Derivatives of beta-adrenergic antagonists. N-Nitrosopropranolol and N-hydroxypropranolol and its aldonitrone.

Potential precursors to chemically reactive species derived from the beta-adrenergic antagonist propranolol were synthesized and tested for mutagenicity in the Ames Salmonella assay. N-Hydroxypropranolol (1), the corresponding aldonitrone, 3-(1-naphthoxy)-2-hydroxypropionaldehyde N-isopropylnitrone (2), and N-nitrosopropranolol (3) were prepared and tested. N-Hydroxypropranolol (1) was obtained by direct alkylation of 3-(1-naphthoxy)-1-bromo-2-propanol with N-isopropylhydroxylamine and isolated as its neutral oxalate or HBr salt. The aldonitrone (2) was obtained by mercuric oxide oxidation of

Retrograde amnesia produced by electrical stimulation of the amygdala: attenuation with adrenergic antagonists.

Subseizure electrical stimulation of the amygdala produced retrograde amnesia for a visual discrimination shock-motivated task. Animals pretreated with the alpha-adrenergic antagonist phenoxybenzamine, or the beta-adrenergic antagonist propranolol, did not develop amnesia. The findings indicate that adrenergic antagonists attenuate amnesia produced by amygdala stimulation for visual discrimination training. These results are consistent with previous evidence indicating that adrenergic antagonists attenuate the amnesias produced by a variety of agents, and thus, suggest that adrenergic mechanis

Design and synthesis of aryloxypropanolamine as β3-adrenergic receptor antagonist in cancer and lipolysis.

β-adrenergic receptors (β-ARs) are broadly distributed in various tissues and regulate a panel of important physiological functions and disease states including cancer. Above all, β3-adrenergic receptor (β3-AR) plays a significant role in regulating lipolysis and thermogenesis in adipose tissue. In this study, we designed and synthesized a series of novel L-748,337 derivatives as selective human β3-AR antagonists. Among all the tested L-748,337 analogs, compound 23d was found to display 23-fold more potent β3-AR antagonist activity (EC50 = 0.5117 nM) than L-748,337 (EC50 = 11.91 nM). In vivo,

Sustained ventricular unloading action of the alpha-adrenergic antagonist nicergoline in the dog.

Preliminary studies indicate that nicergoline, a new alpha-antagonist, can lower HR at doses which decrease arterial BP. The present animal study was designed to quantitate the systemic and carotid (pulsed Doppler) hemodynamic and renin release consequences of nicergoline ventricular unloading, and to investigate the hemodynamic mechanisms of hypotension. In 11 anesthetized dogs, nicergoline infusion induced progressive, moderate, and prolonged hypotension associated with reduced vascular resistance and capacitance. Hypotension resulted from decreased HR and cardiac output, without an increase

The influence of adrenergic receptor antagonists on the amnestic and antiamnestic actions of adrenaline and tyramine.

The posttraining IP administration of adrenaline (epinephrine) HCl (5.0 micrograms/kg) or tyramine HCl (1.0 mg/kg) causes retrograde amnesia for a one-way step-down inhibitory avoidance task in rats. The effect is cancelled by the simultaneous injection of the alpha 2-adrenergic receptor antagonist yohimbine HCl (2.0 mg/kg) or of the beta 1 - beta 2 blocker propranolol HCl (2.0 mg/kg). The amnestic effect of posttraining adrenaline or tyramine is counteracted by the administration of adrenaline or tyramine prior to testing: each drug has a greater antiamnestic effect against itself than agains

Alpha-adrenergic antagonists: correlation of the effect on intraocular pressure and on alpha 2-adrenergic receptor binding specificity in the rabbit eye.

