Five undescribed ent-labdane diterpenoid glucosides from Andrographis paniculata and their anti-inflammatory activity.

A phytochemical investigation on the leaves of medicinal plant Andrographis paniculata resulted in the isolation of five previously undescribed ent-labdane diterpenoid glucosides (1-5), along with ten known ones (6-15). Their chemical structures with absolute configurations were thoroughly elucidated on the basis of comprehensive spectroscopic analysis, single-crystal X-ray crystallographic experiment and acid hydrolysis. All the isolated compounds were measured for the production of nitric oxide levels in LPS-induced RAW 264.7 macrophages to assess their anti-inflammatory activity. Compounds

Unveiling the anti-giardial properties of Andrographis paniculata leaf extract through in vitro studies.

Giardiasis, caused by the parasite Giardia lamblia, is a prevalent and serious public health concern, particularly affecting children worldwide. The primary mode of transmission for the parasite is through contaminated food and water sources and often leads to the onset of diarrhoea in infected individuals. However, the present medications for Giardiasis treatment often come with numerous side effects, while the growing problem of drug resistance adds a significant complication. Therefore, there is an urgent need for alternative treatments. In this study, we explored the in-vitro potential of

Molecular cloning and functional characterization of multiple NADPH-cytochrome P450 reductases from Andrographis paniculata.

Andrographis paniculata (Burm.f.) Wall. ex Nees is widely used as medicinal herb in Southern and Southeastern Asia and andrographolide is its main medicinal constituent. Based on the structure of andrographolide, it has been proposed that cytochrome P450 enzymes play vital roles on its biosynthesis. NADPH:cytochrome P450 reductase (CPR) is the most important redox partner of multiple P450s. In this study, three CPRs were identified in the genomic data of A. paniculata (namely ApCPR1, ApCPR2, and ApCPR3), and their coding regions were cloned. They varied from 62% to 70% identities to each other

A theoretical framework for extrapolating Andrographis paniculata mechanisms to the novel CIRDD and IROD diabetes clusters.

The newly described CIRDD and IROD clusters of Type 2 Diabetes represent clinically meaningful phenotypes, yet no subtype-specific molecular or omics datasets currently exist to define their mechanisms. This study introduces a predictive extrapolation framework to infer how phytocompounds from Andrographis paniculata may interact with these subtypes by mapping established pathways of insulin resistance, β-cell dysfunction, and obesity-driven inflammation onto CIRDD- and IROD-relevant axes. From the broader T2DM network, ten hub proteins (INS, AKT1, TNF, IL6, MMP9, and others) were prioritized

A Diterpenoid of the Medicinal Plant Andrographis paniculata Targets Cutaneous TRPV3 Channel and Relieves Itch.

Transient receptor potential vanilloid subtype 3 (TRPV3) is an ion channel implicated in skin physiology and itch. TRPV3 inhibitors can present a novel strategy for combating debilitating itch conditions, and medicinal plants are a natural pool of such compounds. Here, we report the isolation of a TRPV3-inhibiting compound from Andrographis paniculata, a medicinal plant with anti-inflammatory properties whose bioactive components are poorly characterized in terms of molecular targets. Using 1H and 13C NMR and high-resolution mass spectrometry, the compound was identified as a labdane-type dite

Attenuation of Pseudomonas aeruginosa quorum sensing, virulence and biofilm formation by extracts of Andrographis paniculata.

Quorum-sensing (QS) is known to play an essential role in regulation of virulence factors and toxins during Pseudomonas aeruginosa infection which may frequently cause antibiotic resistance and hostile outcomes of inflammatory injury. Therefore, it is an urgent need to search for a novel agent with low risk of resistance development that can target QS and inflammatory damage prevention as well. Andrographis paniculata, a herbaceous plant under the family Acanthaceae, native to Asian countries and also cultivated in Scandinavia and some parts of Europe, has a strong traditional usage with its k

Assessment of cytotoxicity of leaf extracts of Andrographis paniculata and Aspilia africana on murine cells in vitro.

People, especially in rural environments and recently, those dissatisfied with conventional medicine use medicinal plants for their therapeutic effects. Two or more plants are usually combined for such use and these combinations cause interactions that may be safe but could also be detrimental to health. Used singly, Andrographis paniculata and Aspilia africana are considered safe plants with low toxicities but little is known about their safety in combined therapy. This study assessed the cytotoxic activities of the aqueous, methanol and chloroform extracts of the two plants in a 1:1 combinat

Andrographis paniculata: a critical appraisal of extraction, isolation and quantification of andrographolide and other active constituents.

Andrographispaniculata, referred to as 'Kalmegh' in ancient texts, is regarded as a nostrum of the modern world. Oral administration of the leaves of this plant is effective in the treatment of upper respiratory tract infections, liver toxicity and a variety of other ailments. The practices of adulteration in the original material of this drug have made it imperative to develop techniques to verify the authenticity of the products of this plant for quality assurance. Andrographolide, a bitter diterpenoid, is the major active principle present in the plant, and therefore its determination and q

Enhancing agronomic traits and bioactive compound accumulation in Thai Andrographis paniculata via EMS-induced variation.

BACKGROUND: Andrographis paniculata is a self-pollinating medicinal plant with limited natural genetic diversity, which restricts the development of improved cultivars with desirable agronomic traits and stable accumulation of key bioactive compounds, particularly andrographolide (AP1). To overcome this limitation, ethyl methanesulfonate (EMS) mutagenesis was applied to native Thai germplasm to induce phenotypic and phytochemical variation and to generate novel genetic resources for breeding improvement and gene function analysis. RESULTS: Optimal EMS treatment conditions (1.0% and 1.5% EMS fo

Evaluation of andrographolide from Andrographis paniculata against drug-resistant and H37Rv strains of Mycobacterium tuberculosis.

Tuberculosis (TB) remains a major global health concern, particularly due to the emergence of multidrug-resistant (MDR) Mycobacterium tuberculosis strains. While previous studies have suggested andrographolide as a potential antimycobacterial agent based on in silico predictions, limited empirical evidence exists on its direct efficacy against MDR-TB. This study systematically evaluates the antimycobacterial activity of andrographolide through the microbroth dilution method against M. tuberculosis H37Rv and three distinct MDR strains. The minimum inhibitory concentrations (MICs) were determine
