Reduction of exposure to laboratory animal allergens in a research laboratory.

OBJECTIVES: The purpose of this study was to determine exposure levels in the laboratory during different tasks and evaluate the effectiveness of safety equipment used to reduce personal exposure. METHODS: Personal and stationary air samples were collected during different tasks in a laboratory animal facility in which several allergen reduction strategies had been implemented. Mouse urinary allergen concentrations were measured using a polyclonal sandwich enzyme-linked immunosorbent assay. Sera from the personnel (n = 29) were analysed every 6 months for the presence of specific antibodies a

Laboratory animal allergy: an update.

Allergic reactions are among the most common conditions affecting the health of workers involved in the care and use of research animals. Between 11 and 44% of the individuals working with laboratory animals report work-related allergic symptoms. Of those who become symptomatic, 4 to 22% may eventually develop occupational asthma that can persist even after exposure ceases. Allergic symptoms consist of rashes where animals are in contact with the skin, nasal congestion and sneezing, itchy eyes, and asthma (cough, wheezing, and chest tightness). The generation of immunoglobulin E (IgE) antibodi

Allergy in laboratory animal workers.

179 people working with small laboratory animals were studied by questionnaire, lung-function tests, skin tests with common allergen extracts and extracts of the serum and urine of five animal species, and serology. 49 people had symptoms related to animal contact, most commonly rhinitis but also asthma and skin rashes. Skin reactivity to animal extracts was related to symptoms, particularly asthma, but many people with rhinitis only had negative skin tests. Positive skin tests without symptoms were rare. 3 people had additional late skin reactions. Atopic individuals were no more likely than

[Laboratory animal; allergy; asthma].

Laboratory animal allergy (LAA) may develop when susceptible persons are exposed to allergens produced by laboratory animals. LAA is associated with exposure to urine, fur, and salivae of rats, guinea pigs, dogs and rabbits. Approximately 30% of persons who are exposed to laboratory animals may develop LAA and some will also develop asthma. LAA is most likely to occur in persons with previously known allergies, especially to domestic pets. The majority of LAA sufferers experience symptoms within six months their first exposure to laboratory animals; almost all develop symptoms within three yea

Animal allergen sensitization in veterinarians and laboratory animal workers.

BACKGROUND: Animals secrete allergens into the environment and exposure to these in the workplace may cause sensitization. AIMS: To identify the frequency of animal allergen sensitization and symptoms in animal workers. METHODS: Using skin prick tests (SPT), we assessed sensitization to 15 mammal and bird allergens in animal workers and controls. We also recorded symptoms and pulmonary function tests. RESULTS: There were 100 animal workers and 50 controls included in the study. Thirty-six per cent of animal workers and 10% of controls had positive SPT (P < 0.001, odds ratio [OR] = 5.1, 95%

Laboratory studies of a lymphocytic choriomeningitis virus outbreak in man and laboratory animals.

Investigation of an outbreak of prolonged febrile illness in medical center personnel at the University of Rochester School of Medicine and Dentistry revealed lymphocytic choriomeningitis (LCM) virus to be the causative agent. Syrian or golden hamsters (Mesocricetus auratus) were found to be the only animals involved in maintaining the virus and were the source of human infections. Isolations of LCM virus were made from autopsy specimens of 13 of 46 (28%) golden hamsters. Virus isolations were made from 22 of 28 (79%) frozen specimens of 11 tumor lines transplanted repeatedly in golden hamster

Laboratory animals and respiratory allergies: the prevalence of allergies among laboratory animal workers and the need for prophylaxis.

OBJECTIVE: Subjects exposed to laboratory animals are at a heightened risk of developing respiratory and allergic diseases. These diseases can be prevented by simple measures such as the use of personal protective equipment. We report here the primary findings of the Laboratory Animals and Respiratory Allergies Study regarding the prevalence of allergic diseases among laboratory animal workers, the routine use of preventive measures in laboratories and animal facilities, and the need for prevention programs. METHODS: Animal handlers and non-animal handlers from 2 Brazilian universities (Unive

CIRS-LAS - a novel approach to increase transparency in laboratory animal science for improving animal welfare by reducing laboratory animal distress.

The 3Rs principle is highly topical in animal-based research. These include, above all, new scientific methods for conducting experiments without an animal model, by using non-animal models (Replace), reducing the number of laboratory animals (Reduction) or taking measures to keep the stress on the laboratory animal as low as possible (Refinement). Despite numerous modern alternative approaches, the complete replacement of animal experiments is not yet possible. The exchange in the team about the daily work with laboratory animals, about open questions and problems, contributes to a reflection

Improving the Reliability of Tinnitus Screening in Laboratory Animals.

