The effects of cold temperature on the development, microbiome, and transcriptome of the sea anemone Nematostella vectensis.

Thermal conditions impact essentially all aspects of the physiology for ectotherms. While the effects of high temperatures have been widely studied, cold temperature effects on aquatic invertebrates and their microbial communities have been poorly characterized. To determine the diverse effects of exposure to cold temperatures, we assessed acute and long-term impacts of ecologically relevant low temperatures on the development, microbiome, and gene expression of the sea anemone Nematostella vectensis. Two hours post fertilization, embryos were exposed to temperatures from 4°C to 35°C and devel

Differential metabolic responses in bold and shy sea anemones during a simulated heatwave.

As climate change-induced heatwaves become more common, phenotypic plasticity at multiple levels is a key mitigation strategy by which organisms can optimise selective outcomes. In ectotherms, changes to both metabolism and behaviour can help alleviate thermal stress. Nonetheless, no study in any ectotherm has yet empirically investigated how changing temperatures affect among-individual differences in the associations between these traits. Using the beadlet anemone (Actinia equina), an intertidal species from a thermally heterogeneous environment, we investigated how individual metabolic rate

Severe Toxic Skin Reaction Caused by a Common Anemone and Identification of the Culprit Organism.

In a marine envenomation, identification of the culprit organism can be difficult. In this case report, we present our method to identify snakelocks anemone (Anemonia viridis or formerly Anemonia sulcata) as the culprit of a severe toxic skin reaction. A. viridis is one of the most common anemones of the Mediterranean Sea and the North Atlantic Ocean. It lives at a depth of up to 10 m. It is a member of the phylum Cnidaria, which includes jellyfish, anemones, hydroids, and corals. They have toxic organelles called cnidocysts that have the capacity to inject venom with microscopic harpoon-like

Mercury distribution, methylation and volatilization in microcosms with and without the sea anemone Bunodosoma caissarum.

Mercury (Hg) has a complex biogeochemical cycle in aquatic environments. Its most toxic form, methylmercury (MeHg), is produced by microorganisms. This study investigated how the sea anemone Bunodosoma caissarum affects Hg distribution, methylation and volatilization in laboratory model systems. (203)Hg was added to microcosms and its distribution in seawater, specimens and air was periodically measured by gamma spectrometry. MeHg was measured by liquid scintillation. After the uptake period, specimens had a bioconcentration factor of 70 and in microcosms with and without B. caissarum, respect

Outcomes of infections of sea anemone Aiptasia pallida with Vibrio spp. pathogenic to corals.

Incidents of coral disease are on the rise. However, in the absence of a surrogate animal host, understanding of the interactions between coral pathogens and their hosts remains relatively limited, compared to other pathosystems of similar global importance. A tropical sea anemone, Aiptasia pallida, has been investigated as a surrogate model to study certain aspects of coral biology. Therefore, to test whether the utility of this surrogate model can be extended to study coral diseases, in the present study, we tested its susceptibility to common coral pathogens (Vibrio coralliilyticus and Vibr

EPR study of the sea anemone cytolysin, equinatoxin II, cytotoxicity on V-79 cells.

Electron paramagnetic resonance (EPR) was used to study the effect of equinatoxin II (EqT II), a cytolytic protein isolated from the sea anemone Actinia equina L., on membrane fluidity and cell metabolism of V-79 cells; the reduction of the spin probe incorporated into the cell membranes as well as the oxygen consumption in the cell suspension were measured. The results were compared with the results obtained by the cell viability study. Under the influence of EqT II (less than 37.5 micrograms/10(6) cells) no significant changes in cell membrane fluidity were observed, while reduction kinetics

The probable origin of an anemone cell tumor: metastatic transitional cell carcinoma producing HCG.

A neoplasm of unknown origin in cervical and axillary lymph nodes was diagnosed as anemone cell tumor by ultrastructural examination. Three years after the initial diagnosis of anemone cell tumor, a high-grade transitional cell carcinoma of the bladder was discovered. The results of immunoperoxidase staining of the cervical lymph node, axillary lymph node, and bladder tumors for keratin, carcinoembryonic antigen, and human chorionic gonadotropin (HCG) strongly suggest that the anemone cell tumors in this case represent metastases of bladder carcinoma cells capable of producing HCG.

