A case of chronic sinus disease due to morphologically atypical Aspergillus flavus is described. Multiple fungal isolates sporulated poorly or not at all, displaying unusual color and microscopic morphology, including the absence of typical vesicles and phialides, which caused the isolates to resemble several other fungal genera superficially. The patient received multiple antifungal therapies over at least 10 years with various azole drugs, including voriconazole, itraconazole, and posaconazole. We speculate that this lengthy exposure to azole antifungal drugs may have caused or promoted the

Fungal involvement in rhinosinusitis is classified into four major forms: allergic, mycetoma, chronic invasive (indolent) and acute invasive (fulminant). It can become life threatening if not diagnosed and treated properly. The preliminary diagnosis is usually made by nasal endoscopy and computed tomography (CT) imaging, but tissue biopsy and culture are of vital importance in confirming the disease and in planning treatment. We present a case of allergic fungal rhinosinusitis (AFS) caused by Aspergillus flavus. The clinical manifestation of the disease was the presence of an extensive left na

Aspergillus flavus produces aflatoxin B1, a Group 1 carcinogen that causes severe economic and agricultural losses and can lead to invasive aspergillosis in humans. During its growth, A. flavus develops hyphae that form a continuous mycelium, which is associated with host colonization and toxin production. Transcription factors are considered regulators of fungal development and pathogenicity. Among these families, bZIP proteins are associated with growth, metabolism, and stress tolerance. In A. fumigatus, the bZIP transcription factor ZipD regulates calcium signaling pathways and cell wall in

A patient with granulomatous mediastinitis due to Aspergillus flavus is described. A 22 year old black man presented with cough, fever and a right hilar mass. Mediastinal biopsies revealed granulomatous fibrosing mediastinitis with fungal elements compatible with aspergillus species. A flavus was isolated on culture of this material and later from sputum and bronchial washings. Studies of the patient's immune status revealed normal humoral and cellular immunity. No underlying neoplasm was found. The patient was treated with amphotericin B and 5-fluorocytosine but esophageal and superior vena c

Aspergillus flavus and Aspergillus parasiticus are important plant pathogens and causal agents of pre- and postharvest rots of corn, peanuts, and tree nuts. These fungal pathogens cause significant crop losses and produce aflatoxins, which contaminate many food products and contribute to liver cancer worldwide. Aqueous preparations of Tulbaghia violacea (wild garlic) were antifungal and at 10 mg/ml resulted in sustained growth inhibition of greater than 50% for both A. flavus and A. parasiticus. Light microscopy revealed that the plant extract inhibited conidial germination in a dose-dependent

INTRODUCTION: The generally recognized as safe (GRAS) fungus Aspergillus oryzae has been used for millennia in the production of traditional Asian fermented foods and beverages. Domestication has led to genomic and phenotypic adaptations that distinguish A. oryzae from its wild relative, Aspergillus flavus. While differences between these species have been partially characterized, their comparative production of volatile compounds during food fermentation remains poorly understood.
METHODS: We evaluated alpha-amylase activity, aflatoxin production, taste attributes using an electronic tongue,

Invasive pulmonary aspergillosis is typically caused by a single Aspergillus species, most frequently Aspergillus fumigatus. Here we report that a lung transplant recipient developed invasive aspergillosis due to a mixed infection caused by Aspergillus flavus and A. fumigatus. The implications for this unusual finding are discussed.

The in vitro susceptibility of 105 clinical and environmental strains of Aspergillus fumigatus and Aspergillus flavus to antifungal drugs, such as amphotericin B, azoles, and echinocandins was evaluated by the broth microdilution method proposed by the European Committee on Antimicrobial Susceptibility Testing (EUCAST). Following the EUCAST-proposed breakpoints, 20% and 25% of the clinical and environmental isolates of A. fumigatus, respectively, were found to be resistant to itraconazole (Minimal Inhibitory Concentration, MIC>2.0mg/L). Voriconazole showed good activity against A. fumigatus an

Aspergillus flavus is a ubiquitous filamentous fungus that poses significant threats as both a causative agent of invasive aspergillosis and a major source of crop contamination due to production of aflatoxin B1 (AFB1). Sugars are essential for fungal metabolism, cell wall biosynthesis, and virulence, yet sugar transporters (STPs) in A. flavus remain largely uncharacterized. In this study, we systematically investigated three putative STP genes (G4B84_001982, G4B84_005374, and G4B84_009351) by comprehensive functional characterization of gene deletion mutants. Growth assays revealed that G4B84

