Metabolic engineering of Komagataella phaffii for the efficient utilization of methanol.

BACKGROUND: Komagataella phaffii, a type of methanotrophic yeast, can use methanol, a favorable non-sugar substrate in eco-friendly bio-manufacturing. The dissimilation pathway in K. phaffii leads to the loss of carbon atoms in the form of CO2. However, the ΔFLD strain, engineered to lack formaldehyde dehydrogenase-an essential enzyme in the dissimilation pathway-displayed growth defects when exposed to a methanol-containing medium. RESULTS: Inhibiting the dissimilation pathway triggers an excessive accumulation of formaldehyde and a decline in the intracellular NAD+/NADH ratio. Here, we desi

An enzymic assay for the specific determination of methanol in serum.

This method for the specific determination of methanol in serum is based on the following two reactions: (formula; see text) Alcohol oxidase is not specific: it converts all lower alcohols to their corresponding aldehydes; however, formaldehyde dehydrogenase is specific and thus the transformation of NAD+ to NADH (which is used to monitor the reaction) proceeds only if methanol is originally present in the sample. The method was automated with a Roche COBAS FARA centrifugal analyzer. The calibration curve is linear between 0.6 and 12 mmol/L. The detection limit is about 0.6 mmol/L. The CV is 4

STEAP Proteins: Roles in disease biology and potential for therapeutic intervention.

Iron and copper are essential metal ions, and maintaining their metabolic balance is critical for organismal health. The Six-Transmembrane Epithelial Antigen of the Prostate (STEAP) protein family, comprising STEAP1, STEAP2, STEAP3, and STEAP4, plays a vital role in cellular metal homeostasis. These proteins are located on the cell membrane and are characterized by six transmembrane domains. With the exception of STEAP1, the STEAP proteins function as metal oxidoreductases due to their F420H2:NADP+ oxidoreductase (FNO)-like domain. However, STEAP1 contributes to metal metabolism through its he

Structural analysis of an anthrol reductase inspires enantioselective synthesis of enantiopure hydroxycycloketones and β-halohydrins.

Asymmetric reduction of prochiral ketones, particularly, reductive desymmetrization of 2,2-disubstituted prochiral 1,3-cyclodiketones to produce enantiopure chiral alcohols is challenging. Herein, an anthrol reductase CbAR with the ability to accommodate diverse bulky substrates, like emodin, for asymmetric reduction is identified. We firstly solve crystal structures of CbAR and CbAR-Emodin complex. It reveals that Tyr210 is critical for emodin recognition and binding, as it forms a hydrogen-bond interaction with His162 and π-π stacking interactions with emodin. This ensures the correct orient

[Methanol dehydrogenase, a key enzyme of one-carbon metabolism: a review].

One-carbon compounds such as methanol and methane are cheap and readily available feedstocks for biomanufacturing. Oxidation of methanol to formaldehyde catalyzed by methanol dehydrogenase (MDH) is a key step of microbial one-carbon metabolism. A variety of MDHs that depend on different co-factors and possess different enzymatic properties have been discovered from native methylotrophs. Nicotinamide adenine dinucleotide (NAD)-dependent MDHs are widely used in constructing synthetic methylotrophs, whereas this type of MDH usually suffers from low methanol oxidation activity and low affinity to

Identification of a Hexenal Reductase That Modulates the Composition of Green Leaf Volatiles.

Green leaf volatiles (GLVs), including six-carbon (C6) aldehydes, alcohols, and esters, are formed when plant tissues are damaged. GLVs play roles in direct plant defense at wound sites, indirect plant defense via the attraction of herbivore predators, and plant-plant communication. GLV components provoke distinctive responses in their target recipients; therefore, the control of GLV composition is important for plants to appropriately manage stress responses. The reduction of C6-aldehydes into C6-alcohols is a key step in the control of GLV composition and also is important to avoid a toxic b

Effects of cyanamide and clofibrate on the enzymes of ethanol oxydation and on ethanol consumption in the rat.

