Spatial Distribution and Morphology of CaMKII-Expressing Amacrine Cells in Marmoset, Macaque, and Human Retina.

Over 30 types of amacrine cells have been described in the primate retina, yet few are well characterized. Here, we investigated amacrine cells expressing the alpha subunit of calcium/calmodulin-dependent protein kinase II (CaMKII) in the retinas of human, macaque (Macaca fascicularis, Macaca nemestrina), and marmoset (Callithrix jacchus) monkeys using immunohistochemistry and intracellular injections, with a focus on displaced amacrine cells (dACs) in the ganglion cell layer. The spatial density of CaMKII-positive dACs decreases with the distance from the fovea, but in the peripheral temporal

Dysregulated energy and protein homeostasis and the loss of GABAergic amacrine cells in aging retina.

Aging is a major risk factor for the development or the worsening of retinal degenerative conditions. The intricate network of the neural retina determined that the retinal aging is a complicated process. The aim of this study is to delineate the transcriptomic changes of major retinal neurons during aging in C57BL/6 mice at single-cell level. We analyzed the transcriptional profiles of the photoreceptor, bipolar, amacrine, and Müller glial cells of 1.5-2 and 24-30 months old mice using single-cell RNA sequencing technique. We selectively confirmed the differences in gene expression using immu

Complexity and scaling properties of amacrine, ganglion, horizontal, and bipolar cells in the turtle retina.

In the present study we have evaluated the complexity and scaling properties of the morphology of retinal neurons using fractal dimension as a quantitative parameter. We examined a large number of cells from Pseudemys scripta and Mauremys caspica turtles that had been labeled using Golgi-impregnation techniques, intracellular injection of Lucifer Yellow followed by photooxidation, intracellular injection of rhodamine conjugated horseradish peroxidase, or intracellular injection of Lucifer Yellow or horseradish peroxidase alone. The fractal dimensions of two-dimensional projections of the cells

Starburst amacrine cells form gap junctions in the early postnatal stage of the mouse retina.

Although gap junctional coupling in the developing retina is important for the maturation of neuronal networks, its role in the development of individual neurons remains unclear. Therefore, we herein investigated whether gap junctional coupling by starburst amacrine cells (SACs), a key neuron for the formation of direction selectivity, occurs during the developmental stage in the mouse retina. Neurobiotin-injected SACs coupled with many neighboring cells before eye-opening. The majority of tracer-coupled cells were retinal ganglion cells, and tracer coupling was not detected between SACs. Amacrine cells in Necturus retina: evidence for independent gamma-aminobutyric acid- and glycine-releasing neurons.

About one-half of on-off ganglion cells have inhibitory postsynaptic potentials (IPSP's) which are blocked by strychnine, while the remainder have IPSP's which are blocked by picrotoxin or bicuculline. These antagonists do not abolish light activity of the presynaptic inhibitory neuron, the amacrine cell. The existence of separate gamma-aminobutyric acid- and glycine-releasing amacrine cells is implied by these results.

A role for the enkephalin-immunoreactive amacrine cells of the chicken retina in adaptation to light and dark.

The functional state of the amacrine cells which contain enkephalin-, neurotensin- and somatostatin-like immunoreactivity of the chicken retina was monitored by measuring the rate of change in the levels of [Leu]enkephalin-like immunoreactivity in the retina. Dark-adapted birds were exposed to lights of different intensities for 12 h. At light levels of < or = 0.03 microW/cm2, the ENSLI amacrine cells were highly active but, by 0.08 microW/cm2, they reached a state of maximum inactivation. Thus, the ENSLI amacrine cells act as flip-flop devices, inactivated by critical levels of light, which c

The Atoh1-Cre Knock-In Allele Ectopically Labels a Subpopulation of Amacrine Cells and Bipolar Cells in Mouse Retina.

The retina has diverse neuronal cell types derived from a common pool of retinal progenitors. Many molecular drivers, mostly transcription factors, have been identified to promote different cell fates. In Drosophila, atonal is required for specifying photoreceptors. In mice, there are two closely related atonal homologs, Atoh1 and Atoh7 While Atoh7 is known to promote the genesis of retinal ganglion cells, there is no study on the function of Atoh1 in retinal development. Here, we crossed Atoh1Cre/+ mice to mice carrying a Cre-dependent TdTomato reporter to track potential Atoh1-lineage neuron

Gbx2 and Robo receptors control amacrine cell dendrite stratification in the mammalian retina.

Within the neuronal classes of the retina, amacrine cells (ACs) exhibit the greatest neuronal diversity in morphology and function. We show that the selective expression of the transcription factor Gbx2 is required for dendritic stratification of an individual AC subtype in the mouse retina. We identify Robo1 and Robo2 as downstream effectors that when deleted, phenocopy the dendritic misprojections seen in Gbx2 mutants. Slit1 and Slit2, the ligands of Robo receptors, are localized to the upper layers of the inner plexiform layer where we observe the dendritic misprojections in both Gbx2 and R

The effect of experimental glaucoma and optic nerve transection on amacrine cells in the rat retina.

PURPOSE: To detect alterations in amacrine cells associated with retinal ganglion cell (RGC) depletion caused by experimental optic nerve transection and glaucoma. METHODS: Intraocular pressure (IOP) was elevated unilaterally in 18 rats by translimbal trabecular laser treatment, and eyes were studied at 1 (n = 6), 2 (n = 5), and 3 (n = 7) months. Complete optic nerve transection was performed unilaterally in nine rats with survival for 1 (n = 4) and 3 (n = 5) months. Serial cryosections (five per eye) were immunohistochemically labeled with rabbit anti-gamma-aminobutyric acid (GABA) and anti-

An immunocytochemical study on specific amacrine cell subpopulations in the rat retina after ischemia.

