The phospholipid- and Ca(2+)-binding protein annexin A5 (ANXA5) is the most abundant membrane-associated protein of ~P23 mouse vestibular hair bundles, the inner ear's sensory organelle. Using quantitative mass spectrometry, we estimated that ANXA5 accounts for ~15,000 copies per stereocilium, or ~2% of the total protein there. Although seven other annexin genes are expressed in mouse utricles, mass spectrometry showed that none were present at levels near ANXA5 in bundles and none were upregulated in stereocilia of Anxa5(-/-) mice. Annexins have been proposed to mediate Ca(2+)-dependent repai

Thrombophilia and impaired placental vasculature are a major cause of adverse pregnancy outcome. In 2007, a new hereditary factor for obstetric complications and recurrent pregnancy loss (RPL) was identified as a sequence variation in the core promoter of the annexin A5 gene, ANXA5, called the M2 haplotype. M2 carriership has been demonstrated in couples with recurrent miscarriage and its origin is embryonic rather than specifically maternal, confirmed by subsequent papers. The M2 haplotype is the first report of a hereditary factor related to pregnancy pathology caused by embryonic-induced an

After insemination, a subpopulation of sperm reaches the oviducts and binds to isthmic epithelial cells to form a "sperm reservoir". Our objective was to explore the role of annexin A5 (ANXA5), a protein that binds with high affinity to phosphatidylserine (PS), in the formation of the sperm reservoir in pigs. Phosphatidylserine was detected on the head of approximately 10 % of boar sperm at ejaculation. Porcine ANXA5 was immunodetected with a strong signal on luminal cilia in the isthmus and in derived isthmic epithelial spheroids (IES). Exogenous PS between 0.01 and 0.1 µg/mL and recombinant

The M2 haplotype of the annexin A5 gene is a well-recognized predisposition factor for recurrent spontaneous abortion (RSA). A recent publication by Nagirnaja et al. (2015) in PLoS One discusses the risk role of the M2 haplotype for RSA in cases compared with controls of North European extraction and arrives at a negative result. As a number of previous and fairly recent studies have supported the proposed involvement of the M2 haplotype in the cause of idiopathic RSA, this commentary aims to highlight problematic issues in the above publication. It is the opinion of the authors that the study

In situ generation of autologous tumor vaccines from dying tumor cells offers a promising route for personalized cancer immunotherapy, yet is often limited by phosphatidylserine (PS)-mediated immunosuppressive clearance that can restrict downstream immune activation and favor M2-like macrophage polarization. Here, we report a sequentially orchestrated nanovaccine (NT@ANX5-P) that couples photodynamic therapy (PDT)-induced immunogenic tumor-cell damage with on-demand PS masking to enhance in situ tumor vaccination. The nanovaccine consists of a photosensitizer core cloaked with genetically engi

Phosphatidylserine (PtdSer) is exposed on the external leaflet of the plasma membrane during apoptosis. The protein annexin A5 (anxA5) shows high affinity for PtdSer. When anxA5 binds to the PtdSer-expressing membranes during apoptosis, it crystallizes as an extended two-dimensional network and activates thereby a novel portal of cell entry that results in the internalization of the PtdSer-expressing membrane patches. This novel pathway of cell entry is potentially involved in the regulation of the surface expression of membrane receptors. In this study we report the regulation of surface expr

BACKGROUND AND OBJECTIVES: Preeclampsia (PE) is a leading cause of maternal and perinatal morbidity and mortality, yet effective targeted therapies remain limited. This study aimed to investigate the therapeutic potential of annexin A5 (ANX A5) in PE and to elucidate the underlying mechanisms based on metabolomic profiling.
METHODS: A PE-like mouse model was established by intraperitoneal injection of lipopolysaccharide (LPS, 20 μg/kg/day) from gestational day (GD) 7.5 to 17.5, followed by intravenous administration of ANX A5 (50 μg/kg/day) in the treatment group. In vitro, LPS-stimulated HTR

Annexin A5 is a placental anti-coagulant protein that contains four nucleotide substitutions (M2 haplotype) in its promoter. This haplotype is a risk factor for recurrent spontaneous abortion (RSA). The influence of the M2 haplotype in the gestational timing of spontaneous abortions, paternal risk and relationships with known risk factors were investigated. European couples (n = 500) who had experienced three or more consecutive spontaneous abortions, and two fertile control groups, were selected for this study. The allele frequency of M2 was significantly higher among patients who had experie

Polycystin-1 is the gene product of PKD1, the first gene identified to be causative for the condition of autosomal dominant polycystic kidney disease (ADPKD). Mutations in PKD1 are responsible for the majority of ADPKD cases worldwide. Polycystin-1 is a protein of the transient receptor potential channels superfamily, with 11 transmembrane spans and an extracellular N-terminal region of approximately 3109 amino acid residues, harboring multiple putative ligand binding domains. We demonstrate here that annexin A5 (ANXA5), a Ca(2+) and phospholipid binding protein, interacts with the N-terminal

OBJECTIVE: Twin pregnancies are accompanied by various perinatal complications. The study of placentae is important for understanding the underlying pathogenesis and pathologies. Annexin A5 (ANXA5) is an anti-thrombotic agent, which is expressed by the placental trophoblasts. Apelin is a novel adipokine with a significant role in angiogenesis of the placenta. The aim of the present study was to compare the gross, histopathological findings and immunohistochemical expression of ANXA5 and apelin of twin placentae with those of matched controls. These findings were also correlated with the clinic

