Postmenopausal osteoporosis is the most common form of primary osteoporosis, and the incidence of the condition is rapidly increasing. In consideration of the limitations of current therapeutic options for the treatment of postmenopausal osteoporosis, there is an urgent need to develop safer alternatives. Anacardic acid, a natural phenolic acid compound extracted from cashew nut shell, possesses potent antitumor and anti-inflammatory effects and inhibits NF-κB signaling. However, its effect on osteoclasts remains unknown. This study reports the first evidence for the antiosteoclastogenic and a

The inhibitory and bactericidal activities of anacardic acid and totarol, alone and in combination with methicillin, were investigated against methicillin-resistant Staphylococcus aureus (MRSA). The growth of two MRSA strains was inhibited by 6 x 25 microg ml-1 of anacardic acid and 0 x 78 microg ml-1 of totarol. The time-kill curve study showed that these two compounds were bactericidal against MRSA. Anacardic acid killed MRSA cells more rapidly than totarol, and no viable cells were detected after being exposed to 6 x 25 microg ml-1 of anacardic acid for 6 h. Anacardic acid showed bactericid

The molluscicide activity of hexanic extract from Anacardium occidentale L. (cashew) nut shell, of copper (II) complex, of lead (II) complex and anacardic acid has been compared in the laboratory in an attempt to obtain better stability than anacardic acid. This was obtained from the hexanic extract of the cashew nut shell by precipitation with lead (II) hydroxide or cupric sulfate plus sodium hydroxide or (II) cupric hydroxide followed by treatment of lead (II) complex with a diluted solution of sulfuric acid. Ten products of the mixture obtained were tested on adults snails of Biomphalaria g

SUMOylation is a critical post-translational modification pathway implicated in various ailments, such as diabetes, neurodegenerative disorders, infectious diseases, and cancer. Dysregulation of SUMOylation affects different hallmarks of cancer, including metastasis, angiogenesis, invasion, and proliferation, making it an important therapeutic target. While anacardic acid (AA) has been shown to inhibit the essential SUMO E1 enzyme (IC50 = 2.1 μM), its clinical potential is limited by high lipophilicity and poor bioavailability, stemming primarily from its C15 alkyl chain. This study is therefo

Cardiac hypertrophy is a complex process induced by the activation of multiple signaling pathways. We previously reported that anacardic acid (AA), a histone acetyltransferase (HAT) inhibitor, attenuates phenylephrine (PE)-induced cardiac hypertrophy by downregulating histone H3 acetylation at lysine 9 (H3K9ac). Unfortunately, the related upstream signaling events remained unknown. The mitogen-activated protein kinase (MAPK) pathway is an important regulator of cardiac hypertrophy. In this study, we explored the role of JNK/MAPK signaling pathway in cardiac hypertrophy induced by PE. The mice

Anacardic acids (AAs) are a class of alkyl phenols widely distributed in the Anacardiaceae family and known for their diverse biological properties. However, their potential against type 2 diabetes mellitus (T2DM) has never been thoroughly investigated. In this study, a fraction isolated from a Pistacia vera shell extract, enriched in AAs, exhibited potent inhibitory activity against α-amylase (α-Amy) and α-glucosidase (α-Glu). Pure (15:0)-AA and (17:1)-AA also acted as strong inhibitors of both enzymes, displaying IC₅₀ values significantly lower than those of the marketed anti-diabetic drug a

BACKGROUND: The expression of cardiac genes is precisely regulated, and any perturbation may cause developmental defects. In a previous study, we demonstrated that alcohol consumption during pregnancy could lead to uncontrolled expressions of cardiac genes and eventually result in cardiac dysplasia. However, the underlying mechanisms remain unclear. In the present study, we have investigated the alcohol-induced cardiac hypertrophy and its potential mechanisms. Furthermore, the protective effect of anacardic acid against the alcohol-induced cardiac hypertrophy has been explored in experimental

BACKGROUND: Myocardial fibrosis (MF), a hallmark of cardiovascular diseases (CVDs) such as myocardial infarction (MI), drives progressive cardiac dysfunction and adverse remodeling. Histone acetylation is a critical epigenetic regulator in cardiovascular pathology. Anacardic acid (AA), a histone acetyltransferase inhibitor (HATi) with pleiotropic bioactivities, has been studied in various disease contexts; however, its antifibrotic efficacy and mechanisms in MF remain unclear.
METHODS: In vivo, a mouse model of post-infarction MF was established by permanent left anterior descending (LAD) lig

Amphipterygium adstringens is a plant traditionally used to treat gingivitis, gastric ulcer and even gastric cancer but the mechanism involved in the regulation of the immune response is not elucidated yet. The 6-pentadecylsalicylic acid (6SA) is the main anacardic acid found in A. adstringens. In order to evaluate the immune-modulatory abilities of 6SA, we used mouse splenocytes and determined the phosphorylation of the transcription factor NF-κB and MAP kinases ERK1/2, JNK and p38 in helper and cytotoxic T cells, natural killer (NK) cells and F4/80(+) macrophages. Treatment with 6SA was not

