Six adults heterozygous for phenylketonuria (PKU) ingested eight successive servings of unsweetened and aspartame (APM)-sweetened beverage at 1-hour intervals in a randomized, balanced, crossover design. In one part, the eight beverage servings were not sweetened. In the other, each of the eight beverage servings provided 600 mg of APM, a dose equivalent to the amount provided by 36 oz of an APM-sweetened diet beverage. Plasma aspartate concentration was not significantly increased after ingestion of unsweetened or APM-sweetened beverage. Similarly, ingestion of the unsweetened beverage had no

In aqueous solution, aspartame can cyclicize to form its corresponding diketopiperazine (3-carboxymethyl-6-benzyl-2,5-diketopiperazine; DKP) and methanol. We measured plasma and urinary concentrations of DKP in samples obtained from six normal adult subjects ingesting 2.2 mg DKP/kg body weight. The DKP was administered as part of a dose of 200 mg aspartame/kg body weight. DKP concentrations in plasma were below the detection limit (less than 1 microgram/ml) of the high-pressure liquid chromatographic method at each time interval after ingestion at which they were measured. Mean (+/- SD) total

This work presents a novel bola-type peptide lipid which can gelate water, organic solvents, and water/organic-solvent mixtures. In its molecular structure, an amphiphilic dipeptide aspartame (L-α-aspartyl-L-phenylalanine methyl ester) is connected at both ends of an alkylene linker. The different morphologies in the hydrogel (helical nanotapes) and the organogel (tape-like nanostructures) were visualized by energy-filtering transmission electron microscopy (EF-TEM) and energy-filtering scanning electron microscopy (FE-SEM), and the molecular arrangement was examined using X-ray diffraction (X

The use of aspartame as an artificial sweetener is prevalent in a wide range of everyday food products, potentially leading to health complications such as obesity, diabetes mellitus, autism spectrum disorders, and neurodegeneration. Aspartame has also been detected in natural water bodies at a concentration of 0.49 μg/L, yet research on its ecotoxicological effects on aquatic life remains scarce. This study aimed to investigate the potential negative effects of environmentally relevant concentrations of aspartame on the development of various tissues and organs in zebrafish embryos. We used a

Previously, we reported that oral stimulation with Gymnema sylvestre (GS), a plant that selectively suppresses sweet taste sensation in humans, delayed gastric emptying and glycemic response during and after oral glucose ingestion. It is unclear whether these responses are triggered by sweet taste sensing per se. We tested the hypothesis that the effects of sweet taste sensing involving a low-energy sweetener, aspartame, alters gastric emptying, blood glucose, and plasma insulin responses during and after the prandial phase. Nine participants rinsed their mouths with either 25 mL of water (con

BACKGROUND: Aspartame, a widely used non-nutritive sweetener, has been associated with potential neurotoxicity. However, it remains unclear whether aspartame consumption can exacerbate cerebral ischemia-reperfusion injury (CIRI), a major contributor to stroke outcomes, and the underlying mechanisms are poorly understood.
METHODS: In vivo, male C57BL/6 J mice were given free access to 0.1% or 0.2% (w/v, equivalent to human acceptable daily intake level) aspartame, for 7 days, followed by bilateral common carotid artery occlusion (BCCAO) to induce CIRI. In vitro, hippocampal neural stem cells (

Aspartame is a widely used non-nutritive sweetener suspected of having immunotoxic effects. This study evaluated the protective role of sesame oil against aspartame-induced toxicity in Swiss albino mice. Male Swiss albino mice were divided into four groups (n = 40): control, aspartame-treated (40 mg/kg/day), aspartame + sesame oil, and sesame oil alone. Over 60 days, aspartame (ASP) administration resulted in increased body weight, feed, and water intake, alongside a reduction in relative organ weights. Hematological analysis showed a significant decline in WBCs, eosinophils, and monocytes in

Conflicting reports exist concerning the effect aspartame (APM, l-aspartyl-l-phenylalanine methyl ester) has upon brain biogenic amines. In the following study, in vivo voltammetry was utilized to measure evoked extracellular dopamine (DA) levels in the striatum of rats in order to assess APM's effect. Time-course experiments revealed a significant decline in evoked extracellular DA levels within 1h of a single systemic dose (500mg/kg i.p.) when compared to vehicle-injected controls. The effect was frequency dependent and showed a significant decrease utilizing high frequency stimulation param

An important adaptation for diabetes patients is the consumption of artificial sweetener aspartame, which is present in more than thousands of food products and medicines. Several studies have shown diverse side effects of aspartame, and it is also known to form amyloid-like cytotoxic and cross-seeding-competent nanostructures. Here, we reveal a direct crosstalk between aspartame and insulin that induces their cross-seeding and coaggregation events. Insulin's cross-β structure possesses high affinity for aspartame and triggers its self-assembly into cytotoxic nanostructures. Likewise, aspartam

Aspartame, a widely used artificial sweetener, remains controversial due to neurotoxic risks from its metabolites-phenylalanine, aspartic acid, and methanol. While epidemiological studies link artificial sweeteners to cerebrovascular disease, the molecular mechanisms connecting aspartame to ischemic stroke are unclear. This study integrates network toxicology and molecular docking to identify key targets and pathways. Potential aspartame targets were predicted using STITCH, SwissTargetPrediction, and SEA databases, while ischemic stroke-related genes were retrieved from GeneCards, OMIM, and TT

This study investigates the impact of aspartame consumption on pubertal timing in females. The research employs both human and rat models to explore underlying mechanisms. In the Taiwan Pubertal Longitudinal Study (2018-2022), 858 girls aged 6-12 were assessed for aspartame intake and puberty outcomes. Concurrently, female Sprague Dawley rats were exposed to low (30 mg/kg) or high (60 mg/kg) doses of aspartame from prenatal to postnatal stages. Results demonstrate that aspartame disrupts the hypothalamic-pituitary-gonadal (HPG) axis in rats, causing mitochondrial dysfunction and reduced ovaria
