Characterization of microbial community in the leachate associated with the decomposition of entombed pigs.

Foot and mouth disease (FMD) is one of the acute infectious diseases in hoofed and even-toed mammals, including pigs, and it occurs via acute infection by Aphthovirus. When FMD is suspected, animals around the location of origin are typically slaughtered and buried. Other methods such as rendering, composting, and incineration have not been verified in practice in Korea. After the FMD incident, the regular monitoring of the microbial community is required, as microorganisms greatly modify the characteristics of the ecosystem in which they live. This is the result of their metabolic activities

Development and application of a dual RT-qPCR assay for the differential detection of bovine rhinitis virus genotypes A and B.

Bovine rhinitis virus (BRV) is an important pathogen causing bovine respiratory disease complex (BRDC). To facilitate the rapid and simultaneous detection of bovine rhinitis A virus (BRAV) and bovine rhinitis B virus (BRBV), specific primers and TaqMan probes targeting the viral 3D gene were designed and synthesized. Following the optimization of primer and probe concentrations and annealing temperatures, as well as comprehensive evaluations of specificity, sensitivity and reproducibility, a dual real-time RT-PCR (RT-qPCR) assay was successfully developed for the reliable differential detectio

Ultrastructural and replicative features of foot-and-mouth disease virus in persistently infected BHK-21 cells.

Persistent foot-and-mouth disease (FMD) virus infection in vitro has been studied in a chronically infected cloned BHK-21 cell line. Virus growth during serial cell passages was followed by infectivity assay and immunocytochemical staining. Only a small percentage of cells (0.006-6%) was found to harbour virus during persistence. Light and electron microscopy showed the presence of cytoplasmic protuberances ("blebs") at the surface of persistently infected cells. The curing of cell cultures was achieved by passaging them in the presence of polyvalent immune serum. The absence of virus in cured

The localization of persistent foot and mouth disease virus in the epithelial cells of the soft palate and pharynx.

After contact with foot and mouth disease virus (FMDV), cattle may become persistently infected, regardless of their pre-existing immune status or whether they develop clinical disease. The cellular sites of FMDV persistence have not previously been determined. The use of in-situ hybridization in combination with tyramide signal amplification (TSA) provided the first direct evidence that FMDV RNA is localized within the epithelial cells of the soft palate and pharynx during persistent infection, indicating that these cells remain persistently infected after contact with FMDV.

The early pathogenesis of foot-and-mouth disease in pigs infected by contact: a quantitative time-course study using TaqMan RT-PCR.

Foot-and-mouth disease (FMD) is a highly contagious, economically important virus disease of cloven-hoofed animals. The objective of the present study was to examine the early pathogenesis of FMD in pigs by a quantitative time-course study. Under experimental conditions, recipient pigs were infected by contact with donor pigs affected by FMD. Every 24 h from day 1 to day 4 after exposure, two recipient pigs were selected randomly, killed and necropsied. A range of tissues were analysed by a quantitative TaqMan RT-PCR method and by titration of FMD virus on primary bovine thyroid cells. The tit

Evaluation of techniques to demonstrate foot-and-mouth disease virus in bovine tongue epithelium: comparison of the sensitivity of cattle, mice, primary cell cultures, cryopreserved cell cultures and established cell lines.

Tongue epithelia infected with each of the 7 serotypes of foot-and-mouth disease virus (FMDV) were used to evaluate in vivo and in vitro systems for the detection of FMDV. Cattle inoculated by the intradermal route in the tongue (IDL) and suckling mice inoculated intraperitoneally were compared for susceptibility to FMDV with freshly prepared bovine thyroid cell cultures; cultures from cryopreserved bovine thyroid, bone marrow, mammary gland, myocardium, tongue, ovary and kidney cells; cultures from cryopreserved embryonic ovine kidney, newborn ovine kidney, ovine testicle, bone marrow, and ch

Developing Vaccines Against Foot-and-Mouth Disease: a Biotechnological Approach.

Foot-and-mouth disease (FMD) is a contagious viral disease of livestock with significant economic effect. It is prevalent in various regions of Asia, Africa, and South America. The causative agent of this disease is called foot-and-mouth disease virus (FMDV), which is a member of Aphthovirus genus. Vaccination is an effective technique to prevent the complications of FMD and to eradicate the disease in contaminated regions. Attempts are being made since the 1930s to develop potent vaccines against FMD. The history of vaccination against FMD has documented various types of vaccines including in

[Several criteria of evaluation of foot-and-mouth disease virus reproduced in cell cultures in suspension].

The study of the plaques produced by viruses Asia and O1 reveals different properties which may depend on the virus or on the cell or its method of culture. Antigenic differences and subsequent immunological differences correspond to differences in plaques. On the other hand, the cell line in suspension evolves both in its morphology and its receptivity during the passages. All these variations imply the observations of quantitative and qualitative criteria of evaluation in order to prepare vaccines from viruses similar to viruses specific to a given epizooty.

[Influence of a hypertonic medium on cell susceptibility to foot-and-mouth disease virus].

The influence of hypertonic medium on the relationship between two cell clones of IB-RS-2 swine line and the foot-and-mouth disease virus was studied. Although the number of infected cells was increased by the cell treatment with the hypertonic medium, before or during the time of virus adsorption and penetration onto the cells, showed by the plaque number, the viral replication was partially inhibited as showed by the plaque size as by the viral yield in one-cycle of infection. On the other hand, the cell treatment with that medium after virus adsorption and penetration time inhibited as the

Type-independent detection of foot-and-mouth disease virus by monoclonal antibodies that bind to amino-terminal residues of capsid protein VP2.

