Metabolic reprogramming and signalling networks in microglial activation: Implications for neurodegeneration.

Microglia, the brain's innate immune cells, undergo a complex activation process characterized by metabolic reprogramming and inflammatory signalling that transcends the classical M1/M2 polarization framework. Recent advances in single-cell technologies have unveiled remarkable microglial heterogeneity, including the emergence of disease-associated microglia (DAM), which play critical roles in neurodegeneration. This study synthesizes recent findings from single-cell transcriptomics, spatial proteomics, and metabolomics to provide an integrated perspective on microglial activation. We focus on

Synthesis and evaluation of tofacitinib analogs designed to mitigate metabolic activation.

Tofacitinib (TFT), a JAK inhibitor used for the treatment of rheumatoid arthritis and other diseases, is associated with severe liver injury that is believed to be caused by its reactive aldehyde or epoxide metabolites. In this study, we synthesized six tofacitinib analogs designed to avoid the formation of reactive metabolites and evaluated their JAK3 inhibitory activity, metabolic stability, CYP3A time-dependent inhibition, and cytotoxicity. Our data indicated that purine analog 3, which showed little inhibition of CYP3A and cytotoxicity and inhibited JAK3 in the nanomolar range, could be a

Gender differences in microsomal metabolic activation of hepatotoxic clivorine in rat.

The gender differences in the in vitro microsomal metabolic activation of hepatotoxic clivorine, a representative naturally occurring hepatotoxic otonecine type pyrrolizidine alkaloid, in Sprague-Dawley rats and their relation to the gender differences in susceptibility to clivorine intoxication were reported in the present study. Clivorine-induced liver damage in the male rat via metabolic activation to form the reactive pyrrolic ester followed by covalent binding to liver tissue constituents has been reported previously by our research group. The present study demonstrated, for the first tim

Release of Cr(III) from Cr(III) picolinate upon metabolic activation.

Hexavalent and trivalent chromium are released into the environment from a number of different industrial activities. It is known that Cr(VI) can be reduced and subsequently complexed by humic acids to produce Cr(III) humic acid complexes in the soil and aquatic environments. The metabolic fate of Cr(III) humic acid complexes and other Cr(III) organic complexes in mammalian systems is unknown. Therefore, Cr(III) picolinate was chosen as a model complex for Cr(III) humic acid complexes and other environmentally relevant Cr(III) complexes. Both human hepatocyte microsomes and primary cultures of

PKC-dependent delayed metabolic preconditioning is independent of transient MAPK activation.

In this study we used an in vitro model of delayed preconditioning to investigate activation of mitogen-activated protein kinases (MAPKs) and their potential role in protection. Neonatal rat cardiomyocytes were preconditioned using a buffer containing glycolytic inhibitors and low pH (minimal metabolic preconditioning; MMPC) consisting of modified Krebs buffer, 10 mM 2-deoxyglucose, and 20 mM lactate, pH 6.8, for 2 h followed by 24 h of simulated reperfusion before lethal simulated ischemia (LSI). MAPK activation during the MMPC protocol was determined using phospho-specific antisera and the e

Metabolic activation of sennoside C in mice: synergistic action of anthrones.

Sennosides A and C directly injected into the caecum of mice showed equal purgative activity. Intracaecal administration reduced time to onset of diarrhoea induced by sennoside C from about 3 h after oral administration to about 24 min. At 2.3 h after oral administration of sennoside C, nearly equimolar amounts of aloe-emodin anthrone and rhein anthrone were detected in the large intestine of mice. The purgative effect of oral sennoside C could be reduced by pretreating mice with chloramphenicol. This was observed as a decreased formation of total anthrones in the large intestine. Both anthron

The effect of chemical substitution on the metabolic activation, metabolic detoxication, and pharmacological activity of amodiaquine in the mouse.

The adverse reactions associated with the antimalarial amodiaquine (AQ), agranulocytosis and hepatotoxicity, have been attributed to the bioactivation of the drug to a quinone imine metabolite. Therefore the effect of chemical modification on the metabolism of AQ was studied, with particular reference to the prevention of bioactivation and the introduction of glucuronidation. Glutathione conjugates of AQ and desethylAQ were eliminated in bile after intraportal administration of [3H]AQ (54 mumol/kg, 20 microCi/kg) to anesthetized male CD1 mice. Thioether conjugates excreted into bile over 3 h a

Non-Negligible Source of PFOS: Biochemical Fate and Metabolic Activation of Precursors.

The widespread detection of PFOS precursors in the environment and human matrices raises a new wave of health concerns. However, their absorption, distribution, metabolism, and excretion (ADME) fates and toxicity profiles in mammals remain largely unexplored. Here, we investigated the toxicokinetic, metabolic transformation, and toxicity of three precursors, N-MeFOSA, N-EtFOSA, and N-EtFOSAA, by integrating toxicokinetic modeling in mice, rat liver microsome metabolism, and hepatocellular toxicity assays. N-MeFOSA and N-EtFOSA were rapidly eliminated in vivo and exhibited high efficiencies of

In vitro and in vivo metabolic activation of diaveridine mediated by CYP3A.

