Acute myeloid leukemia in a genetically related captive population of axolotls (Ambystoma mexicanum).

Axolotls (Ambystoma mexicanum) are critically endangered paedomorphic salamanders with remarkable regenerative capacity. While nearly extinct in the wild, they are commonly maintained in captivity as companion animals, zoological display animals, and biomedical research colonies, where they serve as an animal model for genetics, developmental biology, and regenerative medicine. This report documents a case series of spontaneous myeloid leukemia in genetically related, co-housed adult axolotls in a zoological collection that resulted in 50% overall mortality over 38 months. Affected axolotls ex

Thyroid hormone receptors and iodothyronine deiodinases in the developing Mexican axolotl, Ambystoma mexicanum.

The Mexican axolotl, Ambystoma mexicanum, is a neotenous salamander that rarely undergoes anatomical metamorphosis, but can be induced to do so by administration of thyroxine (T4). The neoteny appears to be due primarily to low levels of plasma T4 secondary to a low rate of secretion of thyroid-stimulating hormone. However, other factors may also be involved. In anuran amphibia, metamorphosis is accompanied by alterations in thyroid hormone receptor concentration and marked changes in the activities of the iodothyronine deiodinase systems, all of which contribute to enhancing peripheral sensit

A comparison of the ultrastructure of the cornea of the pre- and post-metamorphic axolotl (Ambystoma mexicanum, Amphibia).

The corneal ultrastructure of the pre- and post-metamorphic stages of the neotenic axolotl Ambystoma mexicanum is examined using light microscopy and both scanning and transmission electron microscopy to reveal whether there are any morphological changes associated with a switch in lifestyle. Although the complement of corneal layers remains the same, there are significant quantitative changes in corneal, epithelial and stromal thickness, epithelial and endothelial cell size and density, and the thickness of Bowman's layer and Desçemet's membrane. Microholes in the epithelium and vertical sutu

Peripheral projections of nervus terminalis LHRH-containing neurons in the tiger salamander, Ambystoma tigrinum.

The peripheral projections of the nervus terminalis (NT) have been difficult to examine due to the weak immunoreactivity of the processes to various antibodies. We performed two experimental manipulations in the tiger salamander in an attempt to increase the luteinizing hormone-releasing hormone-immunoreactive (LHRH-ir) labelling in the peripheral processes of the NT:1) the NT was sectioned centrally, or 2) a 100 mg melatonin pellet was embedded subcutaneously for 3 days prior to sacrifice. Following these manipulations, animals were sacrificed and tissue was processed with standard immunocyto

Role of non-homologous end joining DNA repair during limb regeneration in Ambystoma mexicanum.

BACKGROUND: Ambystoma mexicanum is renowned for its extraordinary regenerative capacity. Regardless of whether progenitor cells arise via dedifferentiation or stem cell activation, these cells must replicate DNA to proliferate before they ultimately differentiate. This rapid cell division increases the likelihood of DNA damage, requiring efficient repair mechanisms. RESULTS: We focused on the non-homologous end joining pathway, responsible for repairing double-strand DNA breaks, and investigated the effects of 2 key components-KU70/80 and LIGIV- using chemical inhibitors STL127705 and SCR130,

Differential effects of 3,5-T2 and T3 on the gill regeneration and metamorphosis of the Ambystoma mexicanum (axolotl).

Thyroid hormones (THs) regulate tissue remodeling processes during early- and post-embryonic stages in vertebrates. The Mexican axolotl (Ambystoma mexicanum) is a neotenic species that has lost the ability to undergo metamorphosis; however, it can be artificially induced by exogenous administration of thyroxine (T4) and 3,3',5-triiodo-L-thyronine (T3). Another TH derivative with demonstrative biological effects in fish and mammals is 3,5-diiodo-L-thyronine (3,5-T2). Because the effects of this bioactive TH remains unexplored in other vertebrates, we hypothesized that it could be biologically a

Lethal and sublethal effects of atrazine, carbaryl, endosulfan, and octylphenol on the streamside salamander (Ambystoma barbouri).

Agricultural contaminants may be contributing to worldwide amphibian declines, but little is known about which agrichemicals pose the greatest threat to particular species. One reason for this is that tests of multiple contaminants under ecologically relevant conditions are rarely conducted concurrently. In this study, we examined the effects of 37-d exposure to the agrichemicals atrazine (4, 40, and 400 micrograms/L), carbaryl (0.5, 5, and 50 micrograms/L), endosulfan (0.1, 1, and 10 micrograms/L for 31 d and 0.1, 10, and 100 micrograms/L for the last 6 d), and octylphenol (5, 50, and 500 mic

Effects of the antiepileptic drug valproic acid on the development of the axolotl (Ambystoma mexicanum): histological investigations.