Potential-dependent effects of sea anemone toxins and scorpion venom on crayfish giant axon.

Effects of two kinds of sea anemone toxin (Parasicyonis actinostoloides and Anemonia sulcata) and scorpion venom (Leiurus quinquestriatus) on crayfish giant axons were examined electrophysiologically. All toxins acted on the axon in a similar manner to prolong the falling phase of the action potential. In all cases the development of toxicity was reduced when the nerve membrane was depolarized by a current injection. However, the ranges of membrane potential where the significant reduction in toxicity took place were different for each toxin. The action of Parasicyonis toxin was also suppresse

Oxylipin-mediated metabolic signatures of symbiosis homeostasis and thermal stress in a model sea anemone.

Oxylipins are oxygenated products of fatty acids proposed to exert a regulatory role in cnidarian-dinoflagellate symbiosis; however, this has not been investigated in detail. We integrated physiological measurements and molecular phenotyping with comparative transcriptome mining to examine how the symbiotic cnidarian model, the sea anemone Aiptasia (i.e. Exaiptasia diaphana), and its dinoflagellate symbiont Breviolum minutum respond to symbiosis and thermal stress. We performed lipidomics in combination with the quantification of oxylipins, including octadecanoids, eicosanoids, and docosanoids

Heterotrophy and symbiosis affect energy reserves for pedal lacerates in the sea anemone Exaiptasia diaphana.

Nutrient exchange between corals and their dinoflagellate symbionts is the foundation of the stable symbiosis that underpins coral reef ecosystem success. The cnidarian-dinoflagellate holobiont engages in both autotrophy (photosynthates supplied by the symbiont) and heterotrophy (feeding by the host on microscopic organisms and particulate matter) to meet their nutritional demands. While considerable research has been devoted to understanding nutrient dynamics in adult corals and other symbiotic cnidarians, less is known about how the combination of heterotrophy and autotrophy influences nutri

Purification and characterization of gigantoxin-4, a new actinoporin from the sea anemone Stichodactyla gigantea.

A new Cytolysin, termed as Gigantoxin-4, was isolated from the sea anemone Stichodactyla gigantea and found to be highly homologous with Cytolysin-3 (HMg III) from Heteractis magnifica, RTX-A from Radianthus macrodactylus, and Sticholysin-1 (St I) and Sticholysin-2 (St II) from Stichodactyla helianthus (homology 82%, 86%, 82% and 86% respectively). Its 20 N-terminal residues were identified and the full-length cDNA sequence was obtained by reverse transcription-polymerase chain reaction (RT-PCR). Multiple sequence alignments with other Cytolysins of the actinoporin family clearly indicated tha

When positive selection of neurotoxin genes is missing. The riddle of the sea anemone Nematostella vectensis.

Rapid evolution driven by positive Darwinian selection appears in toxins of vipers, scorpions, and marine snails. Although the vast phylogenetic distances between these animals suggest that this phenomenon is common, the recent release of the genome of Nematostella vectensis (Starlet anemone) as a collection of contigs portrays another extreme. Besides potassium channel toxin domains, which resemble potassium channel blockers, embedded in various genes, only one gene family encoding for sodium channel neurotoxins has been found, and the putative mature product of 10 family members is identical

A new toxin from the sea anemone Condylactis gigantea with effect on sodium channel inactivation.

A new peptide toxin exhibiting a molecular weight of 5043Da (av.) and comprising 47 amino acid residues was isolated from the sea anemone Condylactis gigantea. Purification of the peptide was achieved by a multistep chromatographic procedure monitoring its strong paralytic activity on crustacea (LD(50) approx. 1microg/kg). Complete sequence analysis of the toxic peptide revealed the isolation of a new member of type I sea anemone sodium channel toxins containing the typical pattern of the six cysteine residues. From 11kg of wet starting material, approximately 1g of the peptide toxin was isola

Biochemical and physiological analyses of a hemolytic toxin isolated from a sea anemone Actineria villosa.

