The newly described CIRDD and IROD clusters of Type 2 Diabetes represent clinically meaningful phenotypes, yet no subtype-specific molecular or omics datasets currently exist to define their mechanisms. This study introduces a predictive extrapolation framework to infer how phytocompounds from Andrographis paniculata may interact with these subtypes by mapping established pathways of insulin resistance, β-cell dysfunction, and obesity-driven inflammation onto CIRDD- and IROD-relevant axes. From the broader T2DM network, ten hub proteins (INS, AKT1, TNF, IL6, MMP9, and others) were prioritized

INTRODUCTION: Andrographis paniculata (AP) has been approved by the Thai government for the treatment of mild cases of COVID-19 patients. Increasing use of AP products requires quality control to ensure efficacy and safety. At present, there is no requirement for dissolution test of AP products in the Thai Herbal Pharmacopoeia (THP).
OBJECTIVE: This work aimed to examine the contents and dissolution profiles of active diterpenoids, andrographolide (AP1), 14-deoxy-11,12-didehydroandrographolide (AP3), neoandrographolide (AP4), and 14-deoxyandrographolide (AP6) in AP capsules available in Thai

Transient receptor potential vanilloid subtype 3 (TRPV3) is an ion channel implicated in skin physiology and itch. TRPV3 inhibitors can present a novel strategy for combating debilitating itch conditions, and medicinal plants are a natural pool of such compounds. Here, we report the isolation of a TRPV3-inhibiting compound from Andrographis paniculata, a medicinal plant with anti-inflammatory properties whose bioactive components are poorly characterized in terms of molecular targets. Using 1H and 13C NMR and high-resolution mass spectrometry, the compound was identified as a labdane-type dite

BACKGROUND: Isoniazid (INH) and Rifampicin (RIF) are the common drugs causing hepatitis in patients undergoing Antitubercular therapy (ATT). This often results in discontinuation of the therapy or change in the treatment regimen. Free radical injury by drug metabolites has been postulated as cause of liver damage. The herbal extracts of Phyllanthus niruri and Andrographis paniculata possess antioxidant activity.
METHODS: Hepatotoxicity was induced in Sprague Dawley rats by administering Isoniazid (100 mg/kg, po) and Rifampicin (100 mg/kg, po) combination for 14 days. Each group of rats were s

Quorum-sensing (QS) is known to play an essential role in regulation of virulence factors and toxins during Pseudomonas aeruginosa infection which may frequently cause antibiotic resistance and hostile outcomes of inflammatory injury. Therefore, it is an urgent need to search for a novel agent with low risk of resistance development that can target QS and inflammatory damage prevention as well. Andrographis paniculata, a herbaceous plant under the family Acanthaceae, native to Asian countries and also cultivated in Scandinavia and some parts of Europe, has a strong traditional usage with its k

Leaves of Andrographis paniculata and Orthosiphon stamineus were extracted with water, ethanol, methanol and chloroform to assess their potential as antibacterial and antioxidant agents. High performance liquid chromatography analysis showed that the methanolic extracts of A. paniculata and O. stamineus leaves gave the highest amounts of andrographolide and rosmarinic acid, respectively. These leaf extracts exhibited antimicrobial and antioxidant activities and, at the highest concentration tested (200 mg/mL), showed greater inhibitory effects against the Gram positive bacteria Bacillus cereus

People, especially in rural environments and recently, those dissatisfied with conventional medicine use medicinal plants for their therapeutic effects. Two or more plants are usually combined for such use and these combinations cause interactions that may be safe but could also be detrimental to health. Used singly, Andrographis paniculata and Aspilia africana are considered safe plants with low toxicities but little is known about their safety in combined therapy. This study assessed the cytotoxic activities of the aqueous, methanol and chloroform extracts of the two plants in a 1:1 combinat

Andrographispaniculata, referred to as 'Kalmegh' in ancient texts, is regarded as a nostrum of the modern world. Oral administration of the leaves of this plant is effective in the treatment of upper respiratory tract infections, liver toxicity and a variety of other ailments. The practices of adulteration in the original material of this drug have made it imperative to develop techniques to verify the authenticity of the products of this plant for quality assurance. Andrographolide, a bitter diterpenoid, is the major active principle present in the plant, and therefore its determination and q

