Palmar and plantar keratoses developed in seven patients many years after ingeston of trivalent inorganic arsenic. Six had basal cell carcinoma (superficial multicentric type in five), carcinoma "in situ" or squamous cell carcinoma of the skin. Two had systemic carcinoma--one, bilateral breast adenocarcinoma and one, carcinoma of the colon. From these observations and from the findings of a review of the literature, there seems no question that long-term arsenic ingestion can cause palmar and plantar keratoses and skin cancer, particularly basal cell carcinoma of the superficial multicentric t

A novel postcolumn photooxidation reactor with remarkably high efficiency was developed. It was installed into a system of liquid chromatography (LC) combined with inductively coupled plasma mass spectrometry (ICPMS) using a hydride generation (HG) technique to enable sensitive speciation of organic arsenicals. An LC eluent was passed into a reaction tube that runs through a low-pressure mercury lamp of a high-efficiency photooxidation (HEPO) reactor. It provided efficient irradiation with vacuum ultraviolet light at 185 nm, which induced oxidative decomposition of persistent organic arsenical

1. Toxic doses of all arsenicals of which we have any knowledge produce definite pathological changes in the adrenals of guinea pigs. These changes include congestion, hemorrhage, disturbances in the lipoid content, cellular degenerations and necroses, and reduction in the chromaffin content. 2. The character and severity of the injury produced by different arsenicals varies with the chemical constitution of the compounds. 3. From these facts, we believe that adrenal injury is an important factor in arsenical intoxication and suggest that therapeutic doses of some arsenicals may produce adrena

A validated method of anion exchange liquid chromatography coupled to inductively coupled plasma mass spectrometry (LC-ICP-MS) was applied to determine arsenic species in human urine, particularly for occupational and environmental health investigations. In addition to the chromatographic peaks of arsenobetaine (As-B), dimethylarsinate (DMA), methylarsonate (MA), As(III) and As(V), which occur regularly, we have observed an additional As peak overlapping with that of As(V) in several routine analyses. Further investigations identified this unknown As compound as p-arsanilic acid, which had con

A case of arsenic poisoning in cattle is reported, in which six out of fifteen animals died. The source of poisoning was traced to a substance in the loft of the hay-barn, which probably consisted of the remains of a pesticide which had been there for several years. This had become liquefied as the result of a leak in the roof and had found its way into the hay. The arsenic content of the substance ranged from 17 to 37 per cent, that of the hay being up to 0.3 per cent. The concentration of arsenic in the organs, blood, faeces and urine of the animals were indicative of poisoning. The main cli

Arsenic exposure remains a leading environmental health concern. MicroRNA (miRNA) dysregulation may provide a mechanistic link to arsenic-related diseases, yet the associations of arsenic species and arsenic methylation capacity with miRNA profiles in plasma and leukocytes remain largely unexplored. In a community-based population of 160 Chinese adults, we characterized the miRNA signatures of urinary arsenic species [arsenate, As(V); arsenite, As(III); monomethylarsonate, MMA; dimethylarsinate, DMA], their summed concentration (ƩAs), and arsenic methylation capacity. We further explored their

Realgar (As(4)S(4)) and cinnabar (HgS) are frequently included in traditional Chinese medicines and Indian Ayurvedic medicines. Both As and Hg are well known for toxic effects, and their safety is of concern. The aim of this study was to compare chronic nephrotoxicity of An-Gong-Niu-Huang Wan (AGNH), realgar and cinnabar with common arsenicals and mercurials. Mice were orally administrated with AGNH (3 g/kg, 6-fold of clinical dose), cinnabar (0.3 g/kg, amount in AGNH) and realgar (0.3 g/kg, amount in AGNH), HgCl(2) (0.118 mmol/kg, 1/10 of cinnabar), MeHg (0.012 mmol/kg, 1/100 of cinnabar), Na

Introduction: The toxicity of arsenic is widely recognized globally, mainly harming human health by polluting water, soil, and food. However, its formulations can also be used for the clinical treatment of diseases such as leukemia and tumors. Arsenic has been used as a drug in China for over 2,400 years, with examples such as the arsenic-containing drug realgar mentioned in Shennong's Herbal Classic. We have reviewed references on arsenic over the past thirty years and found that research has mainly focused on clinical, pharmacological, and toxicological aspects. Results and Discussion: The f

Chemical analyses of metal (Cr, Mn, Fe, Co, Ni, Cu, Zn, As, Mo, Ba, and Pb) concentrations in hair, nails, and urine of Cambodians in arsenic-contaminated areas who consumed groundwater daily showed elevated levels in these biomarkers for most metals of toxicological interest. The levels of metals in biomarkers corresponded to their levels in groundwater, especially for As, whose concentrations exceeded the WHO guidelines for drinking water. About 75.6% of hair samples from the population in this study contained As levels higher than the normal level in unexposed individuals (1 mg·kg(-1)). Mos