Transient retinal ischemia leads to the loss of neurons in the inner retina. In an accompanying paper [F. Dijk, S. Van Leeuwen, W. Kamphuis, Differential effects of ischemia/reperfusion on amacrine cell subtype-specific transcript levels in the rat retina, Brain Res., 1026 (2004) 194-204] we present the results of a study on the effects of experimentally induced retinal ischemia on transcript levels of genes expressed by distinct subpopulations of amacrine cells. In response to 60-min ischemia, three different patterns of changes in transcript levels were found, indicating a differential vulne

Comparison of the responses of AII amacrine cells in the dark- and light-adapted rabbit retina.

We studied the light-evoked responses of AII amacrine cells in the rabbit retina under dark- and light-adapted conditions. In contrast to the results of previous studies, we found that AII cells display robust responses to light over a 6-7 log unit intensity range, well beyond the operating range of rod photoreceptors. Under dark adaptation, AII cells showed an ON-center/OFF-surround receptive-field organization. The intensity-response profile of the center-mediated response component followed a dual-limbed sigmoidal function indicating a transition from rod to cone mediation as stimulus inten

Expression of GluA2-containing calcium-impermeable AMPA receptors on dopaminergic amacrine cells in the mouse retina.

PURPOSE: The neuromodulator dopamine plays an important role in light adaptation for the visual system. Light can stimulate dopamine release from dopaminergic amacrine cells (DACs) by activating three classes of photosensitive retinal cells: rods, cones, and melanopsin-expressing intrinsically photosensitive retinal ganglion cells (ipRGCs). However, the synaptic mechanisms by which these photoreceptors excite DACs remain poorly understood. Our previous work demonstrated that α-amino-3-hydroxyl-5-methyl-4-isoxazole-propionate (AMPA) receptors contribute to light regulation of DAC activity. AMPA

We investigated the cellular mechanisms responsible for lowered levels of retinal dopamine in an oxygen-induced retinopathy (OIR) mouse model of ROP. METHODS: Retinopathy was induced by exposing mice to 75% oxygen from postnatal day 7 (P7) to P12. Oxygen-induced retinopathy and age-matched control mice were euthanized at P12, P17, P25, or P42 to P50. Immunohistochemistry, electrophysiology, and biochemical approaches were used to determine the effect of OIR on the struc

Light-evoked synaptic activity of retinal ganglion and amacrine cells is regulated in developing mouse retina.

Recent studies have shown a continued maturation of visual responsiveness and synaptic activity of retina after eye opening, including the size of receptive fields of retinal ganglion cells (RGCs), light-evoked synaptic output of RGCs, bipolar cell spontaneous synaptic inputs to RGCs, and the synaptic connections between RGCs and ON and OFF bipolar cells. Light deprivation retarded some of these age-dependent changes. However, many other functional and morphological features of RGCs are not sensitive to visual experience. To determine whether light-evoked synaptic responses of RGCs undergo dev

Specialized neuropeptide Y- and glucagon-like immunoreactive amacrine cells in the peripheral retina of the turtle.

There are many regional differences in cell morphology and neurochemistry in the retina. This study examined a specialized population of neuropeptide Y- and glucagon-like immunoreactive amacrine cells in the peripheral retina of the turtle. Some of the dendritic processes from these peptidergic amacrine cells formed a dense circumferentially oriented nerve fiber plexus which ran parallel to the ora serrata. Collaterals from this plexus projected into and innervated the nonpigmented ciliary epithelium in the pars plana region of the ciliary body. Electron microscopy revealed that the neuropepti

α-synuclein overexpression in the retina leads to vision impairment and degeneration of dopaminergic amacrine cells.

The presence of α-synuclein aggregates in the retina of Parkinson's disease patients has been associated with vision impairment. In this study we sought to determine the effects of α-synuclein overexpression on the survival and function of dopaminergic amacrine cells (DACs) in the retina. Adult mice were intravitreally injected with an adeno-associated viral (AAV) vector to overexpress human wild-type α-synuclein in the inner retina. Before and after systemic injections of levodopa (L-DOPA), retinal responses and visual acuity-driven behavior were measured by electroretinography (ERG) and a wa

Junctional conductance of retinal AII amacrine cell electrical synapses is decreased by NMDA receptors.

Retinal AII amacrine cells are extensively coupled together by electrical synapses. Changes to the strength of these synapses affect how signals are routed through rod and cone retinal pathways during scotopic and photopic vision. Plasticity at these electrical synapses has not, to date, been characterized using electrophysiological approaches. We investigated the effects of NMDA receptor (NMDAR) activation on electrical coupling between AII cells using dual whole-cell patch-clamp electrophysiology in mouse retinal slices. NMDAR activation substantially decreased junctional conductance between

Functionally distinct GABAergic amacrine cell types regulate spatiotemporal encoding in the mouse retina.

GABA (γ-aminobutyric acid) is the primary inhibitory neurotransmitter in the mammalian central nervous system. GABAergic neuronal types play important roles in neural processing and the etiology of neurological disorders; however, there is no comprehensive understanding of their functional diversity. Here we perform two-photon imaging of GABA release in the inner plexiform layer of male and female mice retinae (8-16 weeks old) using the GABA sensor iGABASnFR2. By applying varied light stimuli to isolated retinae, we reveal over 40 different GABA-releasing neuron types. Individual types show la
