The potencies in producing muscle relaxation, and in antagonizing apomorphine-induced climbing and hypothermia in mice, were examined for chlorpromazine, levomepromazine and their main metabolites, and for fluphenazine and 7-hydroxy fluphenazine. 3-Hydroxy chlorpromazine was more potent than chlorpromazine in antagonizing apomorphine-induced climbing, while levomepromazine and 3-hydroxy levomepromazine were equipotent in this test. The 3-hydroxy metabolites of chlorpromazine and levomepromazine were more potent than the parent compounds in antagonizing hypothermia, and had significantly weaker

BACKGROUND: Apomorphine sublingual film (SL-APO) and subcutaneous apomorphine (SC-APO) have been used for the treatment of OFF episodes in Parkinson's disease (PD). No study has prospectively compared efficacy and safety of these formulations.
OBJECTIVE: To compare SL-APO with SC-APO for treatment of OFF episodes in PD.
METHODS: An open-label, randomized, crossover study assessed SL-APO versus SC-APO in patients with PD and OFF episodes (N = 113). Doses were optimized in randomly assigned order. SL-APO dose initiation (10 mg) occurred in clinic; further dose optimization (15-30 mg; 5-mg incr

Cognitive rigidity (CR) refers to inadequate executive adaptation in the face of changing circumstances. Increased CR is associated with a number of psychiatric disorders, for example, obsessive-compulsive disorder, and improving cognitive functioning by targeting CR in these conditions, may be fruitful. Levetiracetam (LEV), clinically used to treat epilepsy, may have pro-cognitive effects by restoring balance to neuronal signalling. To explore this possibility, we applied apomorphine (APO) exposure in an attempt to induce rigid cue-directed responses following a cue (visual pattern)-reward (s

A majority of advanced Parkinson's disease (PD) patients on oral levodopa experience motor fluctuations, including sudden OFF and delayed ON periods. Fast-acting rescue medications are a vital part of the clinician's armamentarium in the treatment of motor fluctuations. Sublingual apomorphine is the first sublingual rescue medication on the market for the treatment of OFF times in PD.Materials and Methods: Here, we review the development and pharmacology of apomorphine in the treatment of PD as well as the safety and efficacy of sublingual apomorphine established in clinical trials. Finally, w

AIM: To report the case of a patient, who in the context of an anti-Parkinsonian therapy, developed addiction to apomorphine.
METHODS: Clinical case description.
RESULTS: Apomorphine is a dopaminergic agonist that acts directly on D2 receptors. It has been used in alcoholism, male sexual dysfunction and with diagnostic and therapeutic purposes in Parkinson's disease (PD).
CONCLUSIONS: The present work describes the case of a woman with PD who developed a loss of control over the consumption of apomorphine that resulted in a significant impairment of her functioning. PD patients with high fr

RATIONALE: Classification of pigs based on the degree of resistance they display in a so-called "backtest" seems, to a certain extent, predictive for their coping strategy.
OBJECTIVE: The present study examined whether, as found in rodents, the behavioral response to apomorphine of pigs relates to individual coping characteristics.
METHODS: During the suckling period pigs were subjected to the backtest. In this test, each pig is restrained on its back for 1 min and the resistance (i.e. number of escape attempts) is scored. Pigs classified as low-resisting (LR, n=10) or high-resisting (HR, n=

1. The goal was to verify if central or peripheral sulphated cholecystokinin octapeptide (CCK8) injections can modulate apomorphine (APO)-induced stereotyped behavior. Experiments were designed to determine the involvement of cholecystokinin receptor subtypes as well. 2. Animals which received CCK8 (0.0725, 0.145 and 14.5 nmol, icv) showed a significant (p < 0.05) decrease in APO (0.6 mg/kg, sc)-induced stereotyped behavior. 3. No other statistically significant difference was observed among groups. Since ip CCK8 (1.16 or 2.32 nmol/kg) injections did not interfere with APO-induced stereotypy,

We evaluated the effects of the dopaminergic agent apomorphine on defecation and anorectal function in patients with Parkinson's disease (PD). A gastrointestinal symptom survey, extrapyramidal assessment, defecating proctogram, and anorectal manometric study were performed in 8 subjects with PD. Basal studies showing abnormalities were repeated following apomorphine administration. Prior defecographic abnormalities were normalized following apomorphine injection in 1 of 3 subjects and significant improvements in manometric parameters were observed in all 5 subjects who underwent repeat anorect

Dopamine, infused into the hypothalamus or injected into the 3rd cerebral ventricle of chickens pretreated with mebanazine, induced behavioural and electrocortical sleep, suppressed motor activity and lowered body temperature and CO2 elimination, effects prevented by phenoxybenzamine but not spiperone. The hypothermic effects of dopamine were intensified by testing chicks below thermoneutrality. Ampomorphine, infused into the hypothalamus, injected into the 3rd cerebral ventricle or given intravenously, evoked behavioural and electrocortical arousal in the intact chicken and in the encéphale i

