Replacing alkyl organometallic reagents with alkyl bromides to achieve desulfurative alkylation of thioethers has been a long-standing challenge in desulfurative functionalization of organosulfur compounds. Herein, we report a copper-catalyzed reductive tertiary alkylation of β-acyl allylic sulfides with α-carbonyl alkyl bromides under mild reaction conditions. The reaction accommodates a broad substrate scope with good functional group compatibility, providing a direct route to install diversified (sp3)-carbon quaternary centers at the allylic position of α-branched enones. Further synthetic

Palladium-catalyzed C-H activation reactions are typically viewed as positionally static, with the site of reactivity dictated by the location of the initial metalation event. Herein, we report a strategy that overcomes this constraint by enabling migration of a σ-alkyl palladium intermediate prior to carbon-carbon bond formation via a previously inaccessible through-space alkyl-to-alkyl palladium shift. This two-component annulation of 2-bromoanilines or 2-bromophenols with allylic bromides proceeds through the formation of an alkyl-Pd species that undergoes translocation across the carbon fr

Reformatsky reagents are commonly employed with activated electrophiles, such as aldehydes, ketones, or activated alkyl halides. However, their limited nucleophilicity remains a considerable challenge for direct reactions with unactivated alkyl halides, typically necessitating transition metal catalysis. Here, we present a transition-metal-catalyst-free approach that facilitates direct nucleophilic substitution between Reformatsky reagents and diverse unactivated alkyl halides, which enables formal reductive cross-electrophile coupling via a one-pot process. Mechanistic studies reveal the pivo

Both olefins and arenes are abundant and versatile feedstock chemicals. Although the electrophilic alkylation of arenes constitutes an advanced technology, the corresponding alkylation of olefins is rather undeveloped. Here, we report an asymmetric catalytic benzylation and allylation of unactivated olefins with acetates as electrophiles. The reaction is catalyzed by a single imidodiphosphorimidate organocatalyst operating as a silylium Lewis acid. Mechanistic experiments support an enantioconvergent SN1 mechanism featuring stereoablative ion pair formation, resulting in a dynamic kinetic reso

A highly practical and efficient Cp*Co(III)-catalyzed C-H alkylation/alkenylation reaction of anilides with maleimides and acrylates was developed, during which a weakly coordinating amide carbonyl group functioned as the directing group. This approach features high efficiency, good functional group tolerance, and broad substrate scope, and a variety of 3-substituted succinimides and ortho-alkenylated anilides were synthesized in moderate to excellent yields. Furthermore, the reaction is highly selective, affording mono-ortho-alkylated/alkenylated products only. In addition, synthetic transfor

An electrochemical method for the decarboxylative alkylation of β-ketoacids with phenol derivatives has been developed. The protocol was carried out in readily available unseparated cells at room temperature in the absence of catalysts and oxidants. The corresponding aryl ketones were obtained in satisfactory yields without additional electrolytes, and were easy to produce in gram-scale synthesis. Based on control experiments and cyclic voltammetry, a plausible reaction mechanism was proposed.

Motivated by the scarcity of enantioselective direct intermolecular α-alkylation reactions of ketones with simple alkyl halides, we report a photo-organocatalytic process to access diethyl 2-(2-oxocyclohexyl)malonate and derivatives in good yield and enantioselectivity. The reaction design is based on highly abundant and nature-derived 9-amino-9-deoxy-epi-cinchona alkaloids to activate ketones as transient secondary enamines, which exist unfavorably in equilibrium with imines. These condensed species can serve as powerful photoinitiators via direct photoexcitation. This concept provides access

A single-atom catalyst with generally regarded inert Zn-N4 motifs derived from ZIF-8 is unexpectedly efficient for the activation of alcohols, enabling alcohol-mediated alkylation and transfer hydrogenation. C-alkylation of nitriles, ketones, alcohols, N-heterocycles, amides, keto acids, and esters, and N-alkylation of amines and amides all go smoothly with the developed method. Taking the α-alkylation of nitriles with alcohols as an example, the α-alkylation starts from the (1) nitrogen-doped carbon support catalyzed dehydrogenation of alcohols into aldehydes, which further condensed with nit

Sulfilimines are valuable compounds in both organic synthesis and pharmaceuticals. In this study, we present a copper-catalyzed sulfur alkylation of sulfenamides with N-sulfonylhydrazones. In contrast to prior findings, hydrazones derived from aldehydes act as donor-type carbene precursors, effectively engaging in coupling with sulfenamides via a copper catalyst, demonstrating exclusive S selectivity. The utility of the protocol was highlighted in the rapid access to a wide range of sulfoximine derivatives.