Six alpha-adrenergic antagonists, which have a range of selectivities for alpha 1- and alpha 2-adrenoreceptor subtypes, were compared with respect to their ability to reduce intraocular pressure (IOP) after topical application to the rabbit eye, and their affinity and selectivity for alpha 2-adrenoreceptors, as determined by binding to membranes prepared from rabbit iris-ciliary body. A routine assay for alpha 2-adrenoreceptors using [3H]-rauwolscine was developed for this purpose. ICB contained 200-300 fmol (mg protein)-1 alpha 2-adrenoreceptors which represents approximately two-thirds of th

β2-Adrenergic receptor antagonists are not protective against spinal cord injury-induced bone loss.

Bone loss is an underestimated consequence of spinal cord injury (SCI), that manifests rapidly and is resistant to currently available treatments including exercise, functional electrical stimulation, anabolic agents, and bisphosphonates. While changes in adrenergic signaling have been linked to bone loss, it remains unclear whether maladaptive sympathetic signaling drives SCI-induced bone loss. We evaluated the effects of sympathetic blockade on SCI-induced bone loss using primary bone marrow osteoclast cultures and an in vivo rat model of a moderate T11 spinal contusion injury using young (3

Alpha1-adrenergic antagonists block 6-nitrodopamine contractions on the rat isolated epididymal vas deferens.

6-nitrodopamine (6-ND) is released from rat isolated vas deferens and modulates electrical-field stimulation (EFS) contractions of the rat isolated epididymal vas deferens (RIEVD) via a specific receptor which is blocked by tricyclic antidepressants. Here, the effects of selective α1-adrenergic receptor antagonists on RIEVD contractions induced by 6-ND, dopamine, noradrenaline, adrenaline and EFS were investigated. Doxazosin and tamsulosin (3-10 nM) caused significant rightward shifts of the concentration-response curve to 6-ND, but had no effect on dopamine-, noradrenaline- and adrenaline-ind

Effects of Adrenergic Agonists and Antagonists on Cardiopulmonary Function During Normobaric Hypoxia in Rat.

Pulmonary edema (PE) is an issue widely noted in acute exposure to hypoxia as seen in high altitude climbers, yet the etiology of this is not defined. Previous studies in rats showed that both hypoxia and strong sympathetic activation may induce PE. As acute exposure to hypoxia is accompanied by sympathetic activation, we assume that this may impair pulmonary circulation and contribute to the development of hypoxic PE. The aim of the present study was to investigate the effects of adrenergic agonists and antagonists as models for overstimulation and suppression, respectively, of sympathetic ac

Quantitative relationship between beta-adrenergic receptor number and physiologic responses as studied with a long-lasting beta-adrenergic antagonist.

The aminobenzyl analog of propranolol, 1- (p-amino-alpha,alpha-dimethylphenethylamino)-3-(1-naphthoxy)-2- propanol, was synthesized and found to be a potent beta-adrenergic blocking agent. The beta-adrenergic receptors of cultured rat C6 glioma cells (2B clone) as assessed by [(125)I]iodohydroxybenzylpindolol binding were decreased 50 and >95% after pretreatment with 8 nM and 1 muM aminobenzylpropranolol, respectively. Unlike propranolol, aminobenzylpropranolol displayed a prolonged blockade of receptors that was maintained during several hours of washing. [(125)I]Iodohydroxybenzylpindolol sat

Selective β2-adrenergic Antagonist Butoxamine Reduces Orthodontic Tooth Movement.

Recently, involvement of the sympathetic nervous system in bone metabolism has attracted attention. β2-Adrenergic receptor (β2-AR) is presented on osteoblastic and osteoclastic cells. We previously demonstrated that β-AR blockers at low dose improve osteoporosis with hyperactivity of the sympathetic nervous system via β2-AR blocking, while they may have a somewhat inhibitory effect on osteoblastic activity at high doses. In this study, the effects of butoxamine (BUT), a specific β2-AR antagonist, on tooth movement were examined in spontaneously hypertensive rats (SHR) showing osteoporosis with

Combined use of alpha-adrenergic and muscarinic antagonists for the treatment of voiding dysfunction.

