Overview of porcine interstitial and bronchointerstitial pneumonia: infectious and non-infectious causes.

Interstitial pneumonia in swine is an important respiratory condition in which inflammation and thickening of the pulmonary interstitium result in impaired gas exchange and respiratory distress. The multifactorial etiology of the disease includes viral, bacterial, and parasitic pathogens, as well as environmental or toxic insults. Clinical signs range from acute respiratory distress and fever to chronic weight loss and reduced growth performance, contributing to economic losses due to decreased productivity and increased mortality. Pathogenesis typically begins with a primary insult, commonly

Microbial communities in the tonsils of healthy pigs.

The tonsils of mammals such as humans and pigs are colonized with an extensive microbiota and are frequently the site for asymptomatic carriage of bacterial pathogens. The goal of this study was to determine the composition of the microbial community of the tonsils in healthy pigs. Tonsils were collected from eight pigs from two different healthy herds. Samples of the tonsils from each pig were used for culture dependent and culture independent identification of the microbial community. Aerobic cultivation identified Pasteurella multocida, Actinobacillus spp., Staphylococcus aureus, Staphyloco

Exposure of feral swine (Sus scrofa) in the United States to selected pathogens.

Feral swine (Sus scrofa) are widely distributed in the United States. In 2011 and 2012, serum samples and tonsils were recovered from 162 and 37 feral swine, respectively, in the US to evaluate exposure to important swine endemic pathogens. Antibodies against porcine reproductive and respiratory syndrome virus (PRRSV) and porcine circovirus type 2 (PCV2) were found in 2.5% and 25.3% of tested sera, respectively. Positive serological reactions against Mycoplasma hyopneumoniae and Actinobacillus pleuropneumoniae have been detected in 19.7% and 69.7% of animals. More than 15% of animals presented

Use of recombinant ApxIV in serodiagnosis of Actinobacillus pleuropneumoniae infections, development and prevalidation of the ApxIV ELISA.

Actinobacillus pleuropneumoniae is the etiological agent of porcine pleuropneumonia, which causes worldwide severe losses in pig farming. The virulence of the 15 serotypes of A. pleuropneumoniae is mainly determined by the three major RTX toxins ApxI, ApxII and ApxIII, which are secreted by the different serotypes in various combinations. A fourth RTX toxin, ApxIV, is produced by all 15 serotypes only during infection of pigs, but not under in vitro conditions. Pigs infected with A. pleuropneumoniae show specific antibodies directed against ApxIV. In contrast, antibodies against the other thre

Retrospective Analysis of the Detection of Pathogens Associated with the Porcine Respiratory Disease Complex in Routine Diagnostic Samples from Austrian Swine Stocks.

The diagnostic workup of respiratory disease in pigs is complex due to coinfections and non-infectious causes. The detection of pathogens associated with respiratory disease is a pivotal part of the diagnostic workup for respiratory disease. We aimed to report how frequently certain viruses and bacteria were detected in samples from pigs with respiratory symptoms in the course of routine diagnostic procedures. Altogether, 1975 routine diagnostic samples from pigs in Austrian swine stocks between 2016 and 2021 were analysed. PCR was performed to detect various pathogens, including porcine repro

RPA-CRISPR/Cas12a-Based Detection of Haemophilus parasuis.

Haemophilus parasuis (H. parasuis, HPS) is a prominent pathogenic bacterium in pig production. Its infection leads to widespread fibrinous inflammation in various pig tissues and organs, often in conjunction with various respiratory virus infections, and leads to substantial economic losses in the pig industry. Therefore, the rapid diagnosis of this pathogen is of utmost importance. In this study, we used recombinase polymerase amplification (RPA) and clustered regularly interspaced short palindromic repeats (CRISPR) technology to establish a convenient detection and analysis system for H. par

Culturomics of the pig tonsil microbiome identifies new species and an untapped source of novel antimicrobials.

