Efficient degradation of aflatoxin B1 by Arthrobacter citreus Soilh02: Characterization, degradation products, and detoxification mechanism.

Aflatoxin B1 (AFB1) is a highly toxic mycotoxin prevalent in food and feed that poses a grave threat to human and animal health. In this study, an AFB1- degrading strain of Arthrobacter citreus, designated Soilh02, was isolated. Under optimal conditions (37 °C, pH 8.0, 72 h), the presence of Cu2+ significantly enhanced AFB1 degradation, achieving a maximum degradation rate of 93.96%. Ultra-high-performance liquid chromatography-tandem mass spectrometry (UHPLC-QTOF) identified two novel degradation products with molecular formulas C18H 20O7 and C16H14O6, and proposed plausible structures for th

Evaluation of Single and Joint Toxicity of Perfluorinated Carboxylic Acids and Copper to Metal-Resistant Arthrobacter Strains.

Perfluorocarboxylic acid compounds (PFCAs) and copper have been regarded as ubiquitous environmental contaminants in aquatic ecosystems worldwide. However, data on their possible joint toxic effects on microorganisms are still lacking. To study the combined effects of four PFCAs with different carbon chain lengths and copper, a series of experiments were conducted to explore the acute toxicity of these PFCAs in the absence and presence of copper on a metal-resistant Arthrobacter strain GQ-9 by microcalorimetry. The thermokinetic parameters, including growth rate constant (k), inhibitory ratio

Regulation of 3-ketosteroid-1-en-dehydrogenase activity of Arthrobacter globiformis cells by a respiratory chain.

It has been shown that 3-ketosteroid-1-en-dehydrogenase localized in a cytoplasmic membrane donates reducing equivalents to a respiratory chain directly which passes them over to oxygen. Microbial hydrocortisone oxidation is coupled with energy generation in the form of the H+ transmembrane potential. Electron transfer via a respiratory chain is the limiting stage in the process of hydrocortisone 1-en-dehydrogenation.

Recombinant Arthrobacter β-1, 3-glucanase as a potential effector molecule for paratransgenic control of Chagas disease.

BACKGROUND: Chagas disease is most often transmitted to humans by Trypanosoma cruzi infected triatomine bugs, and remains a significant cause of morbidity and mortality in Central and South America. Control of Chagas disease has relied mainly on vector eradication. However, development of insect resistance has prompted us to develop a paratransgenic strategy to control vectorial transmission of T. cruzi. Here, the potential role of recombinant endoglucanases as anti-trypanosomal agents for paratransgenic application is examined. The surface of T. cruzi is covered by a thick coat of mucin-like

Discovery and structural analysis of glycoside hydrolase family 176 α-1,2 glucosidase from Arthrobacter humicola A8F5.

Glycoside hydrolases (GHs) exhibit remarkable specificity dictated by the structural configuration of their target glycosidic linkages. While enzymes that process α-1,4- and α-1,6-linkages in starch or glycogen are well-characterized, those acting on less common bonds, such as α-1,2-glucosidic linkages, remain largely underexplored. In this study, we report the discovery and structural elucidation of a novel α-1,2-glucosidase from Arthrobacter humicola A8F5 (A8F5 glucosidase), representing a newly uncovered activity within the poorly characterized GH176 family. Biochemical characterizations re

Identification and characterization of cytochrome P450 1232A24 and 1232F1 from Arthrobacter sp. and their role in the metabolic pathway of papaverine.

Cytochrome P450 monooxygenases (P450s) play crucial roles in the cell metabolism and provide an unsurpassed diversity of catalysed reactions. Here, we report the identification and biochemical characterization of two P450s from Arthrobacter sp., a Gram-positive organism known to degrade the opium alkaloid papaverine. Combining phylogenetic and genomic analysis suggested physiological roles for P450s in metabolism and revealed potential gene clusters with redox partners facilitating the reconstitution of the P450 activities in vitro. Extracellular urate oxidase from Arthrobacter creatinolyticus SA1: statistical optimization, purification, and in-vitro evaluation of antihyperuricemic activity.

Urate oxidase (uricase; EC 1.7.3.3) catalyzes the oxidation of uric acid to allantoin, a highly soluble metabolite that is readily excreted. Lack of functional urate oxidase in human's leads to hyperuricemia, which is associated with gout, tumor lysis syndrome and renal complications. The present study reports statistical optimization, purification, and comprehensive characterization of a novel extracellular urate oxidase secreted by Arthrobacter creatinolyticus SA1. Plackett-Burman Design screening followed by Central Composite Design identified inoculum volume, initial pH, peptone, and yeast

Heavy metal resistance genes and plasmid-mediated quinolone resistance genes in Arthrobacter sp. isolated from Brazilian soils.

Arthrobacter sp. are Gram-positive bacilli commonly obtained from soil and in the hospital environment. These species have been reported to cause several types of infection. Heavy metals are a threat to the ecological system due to their high-levels of toxicity and the fluoroquinolones are antimicrobials widely used for the treatment of different bacterial infections. The aim of this study was to investigate the resistance to fluoroquinolone and heavy metals, the presence of plasmid-mediated resistance (PMQR) genes and heavy metals resistance (HMR) genes and the presence of plasmids in Arthrob

Influence of Al on growth, cell size and content of intracellular water of Arthrobacter sp. PI/1-95.

A distinct inhibition of the growth of a typical soil-born bacterium, Arthrobacter sp. PI/1-95, by aluminium was established. The content of intracellular water and (consequently) the cell size of the bacterium significantly increased when cells came into contact with aluminium. Both parameters were also increased when applying HCl, but Al caused the greater change, so that a distinct discrimination between the effects of aluminium on the one hand and a sole pH-effect on the other hand was possible. The presented results point to an osmo-regulative disorder connected with a malfunction of cell

Characterization of indochrome C: a novel blue pigment with cryoprotective potential from Arthrobacter antioxidans QL17T isolated from Mount Everest.

A water-soluble blue pigment, indochrome C, was isolated from Arthrobacter antioxidans QL17T and structurally characterized as a glycosylated diazaindophenol (C20H23N3O12) bearing a C-linked pyranose moiety. Its biosynthesis was strongly cold-enhanced, with yields at 5 °C and 15 °C approximately threefold higher than those at 25 °C. Pigment-deficient and overproducing mutants generated by heavy-ion beam mutagenesis confirmed its role in freeze-thaw protection. Indochrome C significantly improved bacterial survival during freezing, reduced intracellular oxidative stress, and exhibited strong AB