PURPOSE: We provide an overview of the medical literature supporting the combined use of muscarinic and alpha-adrenergic antagonist therapy for the treatment of voiding dysfunction. MATERIALS AND METHODS: The MEDLINE database (1966 to 2004) of the United States National Library of Medicine was searched for pertinent studies. RESULTS: Although the mechanism of action of alpha-adrenergic antagonist therapy for voiding dysfunction has traditionally been assumed to be relaxation of the periurethral, prostatic and bladder neck smooth muscle, substantial evidence supports action at extraprostatic

Mechanism of the lipid-mobilizing effect of alpha-2 adrenergic antagonists in the dog.

Alpha-2 adrenergic antagonists may constitute putative antiobesity agents. These drugs may act: 1) by blockade of the antilipolytic alpha-2 adrenoceptor on fat cell membranes and 2) by activation of the sympathetic adrenergic system (blockade of central and presynaptic alpha-2 adrenoceptors). Studies were undertaken in the dog, a species possessing fat cell alpha-2 and beta-adrenoceptors in order: 1) to define and compare the metabolic and endocrinological impacts of recently discovered alpha-2 antagonists [idazoxan and SK&F 86,466 (6-chloro-N-methyl-2,3,4,5-tetrahydro-1H-3-benzazepine)] and o

Treatment of mild and moderate hypertension with medroxalol, an alpha- and beta-adrenergic antagonist.

Medroxalol, a new antihypertensive agent with alpha- and beta-adrenoreceptor blocking properties in both animals and humans, was administered in a single-blind study for 12 weeks to 29 patients with mild and moderate hypertension (standing blood pressure: 188-130/130-100 mm Hg). After 4 weeks of placebo administration, treatment with oral medroxalol was begun. Six weeks later, half the subjects added hydrochlorothiazide, 12.5 mg twice daily, to medroxalol for an additional 6 weeks, and the other half added placebo. During the final 4-week period medroxalol, but not hydrochlorothiazide, was dis

Adrenergic agonists and antagonists as antiglaucoma agents: a literature and patent review (2013-2019).

Introduction: Glaucoma is a neurodegenerative disease of the eye characterized by selective retinal ganglion cell loss that provokes progressive defects in the visual field. Elevated intraocular pressure (IOP) is an important contributor for the progression of glaucoma. The current therapeutic arsenal for reducing IOP includes prostaglandin analogs, β-blockers, carbonic anhydrase inhibitors, α-adrenergic agonist, miotics, rho-kinase inhibitors and combinations thereof, generally administered as eye drops. Areas covered: This manuscript reviews the state of art on adrenergic modulators for trea

Effects of catecholamine synthesis inhibitors and adrenergic receptor antagonists on restraint-induced LH release.

Acute stress is known to increase LH secretion and the release of central norepinephrine (NE) in intact rats. Studies were performed to analyse the ole of catecholamines in acute stress-induced LH release in male rats. Injection of alpha-methyl-p-tyrosine (alpha MPT) and diethyldithiocarbamate (DDC), catecholamine synthesis inhibitors, significantly decreased both hypothalamic concentration of NE and serum LH. Restraint for 30 min evoked an increase in serum LH in saline-treated rats, whereas alpha MPT and DDC administration blocked the stress-induced LH release. The effects of alpha 1-, alpha

Effects of beta-adrenergic antagonists on salivary secretory function in individuals of different ages.

The purpose of this study was to assess the effects of chronic beta-adrenergic antagonists on parotid and submandibular gland secretions in men and women of different ages. Unstimulated and stimulated saliva flow rates, total protein concentrations, and protein secretion rates were compared from medicated (various beta-antagonists, n = 25) and control (n = 60) subjects. Age-related decreases were found in unstimulated parotid saliva flow rate (p = .011) and protein secretion rate (p = .04), unstimulated submandibular salivary flow rate (p = .005) and protein secretion rate (p = .010), and in s