The characterization of monoclonal antibodies raised against the foot-and-mouth disease virus isolates A22 Iraq/1964, Asia1 Shamir-Israel/1989, and SAT1 Zimbabwe/1989 with regard to neutralizing activity and sensitivity of their epitopes for treatment with trypsin, resulted in the identification of one non-neutralizing antibody in each panel that binds to a trypsin-sensitive epitope. Furthermore, each of these antibodies recognized 27 isolates of different provenance, representative of six serotypes. These antibodies are recommended for type-independent antigen detection by ELISA. The epitopes

Differentiating foot-and-mouth disease virus-infected from vaccinated animals with baculovirus-expressed specific proteins.

We had shown in preliminary studies with a small number of animals that antibodies against 2C could be detected in cattle and pigs which had been infected with FMDV but not in animals which had been vaccinated against the disease. To determine whether this test was generally applicable, sera from several hundred animals which had been vaccinated with different products in many countries have been tested in an ELISA using baculovirus expressed 2C. Our results show that only 1-2% of the sera gave a positive reaction by this method. In contrast, 100% of sera from convalescent animals gave a posit

[Trials to obtain conjugates for the passive hemagglutination test with chromium chloride and their use in studying foot-and-mouth disease viruses].

Experiments were carried out to produce conjugates for the hemagglutination-inhibition reaction with the aid of chromic chloride. Studies with reference F.M.D. viruses and with CFT revealed that the antibody erythrocyte diagnostic agents were highly specific, and were stable in storage at +4 degrees C and in freeze-drying. It was found that the conjugates obtained with F.M.D. antigens possessed high specificity, however, were not stable and 7 to 10 days later lost their activity. Tests with the hemagglutination-reaction showed that immunity itself could be studied in cattle treated with F.M.D.

Antigenic analysis of SAT 2 serotype foot-and-mouth disease virus isolates from Zimbabwe using monoclonal antibodies.

This paper compares strains of foot-and-mouth disease (FMD) serotype SAT (South African Territories) 2 viruses isolated from Zimbabwe and other African countries using monoclonal antibodies (MAb). A sandwich-ELISA was used to examine the relative binding of anti-SAT 2 MAb to the various viruses. The MAb-binding profiles of viruses isolated from field samples were compared using hierarchical cluster analysis. Viruses were obtained from game animals, mainly African buffalo (Syncerus caffer) which is the natural host and reservoir for SAT serotypes in Africa, and from cattle showing clinical sign

Abortions in dromedaries (Camelus dromedarius) caused by equine rhinitis A virus.

A virus was isolated from aborted dromedary (Camelus dromedarius) fetuses during an abortion storm in Dubai, United Arab Emirates. Laboratory investigations showed the causative agent to be indistinguishable from equine rhinitis A virus (ERAV), a picornavirus. Two pregnant dromedaries experimentally infected with the camel virus isolate both aborted and an identical virus was reisolated from both fetuses, thus confirming the diagnosis. The extremely high prevalence of antibody (>90 %) and the high titres recorded against ERAV in the dromedary herd clearly showed that ERAV does infect dromedari

Ihalation, persistence and dispersal f foot-and-mouth disease virus by man.

Sampling of human subjects, who had been in contact with animals infected with foot-and-mouth disease (FMD) virus, showed that virus could be recovered from the nose, throat, saliva and from air expelled during coughing, sneezing, talking and breathing. The amounts of virus recovered paralleled those collected with a large-volume sampler and multistage impinger and these findings confirmed that the highest recovery of airborne virus was from infected pigs followed by cattle and sheep. More virus was found in the noses of those examining infected animals than in those operating the samplers, bu

Binary ethylenimine as an inactivant for foot-and-mouth disease virus and its application for vaccine production.

Foot-and-mouth disease virus was inactivated with binary ethylenimine formed apart from or directly in the virus suspension by the cyclization of 2-bromoethylamine hydrobromide or 2-chloroethylamine hydrochloride under alkaline conditions. The inactivation rates with binary ethylenimine prepared apart from the virus suspension in dilute sodium hydroxide with either 2-bromoethylamine hydrobromide or 2-chlorethylamine hydrochloride were higher than with pure ethylenimine. When binary ethylenime was prepared directly in the virus suspension only 2-bromoethylamine hydrobromide gave acceptable inac

Molecular analysis of peptides from the GH loop of foot-and-mouth disease virus C-S30 using surface plasmon resonance: a role for kinetic rate constants.

A foot-and-mouth disease virus (FMDV) field variant, isolate C-S30 (also named C(1)-Barcelona), is known to contain four changes within the main antigenic site A (GH loop of capsid protein VP1, residues 136-150), at least one of which (Leu147-->Val) involves a highly conserved position, critical for antibody recognition in the reference strain C-S8c1. However, immunoenzymatic analysis of FMDV C-S30 showed it was recognised by 4C4, a monoclonal antibody that specifically targets site A. This remarkable behaviour has led us to analyse the individual and combined contributions of the four mutatio

[Combined vaccines including 1 or several anti-foot-and-mouth valences].

Combined vaccines are used for many human and animal diseases. However, we have very few examples of combinations containing anti-foot-and-mouth valence which would allow easy immunization against differnet diseases without additional handling. Until now foot-and-mouth vaccination has been applied simultaneously with anti-brucellic vaccination (Baldoni, Renoux), andi-anthrax (Nobili) and vaccination against swine fever and Aujeszky's disease (Lysenko). During the last ten years we have particularly studied the association of anti-foot-and-mouth disease vaccines with anti-brucella and anti-rabi