The action of cyanamide and of clofibrate was studied on the voluntary drinking behaviour of alcohol intoxicated animals and also on the enzymes of ethyl oxydation. Both substances reduce alcohol consumption by about 35% and cause metabolic modifications: inhibition of aldehyde dehydrogenase and of catalase by cyanamide; activation of alcohol- and aldehyde-dehydrogenases by clofibrate. These effects are constantly found in most of the tissues of the rat. The results are discussed bearing in mind the relationships between alcohol metabolism, acetaldehyde metabolism, the toxicity of alcohol and

Structure-Based Redesign of a Methanol Oxidase into an "Aryl Alcohol Oxidase" for Enzymatic Synthesis of Aromatic Flavor Compounds.

Alcohol oxidases (AOxs) catalyze the aerobic oxidation of alcohols to the corresponding carbonyl products (aldehydes or ketones), producing only H2O2 as the byproduct. The majority of known AOxs, however, have a strong preference for small, primary alcohols, limiting their broad applicability, e.g., in the food industry. To broaden the product scope of AOxs, we performed structure-guided enzyme engineering of a methanol oxidase from Phanerochaete chrysosporium (PcAOx). The substrate preference was extended from methanol to a broad range of benzylic alcohols by modifying the substrate binding p

Discovery, production and characterization of bacterial aryl-alcohol oxidases.

Alcohol oxidation is a pivotal reaction in synthetic chemistry. Classical procedures typically involve the use of hypervalent reagents and organic solvents that often lead to poor selectivity, overoxidation issues and low atom-economy processes. The implementation of biocatalytic methods for this reaction is particularly appealing and several enzyme types have shown to catalyze this process although equilibrium issues, cofactor and coenzyme recycling or the use of external mediators limit the uptake of biocatalytic oxidations in large scale. Alcohol oxidases catalyze the non-reversible aerobic

The microbial oxidation of methanol. The alcohol dehydrogenase of Pseudomonas sp. M27.

1. No primary hydrogen acceptor other than phenazine methosulphate has been found for the alcohol dehydrogenase from Pseudomonas sp. M27. 2. None of a wide range of vitamins or cofactors has any effect on the activity of the enzyme. 3. The enzyme is far less sensitive to metal-chelating agents and thiol reagents than are other alcohol dehydrogenases. 4. Methanol is oxidized at least as fast as other alcohols by this enzyme and its well-defined substrate specificity is different from that of other alcohol dehydrogenases. Only primary alcohols are oxidized; the general formula for an oxidizable

Enzymatic Cleanup of Formaldehyde in Aqueous Solutions.

Numerous methods have been developed to address gaseous formaldehyde pollution, but most of them cannot be applied directly to eliminate the pollution of formaldehyde in aqueous solutions. Formaldehyde in aqueous solutions can be leached from formaldehyde-containing solid materials (e. g., food, wood, clothes, resins) and absorbed from gaseous formaldehyde by water. Here we developed an enzymatic cleanup technique - the reconstitution of an enzyme cocktail consisting of three coenzyme-free oxidoreductases (i. e., formaldehyde dismutase, methanol oxidase, and formate oxidase) and catalase for t

Iterative and data-driven ortholog mining enables reliable discovery of stereoselective ketoreductases.

Ketoreductases (KREDs) enable stereoselective alcohol synthesis and are widely used industrially. Yet identifying process-compatible and selective enzymes for non-native substrates remains challenging, requiring extensive screening. Here, we present an iterative ortholog mining strategy to explore functional space. Starting from a known KRED, we sample its orthogroup across 48 evolutionarily distant variants, identifying five enzymes producing pharmaceutically relevant alcohols (de/ee 42.8% to >99%). Refined sampling of 60 phylogenetically close orthologs yields further improved ketoreductases

Rational Modification of Carbonyl Reductase from Candida parapsilosis ATCC 7330 to Improve the Yield of 2-Phenethyl Ethanol and (4-Chlorophenyl) (Pyridin-2-yl) Methanol.

