Z-Selective Alkyne Semireduction by an Organic Photoreductant.

The semireduction of alkynes is an indispensable transformation for the synthesis of Z-alkenes. Although extensive studies with mostly transition metal-based catalysts have enabled it to produce a broad scope of alkenes, a SciFinderTM search and a robustness screen of Lindlar's catalyst indicate that highly Lewis-basic nitrogen- and sulfur-containing motifs, which are prevalent in drugs, remain problematic. Herein, we report an alkyne semi-reduction using an organic photoreductant that tolerates all 10 of the most common Lewis-basic motifs in drugs. The reaction reduces terminal alkyl and aryl

Iodine-Catalyzed Carbonyl-Alkyne Metathesis Reactions.

The reaction between aldehydes or ketones and alkynes-the carbonyl-alkyne metathesis-constitutes a very useful strategy for the synthesis of α,β-unsaturated carbonyls. We now demonstrate that iodine is a highly efficient catalyst for both the intra- and intermolecular metathesis reaction in very small concentrations (0.1-1 mol %). Our protocol outperforms other catalytic systems, is operationally very simple, cheap, metal-free, and tolerates a large variety of functional groups (e. g., -CN, -CO2Me, -Br, -OH) at very low catalyst loadings. We can furthermore show that iodine-catalyzed carbonyl-

Recent advances in the application of alkynes in multicomponent reactions.

Alkynes have two active positions to carry out chemical reactions: C[triple bond, length as m-dash]C and C-H. These two positions are involved and activated in different reactions using different reagents. In this study, we investigated the reactions of alkynes that are involved in multi-component reactions through the C-C and C-H positions and examined the progress and gaps of each reaction by carefully studying the mechanism of the reactions. Firstly, we investigated and analyzed the reactions involving the C[triple bond, length as m-dash]C position of alkynes, including the reactions betwee

Electrochemical Controllable Divergent Carboxylation and Hydrocarboxylation of Alkynes with CO2.

Carboxylation and hydrocarboxylation of alkynes with CO2 for the synthesis of value-added and versatile carboxylic acids are of great importance. However, harsh reaction conditions, poor selectivity, practicability, and sustainability have always restricted its application. Electrochemical synthesis has been considered as a practical and environmentally friendly organic synthesis strategy via a tunable redox process. Hence, herein, an efficient electrochemical controllable divergent carboxylation and hydrocarboxylation of alkynes with CO2 was achieved to overcome the limitation of traditional

Regio- and Stereoselective Hydroelementation of SF5 -Alkynes and Further Functionalizations.

Herein is described a fully regio- and stereoselective hydroelementation reaction of SF5 -alkynes with N, O and S-nucleophiles and further functionalization of the corresponding Z-(hetero)vinyl-SF5 intermediates, a suitable platform to access α-SF5 ketones and esters, β-SF5 amines and alcohols under mild reaction conditions. Experimental and computational comparative studies between SF5 - and CF3 -alkynes have been performed to highlight and explain the difference of reactivity and selectivity observed between these two fluorinated motifs.

Copper-catalyzed radical-mediated intermolecular cyanomonofluoroalkylation of alkenes and alkynes.

A novel copper-catalyzed three-component reaction of monofluoroalkylation agents, alkenes and TMSCN is described. In addition, alkynes could also be compatible with the reaction system to obtain three-component products for the first time with moderate yield and excellent E/Z stereoselectivity. This reaction provides a facile method for the synthesis of cyanomonofluoroalkyl compounds, which may serve as potentially useful organic intermediates for further transformations. Preliminary mechanistic investigation indicated that monofluoroalkyl free radicals were involved in the cyanomonofluoroalky

Photoinduced Copper-Catalyzed Monofluoroalkylation of Terminal Alkynes to Propargylic Fluorides.

A photoinduced copper-catalyzed strategy for the monofluoroalkylation of alkynes with readily available monofluoroalkyl triflates was developed. It provides a new protocol to access valuable propargyl fluoride compounds via C-C bond formation by avoiding the use of highly toxic fluorination reagents. This reaction proceeded under mild conditions to afford propargyl monofluorides in moderate to high yields. Preliminary mechanistic studies reveal that a ligand-matched alkynyl copper complex might be the key photoactive substance.

Iron- and Cobalt-Catalyzed Asymmetric Hydrofunctionalization of Alkenes and Alkynes.

Transition metal catalyzed asymmetric hydrofunctionalization of readily available unsaturated hydrocarbons presents one of the most straightforward and atom-economic protocols to access valuable optically active products. For decades, noble transition metal catalysts have laid the cornerstone in this field, on account of their superior reactivity and selectivity. In recent years, from an economical and sustainable standpoint, first-row, earth-abundant transition metals have received considerable attention, due to their high natural reserves, affordable costs, and low toxicity. Meanwhile, the e

Electron-Deficient Alkyne Lipids Enable Efficient Synthesis of Comparable Polymer Lipids via Copper-Free Azide-Alkyne Cycloaddition.

Polymer lipids (PLs) are essential components of liposomes and lipid nanoparticles (LNPs) for drug and gene delivery, providing colloidal stabilization and defining the biological interface. While poly(ethylene glycol) (PEG)-based PLs are the current standard, they are suspected to be responsible for rare adverse reactions, e. g. to LNP-based Covid-19 vaccines. Therefore, PLs based on alternative stealth polymers are being intensively investigated for their use in LNPs. However, alternative PLs often lack comparability due to different synthesis protocols and are often not easily accessible. H

Gold Single-Atom Catalysts Enable Selective Shuttle Hydrochlorination of Alkynes.

