Characterization of a novel Glutaminase-free L-asparaginase from Bifidobacterium thermophilum.

L-asparaginase (ASNase), as a pivotal amidohydrolase enzyme, has been used in removing acrylamide in food processing and treating acute lymphoblastic leukemia in clinic. In this study, a novel ASNase from Bifidobacterium thermophilum (BtASNase) was successfully cloned and heterologously expressed in Escherichia coli. BtASNase was identified to share maximum 40% structural similarity with other ASNases in PDB database. The purified BtASNase with monomeric size about 35 kDa had the highest specific activity (554.82 IU/mg) at 55 °C and pH 8.0. Further investigation indicated that BtASNase showed

N6-adenosine methylation of mRNA integrates multilevel auxin response and ground tissue development in Arabidopsis.

N6-methyl adenosine (m6A) is a widespread internal mRNA modification impacting the expression of numerous genes. Here, we characterize auxin-related defects among the pleiotropic phenotypes of hypomorphic Arabidopsis thaliana mutants with impaired m6A status and reveal that they show strong resistance to exogenously applied auxin. By combining major published m6A datasets, we propose that among high-confidence target transcripts emerge those encoding the main components required for auxin signaling, including the TIR1/AFB auxin receptors and ARF transcriptional regulators. We also observe subt

Unveiling the environmental fate and risks of non-heterocyclic sulfacetamide: From a novel degradation mechanism to microecological effects.

The presence of sulfonamides (SAs) in the environment has been demonstrated to be a significant factor in pollution. Despite extensive research on heterocyclic SAs, the degradation mechanisms and ecological effects of non-heterocyclic sulfacetamide (SA) remain poorly understood. Our previous study isolated a challenging-to-culture strain, Leucobacter sp. HA-1, known for its SAs-degrading capacity, contains the conserved gene sadA. However, SA was not detected within the degradation spectrum linked to monooxygenase SadA activity. This indicates that other genes may be involved in SA degradation

Endocannabinoids acting at cannabinoid-1 receptors regulate cardiovascular function in hypertension.

BACKGROUND: Endocannabinoids are novel lipid mediators with hypotensive and cardiodepressor activity. Here, we examined the possible role of the endocannabinergic system in cardiovascular regulation in hypertension. METHODS AND RESULTS: In spontaneously hypertensive rats (SHR), cannabinoid-1 receptor (CB1) antagonists increase blood pressure and left ventricular contractile performance. Conversely, preventing the degradation of the endocannabinoid anandamide by an inhibitor of fatty acid amidohydrolase reduces blood pressure, cardiac contractility, and vascular resistance to levels in normote

Deletion of the amidase domain of endolysin LysRODI enhances antistaphylococcal activity in milk and during fresh cheese production.

Milk contamination with Staphylococcus aureus can lead to food poisoning in consumers. One strategy to minimize this risk is the use of phage-derived lysins, which are innocuous for humans and do not readily select for resistant variants. However, it remains necessary to find new candidate lysins and define the conditions for their utilization. This study compares the potential of LysRODI and its derivative LysRODIΔAmi (lacking the amidase domain), which displays high activity and storage stability, to successfully decrease staphylococcal contamination in milk under different conditions. Metabolic Targeting of ASAH1 in Combination with Carnosine as a Therapeutic Strategy for TNBC.

Triple-negative breast cancer (TNBC) is a highly aggressive breast cancer subtype with limited targeted therapies, leading to higher mortality compared with other breast cancer subtypes. In our previous study we identified N-acyl sphingosine amidohydrolase 1 (ASAH1) to be overexpressed in TNBC cells and demonstrated its role in promoting TNBC growth and progression. ASAH1 is a metabolic enzyme that converts ceramide into sphingosine and free fatty acids, thereby creating a favorable environment for tumor growth. To gain a more comprehensive understanding of the metabolic alterations associated

ASAH2 deficiency affects sphingolipid homeostasis and neuromotor control, causing a progressive neurological disorder.

Sphingolipids are integral components of cell membranes and modulate cell survival, proliferation, and apoptosis. ASAH2 is a brain- and gut-enriched gene encoding the neutral N-acylsphingosine amidohydrolase 2, a poorly characterized member of the human ceramidase family. This enzyme plays a pivotal role in maintaining the sphingolipid homeostasis, which is crucial for neurogenesis and synaptic function in the central and peripheral nervous systems. In fact, a dysregulated sphingolipid metabolism is associated with progressive neurological conditions, including Alzheimer disease and Parkinson

A novel case report of spinal muscular atrophy with progressive myoclonic epilepsy from Iran.

