Vector competence of Amblyomma americanum (Acari: Ixodidae) for Rickettsia rickettsii.

Rickettsia rickettsii - the etiologic agent of Rocky Mountain spotted fever (RMSF) - is widely spread across the Americas. In the US, Dermacentor spp. ticks are identified as primary vectors of R. rickettsii and Rhipicephalus sanguineus s.l. has been implicated in transmission of this pathogen in several locations in the Southwest. Conversely, ticks of the genus Amblyomma are recognized vectors of RMSF in Central and South America, but not in the US. A. americanum is one of the most aggressive human-biting ticks in the US, whose geographical range overlaps with that of reported RMSF cases. Des

Case of tick-associated rash illness caused by Amblyomma testudinarium.

We report a case of tick-associated rash illness (TARI), a new clinical entity of erythema migrans associated with a tick bite without infection of Lyme borreliosis agent. The patient, a 53-year-old man, went hiking in a mountainous area of Minoh City, Osaka Prefecture in October 2012. An erythematous macule with itching and a biting tick was found on his left thigh 2 days later, which gradually expanded. On the first visit to our department at the fifth day after hiking, an erythematous macule of 10 cm was recognized around the bite site. He had no systemic symptoms, and laboratory data were

Amblyomma parvitarsum (Acari: Ixodidae): localities, hosts and host-parasite ecology.

Only a few aspects of the biology of Amblyomma parvitarsum Neumann are known. Adults of this hard tick species are parasites of South American camelids in the Andean plateau of Argentina, Bolivia, Peru and Chile and also in the Argentine Patagonia, but they have been also rarely found on other artyodactils and two species of birds. The larva has been collected from reptiles in northern Chile, whereas the hosts for the nymph remain unknown. On nine localities included within Altitude Tropical and Perarid Mediterranean ecoregions in northern Chile, we analyzed 237 reptiles, 285 birds, 624 rodent

Density and behavior of capybara (Hydrochoerus hydrochaeris) ticks (Acari: Ixodidae) Amblyomma sculptum and Amblyomma dubitatum with notes on Rickettsia bellii infection: Assessing human exposure risk.

In several urban and peri‑urban areas of Brazil, populations of Amblyomma sculptum and Amblyomma dubitatum ticks are maintained by capybaras (Hydrochoerus hydrochaeris). In some of these areas, this host and these tick species are associated with Brazilian spotted fever (BSF), a lethal human disease caused by the bacterium Rickettsia rickettsii. In this work, we evaluated the risk of human exposure to these tick species using four collection techniques to discern host-seeking behavior. The study was carried out in 10 urban sites inhabited by capybaras in Uberlândia, a BSF-free municipality in

Seasonality of the Rickettsia vectors Amblyomma ovale and Amblyomma sculptum on dogs in Atlantic Forest and Cerrado biomes of Minas Gerais, Brazil: Hypotheses for human rickettsiosis seasonality.

Free-ranging rural dogs in Brazil are parasitized by ticks from indigenous ixodofauna including Amblyomma sculptum and Amblyomma ovale, vectors of Rickettsia rickettsii and Rickettsia parkeri strain Atlantic Rainforest (ARF), respectively. Therefore, dogs are important sentinels and hosts for ticks and tick-borne diseases of public health relevance. This study evaluated abundance, and seasonal dynamics of these tick species infesting free-ranging rural dogs in two municipalities of Minas Gerais State, southeastern Brazil: Araguari (Cerrado biome remnants) and Guaxupé (Atlantic Forest remnants)

Detection of Amblyomma maculatum and Rickettsia parkeri in prairies of Central Missouri.

The destruction and decline of prairie habitats due to landscape repurposing have profoundly impacted the diversity of plant, animal, and insect life. In the Central United States, the reconstruction of prairie habitats from farmland is a widely applied strategy to raise diversity and recreate a healthy, complex ecosystem. In Central Missouri, we examine the consequences of reconstruction efforts on the prevalence of zoonotic diseases and their associated pathogens by performing tick-flagging at two prairie sites. In doing so, we have observed large populations of Dog ticks, Dermacentor variab

Distribution and genetic variation of Amblyomma triste (Acari: Ixodidae) in Argentina.

A study was conducted to investigate the distribution of Amblyomma triste in Argentina under the hypothesis that this tick prevails in riparian localities along the Paraná River and adjacent humid environments from 34° 30' S to 25° 20' S, approximately. Ticks were collected from mammals and vegetation in those environments from November 2008 to October 2012. Additionally, genetic variation was tested from Argentinean, Brazilian, Chilean, and Uruguayan populations of A. triste by comparing sequences of 16S rDNA mitochondrial gene. The hypothesis was not confirmed because A. triste were collecte

A case of tick bite from Amblyomma testudinarium on the glans penis.

A tick which bit the glans penis of a 40-year-old Japanese man was surgically excised together with the skin where the bite occurred. It was identified as Amblyomma testudinarium through the taxonomical investigation of its morphological characteristics. To our best knowledge, our patient is the sixth tick bite from this species of tick recorded in the Chugoku District of Japan.

Amblyomma americanum: a potential vector of Lyme disease in New Jersey.

Amblyomma americanum is a likely secondary vector of Lyme disease in New Jersey. Ticks of this species were removed from the site of the characteristic skin lesion known as erythema chronicum migrans on two patients with the disease, and the Lyme disease spirochete was isolated from nymphs and adults of this species. That A. americanum is a potential vector is supported by its similarities to Ixodes dammini, the known tick vector, in seasonal distribution and host utilization. The extensive range of A. americanum may have great implications for potential Lyme disease transmission outside known

Further insights into the sialome switch of Amblyomma americanum adult females.