Shuttle catalysis enables the transfer of reactive functional groups without relying on highly corrosive reagents. However, it remains largely confined to homogeneous systems, where catalyst recovery and reuse are often challenging. Here, we report a recyclable gold single-atom catalyst (SAC) supported on activated carbon (AC) for the selective shuttle hydrochlorination of terminal alkynes to vinyl chlorides, which are key intermediates in pharmaceutical synthesis. Spectroscopic and catalytic studies indicate that the combination of Au(I) single atoms with the AC carrier is essential for the u

Hydroboration of Alkynes Catalyzed by Fe(II) PCP Pincer Complexes.

A stereo- and regioselective Fe-(II)-catalyzed hydroboration of both terminal and internal alkynes with pinacolborane (HBpin) is described. The hydroboration reaction is in all cases highly syn-selective. The reaction requires no additives and proceeds with a catalyst loading of 1-2 mol % at 45-55 °C. An active pre-catalyst is the bench-stable alkyl Fe-(II) complex [Fe-(PCPCH2-iPr)-(CH2CH2CH3)-(CO)2]. This complex reacts with an excess of HBpin to form the Fe-(II) pinacolborane complex [Fe-(PCPCH2-iPr)-(κ2-H2BPin)-(CO)], which is an equally active pre-catalyst but more difficult to handle, as

Alkynes Hydration in Three-Component Double-Acidic Deep Eutectic Solvents.

In this work, a sustainable method for the hydration of both terminal and internal alkynes is presented, affording the corresponding ketones with complete Markovnikov regioselectivity. The key advantage of this approach lies in the use of recently developed three-component, double-acidic deep eutectic solvents (DESs), designed by our research team, which act as triple-active media-simultaneously serving as solvents, reagents, and catalysts. The reaction setup is straightforward and can be carried out under conventional or dielectric heating, typically providing the desired products in good to

Ni-Catalyzed Cyclotrimerization of Unactivated Internal Alkynes: Ligand-Dependent Regioselectivity.

A nickel-catalyzed [2 + 2 + 2] cyclotrimerization of unactivated internal alkynes is presented, enabling regioselective access to fully substituted benzene derivatives through ligand control. The transformation proceeds under additive-free conditions and tolerates a wide range of aryl, heteroaryl, and aliphatic alkynes. Regioselectivity is governed by the ligand framework, in which trialkylphosphine ligands promote the exclusive formation of 1,2,4-substituted benzenes, whereas the N-heterocyclic carbene ligand IPr favors the formation of 1,3,5-isomers as the major product, although the selecti

Rhodium and Isothiourea Dual Catalysis: Enantiodivergent Transformation of Terminal Alkynes.

A dual rhodium/isothiourea catalytic system was developed for the enantiodivergent transformation of terminal alkynes. Under synergistic rhodium/isothiourea dual catalysis, terminal alkynes can be creatively utilized as precursors for C1-ammonium enolate species, which subsequently participate in [4 + 2] and [2 + 2] annulation reactions with α,β-unsaturated ketimines or ketones, respectively. A wide range of chiral lactams and lactones were obtained in excellent yields and stereoselectivities (up to >20:1 dr, 98% ee).

Stereospecific nickel-catalyzed [4+2] heteroannulation of alkynes with aminophosphanes.

Enantioenriched phosphorus compounds play crucial roles in many fields ranging from catalyst to materials science to drug development. Despite advances in the construction of phosphacycles, incorporation of a P-chirogenic center into heterocycles remains challenging. Here, we report an effective method for the preparation of phosphacycles through nickel-catalyzed [4+2] heteroannulation of internal alkynes with aminophosphanes derived from o-haloanilines. Notably, chiral 2-λ5-phosphaquinolines can be prepared from P-stereogenic substrates via NH/PH tautomeric equilibrium without loss of stereoc

Electrochemical Difunctionalization of Terminal Alkynes: Access to 1,4-Dicarbonyl Compounds.

1,4-Dicarbonyl compounds are versatile scaffolds for the heterocycle synthesis, including the Paal-Knorr reaction. Herein, a feasible electrosynthesis method to access 1,4-dicarbonyl compounds has been developed from simple alkynes and 1,3-dicarbonyl compounds. When the undivided cell is combined with the constant current mode, aryl alkynes containing numerous medicinal motifs with 1,3-dicarbonyl esters or ketones react smoothly. External oxidant and catalyst-free conditions conform to the requirements of green synthesis.

3 d Transition Metal-Catalyzed Hydrogenation of Nitriles and Alkynes.

Selective hydrogenation of nitriles and alkynes is crucial considering the vast applications of reduced products in industries and in the synthesis of bioactive compounds. Particularly, the late 3d transition metal catalysts (manganese, iron, cobalt, nickel and copper) have shown promising activity for the hydrogenation of nitriles to primary amines, secondary amines and imines. Similarly, semihydrogenation of alkynes to E- and Z-alkenes by 3d metals is adequately successful both via the transfer hydrogenation and by using molecular hydrogen. The emergence of 3d transition metals in the select

Amide-Directed Formation of Five-Coordinate Osmium Alkylidenes from Alkynes.

The amide-directed synthesis of five-coordinate osmium alkylidene derivatives from alkynes is reported. These types of complexes, which have been elusive until now because of the tendency of osmium to give hydride alkylidyne species, are prepared by reaction of the dihydride OsH2Cl2(PiPr3)2 (1) with terminal alkynes containing a distal amide group. Complex 1 reacts with N-phenylhex-5-ynamide and N-phenylhepta-6-ynamide to give OsCl2{=C(CH3)(CH2) n NH(CO)Ph}(PiPr3)2 (n = 3 (2), 4 (3)). The relative position of carbonyl and NH groups in the organic substrates has no influence on the reaction. Th