In the amphibian Ambystoma mexicanum, valproic acid (VPA) causes retarded development and malformations including neural tube defects. Some of the observed abnormalities resemble exencephaly. We present the light microscopic characteristics of VPA-induced effects on the developing central nervous system (CNS) as well as on other developing tissues of this species. To induce malformations, various concentrations of VPA were applied to embryos from blastula stage on, either as 24-h pulse or as continuous exposure. In treated embryos the abnormal development of the CNS was indicated by retarded n

Oral administration of GnRH via a cricket vehicle stimulates spermiation in tiger salamanders (Ambystoma tigrinum).

More than 50% of caudates are threatened with extinction and are in need of ex-situ breeding programs to support conservation efforts and species recovery. Unfortunately, many salamander populations under human care can experience reproductive failure, primarily due to missing environmental cues necessary for breeding. Assisted reproductive technologies (ARTs) are a useful suite of techniques for overcoming or bypassing these missing environmental cues to promote breeding. Exogenous hormones are used to stimulate natural breeding behaviors or gamete expression for in-vitro fertilization or bio

Comparative transcriptomics of limb regeneration: Identification of conserved expression changes among three species of Ambystoma.

Transcriptome studies are revealing the complex gene expression basis of limb regeneration in the primary salamander model - Ambystoma mexicanum (axolotl). To better understand this complexity, there is need to extend analyses to additional salamander species. Using microarray and RNA-Seq, we performed a comparative transcriptomic study using A. mexicanum and two other ambystomatid salamanders: A. andersoni, and A. maculatum. Salamanders were administered forelimb amputations and RNA was isolated and analyzed to identify 405 non-redundant genes that were commonly, differentially expressed 24 h

Design and Standardization of a Mandibular Injury Model for the Study of Craniofacial Tissue Regeneration in the Ambystoma mexicanum Model.

The craniofacial complex is an anatomical region that, in humans, can be affected by developmental malformations, trauma, and pathologies such as cancer, leading to the loss of affected tissues. This creates a need to seek strategies for regenerating these lost tissues. This has promoted the study of cellular and molecular mechanisms that orchestrate the regenerative response of injured craniofacial structures in regenerating species, with Ambystoma mexicanum (A. mexicanum) being a species of great interest. In accordance with the above, a reproducible surgical lesion model was designed and st

Exploring the ecological drivers of bacteriophage diversity and functional viral potential in the skin of the axolotl Ambystoma altamirani.

Bacteriophages play important roles in shaping microbial community dynamics across diverse environments. In the amphibian skin, most microbiome studies have focused on bacteria and their interactions with the fungus Batrachochytrium dendrobatidis (Bd), leaving other microbial components, including viruses, largely unexplored. Here, we present the first characterization of the viral community in the amphibian skin microbiome, focusing on ecological drivers of bacteriophage diversity and functional potential in the axolotl Ambystoma altamirani. Using public shotgun metagenomes, we found that the

Neurofilaments are prominent in bullfrog olfactory axons but are rarely seen in those of the tiger salamander, Ambystoma tigrinum.

Bullfrog olfactory axons show variable numbers (0-29) of structurally typical neurofilaments (NFs) about 10 nm in diameter. In studies tracking these NFs through serial sections of axons in cross section, they were found to be discontinuous, with a calculated average length of about 118 microns. In contrast to olfactory axons in bullfrogs, those of the tiger salamander Ambystoma trigrinum rarely show NFs. To be certain that the absence of NFs is a specific characteristic of olfactory axons, pieces of salamander spinal cord, optic nerve, and sciatic nerve were examined and found to contain typi

Adrenergic innervation of the gills, pulmonary arterial plexus, and dorsal aorta in the neotenic salamander, Ambystoma tigrinum.

The presence of adrenergic innervation was investigated in four different vascular segments of the neotenic tiger salamander, Ambystoma tigrinum, by histofluorescent staining for catecholamines. The segments were the respiratory section of the gill, the branchial shunt vessels, a vascular plexus in the pulmonary artery, and the dorsal aorta. No adrenergic fibers were detected in the respiratory section of the gill or the pulmonary arterial plexus. In contrast, the branchial shunt vessels contained both adrenergic varicosities and catecholamine-containing cell bodies. These cells resemble Type

Temporary and selective anosmia in tiger salamanders (Ambystoma tigrinum) caused by chemical treatment of the olfactory epithelium.

Tiger salamanders (Ambystoma tigrinum) were trained to respond to two dissimilar odorants (i.e., cyclohexanone, dimethyl disulfide) but not to a third odorant (n-butanol). When the training criterion was met, the animals were anesthetized and given nasal lavages of saline or odorants in saline. Lavage with cyclohexanone decreased responding to cyclohexanone but not dimethyl disulfide, and vice-versa, in later behavioral tests. Likewise, ethyl acetoacetate lavage produced selective response decrements to cyclohexanone. Lavage with n-butanol had no effect on responding to presentations of either