BACKGROUND: The lone star tick, Amblyomma americanum, is a three-host species widely distributed across North America and a vector for multiple human pathogens. Bites from this tick is also associated with alpha-gal syndrome, underscoring its growing public health importance. Adult female ticks undergo a prolonged feeding cycle divided into preparatory, slow-feeding, and rapid-feeding phases. Tick salivary glands are essential for successful blood feeding, producing a complex mixture of molecules that modulate host hemostasis, immunity, and tissue repair. The dynamic remodeling of the salivary

Transmission of Cytauxzoon felis by injection of Amblyomma americanum salivary glands.

BACKGROUND: Cytauxzoonosis is a life-threatening disease of cats, caused by the tick-borne piroplasmid hemoparasite, Cytauxzoon felis. Current experimental models for cytauxzoonosis rely on either tick transmission or direct injection of infected cat tissues. These models require researchers to directly work with infected ticks or use cats with acute cytauxzoonosis. To improve the feasibility and accessibility, there is a need to establish sharable resources among researchers. In related piroplasmid parasites, sporozoite-based inoculums are routinely produced from tick salivary glands, cryopre

Report of Amblyomma sculptum in a House in a Rickettsia rickettsii Circulation Area.

Amblyomma sculptum is a tick that has medical and veterinary importance as, in Brazil, it is the main vector of Rickettsia rickettsii, a disease affecting humans. The presence of ticks was observed outside a residence in a peri-urban area of the Atlantic Forest region in Brazil, as well as on two dogs that lived there. Eighteen A. sculptum adults were seen walking on a cemented pillar at the porch of the house and sheltering inside the pillar's crevices; meanwhile on the dogs, only Rhipicephalus sanguineus sensu lato ticks were found. It is hypothesized that as the dogs circulated in the fores

Amblyomma sculptum: genetic diversity and rickettsias in the Brazilian Cerrado biome.

Amblyomma sculptum (Ixodida: Ixodidae) Berlese, 1888 is the most important tick vector in Brazil, transmitting the bioagent of the most severe form of spotted fever (SF) in part of the Cerrado (in the states of Minas Gerais and São Paulo). In another part of the Cerrado (Central-West region of Brazil), a milder form of SF has been recorded. However, neither the rickettsia nor the vector involved have been characterized. The aim of the current study was to analyse genetic variation and the presence of rickettsia in A. sculptum in Cerrado, from silent areas and with the milder form of SF. Sample

Discovery of filarial nematode DNA in Amblyomma americanum in Northern Virginia.

Ticks collected in 2011 were screened for the presence of filarial nematode genetic material, and positive samples were sequenced for analysis. Monanema-like filarial nematode DNA was recently discovered in Amblyomma americanum in northern Virginia, marking the first time genetic material from this parasite has been discovered in ticks in the state. Phylogenetic analysis revealed that this material was directly related to a previously discovered filarial nematode in A. americanum populations in Maryland as well as recently identified parasites in Ixodes scapularis from southern Connecticut. Tick paralysis caused by Amblyomma maculatum on the Mexican Pacific Coast.

Tick paralysis is a rare entity in which it is necessary to identify the cause and remove the arthropod to have a rapid remission of symptoms. In the absence of an early diagnosis, the outcome can be fatal, as toxins are released from the tick's saliva as it feeds. To the best of the authors' knowledge, this is the first clinical report of the disease in Mexico and Latin America. A 22-year-old man from a rural area, who was in contact with cattle, developed ascending flaccid paralysis secondary to Amblyomma maculatum tick toxin. He presented flaccid paraplegia and arreflexia that progressed un

First detection of Babesia spp. in Amblyomma mixtum from Veracruz, Mexico.

Tick-borne diseases pose a major threat to cattle production in Mexico, with bovine babesiosis caused by Babesia bigemina and Babesia bovis being among the most severe. While the invasive tick Rhipicephalus microplus is the primary vector, increasing infestations by the native Amblyomma mixtum raise concerns about its potential role in pathogen transmission. This study assessed the diversity and minimum infection rate (MIR) of piroplasmids in R. microplus and A. mixtum across 15 municipalities in Veracruz, Mexico. Ticks were collected from hosts and the environment between June 2023 and March

First molecular evidence of Wolbachia occurrence in Amblyomma sculptum (Acari: Ixodidae).

As the main vector for the bacterium Rickettsia rickettsii in Brazil, the tick Amblyomma sculptum is a parasite of great public health importance in this country. Wolbachia is an endosymbiont bacterium highly widespread among invertebrates and because of its impact on its hosts' biology, form a powerful alternative for pests and disease control. The aim of this study was to investigate the occurrence of this bacterium in A. sculptum. For this, 187 adult ticks collected in two municipalities in the interior of the state of São Paulo, Brazil, were analyzed using molecular techniques and bioinfor

A 24-48 h fed Amblyomma americanum tick saliva immuno-proteome.

BACKGROUND: Multiple tick saliva proteins, the majority of which are unknown, confer tick resistance in repeatedly infested animals. The objective of this study was to identify the 24-48 h fed Amblyomma americanum tick saliva immuno-proteome. The 24-48 h tick-feeding phase is critical to tick parasitism as it precedes important events in tick biology, blood meal feeding and disease agent transmission. Fed male, 24 and 96 h fed female phage display cDNA expression libraries were biopanned using rabbit antibodies to 24 and 48 h fed A. americanum female tick saliva proteins. Biopanned immuno-cDNA
