Influence of additives on the clouding phenomenon of chlorpromazine hydrochloride solutions.

Herein we report the effect of various additives (viz. alcohols, cycloalcohols, amino acids, sugars, ureas) on the clouding phenomenon observed in 50mM chlorpromazine hydrochloride (CPZ) drug solutions (prepared in 10mM sodium phosphate buffer). Long chain alcohols (except octanol), cyclohexanol and allylalcohol increased the cloud point (CP) followed by a decrease with the increase in alcohol concentration but short chain alcohols affected the CP insignificantly. Effect of amino acids depended upon their nature: acidic and salts of basic amino acids increased the CP while basic amino acids de

[Advancements in the genetics of cystinuria].

Cystinuria is an amino acid disease due to a defect of intestinal and renal tubular transport of cystine and various basic amino acids (lysine, arginine and ornithine). The disease is transmitted horizontally according to an autosomal recessive pattern. The overall prevalence is one per 7,000 live births. It is the commonest hereditary disease affecting amino acid transport (MIM 220100). This disease is characterized by excessive urinary excretion of cystine and basic amino acids. From a clinical point of view, almost 50% of homozygotes will develop cystine renal stones with urinary tract infe

Changes in free amino acids in peripheral blood (PB) lymphocytes and polymorphonuclear (PMN) leukocytes after treatment with diazepam.

The effect of single and chronic (15 days) i.p. injections (1.0 and 8.0 mg/kg) of diazepam (DZ) on free amino acid profile in peripheral blood (PB) lymphocytes and polymorphonuclear (PMN) leukocytes of male Wistar Albino rats were investigated. Depletion of some free amino acids was observed in the lymphocytes (mixed T- and B-lymphocytes) and PMN leukocytes (91-95%) neutrophils especially after chronic DZ-treatment. A dose-dependent depletion in the lymphocyte amino acids, Tau, Gly, Ala, Met and Ile, was found after both acute and chronic DZ-treatment. A similar depletion of Tau, Asp, Glu and

Interplay between basic residues of hepatitis C virus glycoprotein E2 with viral receptors, neutralizing antibodies and lipoproteins.

Positively-charged amino acids are located at specific positions in the envelope glycoprotein E2 of the hepatitis C virus (HCV): two histidines (H) and four arginines (R) in two conserved WHY and one RGERCDLEDRDR motifs, respectively. Additionally, the E2 hypervariable region 1 (HVR1) is rich in basic amino acids. To investigate the role(s) of these residues in HCV entry, we subjected to comparative infection and sedimentation analysis cell culture-produced (HCVcc, genotype 2a) wild type virus, a panel of alanine single-site mutants and a HVR1-deletion variant. Initially, we analyzed the effec

Nonenzyme browning and its effect on protein nutrition.

The nonenzyme browning involves the thermal decomposition of sugars, the caramelization, the decomposition of oxi-acids, the so called "Maillard reaction" between amino acids and carbohydrates, the reaction between oxidized fats and proteins, and those alterations which take place by the alkaline treatment of proteins. The Maillard reaction is of secondary importance in the case of foodstuffs and fodders with low carbohydrate contents (meats, meat meal, fish meal). By the heat treatment, the sulphur-containing amino acids of proteins (cystine, methionine) are damaged primarily because of oxida

Application of Analytical Techniques to Chiral Amino Acid Profiling in Clinical Samples: Bridging Chemistry and Disease Research.

Amino acids are the basic building blocks of proteins that are required to control various vital biological processes, including immune response, hormone regulation, metabolic control, and gene expression. Although L-amino acids are the primary components of protein synthesis, their mirror-image counterparts, D-amino acids, exhibit distinct pharmacological and biological characteristics. As a result, reliable identification and measurement of these enantiomers have become critical criteria in biology, chemistry, and medicine. Because changes in D-amino acid levels are closely related to the on

Exploring Helical Folding in Oligomers of Cyclopentane-Based ϵ-Amino Acids: A Computational Study.

The conformational preferences of oligopeptides of an ϵ-amino acid (2-((1R,3S)-3-(aminomethyl)cyclopentyl)acetic acid, Amc5 a) with a cyclopentane substituent in the Cβ -Cγ -Cδ sequence of the backbone were investigated using DFT methods in chloroform and water. The most preferred conformation of Amc5 a oligomers (dimer to hexamer) was the H16  helical structure both in chloroform and water. Four residues were found to be sufficient to induce a substantial H16 helix population in solution. The Amc5 a hexamer adopted a stable left-handed (M)-2.316 helical conformation with a rise of 4.8 Å per t

Therapeutic Effects of Taurine and Histidine Supplementation in Retinal Diseases.

