation of AA was negligible during UV irradiation alone. However, AA could be effectively degraded and mineralized during UV/chlorination due to the generation of hydroxyl radicals (OH). The degradation kinetics of AA during UV/chlorination fitted the pseudo-first order kinetics with the rate constant between AA and OH radicals being determined as 2.11 × 109 M-1 s-1. The degradation rate and mineralization of AA during UV/chlorination were significantly promoted at ac

Acrylamide is a monomer of polyacrylamide, which is widely used in the water treatment process as a flocculant. The degradation kinetics and formation of disinfection by-products (DBPs) during acrylamide chlorination were investigated in this study. The reaction between chlorine and acrylamide followed a pseudo-first-order kinetics. A kinetic model regarding acrylamide chlorination was established and the rate constants of each predominant elementary reaction (i.e., the base-catalyzed reaction of acrylamide with ClO- as well as the reactions of acrylamide with HOCl and ClO-) were calculated as

Potentially toxic acrylamide is largely derived from heat-induced reactions between the amino group of the free amino acid asparagine and carbonyl groups of glucose and fructose in cereals, potatoes, and other plant-derived foods. This overview surveys and consolidates the following dietary aspects of acrylamide: distribution in food originating from different sources; consumption by diverse populations; reduction of the acrylamide content in the diet; and suppression of adverse effects in vivo. Methods to reduce adverse effects of dietary acrylamide include (a) selecting potato, cereal, and o

Male rats receiving acrylamide (ACR) in their drinking water (100 ppm) for a six-week period displayed increased psychomotor stimulation to d-amphetamine (d-A; 1.0 mg/kg SC) under several conditions of handling and drug administration. Following behavioral tests a subset of the animals was sacrificed at 15, 50, 80 and 120 minutes following d-A and the brains removed and dissected for determinations of regional brain levels of several monoamine neurotransmitters and metabolites. ACR rats had elevated levels of 5-hydroxyindoleacetic acid (5-HIAA) in the striatum, septal area, and thalamus. The e

Reducing acrylamide (AA) formation in thermally processed foods, particularly bread, is a major food safety concern. Plant-based additives, including powders and extracts, offer a promising natural strategy for AA mitigation due to their bioactive compounds, such as polyphenols and flavonoids, which can scavenge reactive intermediates, modulate Maillard reactions, and inhibit lipid oxidation. This study systematically evaluated the effects of various plant powders and extracts, including green tea, rosemary, turmeric, buckwheat, and traditional spices, on AA formation in bread. Based on report

Vinpocetine (VIN) is a synthetic drug derived from the natural alkaloid vincamine. The antioxidation and anti-inflammation effects of VIN allow it to be used for multiple therapeutic purposes. So, the research aims to discover the possibility of using VIN to improve the nephrotoxicity of acrylamide (ACR). Twenty-four male albino rats were used in the trial: rats in the control group received 0.5 mL of oral saline, rats in the VIN group received an oral dose of VIN (5 mg/kg), rats in the ACR group received an oral dose of ACR (38.27 mg/kg), and rats in the VIN + ACR group received VIN and then

Acrylamide (ACR) is a widely used environmentally hazardous compound that is known to be neurotoxic, genotoxic, carcinogenic, and reproductive toxicity. It is widely present in soil, water, agents used in chemical industries, and food. It can be distributed to all organs and tissues, and can cause damage to various human systems and those of other animals. Previous metabolomics studies have mainly focused on metabolites in serum and urine, but have lacked comprehensive analysis of major organs and tissues. In the current study, a gas chromatography-massspectrometry method was used to investiga

More than one-third of the calories consumed by U.S. and European populations contain acrylamide, a substance classified as a "probable human carcinogen" based on laboratory data. Thus, it is a public health concern to evaluate whether intake of acrylamide at levels found in the food supply is an important cancer risk factor. Mean dietary intake of acrylamide in adults averages 0.5 microg/kg of body weight per day, whereas intake is higher among children. Several epidemiological studies examining the relationship between dietary intake of acrylamide and cancers of the colon, rectum, kidney, bl

The synthesis of poly(N-isopropyl acrylamide) (pNIPA)-based polymers via the surfactant-free precipitation polymerization (SFPP) method produced thermosensitive nanospheres with a range of distinctive physicochemical properties. Nano- and microparticles were generated using various initiators, significantly influencing particle characteristics, including the hydrodynamic diameter (DH), which varied from 87.7 nm to 1618.1 nm. Initiators, such as potassium persulfate and 2,2'-azobis(2-methylpropionamidine) dihydrochloride, conferred anionic and cationic functionalities, respectively, impacting t

Amyloid β peptide (Aβ) aggregation in the brain represents an initial detrimental episode in the etiology of Alzheimer's disease (AD). Recently, it has been discovered that inhibiting Aβ neurotoxicity by modulating highly toxic Aβ oligomers (AβOs) is more rewarding than reducing the overall amyloid fibril production. In line with this, here, we discussed the efficiency of multifunctional quinazolinone and vanillin acrylamide hybrids (QVA1-5) as modulators of aggregation behavior. The thioflavin T (ThT) assay inferred dose-dependent intensification of Aβ1-42 aggregation by QVA1-5, which may be