Behavioral screening remains a contentious issue for animal studies of tinnitus. Most paradigms base a positive tinnitus test on an animal's natural tendency to respond to the "sound" of tinnitus as if it were an actual sound. As a result, animals with tinnitus are expected to display sound-conditioned behaviors when no sound is present or to miss gaps in background sounds because tinnitus "fills in the gap." Reliable confirmation of the behavioral indications of tinnitus can be problematic because the reinforcement contingencies of conventional discrimination tasks break down an animal's tend

Laboratory diagnosis of Taenia asiatica in humans and animals.

Taenia asiatica is a recently described species known to cause intestinal teniasis in humans and cysticercosis in animals. This species has close morphological resemblance to Taenia saginata and has a life cycle resembling Taenia solium, hence has been posing diagnostic dilemma and had been the reason for its comparatively late discovery. Recent diagnostic tools such as serological and molecular techniques have thrown light on its exact prevalence in the endemic countries. Hence introduction of utilization of these techniques in addition to the routine morphological analysis would be helpful i

Mucus thickness in the gastrointestinal tract of laboratory animals.

OBJECTIVES: The objective of this study was to systematically assess the mucus thickness in the gastrointestinal tract of laboratory animals commonly used in preclinical studies. METHODS: Mucus thickness was studied post-mortem in the rat, rabbit and pig, using cryosections stained by the modified periodic acid Schiff/Alcian blue method. KEY FINDINGS: The mucus thickness in the fundus region of the stomach was higher in the pig (190.7 ± 80.7 µm) than in the rabbit (155.1 ± 85.8 µm) and the rat (31.3 ± 11.4 µm). However, along the small intestine (ileum), mucus was thicker in the rabbit (147.

An aerosolized Brucella spp. challenge model for laboratory animals.

To characterize the optimal aerosol dosage of Brucella abortus strain 2308 (S2308) and B. melitensis (S16M) in a laboratory animal model of brucellosis, dosages of 10(3)-10(10) colony forming units (CFU) were nebulized to mice. Although tissue weights were minimally influenced, total CFU per tissues increased beginning at 10(6)-10(7) CFU dosages, with 10(9) CFU appearing to be an optimal dosage for S16M or S2308 aerosol delivery. At 12 weeks after vaccination with 10(7) CFU of B. abortus strain RB51 (SRB51) or saline (control), mice were challenged intraperitoneally (i.p.) (6.4 x 10(4) CFU) or

Some pregnancy-related effects of artemether in laboratory animals.

Artemether, highly effective in multi-drug-resistant malaria is not routinely available for use in pregnancy due to the lack of adequate research data in animals and man. This study was therefore aimed at investigating some pregnancy-related effects of artemether. Artemether (1.5, 7.5 and 15 mg/kg i.p. daily for 7 days) did not produce changes in rat oestrous cycle. The drug did not prevent or prolong the rate of conception or parturition, cause pre-term delivery and affect litter size. Birth weight and growth rate of pups from artemether-pretreated dams were within the normal range. Artemethe

Laboratory techniques for rabies diagnosis in animals at QSMI.

The Queen Saovabha Memorial Institute (QSMI), Thai Red Cross Society was founded in 1922 to carry out the production of the nervous tissue vaccines, post-exposure prophylaxis (PEP) treatment, research, and laboratory diagnosis. The diagnostic laboratory had replaced the Seller's staining with direct fluorescent test (DFA) as standard technique in 1987 and the mouse inoculation technique (MIT) had been employed as the confirmatory test to ensure the result of the diagnosis. Other techniques conducted at our facility such as tissue culture infection and polymerase chain reaction techniques are l

Halothane anesthesia and aqueous humor dynamics in laboratory animals.

Halothane anesthesia decreased intraocular pressure (IOP) by approximately 4 mm Hg in both normal rabbits and monkeys. The administration of nitrous oxide or 100% oxygen had no effect on IOP. Although PO2 and PCO2 increased following halothane administration in rabbits, similar blood changes occurred without IOP reduction in control rabbits receiving 100% oxygen. Outflow facility in both species as measured by tonography remained unchanged by halothane administration. Aqueous humor flow as estimated by tonography decreased by about 39% in rabbits and 31% in monkeys. Posterior chamber aqueous h

Evaluation of erectile function in laboratory animals: an overview.

INTRODUCTION: Erectile dysfunction (ED) is a common male sexual disorder that diminishes quality of life significantly. Despite extensive research, little is known about the complex physiological mechanisms of ED, in part owing to disparities between animal and human physiology. Comprehensive and objective assessment methods for animal models are crucial to bridge this translational gap. OBJECTIVES: Various experimental approaches for evaluating erectile function in rodent models are explored. METHODS: A search of the medical literature was carried out in PubMed and Medline, focusing on orig
