Further studies on environmental factors that modify the toxicity of nickel to microbes.

The toxicity of nickel (Ni) to mycelial growth of filamentous fungi and to replication of eubacteria, an actinomycete, and yeasts was influenced by various environmental abiotic factors. Sulfide and phosphate reduced the toxicity of Ni by the formation of insoluble salts. The clay minerals, montmorillonite and, to a much lesser extent, kaolinite, and the hydrous oxides of aluminum or manganese reduced the toxicity, presumably by the adsorption of cationic Ni to their net negatively charged surfaces. Amino acids, such as aspartic acid, complex organics, such as tryptone, casamino acids, and yea

NAD-glutamate dehydrogenase from Halobacterium halobium: inhibition and activation by TCA intermediates and amino acids.

A variety of metabolites have been found to elicit a form of inhibition or activation on an NAD-specific glutamate dehydrogenase (NAD-GDH, EC 1.4.1.2) from Halobacterium halobium. The purified halophilic enzyme was tested with several compounds known to be allosteric modifiers of mammalian glutamate dehydrogenases to determine their effects on enzyme activity. GTP, ATP, ADP and AMP did not affect the enzyme, so these effectors of bovine glutamate dehydrogenase do not play a role in the regulation of the halophilic enzyme. However, the halophilic enzyme was subject to strong inhibition by TCA i

Long-term depression requiring tACPD-receptor activation and NMDA-receptor blockade.

In slices from the rat visual cortex, application of the metabotropic glutamate receptor (mGluR) agonist trans-1-aminocyclo-pentane-1,3-dicarboxylic acid (tACPD), whether combined with tetanization or not, produced only a reversible depression but not long-term depression (LTD) of synaptic transmission. In the presence of both tACPD and the NMDA receptor antagonist D-(-)-2-amino-5-phosphonopentanoate, tetanization induced LTD. These findings suggest requirement of tACPD-sensitive mGluR subtypes for inducing a form of LTD in the visual cortex.

Single sensory neurons activate excitatory amino acid receptors in the lamprey spinal cord.

The effects of excitatory amino acid (EAA) antagonists were tested on sensory afferent excitation in the lamprey spinal cord. Paired intracellular recordings were made from mechanosensory neurons (dorsal cells) and second order sensory neurons (giant interneurons). Stimulation of individual dorsal cells evoked mono- and/or polysynaptic excitatory postsynaptic potentials (EPSPs) in giant interneurons. These EPSPs were depressed by the EAA antagonists cis-2,3-piperidine dicarboxylate and kynurenic acid. Small components of the synaptic potentials were due to electrical coupling since they were n

Dicarboxylic aciduria due to medium chain acyl CoA dehydrogenase defect. A cause of hypoglycemia in childhood.

Dicarboxylic aciduria was found during hypoglycemic episode in a 14 months old girl. Her brother had died at the age of 4 years during febrile illness. A ketogenic diet induced in this patient a severe hypoglycemia. Urinary organic acid profile exhibited abnormal excretion of the C6-C10 dicarboxylic acids (adipic-suberic-sebacic) and related metabolites (5 hydroxyhexanoic, hexanoylglycine, suberyl glycine). This pattern suggested a defect in fatty acids beta oxidation. Plasma carnitine values was within control limits. Similar clinical findings and urinary organic acids excretion have been des

Metabolic Myopathies.

PURPOSE OF REVIEW: Metabolic myopathies are genetic disorders that impair intermediary metabolism in skeletal muscle. Impairments in glycolysis/glycogenolysis (glycogen-storage disease), fatty acid transport and oxidation (fatty acid oxidation defects), and the mitochondrial respiratory chain (mitochondrial myopathies) represent the majority of known defects. The purpose of this review is to develop a diagnostic and treatment algorithm for the metabolic myopathies. RECENT FINDINGS: The metabolic myopathies can present in the neonatal and infant period as part of more systemic involvement with

The new mutation, E46K, of alpha-synuclein causes Parkinson and Lewy body dementia.

Familial parkinsonism and dementia with cortical and subcortical Lewy bodies is uncommon, and no genetic defect has been reported in the previously described sibships. We present a Spanish family with autosomal dominant parkinsonism, dementia, and visual hallucinations of variable severity. The postmortem examination showed atrophy of the substantia nigra, lack of Alzheimer pathology, and numerous Lewy bodies which were immunoreactive to alpha-synuclein and ubiquitin in cortical and subcortical areas. Sequencing of the alpha-synuclein gene showed a novel, nonconservative E46K mutation in heter

Targeted serum metabolomics reveals alterations in amino acid and neurotransmitter pathways in Parkinson's disease.

Serum metabolomic profiling in Parkinson's disease (PD) remains underexplored. This study characterized alterations in serum amino acid and neurotransmitter levels in PD using targeted metabolomics and identified associated dysregulated metabolic pathways. A validated ultra-high performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method was employed to quantify 23 amino acids and 3 neurotransmitters in PD patients and controls. Logistic regression models assessed the relationship between motor phenotype and metabolomic profiles. An OPLS-DA model incorporating variable impor

The VC1777-VC1779 proteins are members of a sialic acid-specific subfamily of TRAP transporters (SiaPQM) and constitute the sole route of sialic acid uptake in the human pathogen Vibrio cholerae.

