Antioxidants--carcinogenic and chemopreventive properties.

Synthetic and naturally occurring antioxidants have a wide variety of biological actions in rodents in addition to their primary antioxidant activity. Some of the included biological effects are of direct interest in relation to studies of carcinogenicity and/or modulation of carcinogenesis. Since the synthetic antioxidant BHA was first found to exert carcinogenic potential in rat and hamster forestomach epithelium, many other synthetic and naturally occurring antioxidants have been examined for their ability to induce proliferative activity in the alimentary canal. These studies have revealed

Mitochondria-targeted antioxidants protect Friedreich Ataxia fibroblasts from endogenous oxidative stress more effectively than untargeted antioxidants.

Friedreich Ataxia (FRDA), the most common inherited ataxia, arises from defective expression of the mitochondrial protein frataxin, which leads to increased mitochondrial oxidative damage. Therefore, antioxidants targeted to mitochondria should be particularly effective at slowing disease progression. To test this hypothesis, we compared the efficacy of mitochondria-targeted and untargeted antioxidants derived from coenzyme Q10 and from vitamin E at preventing cell death due to endogenous oxidative stress in cultured fibroblasts from FRDA patients in which glutathione synthesis was blocked. Effects of antioxidants on cancer progression.

Antioxidant supplements are widely marketed for their promised health benefits, including cancer prevention and therapy support. This belief stems from the idea that excessive levels of reactive oxygen species (ROS) cause oxidative damage to cellular macromolecules. However, the effects of antioxidants are highly context-dependent and influenced by the compound type, dosage, and cancer type. While antioxidants may slow tumor progression in specific cancers such as MYC-driven lymphoma, they can accelerate tumor growth, metastasis, and angiogenesis in other settings, including KRAS-driven lung c

Role of Natural Antioxidants in Cancer.

The oxidative stress defined as an event caused by an imbalance between production and accumulation of reactive oxygen species (ROS), which lead to a damage in the structure of proteins, lipids, and DNA. Therefore, the production of ROS may alter the normal physiological process by provoking damage to multiple cellular organelles and processes. Oxidative stress has been linked to heart disease, cancer, respiratory diseases, immune deficiency, stroke, Parkinson's disease, and other inflammatory or ischemic conditions. Antioxidants are substances that can prevent or slow damage to cells and tiss

Nitroxides as Antioxidants and Anticancer Drugs.

Nitroxides are stable free radicals that contain a nitroxyl group with an unpaired electron. In this paper, we present the properties and application of nitroxides as antioxidants and anticancer drugs. The mostly used nitroxides in biology and medicine are a group of heterocyclic nitroxide derivatives of piperidine, pyrroline and pyrrolidine. The antioxidant action of nitroxides is associated with their redox cycle. Nitroxides, unlike other antioxidants, are characterized by a catalytic mechanism of action associated with a single electron oxidation and reduction reaction. In biological condit

Oxidative theory of atherosclerosis and antioxidants.

Atherosclerosis is a multifactorial process that begins early in infancy and affects all the humans. Early steps of atherogenesis and the evolution towards complex atherosclerotic plaques are briefly described. After a brief history of the 'Lipid theory of atherosclerosis', we report the most prominent discoveries on lipoproteins, their receptors and metabolism, and their role in atherogenesis. The main focus is the 'oxidative theory of atherosclerosis', with emphasis on free radicals and reactive oxygen species, lipid peroxidation and LDL oxidation, biological properties of oxidized LDL and t

Antioxidants and vitamins in clinical conditions.

Various reactive oxygen species (ROS) may be produced from normal biochemical, essential metabolic processes or from external sources as exposure to a variety of agents presented in the environment. Lipids, proteins, carbohydrates and DNA are all capable of reacting with ROS and can be implicated in etiology of various human disorders (rheumatoid arthritis, reperfusion injury, atherosclerosis, lung diseases etc.). In the organism damage by ROS is counteracted with natural antioxidants (glutathione peroxidases, superoxide dismutases, catalase, glutathione, ubiquinol, uric acid, and essential mi

Diverse antioxidants protect against acetaminophen hepatotoxicity.

The reactive oxygen species-sensitive transcription nuclear factor-kappaB (NF-kappaB) plays a pivotal role in the development of acetaminophen (APAP) hepatotoxicity. We investigated the efficacy of a diverse series of antioxidants in preventing APAP-induced hepatotoxicity. BALB/c mice were divided into four groups and provided with antioxidants incorporated into chow as follows: (1) control diet; or diet supplemented with (2) S-adenosylmethionine (SAMe); (3) green tea polyphenols (GrTP); or (4) (RS)-n-propylthiazolidine-4(R)-carboxylic acid (PTCA). After 5 days on these diets, the animals were

Lipophilic antioxidants in patients with phenylketonuria.