Andrographis paniculata Nees is an annual erect herb with wide medicinal and pharmacological applications due to the presence of andrographolide and other active chemical constituents. The large-scale cultivation of the kalmegh is not in practice. The aim of this study was to establish sustainable production systems of A. paniculata cv CIM-Megha with the application of different bioinoculants and chemical fertilisers. A. paniculata herb and andrographolide yield in the dried leaves was found to be highest (218% and 61.3%, respectively) in treatment T3 (NPK+Bacillus sp.) compared with T1 (contr

BACKGROUND: Andrographis paniculata is a self-pollinating medicinal plant with limited natural genetic diversity, which restricts the development of improved cultivars with desirable agronomic traits and stable accumulation of key bioactive compounds, particularly andrographolide (AP1). To overcome this limitation, ethyl methanesulfonate (EMS) mutagenesis was applied to native Thai germplasm to induce phenotypic and phytochemical variation and to generate novel genetic resources for breeding improvement and gene function analysis. RESULTS: Optimal EMS treatment conditions (1.0% and 1.5% EMS fo

Tuberculosis (TB) remains a major global health concern, particularly due to the emergence of multidrug-resistant (MDR) Mycobacterium tuberculosis strains. While previous studies have suggested andrographolide as a potential antimycobacterial agent based on in silico predictions, limited empirical evidence exists on its direct efficacy against MDR-TB. This study systematically evaluates the antimycobacterial activity of andrographolide through the microbroth dilution method against M. tuberculosis H37Rv and three distinct MDR strains. The minimum inhibitory concentrations (MICs) were determine

Andrographis paniculata (A. paniculata) showed an anti-depressive effect in rodent models. Zebrafish has recently emerged as a worthy complementary translational model for antidepressant drug discovery study. This study investigates the anti-depressive effect of A. paniculata extract and andrographolide in the chronic unpredictable stress (CUS)- zebrafish model. Four groups of zebrafish (n = 10/group), i.e. control, CUS (stressed, untreated), CUS + A. paniculata (100 mg/L) and CUS + fluoxetine (0.01 mg/L) were assessed in open-field and social interaction tests, 24 h after treatment. After ext

OBJECTIVES: The use of standard antimalarial drugs, such as dihydroartemisinin-piperaquine (DHP) for the treatment of malaria during pregnancy is limited due to the risk of teratogenicity. The alternative is therefore required although few exist. Here we show a phytopharmaceutical drug derived from Andrographis paniculata (AS201-01), which is effective as herbal antimalarial both in vitro and in vivo and may be a suitable alternative when used in complementary treatment with DHP.
METHODS: Plasmodium berghei infected pregnant BALB/c mice were divided into four groups: G1 (negative control), G2

Background/Objectives: Andrographis paniculata is listed in the Thai National List of Essential Medicines. The Thai Herbal Pharmacopoeia specifies the required contents of total lactones and andrographolide for the quality control of the aerial parts of A. paniculata. Effective pre-harvest quality control throughout the cultivation period is essential to ensure compliance with these quality standards. In this study, we aimed to develop a portable smartphone-based colorimetric method for the determination of total lactone content, thereby facilitating in-field quality control of A. paniculata r

Andrographis paniculata, commonly known as kalmegh is a highly valued medicinal plant. Pot-grown plants were subjected to water stress at vegetative, flowering, and fruiting stage by withholding water supply, followed by rewatering to facilitate recovery. Plants at the flowering and fruiting stage were particularly sensitive to drought stress compared to those at the vegetative stage. The plants were analysed for four diterpenoid compounds, namely andrographolide, 14-deoxyandrographolide, neoandrographolide, and andrograpanin. In plants subjected to stress at the vegetative and flowering stage

Neurodegenerative diseases (NDs) affect millions of people worldwide, and to date, Alzheimer's and Parkinson's diseases are the most common NDs. Of the many risk factors for neurodegeneration, the aging process has the most significant impact, to the extent that it is tempting to consider neurodegenerative disease as a manifestation of accelerated aging. However, genetic and environmental factors determine the course of neurodegenerative disease progression. It has been proposed that environmental stimuli influence neuroplasticity. Some clinical studies have shown that healthy lifestyles and t