Sialic acids are nine-carbon amino sugars that are present on all mucous membranes and are often used by bacteria as nutrients. In pathogenic Vibrio the genes for sialic acid catabolism (SAC) are known to be important for host colonization, yet the route for sialic acid uptake is not proven. Vibrio cholerae contains a tripartite ATP-independent periplasmic (TRAP) transporter, SiaPQM (VC1777-VC1779), encoded by genes within the Vibrio pathogenicity island-2 (VPI-2), which are adjacent to the SAC genes nanA, nanE and nanK. We demonstrate a correlation of the occurrence of VPI-2 and the ability o

Integrated bioinformatic and phenotypic analysis of RpoN-dependent traits in the plant growth-promoting bacterium Pseudomonas fluorescens SBW25.

The alternative sigma factor RpoN is a key regulator in the acclimation of Pseudomonas to complex natural environments. In this study we show that RpoN is required for efficient colonization of sugar beet seedlings by the plant growth-promoting bacterium Pseudomonas fluorescens SBW25, and use phenotypic and bioinformatic approaches to profile the RpoN-dependent traits and genes of P. fluorescens SBW25. RpoN is required for flagellar biosynthesis and for assimilation of a wide variety of nutrient sources including inorganic nitrogen, amino acids, sugar alcohols and dicarboxylic acids. Chemosens

The effects of castration on the quality of black Tibetan sheep meat and muscle metabolites.

Castration is a vital technical practice in Tibetan sheep farming on the Qinghai-Tibet Plateau. It reduces fighting among rams, making it easier to manage the herd while they are grazing. However, the impact of castration on the quality of Tibetan sheep meat and muscle metabolites is unclear at present. On this basis, this study examined differences in meat quality and metabolites between castrated (BTC) and non-castrated (BTN) Black Tibetan sheep using targeted and non-targeted metabolomics. Compared with the BTN group, the meat from the BTC group showed reduced drip loss and increased cookin

Four-Component Ugi Reaction for Optical Chirality Sensing and Surface Nanoengineering of Chiral Self-Assemblies.

Using green chemistry to control chirality at hierarchical levels as well as chiroptical activities endows with new opportunities to the development of multiple functions. Here, the four-component Ugi reaction is introduced for the general and precise optical chirality sensing of amines as well as the surface nanoengineering of chiral soft self-assemblies. To overcome the relatively weak Cotton effects, direct synthesis of a folded peptide structure on a rotatable ferrocene core with axial chirality was accomplished from chiral amine, 1,1'-ferrocenyl dicarboxylic acid, formaldehyde and isocyan

Deciphering the metabolic perturbation in hepatic alveolar echinococcosis: a 1H NMR-based metabolomics study.

BACKGROUND: Hepatic alveolar echinococcosis (HAE) is caused by the growth of Echinococcus multilocularis larvae in the liver. It is a chronic and potentially lethal parasitic disease. Early stage diagnosis for this disease is currently not available due to its long asymptomatic incubation period. In this study, a proton nuclear magnetic resonance (1H NMR)-based metabolomics approach was applied in conjunction with multivariate statistical analysis to investigate the altered metabolic profiles in blood serum and urine samples obtained from HAE patients. The aim of the study was to identify the

Integrated omics-based analysis reveals the role of ferulic acid activating a co-production of 4-vinyl guaiacol and fumaric acid by Rhizopus oryzae.

The efficient valorization of lignocellulose-derived phenolics remains a major challenge in sustainable biomanufacturing. Here, an integrated omics approach was employed to elucidate how ferulic acid (FA) activates the co-production of 4-vinyl guaiacol (4-VG) and fumaric acid by Rhizopus oryzae. Co-supplementation of FA with glucose significantly enhanced metabolic performance, increasing fumaric acid yield by 16.9% and inducing simultaneous 4-VG synthesis. Metabolomic profiling revealed that FA reprograms cellular metabolism by reinforcing the tricarboxylic acid cycle and redox pathways while

Excitatory amino acid receptors in commissural nucleus of the NTS mediate carotid chemoreceptor responses.

Stimulation of carotid body chemoreceptors by saline saturated with 100% CO2 elicited an increase in mean arterial pressure, respiratory rate, tidal volume, and minute ventilation (VE). Microinjections of L-glutamate into a midline area 0.5-0.75 mm caudal and 0.3-0.5 mm deep with respect to the calamus scriptorius increased VE. Histological examination showed that the site was located in the commissural nucleus of the nucleus tractus solitarii (NTS). Identification of multispecific organic anion transporter 2 expressed predominantly in the liver.

In the present study, we demonstrate that NLT (novel liver-specific transport protein) is a multispecific organic anion transporter of the liver. The amino acid sequence of NLT shows 42% identity to that of the renal multispecific organic anion transporter, OAT1. When expressed in Xenopus laevis oocytes, NLT mediated uptake of organic anions, such as salicylate, acetylsalicylate, PGE2, dicarboxylates and p-aminohippurate. [14C]Salicylate uptake via NLT was saturable (Km = 88.8 +/- 23.4 microM) and sodium-independent. Expression of the mRNA of NLT was detected in the liver and kidney (liver >>

Long-term alterations in serum metabolite profiles in women with a history of preeclampsia -FINNCARE study.

Preeclampsia (PE) is a multifaceted pregnancy condition which increases the risk of cardiometabolic diseases later in life. Long-term follow-up data on metabolome utilizing novel metabolomic techniques are needed to gain insights into metabolism after PE and non-PE pregnancies for identifying potential areas of investigation that could assist in predicting and managing future health risks. The objective of the current study was to characterize long-term metabolome in women with and without a history of PE. Non-targeted metabolomics (liquid chromatography-mass spectrometry, LC-MS) was applied t
