Mitigation of Cu(II)-induced damage in human blood cells by carnosine: An in vitro study.

Copper (Cu) is an essential micronutrient but human exposure to high level of this metal results in adverse health effects. Oxidative stress is assumed to play a major role in the mechanism of Cu-induced toxicity. The protective role of carnosine, an antioxidant and antiglycating agent, was examined against Cu-induced toxicity in isolated human blood cells. Red blood cells (RBC) were treated with 0.5 mM copper chloride (CuCl2), a Cu(II) compound, either alone or after treatment with carnosine. Incubation of RBC with CuCl2 increased protein oxidation, lipid peroxidation, methemoglobin formation

Unveiling the Role of Mg2+ and Mn2+ in Theaflavin-Mediated Inhibition of Advanced Glycation End Products Formation: Mechanistic Insights from Intermolecular Interaction.

Advanced glycation end products (AGEs) are implicated in the pathogenesis of chronic diseases. This study investigated the inhibitory effect of theaflavin (TA) on AGE formation in a human serum albumin (HSA)-fructose model system, both in the absence and presence of non-cytotoxic Mg2+ or Mn2+. The underlying mechanism was elucidated using multiple spectroscopic techniques and molecular docking. TA significantly inhibited AGE formation, with the enhancement by metal ions following the order: 1.5 μM Mn2+ > 1.5 μM Mg2+ > 1 mM Mg2+. This trend aligned with the binding affinity between HSA and TA d

N-Acetyl Cysteine Mitigates d-Ribose-Induced Protein Glycation and Aggregation Through Multiple Protective Mechanisms.

Advanced glycation end-products (AGEs) arise from non-enzymatic reactions between reducing sugars and proteins, contributing to oxidative stress and metabolic dysfunction. Excessive AGE accumulation is implicated in chronic diabetic complications and may also be relevant to acute metabolic disturbances encountered in emergency medicine. N-acetylcysteine (NAC), a naturally occurring antioxidant found in Allium species, has demonstrated potential to attenuate oxidative and glycation-mediated damage. The effect of NAC on d-ribose-induced glycation of bovine serum albumin (BSA) was investigated us

Multitargeted Modulation of Skin Dullness by HNHT Formulation: Synergistic Inhibition of Melanogenesis, Glycation, and Lipofuscin Deposition.

BACKGROUND: Skin dullness, characterized by pigmentation disorders and yellowing, is a major cosmetic concern in Asian populations. Lipofuscin exhibits ultraviolet (UV)/visible light spectral absorption, reduces skin reflectance, and contributes to dullness, whereas existing therapies targeting melanogenesis and glycation lack comprehensive efficacy. This study aimed to evaluate the multimodal anti-dullness effects of a novel formulation (HNHT: hyaluronic acid, niacinamide, hydrolyzed red algae, and tranexamic acid). METHODS: In vitro models (melanocytes, keratinocytes, fibroblasts, ex vivo s

From Glycation to Toxicity: α-Synuclein Oligomers, Oxidative Stress, and Multi-Targeted Protection by Trans-Ferulic Acid.

Glycation, a spontaneous reaction between sugars and biomolecules, is a central driver of oxidative stress and protein dysfunction in metabolic and neurodegenerative disorders. α-Synuclein (α-Syn), a key protein implicated in Parkinson's disease, is particularly susceptible to modification by reactive carbonyl species such as methylglyoxal (Me) due to its lysine-rich structure. These modifications result in the formation of advanced glycation end products (AGEs), which promote protein aggregation, impair cellular clearance, and induce oxidative and inflammatory damage via the AGE-RAGE axis. Comparative functional evaluation of the atypically modified GlcHMS326 and porphyra-334, Two structurally distinct mycosporine-like amino acids.

Mycosporine-like amino acids (MAAs) are natural compounds widely studied for their photoprotective and antioxidant properties. Typical MAAs consist of one or two amino acids attached to a cyclohexenone or cyclohexenimine ring, whereas atypical MAAs possess unique chemical modifications such as glycosylation and methylation. Recently, we identified an atypical MAA, GlcHMS326, from the cyanobacterium Gloeocapsa sp. BRSZ, characterized by glycosylation, methylation, and hydroxylation. In this study, we compared the chemical stability and biological activities of GlcHMS326 with those of a typical

Glyoxalase I drives epithelial-to-mesenchymal transition via argpyrimidine-modified Hsp70, miR-21 and SMAD signalling in human bronchial cells BEAS-2B chronically exposed to crystalline silica Min-U-Sil 5: Transformation into a neoplastic-like phenotype.

Glyoxalase I (Glo1) is the main scavenging enzyme of methylglyoxal (MG), a potent precursor of advanced glycation end products (AGEs). AGEs are known to control multiple biological processes, including epithelial to mesenchymal transition (EMT), a multistep phenomenon associated with cell transformation, playing a major role in a variety of diseases, including cancer. Crystalline silica is a well-known occupational health hazard, responsible for a great number of human pulmonary diseases, such as silicosis. There is still much debate concerning the carcinogenic role of crystalline silica, main

Functional and Physiological Implications of Oligopeptide Transporters: Potential Targets for Pharmacological Interventions.

