Identification of immunoreactive proteins during acute human giardiasis.

The protozoan Giardia lamblia is a major cause of parasite-induced diarrhea in humans. Humoral immunity has been shown to be important for clearance of the infection, but only a few antigens have been identified. In this study, we focused on the immunoreactivity of nonvariant antigens. Serum samples from 93 patients with acute giardiasis who were infected during a waterborne outbreak in a nonendemic country were screened on 1-dimensional Western blots. Representative serum samples that reacted strongly with proteins of different molecular weights were further analyzed on 2-dimensional Western

TNF-alpha reverses the disease-exacerbating effect of subcutaneous immunization against murine cutaneous leishmaniasis.

Earlier studies have demonstrated that mice injected subcutaneously or intramuscularly with leishmanial antigens develop significantly exacerbated disease compared with unimmunized controls when challenged with the cutaneous protozoan parasites Leishmania major. We report here that this disease enhancement can be prevented, and protective immunity induced, by the incorporation of recombinant tumour necrosis factor (TNF-alpha) in the immunizing inoculum. This effect of TNF-alpha is dose-dependent and is not evident when TNF-alpha and the antigens are injected into separate sites. Furthermore, T

Review on the identification and role of Toxoplasma gondii antigenic epitopes.

Toxoplasma gondii is an obligate intracellular protozoan parasite with a broad range of hosts, and it causes severe toxoplasmasis in both humans and animals. It is well known that the progression and severity of a disease depend on the immunological status of the host. Immunological studies on antigens indicate that antigens do not exert their functions through the entire protein molecule, but instead, specific epitopes are responsible for the immune response. Protein antigens not only contain epitope structures used by B, T, cytotoxic T lymphocyte (CTL), and NK cells to mediate immunological

Effectiveness of manual physical therapy in the treatment of cervical radiculopathy: a systematic review.

BACKGROUND: In sub-Saharan Africa poly-parasite infections are frequently observed in children, and with poly-parasitism modulating immune mechanisms, mediated by cytokines and chemokines, are required to prevent overwhelming inflammation and host tissue damage. We analyzed in children co-infected with helminthes and protozoan parasites their cellular production of regulatory and pro-inflammatory cytokines and chemokines in response to parasite antigens and allergens. METHODS: Intestinal and intravascular parasite infections were detected in stool and urines samples. The in vitro cellular cyt

Epitope mapping and conservation of antigens from the Multi-Cruzi immunoassay platform.

Infection with the protozoan parasite Trypanosoma cruzi can lead to Chagas disease, and diagnosis of infection mostly relies on serological testing, although no gold standard exists. The evaluation of treatment efficacy is also challenging as seronegativization takes decades to be detected. The Multi-Cruzi platform, which provides a serological profile against 15 parasite antigens, has been proposed as promising alternative for confirmatory diagnostic and monitoring of treatment response. To further evaluate this platform, we performed epitope mapping of the Multi-Cruzi antigens with peptide m

How protozoan parasites evade the immune response.

Protozoan pathogens such as Plasmodium, Leishmania, Trypanosoma and Entamoeba are responsible for several of the most widespread and lethal human diseases. Their successful survival depends mainly on evading the host immune system by, for example, penetrating and multiplying within cells, varying their surface antigens, eliminating their protein coat, and modulating the host immune response. Immunosuppression is sometimes caused directly by parasite products and sometimes involves antigenic mimicry, which often appears in association with parasitic diseases. However, one of the most sophistica

Diagnosis of Encephalitozoon cuniculi infection by western blot and the use of cross-reactive antigens for the possible detection of microsporidiosis in humans.

Microsporidia are very primitive, eukaryotic, obligate, intracellular, protozoan parasites. Encephalitozoon cuniculi, a microsporidian originally described from a rabbit infection, has been described in humans as well as in many species of laboratory animals. We report the detection of E. cuniculi by Western blotting in a rabbit with torticollis that was obtained from an Encephalitozoon-free colony. Cross-reactivity of this serum was observed with antigens prepared from several genera of microsporidia. Identical Western blotting patterns were obtained with sera obtained from a rabbit immunized

Antigenic variation as an exploitable weakness of babesial parasites.

Babesia bovis and its bovine host interact in many ways, resulting in a range of disease and infection phenotypes. Host responses to the parasite elicit or select for a variety of responses on the part of the parasite, the full range of which is not yet known. One well-established phenomenon, thought to aid parasite survival by evasion of host adaptive immune responses, is the sequential expansion of antigenically variant populations during an infection, a phenomenon referred to as "antigenic variation". Antigenic variation in B. bovis, like that in the human malarial parasite, Plasmodium falc

Evaluation of two serological tests for Trichomonas vaginalis infection.

Trichomonas vaginalis is a widely prevalent, sexually transmitted protozoan infecting both males and females. Despite its prevalence, little is known about its contribution to the morbidity rates for urogenital-tract infections. Currently accepted diagnostic methods are limited to the demonstration of the organism in fresh material, identification in stained material, or in vitro cultivation of organisms from the urogenital tract. We have evaluated the indirect hemagglutination test and the gel diffusion test for efficacy in detecting antibodies in serum samples drawn from two population group

Antibody response induced by structural proteins from Triatoma virus as potential adjuvants in experimental immunisation models.