Diaveridine (DVD) is an anticoccidial drug and antibacterial synergist used in the livestock and poultry industries worldwide. DVD residues have been reportedly detected in pork and related meat products. Human exposure to DVD through the environment and the food chain has attracted wide attention, due to the abuse of DVD. Liver injury of animals administered with DVD has been observed and reported, but the potential mechanisms are unelucidated so far. This study aimed to reveal the metabolic activation pathway of DVD that may be related to its hepatotoxicity. DVD was found to be metabolized t

Metabolic crisis and TRPM4 activation cause QT prolongation in TANGO2 deficiency disorder.

AIMS: TANGO2 deficiency disorder (TDD), caused by biallelic pathogenic variants in TANGO2, is associated with impaired fatty acid oxidation and metabolic crises that frequently trigger QT prolongation and arrhythmias. The objective was to study the mechanisms responsible for QT prolongation and arrhythmogenesis in TDD. METHODS AND RESULTS: CRISPR/Cas9 were used to generate human-induced pluripotent stem cell cardiomyocytes (hiPSC-CMs) carrying a TANGO2 exon 3-9 deletion (TANGO2-/-). Metabolic activation of flunitrazepam via nitroreduction mediated by aldo-keto reductase 1C3.

Aldo-keto reductases (AKRs) are a superfamily of NAD(P)(H)-dependent oxidoreductases (phase I enzymes) that reduce aldehydes and ketones to primary and secondary alcohols, respectively. Four percent of xenobiotic biotransformation has been attributed to AKRs, although this is likely an underestimate because this is based on their ability to act as carbonyl reductases. AKRs also have an emerging role in nitroreduction. We screened 6 nitro-containing drugs (flunitrazepam, clonazepam, nimesulide, nilutamide, flutamide, and chloramphenicol) as substrates against recombinant human AKR1C1, AKR1C2, A

Rapid transcriptional and metabolic adaptation of intestinal microbes to host immune activation.

The gut microbiota produces metabolites that regulate host immunity, thereby impacting disease resistance and susceptibility. The extent to which commensal bacteria reciprocally respond to immune activation, however, remains largely unexplored. Herein, we colonized mice with four anaerobic symbionts and show that acute immune responses result in dramatic transcriptional reprogramming of these commensals with minimal changes in their relative abundance. Transcriptomic changes include induction of stress-response mediators and downregulation of carbohydrate-degrading factors such as polysacchari

In vivo metabolic effects after acute activation of skeletal muscle Gs signaling.

OBJECTIVE: The goal of this study was to determine the glucometabolic effects of acute activation of Gs signaling in skeletal muscle (SKM) in vivo and its contribution to whole-body glucose homeostasis. METHODS: To address this question, we studied mice that express a Gs-coupled designer G protein-coupled receptor (Gs-DREADD or GsD) selectively in skeletal muscle. We also identified two Gs-coupled GPCRs that are endogenously expressed by SKM at relatively high levels (β2-adrenergic receptor and CRF2 receptor) and studied the acute metabolic effects of activating these receptors in vivo by hig

Acetyl-CoA carboxylase inhibition disrupts metabolic reprogramming during hepatic stellate cell activation.

BACKGROUND & AIMS: Non-alcoholic steatohepatitis (NASH) is a chronic liver disease characterized by hepatic lipid accumulation, inflammation, and progressive fibrosis. Acetyl-CoA carboxylase (ACC) catalyzes the rate-limiting step of de novo lipogenesis and regulates fatty acid β-oxidation in hepatocytes. ACC inhibition reduces hepatic fat content and markers of liver injury in patients with NASH; however, the effect of ACC inhibition on liver fibrosis has not been reported. METHODS: A direct role for ACC in fibrosis was evaluated by measuring de novo lipogenesis, procollagen production, gene

Regioselectivity of metabolic activation of acetylenic steroids by hepatic cytochrome P450 isozymes.

Liver cytochrome P450 monooxygenases (P450), a group of isozymes that catalyze the reductive cleavage of molecular oxygen, dominate hepatic metabolism of xenobiotic lipophilic substances. These P450 enzymes exhibit broad and overlapping substrate specificities, in contrast to the P450 isozymes of the steroid biosynthetic pathways, which are highly substrate specific. Hepatic heme pigments, N-alkylated porphyrins, accumulate following the self-catalyzed destruction of P450 by the metabolic activation of 17 alpha-ethynyl steroids. Acetaminophen nephrotoxicity in the rat: quantitation of renal metabolic activation in vivo.

Renal cortical necrosis induced by acetaminophen (APAP) may be related to generation of reactive intermediates by two mechanisms of metabolic activation, direct P-450 dependent metabolic activation (P-450) or metabolic activation subsequent to deacetylation of APAP to p-aminophenol (PAP). Generation of arylating intermediates by both pathways of metabolic activation was quantified in cyclohexamide (HEX)-pretreated or naive rats in vivo with specifically labeled [14C]APAP. The association of each type of metabolic activation with APAP-induced nephrotoxicity was determined in Fischer 344 (F344)

Spatially Resolved Multiomics Reveals Metabolic Remodeling and Autophagy Activation in Adamantinomatous Craniopharyngiomas.

Adamantinomatous craniopharyngioma (ACP), a benign yet highly recurrent and therapy-resistant intracranial tumor, remains a considerable clinical challenge because of its complex pathological structure, infiltrative growth, and limited treatment options. Here, integrated spatially resolved multiomics is employed-including single-cell spatial transcriptomics via CosMx SMI and spatially resolved metabolomics via AFADESI-MSI, accompanied by bulk metabolomics and functional validation-to unravel the driving factors of ACP progression and recurrence. Analysis results reveal three interdependent bio
