Relationship between biochemical parameters and paraoxonase 1 activity of boar seminal plasma and semen quality.

The aim of this study was to examine the biochemical components and the parameters of antioxidant protection in the seminal plasma (SP) of boars, as well as their relationship with semen quality parameters. Thirty-six boars were included in the study, whose ejaculates were divided into two groups: Group I (good quality semen, > 70% progressively motile sperm, < 20% spermatozoa with abnormal morphology, n = 16), and Group II (poor quality semen, < 70% progressively motile sperm, > 20% spermatozoa with abnormal morphology, n = 20). Significantly higher concentrations of ionized calcium (iCa), to

HDL and Oxidation.

In this chapter, we will focus on HDLs' activity of inhibiting LDL oxidation and neutralizing some other oxidants. ApoA-I was known as the main antioxidant component in HDLs. The regulation of antioxidant capacity of HDL is mainly exhibited in regulation of apoA-I and alterations at the level of the HDL lipidome and the modifications of the proteome, especially MPO and PON1. HDL oxidation will influence the processes of inflammation and cholesterol transport, which are important processes in atherosclerosis, metabolic diseases, and many other diseases. In a word, HDL oxidation might be an effe

Correlation of oxidative stress-related biomarkers with postmenopausal osteoporosis: a systematic review and meta-analysis.

UNLABELLED: Oxidative stress (OS) is implicated in postmenopausal osteoporosis (PO). Results show an imbalance between antioxidative and oxidative markers in PO. Thus, monitoring of OS-related biomarkers and keeping balance between reactive oxygen species and antioxidant system are beneficial to the diagnosis and prognosis of PO. PURPOSE: Oxidative stress (OS) has been implicated in postmenopausal osteoporosis (PO). However, the relations between OS-related markers and PO are controversial. This study aimed to quantitatively and comprehensively assess the roles of OS-related biomarkers in PO.

Ophthalmic drugs: in vitro paraoxonase 1 inhibition and molecular docking studies.

Glaucoma is a neuropathy disorder and is generally treated by drugs. Allergic conjunctivitis is a common ophthalmologic disease. Paraoxonase 1 (PON1) is an organophosphate hydrolyzer and antiatherogenic enzyme. PON1 is known for preventing atherosclerosis through lipid-modifying features, as well as which has decisive actions of antiapoptosis, anti-inflammatory, antithrombosis, and antiadhesion antioxidant activity properties. Thus, reducing the enzyme levels in hyperthyroidism, chronic renal failure, glaucoma, diabetes mellitus, and cardiovascular diseases is a significant risk. This study wa

Why Should Psychiatrists and Neuroscientists Worry about Paraoxonase 1?

BACKGROUND: Nitro-oxidative stress (NOS) has been implicated in the pathophysiology of psychiatric disorders. The activity of the polymorphic antioxidant enzyme paraoxonase 1 (PON1) is altered in diseases where NOS is involved. PON1 activity may be estimated using different substrates some of which are influenced by PON1 polymorphisms. OBJECTIVES: 1) to review the association between PON1 activities and psychiatric diseases using a standardized PON1 substrate terminology in order to offer a state-of-the-art review; and 2) to review the efficacy of different strategies (nutrition, drugs, lifes

Blood and semen paraoxonase-arylesterase activities in normozoospermic and azoospermic men.

Paraoxonase and arylesterase enzymes are corner stones of antioxidant defence. We aimed to compare azoospermic infertile men and normozoospermic individuals with respect to total antioxidant status (TAS), total oxidant status (TOS), oxidative stress index (OSI), paraoxonase and arylesterase levels in the blood and seminal plasma. Two-hundred consecutive infertility patients and voluntarily participated were included. In the normozoospermic group, TAS, PON, arylesterase values were statistically significantly higher when compared with those in the azoospermic group, while lower TOS and OSI leve

Functionality of HDL: antioxidation and detoxifying effects.

High-density lipoproteins (HDL) are complexes of multiple talents, some of which have only recently been recognised but all of which are under active investigation. Clinical interest initially arose from their amply demonstrated role in atherosclerotic disease with their consequent designation as a major cardiovascular disease (CVD) risk factor. However, interest is no longer confined to vascular tissues, with the reports of impacts of the lipoprotein on pancreatic, renal and nervous tissues, amongst other possible targets. The ever-widening scope of HDL talents also encompasses environmental

Review: The role of paraoxonase in cardiovascular diseases.