Amphipterygium adstringens (cuachalalate) contains anacardic acids (AAs) such as 6-pentadecyl salicylic acid (6SA) that show immunomodulatory and antitumor activity with minimal or no secondary adverse effects. By contrast, most chemotherapeutic agents, such as 5-fluorouracil (5-FU) and carboplatin (CbPt), induce myelosuppression and leukopenia. Here, we investigated the myeloprotective and antineoplastic potential of an AA extract or the 6SA as monotherapy or in combination with commonly used chemotherapeutic agents (5-FU and CbPt) to determine the cytoprotective action of 6SA on immune cells

The increasing rise of multidrug-resistant bacteria necessitates an urgent need for the discovery of novel antibacterial agents. Natural products have long been a source for identifying and isolating novel antibacterial agents. Anacardic acids (AAs), a phenolic lipid isolated from solventextracted cashew nutshell liquid (CNSL) of Anacardium occidentale (Family Anacardiaceae), have garnered potential attention for their potent antibacterial properties. Besides Anacardium occidentale, different analogues of AAs have also been isolated from various natural sources. These natural and structurally

Diabetic nephropathy (DN) is a multifactorial disease, so there is a global attitude to use natural products for DN management due to their polypharmacological effects. Anacardic acid (AA) is a natural product from Anacardium occidentale. We aimed to investigate the protective effect of AA on the kidney and pancreas through targeting hyperlipidemia, hyperglycemia, insulin resistance oxidative stress, TNF-α, TGFβR1, and α-SMA signaling. Network pharmacology, Molecular docking, experimental, and histopathological studies were established to evaluate the efficacy of AA on DN management. Biochemic

Chagas disease (CD) is a tropical disease caused by the parasite Trypanosoma cruzi, transmitted by the barber insect. Currently, there are approximately 7 million infected people in the world, and it is estimated that 70 million people could contract this disease. The anacardic acid (AA) showed effectiveness in in silico and in vitro tests. The antichagasic potential of five sulfonamide molecules, derived from anacardic acid, was evaluated from a molecular approach based on the density functional theory (DFT), molecular dynamics (MD), and molecular docking (docking) calculations. Malaria represents a major global health challenge. Excessive activation of inflammatory response is critical for the pathogenesis of severe malaria. The NF-κB signaling pathway regulates inflammatory responses; saturated anacardic acid (SAA) inhibits NF-κB activation. The aim of this study was to evaluate whether SAA modulated inflammatory mediators in BV-2 microglia and THP-1 monocytes, whose overproduction contributes to the development of severe malaria. Survival rate was assessed in cerebral malaria-susceptible CBA mice infected with Plasmodium berghei ANKA and treated or not with SAA. TH

Anacardic acids, a class of secondary compounds derived from fatty acids, are found in a variety of dicotyledonous families. Pest resistance (e.g., spider mites and aphids) in Pelargonium xhortorum (geranium) is associated with high levels (approximately 81%) of unsaturated 22:1 omega 5 and 24:1 omega 5 anacardic acids in the glandular trichome exudate. A single dominant locus controls the production of these omega 5 anacardic acids, which arise from novel 16:1 delta 11 and 18:1 delta 13 fatty acids. We describe the isolation and characterization of a cDNA encoding a unique delta 9 14:0-acyl c

INTRODUCTION: Traumatic brain injury (TBI) triggers neuroinflammation and NLRP3 inflammasome-dependent pyroptosis, causing substantial neuronal damage. Anacardic acid (AA), a natural histone acetyltransferase (HAT) inhibitor, has anti-inflammatory properties related to TBI. This study explored AA's effects on pyroptosis and its neuroprotective potential after TBI.
METHODS: A controlled cortical impact (CCI) mouse model was employed. Neurological recovery was evaluated by mNSS and rotarod tests, lesion volume by H&E staining, and neuronal survival by NeuN/TUNEL immunofluorescence. Inflammatory

A Ginkgo biloba L. fruit extract was prepared and purified. Three groups of guinea pigs were sensitized to the crude extract, anacardic acids 1, and cardanols 2 respectively, using the FCAT method, and the fourth group to urushiol using the epicutaneous route. Each group was tested for reaction to the primary sensitizer and to the different main aromatic compounds isolated from Ginkgo fruits. Anacardic acids were found to be good sensitizers, while cardanols failed to induce allergic contact dermatitis (ACD). No cross-reactions were observed among the compounds tested. Ginkgolic acids 1 seem t

Hydrogenation of crude anacardic acid gave a transparent crystalline product on recrystallization. When reacted with copper nitrate in the presence of pyridine it produced green crystals of a pyridine adduct of a dimeric copper(II) anacardate with the copper acetate structure. The X-ray single crystal structures of both anacardic acid and the copper complex were determined. Magnetic studies have confirmed strong antiferromagnetic coupling between copper(II) centre in the dimer. The exchange coupling constant was determined to be J = -324 cm(-1). The EPR spectra of the polycrystalline product a