Peptide transporters are important plasma membrane proteins that facilitate the cellular uptake of di- and tripeptides in addition to various peptidomimetic drugs. The proton-coupled oligopeptide transporter (POT) family consists of mainly four transporter proteins, which include two H+ -coupled oligopeptide transporters known as PEPT1 (SLC15A1) and PEPT2 (SLC15A2), along with two peptide/histidine transporters referred to as PHT1 (SLC15A4) and PHT2 (SLC15A3). These transporters play an important role in the drug delivery process in mammalian tissues. They are highly expressed in tissues such

Vitamin B6 Modification of Bovine Pericardial Bioprosthetic Heart Valve Leaflets Promotes Blood Outgrowth Endothelial Cell Adhesion.

Bioprosthetic heart valves (BHV), typically fabricated from glutaraldehyde-fixed bovine pericardium (BP), are the most common type of replacement for patients with severe heart valve disease. Despite being preferred, BHV are limited in durability due to structural valve degeneration (SVD), which occurs due to leaflet calcification and non-calcific mechanisms including advanced glycation end product (AGE) accumulation and serum protein infiltration. Another limitation of BHV is characteristically they do not endothelialize, even after years of implantation. Previously we showed pretreating BP w

Antioxidant and Anti-Glycation Potential of H2 Receptor Antagonists-In Vitro Studies and a Systematic Literature Review.

Background: Histamine H2 receptor antagonists are a group of drugs that inhibit gastric juice secretion in gastrointestinal diseases. However, there is evidence to suggest that H2 blockers have a broader spectrum of activity. The antioxidant properties of H2 blockers have not been fully elucidated, and their anti-glycation potential has not been studied to date. Therefore, this is the first study to compare the antioxidant and antiglycation potentials of the most popular H2 antagonists (ranitidine, cimetidine, and famotidine) on protein glycoxidation in vitro. Methods: Bovine serum albumin (BS

Reactive oxygen species induce apoptosis in bronchial epithelial BEAS-2B cells by inhibiting the antiglycation glyoxalase I defence: involvement of superoxide anion, hydrogen peroxide and NF-κB.

Reactive oxygen species (ROS) are implicated in the regulation of apoptosis through a number of distinct mechanisms depending on cell type and stimulation conditions. Glyoxalase I (GI) metabolizes methylglyoxal (MG) and MG-derived advanced glycation end products (AGEs) known to cause apoptosis. This study examined the possible role of GI among the mechanisms of ROS-driven apoptosis in human bronchial epithelial BEAS-2B cells exposed to wood dust and signaling pathways by which these reactive species regulate GI expression. Our results showed that wood dust generated distinct ROS (superoxide an

s-Allyl cysteine, s-ethyl cysteine, and s-propyl cysteine alleviate β-amyloid, glycative, and oxidative injury in brain of mice treated by D-galactose.

The neuroprotective effects of s-allyl cysteine, s-ethyl cysteine, and s-propyl cysteine in D-galactose (DG)-treated mice were examined. DG treatment increased the formation of Aβ(1-40) and Aβ(1-42), enhanced mRNA expression of β-amyloid precursor protein (APP) and β-site APP cleavage enzyme 1 (BACE1), and reduced neprilysin expression in brain (P < 0.05); however, the intake of three test compounds significantly decreased the production of Aβ(1-40) and Aβ(1-42) and suppressed the expression of APP and BACE1 (P < 0.05). DG treatments declined brain protein kinase C (PKC) activity and mRNA expr

Identification of RAGE Inhibitors from Curcuma caesia Roxb: Implications in Treatment against Diabetic Complications.

INTRODUCTION: Diabetic nephropathy (DN) is a progressive renal complication that significantly contributes to end-stage renal disease. Hyperglycaemia contributes to the formation of advanced glycation end-products (AGEs). The interaction between AGEs and their receptor (RAGE) plays a key role in the progression of DN. RAGE activation increases oxidative stress and promotes inflammation, thereby evoking cellular and molecular damage. Together, these events result in kidney injury and varying degrees of proteinuria. This study aims to evaluate the drug-like properties of potential natural compou

Protective effect of unsaturated fatty acids against glycation-induced structural and functional alteration of albumin: Perspectives on diabetic nephropathy.

Diabetic nephropathy (DN) is the leading cause of end-stage renal disease, and the protein glycation pathway has a major role in DN pathogenesis. Accumulation of advanced glycation end products (AGEs) upregulates the Receptor for Advanced Glycation End products (RAGE)-NF-κB pathway, leading to elevated oxidative stress and inflammation. The present study aims to shed light on the anti-glycation mechanism of unsaturated fatty acids (UFAs), such as oleic acid (OA), linoleic acid (LA), and docosahexaenoic acid (DHA), on glycated human serum albumin (G.