INTRODUCTION: Virus-Like Particles (VLPs) are viral protein structures widely used as adjuvants in vaccine formulations due to their ability to stimulate the innate immune response, thereby contributing to the activation of adaptive immunity through the production of different IgG subclasses. The present study evaluated the adjuvant potential of recombinant structural proteins of the Triatoma virus VLPs (TrV-VLPs: VP1, VP2, and VP3) in experimental immunisation protocols for American Cutaneous Leishmaniasis and Chagas disease. METHODS: BALB/c mice were immunised with native antigens of Leishm

Modulation of human dendritic cell activity by Giardia and helminth antigens.

Giardia duodenalis is a common intestinal protozoan parasite known to modulate host immune responses, including dendritic cell (DC) function. Coinfections of intestinal pathogens are common, and thus, DCs may be concurrently exposed to antigens from multiple parasites. Here, we investigated the effects of G. duodenalis products on human monocyte-derived DC function independently and in combination with helminth antigens (Ascaris suum and Trichuris suis). All antigens individually induced an anti-inflammatory phenotype in DCs, reducing lipopolysaccharide (LPS)-induced interleukin (IL)-6, IL-12p

Advancing protozoan vaccine research: A review of promising candidates and emerging technologies.

Protozoan parasites, a diverse group of unicellular eukaryotic organisms, represent a significant challenge to global human and animal health, particularly in regions with constrained healthcare resources. These pathogens are etiological agents of most severe and often fatal diseases, collectively accounting for millions of infections and deaths annually. Despite the profound impact on public health, vaccine development for protozoan diseases has been markedly slower than the advancements in combating other infectious agents. However, recent strides in understanding parasite biology and host-p

Toxoplasma gondii surface antigen 1 (SAG1) interacts in vitro with host cell receptor for activated C kinase 1 (RACK1).

Toxoplasma gondii (T. gondii) surface antigen 1 (SAG1) is crucial for tachyzoite invasion into host cells. However, the role of SAG1 in interaction with host cells remains unknown. The primary objective of this study was to analyze and validate the interaction between SAG1 and host cells. RACK1, an intracellular multifunctional protein, was identified as a SAG1 binding partner in host cells. Furthermore, the expression of RACK1 is manipulated by SAG1, and depletion of RACK1 negatively regulated host cell viability. These results imply that through interaction with RACK1, SAG1 preserves the via

Designing blood-stage vaccines against Babesia bovis and B. bigemina.

The tick-transmitted apicomplexan parasites Babesia bovis and B. bigemina cause significant disease in cattle in many tropical and temperate areas of the world. These parasites present a challenge for vaccine development, and yet provide a system for studying the pathogenesis, mechanisms of protective immunity and regulation of host immune responses associated with intraerythrocytic protozoan parasites in a non-rodent species. In this article, Wendy Brown and Guy Palmer review strategies for identifying candidate vaccine antigens of B. bovis and B. bigemina and for priming immune responses to

[Current methods of immunologic diagnosis in parasitology].

This review of the immunological diagnosis of parasitic diseases defines the various indications, the means of collection and preparation, the various levels of specificity and the choice of parasitic antigen which should be used for immuno-diagnosis. The detection and assay of circulating antibodies relies on the techniques of immuno-precipitation (immunodiffusion, immunoelectrophoresis, electrosyneresis), indirect agglutination (latex and haemagglutination) or the use of labelled compounds (immunofluorescence, enzymo-immunoassay, radio-immunoassay). Their respective advantages and disadvanta

The immunology of experimental Chagas' disease. IV. Production of lesions in rabbits similar to those of chronic Chagas' disease in man.

Eight rabbits that received a single inoculation of trypomastigotes of a virulent strain of Trypanosoma cruzi first developed a transient acute illness associated with parasitemia; later, 4 of these rabbits died with chronic myocarditis and/or with megacolon in the absence of demonstrable parasitemia or encysted parasites in tissues. Two of these rabbits with chronic myocarditis died unexpectedly. Three of the inoculated rabbits that survived the infection were sacrificed 18 months later and showed similar but less severe microscopic lesions. The remaining rabbit is alive and well at the time

Repurposing of Plasmodium falciparum var genes beyond the blood stage.

A commonly observed survival strategy in protozoan parasites is the sequential expression of clonally variant-surface antigens to avoid elimination by the host's immune response. In malaria-causing P. falciparum, the immunovariant erythrocyte-membrane protein-1 (PfEMP1) adhesin family, encoded by var genes, is responsible for both antigenic variation and cytoadherence of infected erythrocytes to the microvasculature. Until recently, the biological function of these variant genes was believed to be restricted to intraerythrocytic developmental stages. With the advent of new technologies, var ge

Regulation of antigenic variation in Giardia lamblia.

Antigenic variation, a clonal phenotypic variation developed by microorganisms, involves the permanent switching of homologous, antigenically different cell surface molecules. In pathogenic microorganisms, antigenic variation is often described as a mechanism to evade the host immune system and therefore is responsible for the generation of chronic and/or recurrent infections. However, antigenic variation has also been involved in expanding host diversity and differential courses of the diseases. The intestinal protozoan parasite Giardia lamblia undergoes antigenic variation through the contin
