Silylacylation of Alkenes through N-Heterocyclic Carbene Catalysis.

The construction of silicon-containing molecules has received increasing attention in recent years. Herein, we report the generation of silyl radicals through NHC catalysis under mild reaction conditions. This methodology offers a novel and convenient route to a diverse range of β-silyl ketones with a broad substrate scope and good functional group compatibility. Both the radical clock and electrochemical studies are consistent with the hypothesis of ground-state SET, and a plausible mechanism for the organocatalytic transformation is proposed.

Radical 1-Fluorosulfonyl-2-alkynylation of Unactivated Alkenes.

Sulfonyl fluorides have found widespread use in chemical biology and drug discovery. The development of synthetic methods for the introduction of the sulfonyl fluoride moiety is therefore of importance. Herein, a transition-metal-free radical 1,2-difunctionalization of unactivated alkenes via FSO2 -radical addition with subsequent vicinal alkynylation to access β-alkynyl-fluorosulfonylalkanes is presented. Alkynyl sulfonyl fluorides are introduced as highly valuable bifunctional radical trapping reagents that also serve as FSO2 -radical precursors. The β-alkynyl-fluorosulfonylalkanes obtained

Oxime ester-enabled anti-Markovnikov hydrosilylation of alkenes.

An oxime ester-enabled anti-Markovnikov hydrosilylation of alkenes is developed. This protocol is operationally simple and atom economical. The reaction shows broad functional group tolerance, such as ketones, amides, esters, epoxides, alcohols and carboxylic acids. Mono-substituted and geminally substituted alkenes could be chemoselectively hydrosilylated.

Dual electrocatalysis enables enantioselective hydrocyanation of conjugated alkenes.

Chiral nitriles and their derivatives are prevalent in pharmaceuticals and bioactive compounds. Enantioselective alkene hydrocyanation represents a convenient and efficient approach for synthesizing these molecules. However, a generally applicable method featuring a broad substrate scope and high functional group tolerance remains elusive. Here, we address this long-standing synthetic problem using dual electrocatalysis. Using this strategy, we leverage electrochemistry to seamlessly combine two canonical radical reactions-cobalt-mediated hydrogen-atom transfer and copper-promoted radical cyan

Chromium-Mediated Stannylcyclopropanation of Alkenes with (Diiodomethyl)stannanes.

A stannyl-group-substituted gem-dichromiomethane species, generated in situ from CrCl2, TMEDA, and tributyl(diiodomethyl)stannane, worked as an efficient stannylcarbene equivalent to promote cyclopropanation of alkenes. The reaction provided synthetically useful stannylcyclopropanes directly from commercially available unactivated alkenes without using potentially flammable alkylzinc and diazo compounds. Structural characterization of stannyl- and germyl-group-substituted gem-dichromiomethane complexes and the effect of group 14 elements containing substituents for cyclopropanation are also de

Highly efficient synthesis of terminal alkenes from ketones.

[reaction: see text] The rhodium(I)-catalyzed methylenation of ketones using trimethylsilyldiazomethane proceeds to give the corresponding alkenes in good yields (60-97%). The use of an excess of 2-propanol and 1,4-dioxane as a solvent were instrumental to obtain the desired alkenes in high yields. Superior results were achieved with the rhodium(I)-catalyzed methylenation in comparison with the standard Wittig reaction.

Electrochemical Aerobic Oxygenation and Nitrogenation of Cyclic Alkenes via C═C Bond Cleavage or Oxygenation and Azidation of Open-Chain Alkenes.

An efficient strategy involving electrochemical C═C double-bond cleavage and functionalization of cyclic alkenes for the synthesis of ketonitriles is described. This transformation features environmentally friendly conditions and utilizes relatively safe TMSN3 as the nitrogenation reagent and molecular oxygen as the oxidant. For the open-chain alkenes, the reaction gave 1,2-difunctionalized products. A wide range of cyclic alkenes and open-chain alkenes were found to be compatible, providing the corresponding ketonitriles and α-azido aromatic ketones in moderate to good yields.

Direct access to 2-imidazolines from unactivated alkenes.

The formation of carbon-nitrogen bonds is crucial for the discovery of pharmaceuticals, materials, and agrochemicals. Nitrogen-containing functional groups, especially heterocycles, are common motifs in biologically active compounds. Among these, 2-imidazolines have emerged as important scaffolds with their own class of biological targets. Herein, we report the development of an oxidative amination process to directly convert unactivated alkenes into 2-imidazolines. The combination of ammonia and a commercially available hypervalent iodine reagent achieves the cleavage of the carbon-carbon dou

Bromonium-catalyzed 1,2-diamination of various conjugated alkenes.

Alkene functionalization via cyclic halonium intermediates is highly effective for stereocontrol, and a catalytic approach further expands this strategy, offering access to more advanced functionalization. Although various studies have utilized halonium species, expanding their scope to electron-deficient or π-conjugated alkenes has remained challenging. In this report, using a bromonium catalytic system, we achieved the stereoselective intermolecular 1,2-diamination of various α,β-unsaturated carbonyl compounds and π-extended alkenes. The direct use of hazardous Br2 was avoided by generating

PdYb/CeO2 Catalyst for Efficient Hydroaminocarbonylation of Alkenes.

