Reversibility of thiamine deficiency-induced partial necrosis and mitochondrial uncoupling by addition of thiamine to neuroblastoma cell suspensions.

Culture of neuroblastoma cells in the presence of low thiamine concentration (16 nM) and of the transport inhibitor amprolium leads to the appearance of signs of necrosis: the chromatin condenses, the oxygen consumption decreases and is uncoupled, the mitochondrial cristae are disorganized, the thiamine diphosphate-dependent dehydrogenase activities are impaired. When 10 microM thiamine are added to these cells, the basal respiration increases, the coupled respiration is restored and mitochondrial morphology is recovered within 1 h. Addition of succinate, which is oxidized via a thiamine dipho

Comparative efficacy of commonly used herbal and orthodox anticoccidial drugs in broiler birds experimentally infected with mixed Eimeria species.

Coccidiosis is a major economic burden in poultry farming worldwide. In Nigeria, herbal anticoccidials are widely used, yet their efficacy compared to orthodox drugs remains unclear. This study evaluates their comparative effects in broilers experimentally infected with mixed Eimeria species. 70 five-week-old broiler birds were employed in this study. The infective dose of the field strain of mixed Eimeria species was determined following standard procedures. Forty-two birds were randomly assigned to six groups (seven birds per group) to compare the anticoccidial effects of herbal and orthodox

[Therapeutic approaches to cryptosporidiosis. A review of the literature].

Cryptosporidiosis is a coccidian infection that usually occurs in children an immunocompromised patients. With the AIDS (Acquired Immunodeficiency Syndrome) epidemic there have been an increased number of clinical cases and still we don't have an optimal therapeutic regimen to eradicate the infection. Since 1907 when the organism was first described, a large amount of anti-infective agents have been used without success. We present herein a review of the new therapeutic approaches, although none of them is satisfactory and new studies are required for the development of an optimal treatment. S

[Ultrastructural study of experimental cerebrocortical necrosis in calves (author's transl)].

Cerebrocortical necrosis (CCN) was experimentally induced in three calves with the thiamine antagonist Amprolium. The calves were followed clinically. At the time of development of typical clinical signs of CCN the calves were killed and necropsied and a complete histopathologic examination was performed. The light microscopic lesions of the cerebral cortex were those of middle laminar necrosis and deep laminar edema. The necrotic zone consisted of fine vesicles and differed only slightly from the edema zone. In the former some neurons were morphologically normal, others showed signs of injury

Commiphora myrrha extract protects pigeons from Eimeria labbeana-like-triggered inflammatory dysregulation.

BACKGROUND: Coccidiosis, caused by Eimeria species, is a major enteric disease in birds, with Eimeria labbeana-like isolates frequently inducing severe intestinal lesions, diarrhea, and reduced weight gain in pigeons. Conventional anticoccidial drugs face limitations due to resistance, residue concerns, and environmental impact, highlighting the need for alternative strategies. Commiphora myrrha (myrrh) is a resinous plant extract rich in bioactive compounds with antioxidant, antimicrobial, antiparasitic, and anti-inflammatory properties. This study evaluated the protective effects of C. myrrh

Substrate transport and drug interaction of human thiamine transporters SLC19A2/A3.

Thiamine and pyridoxine are essential B vitamins that serve as enzymatic cofactors in energy metabolism, protein and nucleic acid biosynthesis, and neurotransmitter production. In humans, thiamine transporters SLC19A2 and SLC19A3 primarily regulate cellular uptake of both vitamins. Genetic mutations in these transporters, which cause thiamine and pyridoxine deficiency, have been implicated in severe neurometabolic diseases. Additionally, various prescribed medicines, including metformin and fedratinib, manipulate thiamine transporters, complicating the therapeutic effect. Despite their physiol

Vitamin deficiencies in cattle.

Deficiencies of vitamins A, D, K, E and thiamin can cause severe limitations in beef production. In particular, vitamin A and E can be common causes of lost profit, secondary to limitations of reproductive and growth potential. Prolonged dry periods will reduce available A and E in pasture forage, as can ensiling and prolonged storage of harvested feedstuffs. Polioencephalomalacia is a thiamin responsive disorder, associated with high concentrate feeding and lush pastures. Antimetabolites, such as amprolium, will cause thiamine deficiency when fed in excess. Recent information has shown improv

Mulberry extract as an ecofriendly anticoccidial agent: in vitro and in vivo application.