BACKGROUND: In humans and pigs, altered composition of the microbiota associated with the epithelium of the palatine tonsils has been associated with bacterial or viral infection and lymphoid tissue inflammation. Tonsil lymphoid tissue is important for immunity and considered an important portal of entry for pathogens such as Streptococcus suis. Little is known about correlations between tonsil-associated microbiota, tonsillar infections, and the species that might confer colonization resistance against pathogens. Here, we describe a large collection of representative bacterial species from th

Effects of a novel feed additive on clinical symptoms and the nasal and cecal microbiome in nursery pigs challenged with PRRSV and Streptococcus suis.

Porcine reproductive and respiratory syndrome virus (PRRSV) is a major endemic disease in swine, causing significant economic losses to the pork industry. PRRSV infection predisposes pigs to secondary infections, leading to a costly and challenging respiratory disease complex. Effective mitigation strategies are needed to reduce negative impacts of PRRSV on swine health and productivity. This study evaluated the efficacy of a novel feed additive (EP) containing a blend of activated medium-chain fatty acids, phytogenic compounds, and probiotics in PRRSV viral load, clinical symptoms, lung lesio

An unusual culprit: Klebsiella pneumoniae causing septicaemia outbreaks in neonatal pigs?

This study investigated the cause of recent outbreaks of septicaemia in neonatal pigs in Australia (Victoria and Queensland). Septicaemia in neonatal pigs is commonly caused by enterotoxigenic E. coli, extraintestinal pathogenic E. coli and beta-haemolytic streptococci. Infrequent causes of septicaemia are Actinobacillus suis and Citrobacter freundii. Therefore, it was quite unexpected when Klebsiella pneumoniae was isolated in predominant growth from multiple organs of pigs dying of septicaemia from six days of age. Two cases in Victoria were associated with multilocus sequence type 25 (ST25)

Nasal Colonizers from Sows in the Federal District of Brazil Showed a Diverse Phenotypic Resistance Profile.

Antimicrobial resistance (AMR) is a major public health concern influenced by antimicrobial use (AMU) in animal production systems. In swine, metaphylactic treatments may contribute to the emergence and dissemination of resistance genes. In this study, we isolated bacteria from the nasal cavities of 50 sows across 10 farms in the Federal District, Brazil. A total of 132 bacterial isolates were obtained and tested for phenotypic resistance to 23 antimicrobials using the disk diffusion method. Resistance was detected against all tested antimicrobials, with an overall resistance rate of 55.6% (16

Prevalence of pseudorabies virus infection and associated infections in six large swine herds in Illinois.

Sera were collected from 6 large farrow-to-finish swine herds infected with pseudorabies virus (PRV) in Illinois. All herds were participating in the Large Herd Cleanup Study, a USDA-initiated project to evaluate the feasibility of eradicating pseudorabies from large farms (greater than 400 sows) by use of a combination of vaccination and management changes. Herd size ranged between 425 and 1,500 breeding females. Between April and July 1990, sera for measurement of PRV antibodies were obtained from 113 to 156 sows and 112 to 162 finishing pigs (body weight greater than 70 kg)/herd. Duplicate

Efficacy of vaccines against bacterial diseases in swine: what can we expect?

This paper discusses what can be expected with regard to efficacy of antibacterial vaccines used in swine, based on the present knowledge of pathogen-host interactions. First, vaccination against bacteria that mainly cause disease by production of exotoxins is considered. Vaccines containing the inactivated toxin or a non-toxic but antigenic recombinant protein derived from the exotoxin can be expected to provide protection against disease. The degree of protection induced by such vaccines varies, however, depending amongst other things on the pathogenesis of the disease. Vaccination against c

Diagnostic and pathological characterization of senecavirus A-associated epidemic transient neonatal losses in swine.

Senecavirus A (SVA) has emerged as a significant viral pathogen of swine, traditionally associated with idiopathic vesicular disease but more recently linked to epidemic transient neonatal losses (ETNL), a syndrome characterized by increased mortality in piglets without consistent vesicular lesions. The mechanisms underlying SVA-associated neonatal mortality remain unclear due to the lack of specific clinical signs, frequent co-infections, and limited tissue characterization. This retrospective study analyzed 186 neonatal submissions (<24 days old) that were tested for SVA by RT-qPCR at the Io
