Expression profile and functional characterization of odorant binding proteins in a forest pest, Dioryctria abietella (Lepidoptera: Pyralidae).

In the forest ecosystem dominated by the Pinaceae plants, this boring pest Dioryctria abietella is subject to a variety of odorants derived from host and nonhost plants, in which olfactory-related proteins enriched in antennae are key behavioral modulators for the orientation of feeding and ovipositing hosts. Here, we addressed the odorant binding protein (OBP) gene family in D. abietella. Expression profiles revealed that the majority of OBPs were abundantly expressed in the antennae at a female-biased level. A male-antenna-biased DabiPBP1 was a strong candidate for detecting type I and type

Is the morphology of Culicoides intersexes parasitized by mermithid nematodes a parasite adaptation? A morphometric approach to Culicoides circumscriptus (Diptera: Ceratopogonidae).

Mermithidae is a family of endoparasitic nematodes known to cause intersexuality in arthropods. Intersexes of the genus Culicoides parasitized by mermithids have been the object of several studies aiming to describe their particular morphology. Culicoides intersexes are specimens with male genitalia and feminized sexually dimorphic structures, i.e. antennae, mouthparts and wings. To date, these specimens have only been described qualitatively and a quantitative approach supported by statistical analysis is lacking. Here we conduct morphometric analyses of sexually dimorphic structures in a sam

Centralized brain networks controlling antennal grooming coordination.

Animals must coordinate their body parts to perform essential behaviors like grooming and locomotion. Here, we investigate the neural basis of body part coordination during antennal grooming in the fly, Drosophila melanogaster. Unilateral and bilateral grooming, targeting one or both antennae, respectively, arise from synchronized movements of the head, antennae, and forelegs. Replay of these kinematics in a biomechanical model suggests that coordination allows for unobstructed, forceful collisions between the foreleg tibiae and antennae, potentially increasing grooming efficiency. Movements o

Caste- and environment-associated differential expression of olfactory receptors in pest ants.

As human society continues to grow and evolve, so does the need for effective pest management strategies. Olfactory-mediated control methods, such as attractant and repellent compounds, are a proposed strategy for mitigating the damaging effects of some insect pests, most notably ants, that rely on olfaction for communication. To develop such compounds, it is first important to comprehensively understand the target species' olfactory transcriptome in order to guide future targeted functional characterization of relevant olfactory proteins. Here, we perform bulk RNA-seq analysis of antennae fro

Identification and expression analyses of the olfactory-related genes in different tissues' transcriptome of a predacious soldier beetle, Podabrus annulatus (Coleoptera, Cantharidae).

We sequenced and analyzed the transcriptomes from different tissues of the soldier beetle, Podabrus annulatus (Coleoptera: Cantharidae), and obtained 75.74 Gb clean reads which were assembled into 95,274 unigenes. Among these transcripts, 25,484 unigenes of highly quality were annotated. Based on annotation and tBLASTn results, we identified a total of 101 candidate olfactory-related genes for the first time, including 11 putative odorant-binding proteins (OBPs), 6 chemosensory proteins (CSP), 50 olfactory receptors (ORs), 25 gustatory receptors (GRs), 6 ionotropic receptors (IRs), and 3 senso

Observations on the ultrastructure of the oral appendages of Glenea cantor Fabricius (Coleoptera: Lamiinae).

Using scanning electron microscopy, we examined the gross and ultrastructure morphology of the mandibles, labial palpi, and mandibular palpi of adult male and female Glenea cantor beetles. The morphology of these parts, both in their gross and ultrastructure, varied significantly between males and females. The lengths of the mandible, labial palpi, and mandibular palpi were clearly noticeably longer in females than in men, which is mostly related to the oviposition mechanism. In terms of the ultrastructural morphology of the mandibles, labial palpi, and mandibular palpi, seven types of sensill

Ultrastructure of the Antennae and Sensilla of Nyssomyia intermedia (Diptera: Psychodidae), Vector of American Cutaneous Leishmaniasis.

The antennal sensilla and the antenna of females Nyssomyia intermedia, one of the main vectors of American cutaneous leishmaniasis, were studied by scanning electron microscopy. The main goal was to characterize the quantity, typology, and topography of the sensilla with particular attention to the olfactory types. The insects were captured in the city of Corte de Pedra, State of Bahia, Brazil, by CDC-type light traps and raised in a laboratory as a new colony. Fourteen well-differentiated sensilla were identified, among six cuticular types: trichoidea, campaniformia, squamiformia, basiconica,

Behavioural and electrophysiological responses of Triatoma dimidiata nymphs to conspecific faecal volatiles.

The behavioural and electrophysiological (electroantennography) responses of the first two instars of Triatoma dimidiata (Hemiptera: Reduviidae) Latreille to fresh and dry faecal headspace volatile extracts from fifth instar conspecific nymphs and synthetic compounds were analysed in this study. Recently emerged nymphs (3-5 days) aggregated around filter paper impregnated with dry faeces and around filter paper impregnated with extracts from both fresh and dry faeces. Older first instars (10-15 days) and second instars aggregated around filter paper impregnated with fresh and dry faeces, and t

Hunger is the best spice: effects of starvation in the antennal responses of the blood-sucking bug Rhodnius prolixus.

Blood-sucking insects strongly rely on olfactory cues to find their vertebrate hosts. As in other insects with different lifestyles, it has been shown that endogenous and exogenous factors modulate olfactory responses. The triatomine bug Rhodnius prolixus is an important vector of Chagas disease and a classical model for studies of physiology and behavior. In this species, the behavioral response to host-derived odorants is modulated by both the time of the day and the starvation. Here I investigated the peripheral neural mechanisms underlying these modulatory effects. For this, I measured the

Neuropeptides in the antennal lobe of the yellow fever mosquito, Aedes aegypti.