BACKGROUND: Low serum ubiquinone-10 concentrations have been described in phenylketonuric patients fed natural-protein-restricted diets. Such low concentrations may be related to increased free radical damage. OBJECTIVE: We evaluated the relation between low serum ubiquinone-10 concentrations and other lipophilic antioxidants (tocopherol and retinol), selenium, glutathione peroxidase activity, and malondialdehyde concentrations as a marker of lipid peroxidation. DESIGN: This was a cross-sectional study of 58 patients with phenylketonuria (aged 2-36 y; median: 13 y) under dietary treatment, 5

Botanical antioxidants for chemoprevention of photocarcinogenesis.

The incidence of non-melanoma skin cancer, consisting of basal- and squamous- cell carcinoma, continues to increase in the United States and elsewhere. Solar ultraviolet (UV) B radiation has been implicated as its main cause. This adverse effect of UVB has become a major human health concern. Therefore, development of novel strategies to reduce the occurrence of skin cancer is a highly desirable goal. Because UV radiation is known to cause excessive generation of reactive oxygen species (ROS) thereby resulting in an oxidative stress condition, the approaches aimed at counteracting ROS producti

[Antioxidants for prophylaxis of eye diseases].

The contemporary literature has widely described the role of free oxygen radicals and their antioxidants in pathogenesis of some eye diseases, mainly cataract, age-related macular degeneration, retinopathy of prematurity and cystic macular oedema. This paper presents publications which stress the importance of antioxidants use in prophylaxis of cataract and age-related macular degeneration. Positive antioxidants role was proved both in experimental research and in clinical observations.

Role of antioxidants in health maintenance.

Free radicals are produced in the body as by products of normal metabolism and as a result of exposure to radiation and some environmental pollutants. Because they are highly reactive, they can damage cellular components and are implicated in a variety of diseases. Free radicals are normally neutralized by efficient systems in the body that include the antioxidant enzymes (superoxide dismutase, catalase, and glutathione peroxidase) and the nutrient-derived antioxidant small molecules (vitamin E, vitamin C, carotenes, flavonoids, glutathione, uric acid, and taurine). In healthy individuals, a d

Asthma, inhaled oxidants, and dietary antioxidants.

The possible influence of dietary antioxidants, especially vitamin C, on the increasing prevalence of asthma is explored. Vitamin C intake in the general population appears to correlate with asthma, suggesting that a diet low in vitamin C is a risk factor for asthma. Epidemiological studies show associations among oxidant exposure, respiratory infections, and asthma in children of smokers. Symptoms of ongoing asthma in adults appear to be increased by exposure to environmental oxidants and decreased by vitamin C supplementation. There is evidence that oxidants produced endogenously by overacti

Mechanisms of oxidation, antioxidants, and atherosclerosis.

The oxidation of LDL by a number of different cell types has been studied and described. Cells may be able to initiate and seed peroxides in LDL but, in the absence of metals or peroxidase activity, may not be able to oxidize the lipoprotein. Monocyte-derived macrophages and neutrophils are able to initiate and propagate lipid peroxidation. The oxidative role of smooth muscle cells and endothelial cells may be restricted solely to the generation of hydrogen peroxide or other oxidants. The above rationale presents a problem in using LDL preparations that already contain peroxides, and in using

Antioxidants Differentially Regulate Non-Photochemical Quenching.

Non-photochemical quenching (NPQ) dissipates excess light energy as heat, limiting reactive oxygen species production during photosynthesis. Exogenous antioxidants are widely reported to improve plant performance under stress, yet their impacts on NPQ are inconsistent across studies, complicating mechanistic interpretation. Here, we systematically compared six antioxidants - ascorbate, chitosan, coenzyme M, reduced glutathione, melatonin, and N-acetyl cysteine - applied to Arabidopsis thaliana seedlings using individual concentration gradients, under standardised pH and illumination regimes. Applications of Antioxidants in Dental Procedures.

As people are paying more and more attention to dental health, various dental treatment procedures have emerged, such as tooth bleaching, dental implants, and dental restorations. However, a large number of free radicals are typically produced during the dental procedures. When the imbalance in distribution of reactive oxygen species (ROS) is induced, oxidative stress coupled with oxidative damage occurs. Oral inflammations such as those in periodontitis and pulpitis are also unavoidable. Therefore, the applications of exogenous antioxidants in oral environment have been proposed. In this arti

Antioxidants and Chronic Obstructive Pulmonary Disease.

Antioxidants represent an attractive therapeutic avenue for individuals with chronic obstructive pulmonary disease (COPD). Cigarette smoke, the major cause of COPD, contains very high concentrations of gaseous and soluble oxidants that can directly induce cell injury and death. Furthermore, particulate matter in cigarette smoke activates lung macrophages that subsequently attract neutrophils. Both neutrophils and macrophages from the lungs of cigarette smokers continuously release large amounts of superoxide and hydrogen peroxide through the nicotinamide adenine dinucleotide phosphate (NADPH)