Spinal muscular atrophy (SMA) is a disorder characterized by decreased motor function due to the muscle atrophy in the background of degenerated anterior horn cells and motor cells of lower cranial nerves nuclei. The most frequent form is inherited as an autosomal recessive trait resulting from mutations in the survival motor neuron gene (SMN-1). On the other hand, a rare variant of this condition, named progressive myoclonic epilepsy subtype (SMA-PME) occurs in the result of a mutation in N-acylsphingosine amidohydrolase-1 gene (ASAH-1). The latter gene is responsible for lysosomal acid-ceram

Cooperative Amidase Systems Control Product Selectivity During Enzymatic Nylon Depolymerization.

Recent advances in enzymatic polyester depolymerization have highlighted the potential of biocatalysis for sustainable plastic recycling. In contrast, efficient enzymatic strategies for polyamide (PA) degradation remain limited. Here, we investigate how complementary amidases influence product formation during the enzymatic treatment of bulk nylon-6 (PA6) and nylon-6,6 (PA66). Guided by previously established substrate preferences, enzyme combinations are designed to couple NylC-mediated oligomer cleavage with downstream conversion by NylB or cyclic oligomer ring-opening by NylA. Enzymatic production of enantiopure amino acids from mono-substituted hydantoin substrates.

Biocatalytic conversion of 5-substituted hydantoin derivatives is an efficient method for the production of unnatural enantiomerically pure amino acids. The enzymes required to carry out this hydrolysis occur in a wide variety of eubacterial species each of which exhibit variations in substrate selectivity, enantiospecificity, and catalytic efficiency. Screening of the natural environment for bacterial strains capable of utilizing hydantoin as a nutrient source (as opposed to rational protein design of known enzymes) is a cost-effective and valuable approach for isolating microbial species wit

Distribution of cyclic imide-transforming activity in microorganisms.

Cyclic imide-transforming activity was found to be widely distributed in bacteria, yeast and molds. This activity was not correlated with cyclic ureide-transforming activity in bacteria, but there was some correlation in yeast and molds. These two activities are probably catalyzed by different enzymes in bacteria. Besides the well-known cyclic ureide transformation, cyclic imide transformation by microorganisms was common.

Infantile spasms as the initial symptom of biotinidase deficiency.

Two patients with biotinidase deficiency had diagnoses of infantile spasms made at 1 month of age. Biotinidase deficiency may be seen early in the neonatal period without the characteristic findings such as alopecia and seborrheic dermatitis. This diagnosis should be considered in patients with infantile spasms.

A Hadal Streptomyces-Derived Echinocandin Acylase Discovered through the Prioritization of Protein Families.

Naturally occurring echinocandin B and FR901379 are potent antifungal lipopeptides featuring a cyclic hexapeptide nucleus and a fatty acid side chain. They are the parent compounds of echinocandin drugs for the treatment of severe fungal infections caused by the Candida and Aspergilla species. To minimize hemolytic toxicity, the native fatty acid side chains in these drug molecules are replaced with designer acyl side chains. The deacylation of the N-acyl side chain is, therefore, a crucial step for the development and manufacturing of echinocandin-type antibiotics. Echinocandin E (ECE) is a n

Recharacterization of EmAGA, a Potential Candidate for Novel ALL Therapeutics.

Aspartylglucosaminidase (AGA) is an amidohydrolase that can hydrolyze the amide bond between N-acetylglucosamine (GlcNAc) and asparagine (Asn), producing N-acetylglucosamine and aspartic acid (Asp). AGA is distributed in both eukaryotes and prokaryotes. In this study, we identified the sequence of AGA in Elizabethkingia meningoseptica (EmAGA), cloned and expressed it in Escherichia coli, and recharacterized its properties, confirming its substrates as aspartylglucosamine (Asn-GlcNAc) and Asn. Key residues affecting its enzymatic activity were predicted through molecular docking and conserved s

Evidence for clinical, genetic and biochemical variability in spinal muscular atrophy with progressive myoclonic epilepsy.

Spinal muscular atrophy with progressive myoclonic epilepsy (SMA-PME) is a recently delineated, autosomal recessive condition caused by rare mutations in the N-acylsphingosine amidohydrolase 1 (acid ceramidase) ASAH1 gene. It is characterized by motor neuron disease followed by progressive myoclonic seizures and eventual death due to respiratory insufficiency. Here we report an adolescent female who presented with atonic and absence seizures and myoclonic jerks and was later diagnosed as having myoclonic-absence seizures. An extensive genetic and metabolic work-up was unable to arrive at a mol