For many insects, including mosquitoes, olfaction is the dominant modality regulating their behavioral repertoire. Many neurochemicals modulate olfactory information in the central nervous system, including the primary olfactory center of insects, the antennal lobe. The most diverse and versatile neurochemicals in the insect nervous system are found in the neuropeptides. In the present study, we analyzed neuropeptides in the antennal lobe of the yellow fever mosquito, Aedes aegypti, a major vector of arboviral diseases. Direct tissue profiling of the antennal lobe by matrix-assisted laser deso

Morphological Characterization of the Sensilla from the Antennal Flagella, Maxillary Palps, and Aculei, and Electroantennogram Responses of Anastrepha obliqua (Diptera: Tephritidae) to Host Volatiles.

Insect chemoreception is essential for locating food, selecting oviposition sites, and detecting infochemicals. In tephritid fruit flies, chemosensory perception occurs primarily through sensilla on the antennal flagella, maxillary palps, and ovipositor. Identifying these sensilla provides insights into olfaction, which may lead to improvements in insect control measures. This study describes the morphology of sensilla in Anastrepha obliqua (Macquart) using scanning electron microscopy, assesses their permeability to crystal violet and silver nitrate, and evaluates antennal electrophysiologica

Identification of chemosensory genes and antennal sensilla in Nassophasis sp. (Coleoptera: Rhynchophorinae).

Phytophagous insects rely on plant volatiles to select and locate hosts for feeding or reproduction and their olfactory system is essential for detecting plant volatiles. The stem-boring pest, Nassophasis sp. damages Dendrobium and causes economic losses. Currently, there are no effective methods for its control. However, understanding the morphological and molecular basis of its olfactory system may identify new pathways for their management and control. In this study, we observed the stemborer's antennal sensilla using scanning electron microscopy, and transcriptome sequencing was undertaken

Transcriptomic analysis of male diamondback moth antennae: Response to female semiochemicals and allyl isothiocyanate.

Female semiochemicals and allyl isothiocyanate (AITC) attract moths, and the moths use odorant-degrading enzymes (ODEs) to break down the excess odor. By identifying antennae-specific ODEs, researchers have established the molecular foundation for odorant degradation and signal inactivation in insects. This enables further exploration of new pest control methods. Currently, the degradation of female semiochemicals and AITC has received limited attention, inspiring this study to identify target ODEs in diamondback moths through transcriptome analysis. Sequencing of antennae from male adults (MA

Antennal and maxillary palp morphology, and sensillar equipment, of the spruce bark beetle predators, Medetera signaticornis and Medetera infumata (Diptera: Dolichopodidae).

Many long-legged Medetera flies are natural enemies of bark beetle pests, which they detect using olfactory cues, likely through olfactory sensilla on the antennae and maxillary palps. Morphological characterisation of olfactory sensilla among insects can provide a basis for future taxonomic, phylogenetic or electrophysiological studies. Scanning electron microscopy was used to describe the morphology of olfactory organs and sensillar equipment of Medetera signaticornis and M. infumata. Three different olfactory sensillum types were found in both fly species, sensilla trichodea, s. basiconica

Identification and analysis of odorant receptors expressed in the two main olfactory organs, antennae and palps, of Schistocerca americana.

Locusts depend upon their sense of smell and provide useful models for understanding olfaction. Extending this understanding requires knowledge of the molecular and structural organization of the olfactory system. Odor sensing begins with olfactory receptor neurons (ORNs), which express odorant receptors (ORs). In insects, ORNs are housed, in varying numbers, in olfactory sensilla. Because the organization of ORs within sensilla affects their function, it is essential to identify the ORs they contain. Here, using RNA sequencing, we identified 179 putative ORs in the transcriptomes of the two m

Comparative antennal transcriptome of Apis cerana cerana from four developmental stages.

Apis cerana cerana, an important endemic honey bee species in China, possesses valuable characteristics such as a sensitive olfactory system, good foraging ability, and strong resistance to parasitic mites. Here, we performed transcriptome sequencing of the antenna, the major chemosensory organ of the bee, using an Illumina sequencer, to identify typical differentially expressed genes (DEGs) in adult worker bees of different ages, namely, T1 (1 day); T2 (10 days); T3 (15 days); and T4 (25 days). Surprisingly, the expression levels of DEGs changed significantly between the T1 period and the oth

Impact of elevated CO2 level and egg quiescence duration on gene expression in the peripheral olfactory system of Aedes aegypti.

Elevation in CO2 can significantly impact the biology of various organisms, affecting life-history traits of both aquatic and terrestrial forms, including disease-vectoring mosquitoes. For mosquitoes, this effect is accentuated by egg quiescence duration, resulting in a change in foraging of adult females. Female mosquitoes rely on their olfactory system for locating resources, such as nectar and blood. This study employs a transcriptomic approach to investigate how a projected elevation in CO2 level, under a worst-case scenario, interacts with extended egg quiescence duration to modulate the

Comparative Development of the Ant Chemosensory System.

The insect antennal lobe (AL) contains the first synapses of the olfactory system, where olfactory sensory neurons (OSNs) contact second-order projection neurons (PNs). In Drosophila melanogaster, OSNs expressing specific receptor genes send stereotyped projections to one or two of about 50 morphologically defined glomeruli [1-3]. The mechanisms for this precise matching between OSNs and PNs have been studied extensively in D. melanogaster, where development is deterministic and independent of neural activity [4-6]. However, a number of insect lineages, most notably the ants, have receptor gen