A species of venomous sea anemone Actineria villosa was recently found inhabiting the coastal areas of Okinawa, Japan. This marine animal produces various proteinous toxins, so that a local health organization was called for medical treatment for those who had accidental contact with this animal. In this study we analyzed the biochemical and physiological properties of hemolytic protein from A. villosa. The toxin purified from the tentacles of the animals was found to be a protein with a molecular weight of approximately 19 kDa. We named this newly found hemolytic toxin of A. villosa, Avt-I. I

Comparison of sea anemone and scorpion toxins binding to Kv1 channels: an example of convergent evolution.

Comparison of data from functional mapping carried out on scorpion and sea anemones toxins blocking currents through voltage-gated potassium channels revealed that, despite their different 3D structures, the binding cores of these toxins displayed some similarities. Further molecular modeling studies suggested that these similarities reflect the use by these toxins of a common binding mode to exert their blocking function. Therefore, scorpion and sea anemone toxins offer an example of mechanistic convergent evolution.

Kalicludines and kaliseptine. Two different classes of sea anemone toxins for voltage sensitive K+ channels.

New peptides have been isolated from the sea anemone Anemonia sulcata which inhibit competitively the binding of 125I-dendrotoxin I (a classical ligand for K+ channel) to rat brain membranes and behave as blockers of voltage-sensitive K+ channels. Sea anemone kalicludines are 58-59-amino acid peptides cross-linked with three disulfide bridges. They are structurally homologous both to dendrotoxins which are snake venom toxins and to the basic pancreatic trypsin inhibitor (Kunitz inhibitor) and have the unique property of expressing both the function of dendrotoxins in blocking voltage-sensitive

PHOTOBIOLOGY OF THE SYMBIOTIC SEA ANEMONE, ANTHOPLEURA ELEGANTISSIMA: DEFENSES AGAINST PHOTODYNAMIC EFFECTS, AND SEASONAL PHOTOACCLIMATIZATION.

The sea anemone Anthopleura elegantissima, which contains photosynthetic symbionts (zooxanthellae), responds both biochemically and behaviorally to the combined environmental stresses of exposure to sunlight and photosynthetically generated hyperbaric O2. Activities of the enzymes superoxide dismutase (SOD) and catalase, which act in concert as defenses against oxygen toxicity, parallel the distribution of chlorophyll. A. elegantissima shows a finely controlled contraction behavior which shades the zooxanthellae and reduces O2 production, but which leaves the body column tissues directly expos

An abundant membrane protein in the NCAM superfamily is required for epithelial organization in sea anemone embryos.

Assembly of cells into epithelial layers marks the early steps of tissue organization and embryonic development in most animals. Although several conserved proteins are known to be essential for epithelialization in bilaterians, it is unclear whether these are sufficient to drive epithelial organization in all metazoans. Using proteomics and knockdown approaches in embryos of the early-branching sea anemone Nematostella vectensis, we identified the Neural Cell Adhesion Molecule (NCAM) NCAM2 to be essential for organization of the primary epithelial layer. NCAM2 belongs to the Immunoglobulin su

Structural and functional characterisation of a novel peptide from the Australian sea anemone Actinia tenebrosa.

Sea anemone venoms have long been recognised as a rich source of peptides with interesting pharmacological and structural properties. Our recent transcriptomic studies of the Australian sea anemone Actinia tenebrosa have identified a novel 13-residue peptide, U-AITx-Ate1. U-AITx-Ate1 contains a single disulfide bridge and bears no significant homology to previously reported amino acid sequences of peptides from sea anemones or other species. We have produced U-AITx-Ate1 using solid-phase peptide synthesis, followed by oxidative folding and purification of the folded peptide using reversed-phas

Enzymatic and structural characterization of a basic phospholipase A(2) from the sea anemone Condylactis gigantea.

This work aimed at the isolation and structural/functional characterization of a phospholipase A(2) (CgPLA(2)) from the extract of the anemone Condylactis gigantea. CgPLA(2) was isolated with a high purity level through three chromatographic steps, showing pI 8.6 and molecular weights of 14,500 and 29,000 for the monomer and dimer, respectively. CgPLA(2) showed a high catalytic activity upon fluorescent phospholipids inducing no direct hemolytic activity. This enzyme, which is Ca(2+)-dependent, showed a lower stability against temperature and pH variations when compared with snake venom enzyme