Amino acids are basic building blocks of structural proteins and enzymes. They also act as signaling molecules and as fuel. They are characterized as essential if sufficient quantities must be supplied exogenously or as non-essential if they can be endogenously synthesized. Appropriate intake of amino acids not only prevents the development of metabolic diseases but also can reduce the progression of some disease states. Amino acids are strongly associated with retinal metabolism in physiology and pathology. Nonetheless, there is a lack of robust clinical studies supporting the benefits of ami

Effects of recombinant imperatoxin A (IpTxa) mutants on the rabbit ryanodine receptor.

Imperatoxin A (IpTxa), a 3.7 kDa peptide from the African scorpion Pandinus imperator, is an agonist of the skeletal muscle ryanodine receptor (RyR1). In order to study the structure of the toxin and its effect on RyR1, IpTxa cDNA was PCR-amplified using 3 pairs of primers, and the toxin was expressed in E. coli. The toxin was further purified by chromatography, and various point mutants in which basic amino acids were substituted by alanine were prepared by site-directed mutagenesis. Studies of single channel properties by the planar lipid bilayer method showed that the recombinant IpTxa was

Novel gain-of-function mutants identify a critical region within CFTR membrane-spanning domain 2 controlling cAMP-dependent and ATP-independent channel activation.

CFTR is an anion channel that has evolved from the mold of an ABC transporter. It possesses specific structural features, including a lateral portal between the cytoplasmic extensions of its transmembrane helices TM4 and TM6. This TM4-TM6 portal is lined by basic residues attracting anions from the cytosol towards the intracellular vestibule. Even though a symmetric, open portal is not observed at the level of the TM10/TM12 interface, basic amino acids are also present at this level, exposed to solvent in the vicinity of the regulatory R region, whose phosphorylation enables channel activation

Hartnup disease. Clinical, pathological, and biochemical observations.

Hartnup disease is a rare genetic disorder of amino acid transport associated with variable and intermittent clinical abnormalities. A family is described in which three siblings had an intermittently progressive neurological disease and two of the affected siblings had the Hartnup-pattern aminoaciduria. Neuropathological examination of one case showed severe diffuse atrophy, generalized neuronal loss in the cortex, and Purkinje cell loss in the cerebellum. In vivo and in vitro studies of intestinal amino acid transport in the surviving sibling indicated a partial defect in the transport of se

Carboxy-terminal conversion of profibrillin to fibrillin at a basic site by PACE/furin-like activity required for incorporation in the matrix.

Fibrillin-1, the main component of 10-12 nm microfibrils of the extracellular matrix, is synthesized as profibrillin and proteolytically processed to fibrillin. The putative cleavage site has been mapped to the carboxy-terminal domain of profibrillin-1, between amino acids arginine 2731 and serine 2732, by a spontaneous mutation in this recognition site that prevents profibrillin conversion. This site contains a basic amino acid recognition sequence (R-G-R-K-R-R) for proprotein convertases of the furin/PACE family. In this study, we use a mini-profibrillin protein to confirm the cleavage in th

Gyrate atrophy of the choroid and retina: amino acid metabolism and correction of hyperornithinemia with an arginine-deficient diet.

Four patients with gyrate atrophy of the choroid and retina were studied, all of whom exhibited the hyperornithinemia characteristic of this disorder. Elevated plasma histidine and diminished plasma lysine and branched-chain amino acids were also noted. The renal clearances of these four amino acids were not sufficiently elevated to explain their low plasma levels. In one subject, an arginine-deficient diet led to progressive reduction in plasma ornithine from 13 times normal to the upper limits of normal, along with the disappearance of ornithinuria and lysinuria. Competitive inhibition and exchange transport between arginine and lysine in the process of absorption from the caecum in pigs.

Competitive inhibition and exchange transport between arginine and lysine in the process of absorption from the caecum in pigs. Acta Physiol. Pol., 1978, 29 (2): 161--165. Arginine competes with lysine in the process of transport across the mucosal epithelium of the pig caecum, and the ability of arginine absorption is much greater than that of lysine. Absorption of large amounts of arginine causes secretion of large amounts of lysine into the intestinal lumen in the process of primary active exchange transport. Lysine is better absorbed from solution containing no other amino acids than from
