Multiple unerupted teeth with amelogenesis imperfecta in siblings.

Amelogenesis imperfecta encompasses a group of inherited abnormalities that are generally considered to primarily affect the formation and/or calcification of enamel. This case report describes the unusual presentation of amelogenesis imperfecta in siblings as multiple unerupted teeth, multiple pulpal calcifications, and multiple dilacerations of roots along with the defect in the enamel. The intent of our report is to highlight a rare co-occurrence of amelogenesis imperfecta with multiple morphologic alterations in siblings.

Molecular analysis for genetic counselling in amelogenesis imperfecta.

OBJECTIVE: To use molecular genetics to establish the mode of inheritance in a family with amelogenesis imperfecta. MATERIALS AND METHODS: The polymerase chain reaction was used to amplify exons of the amelogenin gene on the short arm of the X chromosome. RESULTS: A single base deletion mutation in exon 6 of the amelogenin gene was identified. This mutation was a single base deletion of a cytosine residue - 431delC - in codon 96 of exon 6, introducing a stop codon 30 codons downstream of the mutation in codon 126 of the exon. CONCLUSION: The firm establishment of an X-linked mode of inherit

Dental development in amelogenesis imperfecta: a controlled study.

The present investigation studied dental development in 23 subjects with amelogenesis imperfecta (AI), aged 4.2 to 15.6 years, compared with 46 race-, age-, and sex-matched, healthy, normal controls. The dental ages of the children were assessed from panoramic radiographs using previously established methods. Their chronologic ages at the time of radiographic examination were compared with their dental ages. The results showed that all subjects with AI showed a significant acceleration of dental age of approximately 1.13 +/- 0.78 years compared with control children (P < 0.001). All children w

Ufl1-Mediated UFMylation Sustains Amelogenesis by Stabilizing RUNX2.

UFMylation is a conserved ubiquitin-like post-translational modification that controls protein stability and tissue homeostasis, while its role in amelogenesis remains largely uncharacterized. Here, we investigated the function of UFL1, the sole E3 ligase of the UFMylation pathway, in mammalian enamel development using K14-Cre-mediated epithelial-specific Ufl1 knockout mice. Ufl1 ablation caused severe amelogenesis imperfecta, with impaired enamel deposition, hypomineralization, and progressive tooth damage, accompanied by abnormal cervical loop development. Transcriptomic profiling revealed e

Retinoic Acid Excess Impairs Amelogenesis Inducing Enamel Defects.

Abnormalities of enamel matrix proteins deposition, mineralization, or degradation during tooth development are responsible for a spectrum of either genetic diseases termed Amelogenesis imperfecta or acquired enamel defects. To assess if environmental/nutritional factors can exacerbate enamel defects, we investigated the role of the active form of vitamin A, retinoic acid (RA). Robust expression of RA-degrading enzymes Cyp26b1 and Cyp26c1 in developing murine teeth suggested RA excess would reduce tooth hard tissue mineralization, adversely affecting enamel. We employed a protocol where RA was

Amelogenin-deficient mice display an amelogenesis imperfecta phenotype.

Dental enamel is the hardest tissue in the body and cannot be replaced or repaired, because the enamel secreting cells are lost at tooth eruption. X-linked amelogenesis imperfecta (MIM 301200), a phenotypically diverse hereditary disorder affecting enamel development, is caused by deletions or point mutations in the human X-chromosomal amelogenin gene. Although the precise functions of the amelogenin proteins in enamel formation are not well defined, these proteins constitute 90% of the enamel organic matrix. We have disrupted the amelogenin locus to generate amelogenin null mice, which displa

Hereditary amelogenesis imperfecta. I. Oral health in children.