Aspergillus flavus is a commonly encountered pathogen responsible for fungal rhinosinusitis (FRS) in arid regions. The species is known to produce aflatoxins, posing a significant risk to human health. This study aimed to investigate the aflatoxin profiles of A. flavus isolates causing FRS in Sudan. A total of 93 clinical and 34 environmental A. flavus isolates were studied. Aflatoxin profiles were evaluated by phenotypic (thin-layer and high-performance chromatography) and genotypic methods at various temperatures and substrates. Gene expression of aflD and aflR was also analyzed. A total of

Aspergillus flavus, a ubiquitous and saprophytic fungus, is the second most common cause of aspergillosis worldwide. Several mechanisms contribute to the establishment of the fungal infection. Extracellular vesicles (EVs) have been described as "virulence factor delivery bags" in several fungal species, demonstrating a crucial role during the infection. In this study, we evaluated production of A. flavus EVs and their immunomodulatory functions. We verified that A. flavus EVs induce macrophages to produce inflammatory mediators, such as nitric oxide, tumor necrosis factor alpha (TNF-α), interl

Aspergillus spp. rarely cause mycetomata. We report a patient with diabetes and nephrotic syndrome with Aspergillus flavus mycetoma of the back, with the development of an epidural abscess, diskitis and vertebral osteomyelitis. The patient was successfully treated with decompressive laminectomy and a 14-month itraconazole regimen. Serial serum itraconazole levels and quantitative Aspergillus antigen levels were performed. This is the second reported and first extrapedal case of mycetoma caused by A. flavus.

Aspergillus endocarditis is associated with a very high mortality. Of approximately 67 cases reported in the English language literature, there have been only two known survivors. This report describes a patient with Aspergillus flavus endocarditis after mitral valve annuloplasty who recovered with combined surgical and antifungal therapy. This is the first successfully treated case due to A. flavus and the first involving an annuloplasty ring.

The mycotoxin-producing fungi contamination of foodstuffs and agricultural commodities is a serious problem. In this study, the efficacy of acetic acid (AcA), chitosan (CS), and CS with extra AcA (CS/extra AcA) against Aspergillus flavus (A. flavus) in vitro and in vivo were investigated. Results showed that CS/extra AcA strongly inhibited the mycelial growth and aflatoxins production of A. flavus at lower concentrations than CS (MIC > 16.0 mg/mL) or AcA (MIC: 0.31 %). Significant inhibition was achieved in the presence of 4.0/0.12 and 8.0/0.08 CS (mg/mL)/extra AcA (%). CS/extra AcA exhibited

To get a better understanding of the possible role of proteases in the pathogenesis of fungal keratitis, the extracellular proteases of a clinical isolate of Aspergillus flavus, from a severe case of keratitis, were identified and partially characterized. This strain, designated CU226/88, was grown with a variety of substrates as nitrogen sources, under conditions that would be expected to derepress the production of extracellular proteases. When grown on minimal medium with milk protein as a nitrogen source, the fungus appeared to produce primarily a metalloprotease, which has a zinc cofactor

Aspergillus section Flavi encompasses multiple species, with A. flavus being significant for human health because of its dual role as a major aflatoxin producer and opportunist. However, the mechanisms underlying its virulence remain incompletely understood. This study evaluates the pathogenic potential of A. flavus and its relatives using the Galleria mellonella infection model. Twenty-six A. flavus isolates (clinical and environmental) and 17 relatives/domesticated species were tested in G. mellonella, with larval survival monitored over 7 d. Histology, direct microscopy, and culture were us

PURPOSE: Fungal keratitis (FK) is a major cause of corneal blindness, with substantial variability in disease severity that cannot be fully explained by host factors. This study investigated whether endogenous mycovirus infection contributes to differences in fungal pathogenicity.
METHODS: Fungal isolates from 38 patients with FK were analyzed by metagenomic sequencing. A novel viral RNA identified in an Aspergillus flavus isolate was characterized by sequence and phylogenetic analyses. Virus-cured strains were generated and compared with virus-infected strains using phenotypic assays, transc
