Cellular engineering of Artemisia annua and Artemisia dubia with the rol ABC genes for enhanced production of potent anti-malarial drug artemisinin.

BACKGROUND: Malaria is causing more than half of a million deaths and 214 million clinical cases annually. Despite tremendous efforts for the control of malaria, the global morbidity and mortality have not been significantly changed in the last 50 years. Artemisinin, extracted from the medicinal plant Artemisia sp. is an effective anti-malarial drug. In 2015, elucidation of the effectiveness of artemisinin as a potent anti-malarial drug was acknowledged with a Nobel prize. Owing to the tight market and low yield of artemisinin, an economical way to increase its production is to increase its co

Korean Artemisia species: phylogenomic authentication, ethnopharmacology, and commercial applications.

Artemisia species are integral to traditional Korean medicine and are increasingly used in medicinal and cosmeceutical products. However, taxonomic ambiguity, chemical variability, and limited clinical validation constrain their reliable therapeutic and commercial utilization. This review critically examines how phylogenomic evidence can be integrated with ethnopharmacology and phytochemistry to strengthen the authentication, scientific evaluation, and quality-controlled development of Artemisia taxa occurring in Korea. Its distinctive contribution is a Korea-focused synthesis linking chloropl

Anticancer power of Artemisia annua: A preclinical systematic review.

Cancer remains a leading global health challenge, and treatment efficacy is often limited by resistance to conventional therapies and their associated toxicities. These limitations have intensified the exploration of alternative strategies, particularly natural compounds with antitumor potential such as Artemisia annua (AA). This review aimed to systematically evaluate the antitumor efficacy and mechanisms of action of AA extracts and derivatives in preclinical animal models. A structured literature search was conducted in PubMed, Scopus, and Web of Science for studies published between 2019 a

Discovery of rearranged eudesmane-type sesquiterpenes from Artemisia tournefortiana.

Six undescribed sesquiterpenes artetourneforin F-K (1-6), along with six known compounds (7-12), were isolated from Artemisia tournefortiana. Their structures were established by 1D and 2D NMR spectroscopic data, HRESIMS, ECD, and quantum chemical calculations. Artetourneforin F-K (1-6) represented a rare 4,5-seco undescribed skeleton in natural sesquiterpenes, and a plausible biosynthetic pathway was proposed. The X-ray crystal structures of compounds 7 and 8 were reported for the first time. Compound 9 showed extremely significant anti-inflammatory and cytotoxicity activity against HeLa cell

Green Extraction of Volatile Terpenes from Artemisia annua L.

In the present study, the extraction of volatile terpenes from A. annua with supercritical CO2 (sc-CO2) was optimized by a full factorial design procedure and compared with conventional distillation. The influence of pressure (100-220 bar) and temperature (40-60 °C) on sc-CO2 extraction was investigated to obtain extracts rich in the desired components while maintaining a high yield. Extraction yields (m/m) varied from 0.62% (130 bar/40 °C) to 1.92% (100 bar/60 °C). Monoterpenes were the most abundant constituents of the sc-CO2 extracts, among which artemisia ketone (16.93-48.49%), camphor (3.

In vivo antimalarial efficacy of Artemisia afra powder suspensions.

BACKGROUND: A global death toll of 608,000 in 2022 and emerging parasite resistance to artemisinin, the mainstay of antimalarial chemotherapy derived from the Chinese herb Artemisia annua, urge the development of novel antimalarials. A clinical trial has found high antimalarial potency for aqueous extracts of A. annua as well as its African counterpart Artemisia afra, which contains only trace amounts of artemisinin. The artemisinin-independent antimalarial activity of A. afra points to the existence of other antimalarials present in the plant. However, the publication was retracted due to eth

Anaphylaxis to pine nut: cross-reactivity to Artemisia vulgaris?

The use of pine nuts, the seeds of Pinus pinea, is on the increasing in the modern Mediterranean diet. Little more than 20 cases of allergy to this tree nut have been published, and cross-reactivity with pine pollen, peanut and almond has already been reported. We describe the case of a young boy with several episodes of anaphylaxis after pine nut ingestion. Specific IgE to pine nut and Artemisia vulgaris was demonstrated by skin prick tests and in vitro determination of specific IgE, although no IgE to pine pollen or other nuts was detected. Immunoblotting of Artemisia vulgaris and pine nut r

Artemisia and Ambrosia hypersensitivity: co-sensitization or co-recognition?

BACKGROUND: Ragweed and mugwort have nearly identical flowering periods. Clinical and serological studies showed that ragweed and mugwort sensitization are often associated and this poses relevant clinical problems in patients for whom specific immunotherapy is warranted. OBJECTIVE: To establish whether the concurrent ragweed and mugwort pollen hypersensitivity is the result of co-sensitization or of co-recognition by using purified recombinant allergens. METHODS: Sensitization to ragweed and mugwort pollen was assessed by skin prick test (SPT) in all patients reporting allergic symptoms in

Dried whole plant Artemisia annua as an antimalarial therapy.

Drugs are primary weapons for reducing malaria in human populations. However emergence of resistant parasites has repeatedly curtailed the lifespan of each drug that is developed and deployed. Currently the most effective anti-malarial is artemisinin, which is extracted from the leaves of Artemisia annua. Due to poor pharmacokinetic properties and prudent efforts to curtail resistance to monotherapies, artemisinin is prescribed only in combination with other anti-malarials composing an Artemisinin Combination Therapy (ACT). Low yield in the plant, and the added cost of secondary anti-malarials