An epidemiological study of hereditary enamel defects comprising more than 400 000 children, 3-19 years of age, was performed in 1982-1983 in the middle of Sweden. 105 children were diagnosed as having Hereditary Amelogenesis Imperfecta (HAI). Based on clinical and genetic data a classification of HAI could be carried out based on the hypoplastic and the hypomineralized type. 99 out of the 105 children were available for a thorough clinical and radiographical examination concerning oral health and these children were analysed in terms of carious lesions, restorations, dental plaque, calculus,

Distinctive Amelogenesis Imperfecta in Loeys-Dietz Syndrome Type II.

Loeys-Dietz syndrome (LDS1-6) is caused by mutations along the transforming growth factor-β (TGF-β) signaling pathway and features aortic aneurysms and craniofacial dysmorphology. Mutations that cause LDS can be found in the genes encoding transforming growth factor-β (TGF-β) ligands (TGFB2 and TGFB3), receptors (TGFBR1 and TGFBR2), and signal transducers (SMAD2 and SMAD3). Variable enamel anomalies are seen in patients with LDS, but the most severe enamel defects with advanced attrition have been observed specifically in some patients with mutation in the TGFBR2 gene (LDS2). We used human spe

Adaptive amelogenesis during unimpeded eruption of rat mandibular incisor.

The aim of the study was to see whether the length of the enamel secretion zone in unimpeded rat incisors, measured precisely, is in agreement with the observed decrease in enamel thickness. Unimpeded eruption of mandibular incisors of five experimental and two control rats was induced by cutting off the erupted part of the incisors three times per week for 5 weeks. The length of the zone of enamel secretion in unimpeded and impeded control incisors was measured on longitudinal and serial transverse histological sections of fixed, demineralised and embedded hemimandibles. Impeded contralateral

Amelogenesis Imperfecta with Coronal Resorption: Report of Three Cases.

Intracoronal resorption of the permanent dentition in cases of amelogenesis imperfecta (AI) is a rare finding which poses an added complication to the already complex management of this condition. This paper presents three cases of AI associated with delayed eruption of permanent teeth in which asymptomatic intracoronal resorption occurred. CPD/Clinical Relevance: This paper highlights the fact that teeth affected with amelogenesis imperfecta may undergo asymptomatic intracoronal resorption which is only identifiable radiographically.

Amelogenesis imperfecta: a scanning electron microscopic and histopathologic study.

Amelogenesis imperfecta (AI) is a hereditary defect in enamel formation affecting both primary and permanent dentition. Scanning electron microscopic investigation is one of the most effective methods in diagnosing and identifying the type of amelogenesis imperfecta. The aim of this study was to investigate the ultrastructure of different types of amelogenesis imperfecta enamel. The primary teeth of three children with AI aged 4, 10 and 11-years-old were studied by scanning electron microscopy and irregular enamel, irregularities in enamel crystallites, hypoplastic areas on the enamel surface

Odontogenesis and amelogenesis in interacting lizard-quail tissue combinations.

In this study we examined the possible inductive role of the dental papilla from polyphyodont lizard tooth germs. Flank skin sheets of quail ectoderm enzymatically separated from dermal tissue were recombined with lizard tooth papillae and placed on semisolid medium and cultured for 2 days. Subsequently, the recombinants were removed and placed on the chorioallantoic membrane of chick hosts and incubated for 6 days. After this period of 8 days in explant, control tissues differentiated according to their own phenotypes. Lizard dental papilla alone differentiated as fibroblasts. Quail flank ski

A novel biomimetic strategy for mimicking amelogenesis to repair enamel.

According to the principle of minimal invasiveness in modern dentistry, biomimetic remineralization therapy constitutes a significant strategy for the prevention and treatment of early enamel caries. Based on the three "key events" of amelogenesis in vivo, silk fibroin (SF) combined with carboxymethyl chitosan (CMC) successfully formed an SF/CMC composite, and amorphous calcium phosphate (ACP) was then used to form an SF/CMC-ACP nanocomposite with remineralization properties. In our study, SF was used as a template protein for biomimetic amelogenin, ACP was stabilized with CMC and the reminera