Carbonyl reductases (CRs) are a class of oxidoreductases that convert carbonyl groups into hydroxyl groups. The application of CRs in the refined chemical industry is hindered by an unclear catalytic mechanism and low catalytic activity. In this study, CpCR from Candida parapsilosis ATCC 7330 was engineered via error-prone PCR and semirational design. The T171F mutant exhibited 2-fold enhanced catalytic efficiency toward PAAH, with an extended t1/2 by 7.0 h and elevated T50 by 1 °C. Consequently, 2-phenethyl alcohol (2-PE) production achieved a yield of 91.31%, representing a 2.36-fold improve

Mouse liver dihydrodiol dehydrogenases. Identity of the predominant and a minor form with 17 beta-hydroxysteroid dehydrogenase and aldehyde reductase.

A major and a minor form of dihydrodiol dehydrogenase were co-purified with 17 beta-hydroxysteroid dehydrogenase and aldehyde reductase, respectively, to apparent homogeneity from liver cytosol of male ddY mice. The activities of dihydrodiol dehydrogenase and testosterone dehydrogenase or aldehyde reductase of the two enzyme forms comigrated electrophoretically. The major form of the enzyme oxidized 17 beta-hydroxysteroids and nonsteroidal alicyclic alcohols and reduced 17-ketosteroids and various synthetic carbonyl compounds, showing higher affinity for steroids than for xenobiotics. The acti

From discovery to application: Enabling technology-based optimizing carbonyl reductases biocatalysis for active pharmaceutical ingredient synthesis.

The catalytic conversion of chiral alcohols and corresponding carbonyl compounds by carbonyl reductases (alcohol dehydrogenases), which are NAD(P) or NAD(P)H-dependent oxidoreductases, has attracted considerable attention. However, existing carbonyl reductases are insufficient to meet the demands of diverse industrial applications; hence, new enzymes with functions that can expand the toolbox of biocatalysts are urgently required. Developing precisely controlled chiral biocatalysts is of great significance for the efficient development of a broad spectrum of active pharmaceutical ingredients v

Characterisation of recombinant human fatty aldehyde dehydrogenase: implications for Sjögren-Larsson syndrome.

Fatty aldehyde dehydrogenase (FALDH) is an NAD+-dependent oxidoreductase involved in the metabolism of fatty alcohols. Enzyme activity has been implicated in the pathology of diabetes and cancer. Mutations in the human gene inactivate the enzyme and cause accumulation of fatty alcohols in Sjögren-Larsson syndrome, a neurological disorder resulting in physical and mental handicaps. Microsomal FALDH was expressed in E. coli and purified. Using an in vitro activity assay an optimum pH of approximately 9.5 and temperature of approximately 35 degrees C were determined. Medium- and long-chain fatty

Adh expression in species of the mulleri subgroup of Drosophila.

The locus encoding the NAD-dependent alcohol dehydrogenase (alcohol:NAD oxidoreductase; EC 1.1.1.1) is duplicated in some species of the cactophilic mulleri subgroup of Drosophila. Here we report a survey on the expression throughout development and in different tissues for this system, its regulation using interspecific hybrids, and the effects induced by 2-propanol in 10 species belonging to four different clusters of the above-mentioned subgroup. Our results indicate different expression throughout development and in different tissues, cis regulation in hybrid ovaries, trans (-) regulation

Characterization of the in vitro conversion of dolichol to dolichoate in bovine thyroid.

The enzymic conversion of dolichol into dolichoic acid has been studied in bovine thyroid subcellular fractions using [1-3H]dolichol as a substrate. The presence of conversion activity could be demonstrated in both the mitochondrial- and supernatant fractions. Investigation of cofactor requirements revealed that NAD+ was essential for reaching optimal activity. From kinetic studies Km-values of 3.5-4 microM and 0.29 mM could be calculated for, respectively, dolichol and NAD+ using the mitochondrial fraction as an enzyme source. No inhibitory effects from ethanol or pyrazole were detected sugge