The hydroaminocarbonylation of alkenes is one of the most promising methods for economically utilizing carbon monoxide (CO) to form high-value organic molecules. Here, we reported that a dual-atom catalyst named PdYb/CeO2 was prepared by impregnation method for the one step synthesis of amides from CO, alkenes with amines. Compared with Pd/CeO2, PdYb/CeO2 nearly doubled the amide yield. And the latter exhibited improved chemical selectivity and functional group tolerance toward diverse olefin and amine substrates. Characterization indicated that Pd and Yb species were embedded into the CeO2 su

Iron-Catalyzed Dealkenylative Borylation of Alkenes via Ozonolysis.

Given the exceptional versatility of boronic esters as synthetic linchpins, strategies that directly transform naturally abundant motifs such as alkenes into boron-containing intermediates offer substantial value. Herein, we report the development of a dealkenylative borylation transformation that converts C(sp3)-C(sp2) bonds into C(sp3)-B bonds via ozonolysis and iron catalysis. This protocol proceeds efficiently while tolerating a broad range of functional groups. Mechanistic studies indicate that the borylation reagent bis(catecholato)diborane (B2cat2) serves a dual purpose as both the boro

Chiral oxazolidinones via biocatalytic aziridination of unactivated alkenes.

Chiral oxazolidinones are privileged heterocycles broadly used in asymmetric synthesis (1) and drug discovery (2-3). Conventional synthetic routes rely on a "chiral pool" strategy, where enantiopure amino alcohols are required as key intermediates. Of particular importance are a class of 5-(S)-aminomethyl oxazolidinones that are key scaffolds in next-generation antibiotics targeting multidrug- and extensively drug-resistant Mycobacterium tuberculosis (4-6). While many strategies exist to construct chirality at the 4-position (α-to-nitrogen), methods to install the desired 5-stereocentre (α-to-

Enantioselective Cyanofunctionalization of Aromatic Alkenes via Radical Anions.

Alkene radical ions constitute an integral and unique class of reactive intermediates for the synthesis of valuable compounds because they have both unpaired spins and charge. However, relatively few synthetic applications of alkene radical anions have emerged due to a dearth of generally applicable and mild radical anion generation approaches. Precise control over the chemo- and stereoselectivity in alkene radical anion-mediated processes represents another long-standing challenge due to their high reactivity. To overcome these issues, here, we develop a new redox-neutral strategy that seamle

Nickel-Catalyzed Three-Component 1,2-Carboacylation of Alkenes.

Ketones, prevalent in many biologically significant molecules, require the development of novel methods to synthesize these structures, which is a critical endeavor in organic synthesis. Transition metal catalysis has proven to be an effective method for synthesizing ketones. However, the scope of these substrates remains relatively limited, particularly due to their incompatibility with sensitive functional groups. Herein, we report a Ni-catalyzed three-component 1,2-carboacylation of alkenes, which activates secondary/tertiary alkyl bromides. This method offers significant advantages: simpli

Oxidative rearrangement of 1,1-disubstituted alkenes to ketones.

The Wacker process, which is widely used to convert monosubstituted alkenes into the corresponding methyl ketones, is thought to proceed through a PdII/Pd0 catalytic cycle involving a β-hydride elimination step. This mechanistic scenario is inapplicable to the synthesis of ketones from the 1,1-disubstituted alkenes. Current approaches based on semi-pinacol rearrangement of PdII intermediates are limited to the ring expansion of highly strained methylene cyclobutane derivatives. Herein, we report a solution to this synthetic challenge by designing a PdII/PdIV catalytic cycle incorporating a 1,2

Hydrotrifluoroacetylation of Alkenes via Designer Masked Acyl Reagents.

Because of its impressive ability to promote pharmaceutical activity, the introduction of trifluoromethylacyl (CF3CO) functionality into organic compounds has become an important and growing research area. Although various protocols have been developed to access trifluoroketones, the use of trifluoroacetyl radicals remains virtually undeveloped. Herein, we disclose a novel method for trifluoroacetylation through an umpolung reagent, thereby transforming an electrophilic radical into a nucleophilic radical. The applicability of this transformation is highlighted by large-scale, late-stage react

Light-Induced Iron-Catalyzed Trifluoromethylative Thiolation of Alkenes.

A series of novel trifluoromethylative thiolations of alkene are realized by using visible light as a driving force and iron salts as a catalyst, and 1,2-bis(trifluoromethylated) compounds could be obtained in moderate to good yields. These multicomponent protocols proceed in an atom-economical way with a broad substrate scope. Biologically active chemicals can also be tolerated to provide desired products, suggesting that the catalytic protocol could be viable for late-stage modification in pharmaceutical discovery. At last, flow-setup synthesis of the desired product is successfully applied

Trifluoromethylation of alkenes by visible light photoredox catalysis.

A method for trifluoromethylation of alkenes has been developed employing visible light photoredox catalysis with CF(3)I, Ru(Phen)(3)Cl(2), and DBU. This process works especially well for terminal alkenes to give alkenyl-CF(3) products with only E-stereochemistry. The mild reaction conditions enable the trifluoromethylation of a range of alkenes that bear various functional groups.

Type-2 alkenes mediate synaptotoxicity in neurodegenerative diseases.

Synaptic dysfunction appears to be an early pathogenic event in Alzheimer's disease, amyotrophic lateral sclerosis and Parkinson's disease. Although the molecular mechanism of this synaptotoxicity is not known, evidence suggests that these diseases are characterized by a common pathophysiological cascade involving oxidative stress, lipid peroxidation and the subsequent liberation of alpha,beta-unsaturated carbonyl derivatives such as acrolein and 4-hydroxy-2-nonenal (HNE). A diverse body of in vivo and in vitro data have shown that these soft electrophilic chemicals can cause nerve terminal da