Natural products are ecofriendly agents that can be used against parasitic diseases. Eimeria species cause eimeriosis in many birds and mammals and resistance to available medications used in the treatment of eimeriosis is emerging. We investigated the in vitro and in vivo activity of Morus nigra leaf extracts (MNLE) against sporulation of oocysts and infection of mice with Eimeria papillata. Phytochemical analysis of MNLE showed the presence of seven compounds and the in vitro effects of MNLE, amprolium, DettolTM, formalin, ethanol, and phenol were studied after incubation with oocysts before

Biosynthesized chitosan nanoparticles from Krameria lappacea: a strategy to ameliorate coccidiosis-induced intestinal dysfunction.

BACKGROUND: Coccidiosis is a major parasitic disease caused by Eimeria species that induces intestinal damage, oxidative stress, and impaired nutrient absorption. The risk associated with drug-resistant strains and other adverse effects related to conventional anticoccidial drugs necessitates the development of safer natural alternatives. Thus, in this study, we examined the anticoccidial efficacy of biosynthesized chitosan nanoparticles (CNPs) derived from Krameria lappacea root extract against Eimeria papillata-induced murine coccidiosis. METHODS: Chitosan nanoparticles were biosynthesized

Comparison of the effects of some thiamine analogues upon thiamine transport across the blood-brain barrier of the rat.

The flux of thiamine from the blood into the brain has been measured by a specially devised procedure in which a steady raised level of the vitamin, with or without radioactive labelling, was achieved rapidly and maintained steadily in the circulating blood plasma. This was done by a single rapid I.V. injection followed by a continuous injection given at a rate adjusted according to a pre-determined programme, so as to replace the injected material at the rate at which it had been found to leave the circulation in preliminary experiments. A series of four chemical analogues of thiamine were gi

Residues of antibiotics in yeasts from ethanol production: a possible contamination route for feedingstuffs.

Sugarcane yeast and brewer's yeast from ethanol production are widely used as ingredients of animal feed formulations in Brazil. To avoid the contamination of the must in ethanol production refineries, the use of antibiotics is one of the main preventive treatments. Thus, there is a risk of antibiotic residues carry over from yeast to animal feed. This unintentional addition of antibiotics can produce non-compliant feed products, due to regulatory aspects and their toxicity for animals. The results of an exploratory program to assess the occurrence of over 60 antibiotics and other pharmaceutic

Cerebral encephalomalacia in commercial turkeys.

A flock of 9 1/2-week-old commercial tom turkeys experienced high mortality after consuming a complete feed containing an unidentified toxic substance. Initially, turkeys were found dead. Clinically, the birds were calm and still but became hyperexcitable with noise. A small percentage of birds exhibited torticollis, opisthotonos, circling, ataxia, and blindness. Findings at necropsy and upon microscopic examination were bilaterally symmetrical areas of necrosis of the cerebral hemispheres in the area of the neostriatum that were well demarcated from the surrounding normal neuropil. A feeding

Influence of Eimeria spp. Infection on Chicken Jejunal Microbiota and the Efficacy of Two Alternative Products Against the Infection.

Eimeria spp. are important intestinal pathogens of chickens (Gallus gallus domesticus). Anticoccidial feed additives, chemicals, and ionophores have traditionally been used to control Eimeria infections in broiler production. Thus, the trend toward antibiotic-free and organic production requires new approaches to coccidiosis prevention. Two not mutually exclusive methods are the use of plant extracts with antiparasitic activity and manipulation of the intestinal microbiota by pre- and probiotics. In the present study, birds were inoculated with a combination of Eimeria acervulina, Eimeria maxi

Integrated Experimental and Computational Evaluation of the Gut-Targeted Anticoccidial Potential of Agaricus bisporus Methanolic Extract.

Coccidiosis caused by Eimeria spp. is a major constraint to global poultry production, causing economic losses exceeding USD 13 billion annually. Growing drug resistance from prolonged anticoccidial use highlights the need for safer, eco-friendly alternatives. Edible mushrooms, such as Agaricus bisporus, are rich in bioactive phytochemicals, yet their anticoccidial potential remains underexplored. This study investigated the methanolic extract of A. bisporus (ABE) for inhibitory effects on Eimeria papillata oocyst sporulation and interactions with essential apicomplexan enzymes. ABE was chemic