The objective of this study was to design and evaluate a household-level arsenic education and well water arsenic testing intervention to increase arsenic awareness in Bangladesh. The authors randomly selected 1,000 study respondents located in 20 villages in Singair, Bangladesh. The main outcome was the change in knowledge of arsenic from baseline to follow-up 4 to 6 months after the household received the intervention. This was assessed through a pre- and postintervention quiz concerning knowledge of arsenic. Respondents were between 18 and 102 years of age, with an average age of 37 years;

Inorganic arsenic is a known human carcinogen causing tumors of the skin, urinary bladder, lung, liver, kidney, and possibly other organs. However, the animal models for inorganic arsenic carcinogenesis have been limited and development has been problematic. Gestation is often a period of high sensitivity to carcinogenesis so we investigated inorganic arsenite as a transplacental carcinogen in mice. Pregnant C3H mice were exposed to sodium arsenite (0, 42.5, and 85 ppm as arsenic) in the drinking water for a brief period during gestation (from gestation day 8 to 18), with no further arsenic ex

We determined the concentration and chemical status of arsenic in the placentas of hamsters following continuous exposure via the osmotic minipump to minimally and frankly teratogenic doses of arsenate. Close to 70% of the placental arsenic is bound to macromolecules, two-thirds of which is dialyzable. The remaining 30% of arsenic consists of low molecular weight species, predominantly inorganic arsenic. This mix is the same for minimally teratogenic and frankly teratogenic doses of arsenate.

Arsenic species were quantified to further explore the biotransformation of arsenic and better explain the potential health risks. In this research, the levels of six arsenic species (AsB, AsC, MMA, DMA, As3+, and As5+) were determined in 160 squid samples by HPLC-ICP-MS. AsB was the most predominant speciation in squid tissues. The arsenic species, including AsB, AsC, and MMA, were found in all samples, followed by DMA (49.38%), As3+ (21.25%), and As5+ (2.5%). Results indicated squid samples from offshore had higher levels of AsB, AsC, MMA, and DMA. Pearson's correlation analysis found correl

Combined pollutions of arsenic (As) and copper (Cu) are common in water bodies near mines, non-ferrous metal smelting and power plants. This study investigated the effect of Cu(II) on the absorption and speciation of As(V) by microalgae. We compared the absorption and speciation of arsenic by microalgae (mainly Cyanophyta and Chlorophyta) when exposed to single As(V) with that exposed to As-Cu co-exposure in laboratory. The results showed that in the case of single As(V) exposure, the inhibitory effect of As(V) on microalgae was primarily affected by the exposure time, instead of the concentra

Kidney disease is emerging as an arsenic (As)-linked disease outcome, however further evidence of this association is warranted. Our first objective for this paper was to examine the potential renal toxicity of As exposure in Bangladesh. Our second objective relates to examining whether the previously reported positive association between urinary creatinine (uCrn) and As methylation may be explained by renal function. We had hypothesized that these associations relate to supply and demand for s-adenosylmethionine, the methyl donor for both creatine synthesis and As methylation. Alternatively,

In most mammalian species, arsenic biotransformation occurs primarily by biomethylation and reduction reactions, with dimethylarsinic acid being the predominant metabolite excreted in the urine. Methylenetetrahydrofolate reductase (Mthfr) plays a key role in folate metabolism by channeling one-carbon units between nucleotide synthesis and methylation reactions. In the study on transgenic Mtfhr knockout mice we investigated: (1) whether Mthfr is an important determinant in arsenic biotransformation by performing urinary arsenic speciation, and (2) whether dietary folate deficiency alters arseni

A method for the determination of trivalent arsenicals in urine was examined. Trivalent arsenicals, extracted as complexes with diethylammonium diethyldithiocarbamate (DDDC) into carbon tetrachloride, were determined by liquid chromatography-inductively coupled plasma mass spectrometry (LC-ICP-MS). The trivalent methylated arsenicals monomethylarsonous acid (MMA(III)), dimethylarsinous acid (DMA(III)), and trimethylarsine (TMA) were detected in urine of rats that had received dimethylarsinic acid (DMA(V)) or monomethylarsonic acid (MMA(V)) at concentration of 200 microg ml(-1) in drinking wate

More than 100 million people around the world are exposed to arsenic in drinking water that exceeds the World Health Organization guideline level of 10 μg/L. Ingestion of water and food is the main route of human exposure to arsenic. Arsenic species excreted into urine serve as a useful biomarker of exposure and a potential marker of susceptibility, which requires speciation of arsenic in population studies. We report here the determination of arsenic species in 1873 urine samples from the Health Effects of Arsenic Longitudinal Study (HEALS) in Araihazar, Bangladesh. This double-blind research