The efficient conversion of a C-H bond in the polyether chain to other functional groups provides great opportunities for development of novel applications in many research fields. However, this field is quite underdeveloped due to the key challenge on controlling the selectivity of the C-H bond functionalization over the chain cleavage. In this work, we report a controllable C-H bond alkylation of polyethers under mild conditions via photoinduced iron catalysis. The level of functionalization could be controlled by using different amounts of alkenes and various reaction times, while the molec

We report a visible-light-induced iron-catalyzed α-alkylation of ketones. The photocatalytic system is based on the single diaminocyclopentadienone iron tricarbonyl complex. Two catalytic intermediates of this complex are able to harvest light, allowing the synthesis of substituted aromatic and aliphatic ketones at room temperature using the borrowing hydrogen strategy in the presence of various substituted primary alcohols as alkylating reagents. Preliminary mechanistic studies unveil the role of light for both the dehydrogenation and reduction step.

Reductive alkylation of the A82846 family of glycopeptide antibiotics has the potential of producing seven products. N-Alkylation of the disaccharide amino function can be accomplished selectively, and offers the greatest increase in antibacterial activity. Products resulting from N-alkylation of LY264826 (A82846B) provide the most potent derivatives as compared to other members of this class of antibiotics. Two of these derivatives, LY307599 and LY333328 are approximately 500 times more active than vancomycin against vancomycin-resistant enterococci.

Sulfur(VI)-containing functional groups, such as sulfonamides, sulfinates and sulfonyl fluorides, are ubiquitous in pharmaceuticals, where they confer unique physicochemical properties that enhance metabolic stability, solubility and bioactivity across diverse therapeutic areas. Despite their prevalence, synthetic access to alkyl-sulfur(VI) derivatives remains fragmented, often requiring multi-step sequences from scarce precursors or harsh conditions that limit functional-group tolerance and scalability. Here we report a unified platform that enables divergent synthesis of alkyl-sulfur(VI) com

The incorporation of sp 3 -hybridized carbon centers into planar sp 2 carbon nanostructures is a powerful strategy in solution chemistry for tuning their electronic and structural properties. While on-surface synthesis has demonstrated efficient coupling strategies for sp 2 systems, C s p 3 ─ C s p 2 bond formation on surfaces remains largely unexplored. Here, we report the on-surface analogue of Friedel-Crafts alkylation on a metal surface: an intramolecular C s p 3 ─ C s p 2 coupling reaction based on a specifically designed fluorinated aromatic reactant, with the metal surfac

Ring-cleavage reactions of cyclopropyl alcohols are a valuable tool for the synthesis of linear and α- or β-branched ketones. Here, we describe a visible light-induced ring-opening alkylation of these substrates with benzyl, allyl, and secondary alkyl Katritzky salts. The reaction proceeds under dual photoredox and nickel catalysis in the presence of a titanium(IV) isopropoxide additive. Mechanistically, the ring-opening step starts with generation of the cyclopropoxy radical and is followed by its isomerization to the β-carbonyl radical. This enables the regioselective preparation of β-branch

In modern toxicology, substantial efforts are undertaken to develop alternative solutions for in vivo toxicity testing. The adverse outcome pathway (AOP) concept could facilitate knowledge-based safety assessment of chemicals that does not rely exclusively on in vivo toxicity testing. The construction of an AOP is based on understanding toxicological processes at different levels of biological organisation. Here, we present the developed AOP for liver fibrosis and demonstrate a linkage between hepatic injury caused by chemical protein alkylation and the formation of liver fibrosis, supported b

The inactivation of tetrameric isocitrate lyase from Escherichia coli by 3-bromopyruvate, exhibiting saturation kinetics, is accompanied by the loss of one sulfhydryl per subunit. The substrates glyoxylate and isocitrate protect against inactivation whereas the substrate succinate does not. The modification by 3-bromopyruvate (equimolar to subunits) imparts striking resistance to digestion of isocitrate lyase by trypsin, chymotrypsin, and V8 protease as well as a major decrease in the intensity of tryptophan fluorescence. After alkylation, the sequence Gly-His-Met-Gly-Gly-Lys is found followin

HYPOTHESIS: Incorporation of a glucofuranose linker within alkyl amidopropyl betaine surfactants modulates interfacial, bulk self-assembly and rheological behaviour.
EXPERIMENT: A series of alkyl-β-d-glucuronamidopropyl betaine surfactants were synthesised to investigate how incorporation of a glucuronolactone-derived linker influences micellar curvature, rheology, and interfacial properties.
FINDINGS: Small-angle neutron scattering revealed pronounced reductions in micellar curvature relative to the structurally similar family of alkylamidopropyl betaines, where charge screening was also ob

A radical-mediated oxidative cross-coupling of readily accessible α-alkylated styrenes with 1,3-dicarbonyl compounds utilizing a combination of Cu(OAc)2 and air as a catalytic system is described. Rather than requiring α-halocarbonyl compounds, this efficient approach enables direct installation of tertiary functionalized alkyl motifs to olefins with simple carbonyl derivatives. The novel protocol is characterized with high allylic selectivities via a competing β-H elimination. Both radical-clock and -trapping experiments provided clear-cut evidence for the intermediacy of an α-keto carbon-cen