BACKGROUND & AIM: M2 macrophages are key drivers of immune-mediated liver injury. The aim of this study was to investigate the mechanisms by which Annexin A5 regulates ferroptosis in M2 macrophages and its protective effect against immune liver injury.
METHODS: The colocalization of ferroptosis in M2 macrophages from autoimmune hepatitis (AIH) patients and mice with ConA-induced immune liver injury was analyzed via laser confocal microscopy. The effects of ferritinophagy and autophagosomal-lysosomal fusion on the regulation of ferroptosis in M2 macrophages by Annexin A5 were investigated thro

Expression of phosphatidylserine (PS) on the membrane surface of red blood cells and circulating microparticles (MP) plays an important role in etiology of the hypercoagulable state of sickle cell disease (SCD), as well as in the reduced red cell life span and adhesive interactions between red cells and endothelium. Annexin A5, an intracellular protein abundantly present in endothelial cells and platelets, exhibits high affinity for PS and has been shown to inhibit several of these PS-mediated pathophysiological processes. We determined plasma annexin A5 levels and MP-associated procoagulant a

Annexin A5 (ANXA5) is a small calcium-dependent protein that binds specifically to negatively charged phosphatidylserine as a marker of apoptosis. Previous studies have shown that ANXA5 expression is elevated in hypertensive patients and is closely related to left ventricular systolic function in hypertensive patients, but its specific mechanism of action has not been clarified. GEO database analysis showed that ANXA5 expression was significantly upregulated in hypertensive myocardial hypertrophy. The expression of ANXA5 protein and mRNA was overexpressed, and knockdown of ANXA5 can effectivel

Aims: Annexin A5 (ANXA5) exhibited potent antithrombotic, antiapoptotic, and anti-inflammatory properties in a previous study. The role of ANXA5 in traumatic brain injury (TBI)-induced intestinal injury is not fully known. Main methods: Recombinant human ANXA5 (50 µg/kg) or vehicle (PBS) was administered to mice via the tail vein 30 min after TBI. Mouse intestine tissue was gathered for hematoxylin and eosin staining 0.5 d, 1 d, 2 d, and 7 d after modeling. Intestinal Western blotting, immunofluorescence, TdT-mediated dUTP nick-end labeling staining, and enzyme-linked immunosorbent assays were

Numerous biochemical studies have pointed to an essential role of annexin A5 (AnxA5), annexin A6 (AnxA6), and collagen X in matrix vesicle-mediated biomineralization during endochondral ossification and in osteoarthritis. By binding to the extracellular matrix protein collagen X and matrix vesicles, annexins were proposed to anchor matrix vesicles in the extracellular space of hypertrophic chondrocytes to initiate the calcification of cartilage. However, mineralization appears to be normal in mice lacking AnxA5 and AnxA6, whereas collagen X-deficient mice show only subtle alterations in the gr

Cancer immunotherapies, particularly therapeutic vaccination, do not typically generate robust anti-tumour immune responses. Here, we show that the intratumoral burst release of the protein annexin A5 from intravenously injected hollow mesoporous nanoparticles made of diselenide-bridged organosilica generates robust anti-tumour immunity by exploiting the capacity of primary tumours to act as antigen depots. Annexin A5 blocks immunosuppressive apoptosis and promotes immunostimulatory secondary necrosis by binding to the phagocytic marker phosphatidylserine on dying tumour cells. In mice bearing

The accumulation of senescent cells and their secretion of senescence-associated secretory phenotype (SASP) play important roles in the pathogenesis of idiopathic pulmonary fibrosis (IPF). Small molecules, known as senolytics or senomorphics, have been effective in targeting senescent cells. Although senolytic drugs have been well-studied in pulmonary fibrosis, senomorphics with defined protein targets and potential applications are rarely investigated. In this study, we identified a widely sourced natural product, caffeic acid (CA), to act as a potent senomorphic that effectively inhibits the

Autoantibody profiles in HIV-associated pulmonary arterial hypertension (HIV-PAH) remain poorly characterized. Using a validated proteomic pipeline (two-dimensional immunoblot, mass spectrometry, ELISA), we compared 20 HIV-PAH patients with 20 HIV controls without PAH. Anti-annexin A5 (AnxA5) autoantibodies were present in 55% of HIV-PAH patients versus 0% of controls (P < .001) and anti-14-3-3ε in 35% versus 5% (P = .04), despite comparable HIV duration, CD4 counts, and virological control. Both remained associated with PAH after adjustment for coinfections. These autoantibodies mirror those

The mechanism by which GnRH stimulates annexin A5 expression was examined with LbetaT2 gonadotrope cells. Continuous stimulation with GnRH analog (GnRHa, Des-Gly10 [Pro9]-GnRH ethylamide) transiently elevated LHbeta mRNA expression while maintaining annexin A5 mRNA at high levels for 24 h. GnRH antagonist blocked the effect of GnRHa on annexin A5. While 12-O-tetradecanoyl-phorbol-13 acetate, a protein kinase C activator, increased the expression of annexin A5 mRNA, bisindolylmaleimide, an inhibitor of protein kinase C, suppressed GnRHa-stimulated expression of annexin A5 and LHbeta mRNA. GnRHa