Paraoxonase (PON) is a group of proteins present in three forms (PON1, PON2, PON3) encoded by genes PON1, PON2, and PON3. PON1 and PON3 are plasma enzymes, structurally and functionally related to HDL, while PON2 is characterized by an intracellular location. Many polymorphisms of PON have been observed. The most widely and accurately described is the single nucleotide polymorphism, which impacts the conversion of glutamine (Q) to arginine (R) and has the effect of altering the hydrolytic activity of the PON1 form. Each PON form plays an important role in the human body, and they exhibit antio

Paraoxonase 1 deficiency and hyperhomocysteinemia alter the expression of mouse kidney proteins involved in renal disease.

SCOPE: Hyperhomocysteinemia (HHcy) is associated with kidney disease and leads to atherosclerosis and thrombosis. Paraoxonase 1 (Pon1), a hydrolase that participates in homocysteine (Hcy) metabolism and is carried in the circulation on high-density lipoprotein, has also been linked to kidney disease and atherothrombosis. Pon1-knockout mice are susceptible to atherosclerosis and exhibit a kidney-associated phenotype, polyuria or urine dilution. We hypothesize that HHcy and Pon1 deficiency are toxic to kidney function because they impair metabolic pathways important for normal kidney homeostasis

Robust and Versatile OPH-Cu Phosphate Hybrid Nanoflower for Organophosphorus Compound Degradation.

Organophosphorus hydrolase (OPH), a promising biocatalyst for the degradation of organophosphorus compounds, faces application limitations due to its structural fragility. To address this, we developed a novel bioinorganic hybrid nanoflowers (HNFs) system by integrating OPH with copper phosphate via a self-assembly strategy. The HNFs exhibited significant catalytic robustness across a broad pH and temperature range. This hybrid system demonstrated enhanced tolerance to organic solvents and proteases, retaining 81% of its initial activity after five catalytic cycles. Furthermore, we successfull

HDL Function and Atherosclerosis: Reactive Dicarbonyls as Promising Targets of Therapy.

Epidemiologic studies detected an inverse relationship between HDL (high-density lipoprotein) cholesterol (HDL-C) levels and atherosclerotic cardiovascular disease (ASCVD), identifying HDL-C as a major risk factor for ASCVD and suggesting atheroprotective functions of HDL. However, the role of HDL-C as a mediator of risk for ASCVD has been called into question by the failure of HDL-C-raising drugs to reduce cardiovascular events in clinical trials. Progress in understanding the heterogeneous nature of HDL particles in terms of their protein, lipid, and small RNA composition has contributed to

[Enzymatic activity of paraoxonase depending on polymorphism Q192R of the PON1 gene in patients with primary open-angle glaucoma].

UNLABELLED: It is believed that one of the main blood enzymes that hydrolyzes oxidized lipids incorporated in lipoproteins is the calcium-dependent hydrolase of paraoxonase 1, which has a significant antioxidant effect depending on the polymorphism of the PON1 gene. PURPOSE: To genotype patients with primary open-angle glaucoma (POAG) by the Q192R polymorphism of the PON1 gene in order to identify their genetic predisposition to dyslipidemia and atherosclerosis, as well as to determe the possibility of correcting the reduced activity of the PON1 enzyme in the examined individuals by the compl

Identification of tissue-specific and common methylation quantitative trait loci in healthy individuals using MAGAR.

BACKGROUND: Understanding the influence of genetic variants on DNA methylation is fundamental for the interpretation of epigenomic data in the context of disease. There is a need for systematic approaches not only for determining methylation quantitative trait loci (methQTL), but also for discriminating general from cell type-specific effects. RESULTS: Here, we present a two-step computational framework MAGAR ( https://bioconductor.org/packages/MAGAR ), which fully supports the identification of methQTLs from matched genotyping and DNA methylation data, and additionally allows for illuminatin

Hypochlorous acid is a potent inactivator of human plasminogen at concentrations secreted by activated granulocytes.

BACKGROUND: Myeloperoxidase (MPO) neutrophils have been considered an important pathophysiological factor in oxidative stress. Mainly through generation of hypochlorous acid in the phagosome, unchecked activity may lead to inactivation of important proteins through modification of tyrosine and other residues. This has been shown for low-density lipoprotein, apolipoprotein AI and paraoxonase 1 (PON-1). Notably, plasminogen has 29 tyrosine residues and we previously demonstrated that it can be inactivated by nitration of these residues. METHODS: We hypothesized that plasminogen can also be inac