Eleven patients with levodopa-related motor fluctuations were scored before and after intranasal apomorphine monotherapy, and the motor responses were compared with those with levodopa/carbidopa in this openlabel study. Oral trimethobenzamide was used to prevent apomorphine-induced nausea. Three measures of motor performance were employed: (a) the Unified Parkinson's Disease Rating Scale (UPDRS) motor battery; (b) a timed hand-tapping test; and (c) the Webster's step-seconds test. The magnitude of the motor-score improvement after apomorphine administration was very similar to that after the u

Data from studies indicate that blood glucose levels may be influenced by stimulation of central D1 and D2 dopamine receptors. This is modulated by central dopamine receptor status. We studied apomorphine-induced increase in blood glucose levels in Parkinson's disease compared with those in Parkinsonian syndromes unresponsive to dopaminergic drugs. Results indicate that apomorphine-induced blood glucose level increases are significant only in Parkinson's disease. As in the animal, this could possibly be explained by central dopamine receptor status.

Previous studies have indicated that drug-induced experimental diabetes is associated with increased receptor binding in the rat brain. The purpose of this study was to determine whether the dopamine receptor agonist apomorphine (APO) might produce an accentuated hypothermic response in rats rendered diabetic by alloxan (ALX) treatment. In a previous study, however, the only controls used were ALX-treated rats that failed to develop glycosuria. Therefore, in this study, APO (0.5 mg/kg IP) was administered to ALX-diabetic and non-diabetic as well as saline-treated control rats to ascertain whet

1. The LD50 for a 7-day period following intraperitoneal injection of apomorphine-HCl was calculated to be 158 mg/kg in rainbow trout. 2. Intraperitoneal injection of apomorphine at doses of 60 mg/kg or greater caused vomiting of plastic balls which had been placed in the stomachs of rainbow trout. 3. Apomorphine-induced effects included vomiting, vomiting behavior, toxicity, increased respiration, impaired motor control and equilibrium, and increased aggression. 4. The vomiting control mechanism of trout may be similar to that described in mammals.

Rats with endogenous hyperprolactinaemia induced by pituitary homografts under the kidney capsule were more sensitive to amphetamine and apomorphine with respect to drug-induced stereotypy, while a normal response was noted in drug-induced changes of locomotor activity, rearing and grooming. It is suggested that postsynaptic dopaminergic receptors, probably in the nigro-striatal system, may become selectively supersensitive in the endogenous hyperprolactinaemic animals.

28 patients suffering from idiopathic Parkinson's disease received subcutaneous apomorphine for the treatment of painful dystonia and of off-phases of long duration. After a mean period of 12.4 months 20 patients still used apomorphine. In this time the mean dose per injection had fallen from 2.5 mg to 2.1 mg and the mean number of daily applications from 2.6 to 2.2. Most patients considered dystonias and off-phases to respond satisfactorily to apomorphine. However, in most cases the treatment effect was impaired by side effects, especially nausea.

Apomorphine is a D1 and D2 dopamine receptor agonist with anti-parkinsonian properties qualitatively similar to those seen with L-dopa. It was first used in the treatment of Parkinson's disease by Schwab in the 1950s but owing to its short duration of action, the need for parenteral administration, and adverse reactions including nausea, vomiting, postural hypotension and sedation, it was not widely prescribed. In the early 1970s, Cotzias confirmed its potent anti-parkinsonian effects and that some of its secondary effects were diametrically opposite to those seen with L-dopa. The advent of pe

The diagnosis of Parkinson's disease (PD) is difficult despite the use of strict clinical criteria. Apomorphine (APP) is a strong dopaminergic D1 and D2 agonist which has been used as a diagnostic test for PD by several authors. Sixty patients with a Parkinson's syndrome received subcutaneous doses of APP with diagnostic objectives. Ninety-four percent of the subjects with clinical criteria of PD responded positively to the bolus of subcutaneous APP. The remaining six percent had secondary effects limiting the usefulness of the test. Two point seven percent of the patients with atypical data o

Neurogenic bladder frequently occurs in Parkinson's disease. Detrusor hyperreflexia is a consequence of nigrostriatal dopamine depletion, whereas the cause of hyporeflexia remains unclear. We compared the results of cystometry and urethral profilometry with and without apomorphine (and L dopa) in 12 idiopathic parkinsonians with urinary disorders free of dopaminergic treatment. Whereas hyperreflexic patients improved with apomorphine, and to a lesser extent with L dopa, these drugs had no effect on hyporeflexic patients. These results confirm the role of dopaminergic lesions in the occurrence
