Aphanomyces astaci in Mexico: A new haplotype from dwarf crayfish Cambarellus montezumae.

The crayfish plague is an emerging infectious disease caused by the pathogen Aphanomyces astaci (Oomycota), which is responsible for the decimation of Eurasian freshwater crayfish. This pathogen can coexist with the North American crayfish. These are chronic carriers of the disease as consequence of an immune response that can contain the growth of the pathogen without killing it. The origin of A. astaci locates in the southeastern United States and coincides with the origin of the family Cambaridae. This diverse family of decapods is distributed in North America from southern Canada to Hondur

eDNA monitoring as a tool for evaluating the reintroduction of Austropotamobius pallipes after a crayfish plague outbreak.

The crayfish plague, a severe disease caused by the oomycete Aphanomyces astaci, is responsible for most population declines of susceptible crayfish in Europe. This pathogen has been devastating native populations of Austropotamobius pallipes since the 1970s in the Iberian Peninsula. In this study, we report a massive mortality event in one of the most important Spanish populations of A. pallipes. We aimed to: (i) identify the cause of the mortality, and (ii) evaluate the reintroduction viability of the species. Over the course of six months, we used environmental DNA (eDNA) and traditional tr

Dataset of the de novo assembly and annotation of the marbled crayfish and the noble crayfish hepatopancreas transcriptomes.

Crayfish plague disease, caused by the oomycete pathogen Aphanomyces astaci represents one of the greatest risks for the biodiversity of the freshwater crayfish. This data article covers the de novo transcriptome assembly and annotation data of the noble crayfish and the marbled crayfish challenged with Ap. astaci. Following the controlled infection experiment (Francesconi et al. in Front Ecol Evol, 2021, https://doi.org/10.3389/fevo.2021.647037 ), we conducted a differential gene expression analysis described in (Boštjančić et al. in BMC Genom, 2022, https://doi.org/10.1186/s12864-022-08571-z

Not so fast: giant interneurons control precise movements of antennal scales during escape behavior of crayfish.

High-speed video recordings of escape responses in freely behaving crayfish revealed precisely coordinated movements of conspicuous head appendages, the antennal scales, during tail-flips that are produced by giant interneurons. For tail-flips that are generated by the medial giants (MG) in response to frontal attacks, the scales started to extend immediately after stimulation and extension was completed before the animal began to propel backwards. For tail-flips that are elicited by caudal stimuli and controlled by the lateral giants (LG), scale extensions began with significant delay after t

Self-limiting outbreak of crayfish plague in an Austropotamobius pallipes population of a river basin in the Abruzzi region (central Italy).

Crayfish plague, caused by the oomycete Aphanomyces astaci, is a serious disease of European freshwater crayfish and has eliminated entire populations in several European countries. In September 2011, mortality was observed among the Austropotamobius pallipes population of a river basin in the Abruzzi region (central Italy), and A. astaci DNA was detected by PCR in dead crayfish. A systematic survey was carried out to evaluate the spread and the effects of the plague in the river basin. The source of the outbreak remained unknown since North American crayfish species, which frequently act as s

Characterisation of the noble crayfish immune response to oomycete-derived immunostimulants.

The invasive oomycete pathogen Aphanomyces astaci significantly threatens native European crayfish populations, prompting investigations towards the effects of protective immunostimulation on the immune response of the vulnerable noble crayfish (Astacus astacus). Here, we evaluate the effect of three oomycete-derived immunostimulant treatments: laminarin (β-1,3-glucan found within the Ap. astaci cell wall), inactivated Ap. astaci spores and Ap. astaci hyphal homogenate. Our findings reveal immediate changes in the noble crayfish total haemocyte count (THC), differential haemocyte count (DHC),

The functional role of Daphnia in the host-pathogen interaction of crayfish and the crayfish plague disease agent (Aphanomyces astaci).

Pathogen spores have been recognized as prey with implications for resource dynamics, energy transfer and disease transmission. In aquatic ecosystems, filter-feeders are able to consume such motile forms of pathogens that can cause severe disease in susceptible hosts. The interactions between European crayfish and the crayfish plague pathogen Aphanomyces astaci are of particular conservation interest. In this study, we aim to evaluate the ecological interactions between Ap. astaci, its host Astacus astacus and individuals of the genus Daphnia, filter-feeding planktonic crustaceans. Our focus w

World of Crayfish™: a web platform towards real-time global mapping of freshwater crayfish and their pathogens.

Freshwater crayfish are amongst the largest macroinvertebrates and play a keystone role in the ecosystems they occupy. Understanding the global distribution of these animals is often hindered due to a paucity of distributional data. Additionally, non-native crayfish introductions are becoming more frequent, which can cause severe environmental and economic impacts. Management decisions related to crayfish and their habitats require accurate, up-to-date distribution data and mapping tools. Such data are currently patchily distributed with limited accessibility and are rarely up-to-date. To addr

Functions of potassium channels blocked by low micromolar 4-aminopyridine in the crayfish nervous system.

4-aminopyridine (4-AP) is a potassium channel blocker that has been used to treat patients with multiple sclerosis and Lambert-Eaton disease. The concentration of this drug in the blood of patients was estimated to be in low or submicromolar range. Animal studies have shown that 4-AP at such low concentration selectively blocks a subset of channels in Kv1 or Kv3 families. The crayfish opener neuromuscular junction and ventral superficial flexor (VSF) preparations were used to examine functions of K+ channels blocked by low concentrations of 4-AP. At opener motor axons, intracellular recordings

Cambarus polypilosus, a new species of stream-dwelling crayfish (Decapoda: Cambaridae) from the Western Highland Rim of Tennessee, USA.

Cambarus polypilosus sp. nov. is a stream-dwelling crayfish endemic to tributaries in the Buffalo and lower Duck River drainage, and a reach of the lower Tennessee River in the Western Highland Rim of West-Central Tennessee, U.S.A. The new species is closely allied to the three members of the former subgenus Glareocola, but can be differentiated from each by a combination of characters, including body size, coloration, spination, setation, and form I male gonopod morphology. Several meristic measurements and ratios also differentiate C. polypilosus sp. nov. from Cambarus friaufi, to which it i

Sedentary behavior as a factor in determining lateral line contributions to rheotaxis.

Rheotaxis is a robust, multisensory behavior with many potential benefits for fish and other aquatic animals. Visual (optic flow) cues appear to be sufficient for rheotaxis, but other sensory cues can clearly compensate for the loss of vision. The role of various non-visual sensory systems, in particular the flow-sensing lateral line, is poorly understood, largely because of widely varying methods and sensory conditions for studying rheotaxis. Here, we examine how sedentary behavior under visually deprived conditions affects the relative importance of lateral line cues in two species: one that

New Active Biopolymers and Chitosan-Based Films from Non-Native Crayfish Shell.

Freshwater crayfish impact ecosystems and the food industry. The aim of the study was to valorize shells from the invasive crayfish, Faxonius limosus, to extract novel bioactive biopolymers and prepare biodegradable films, thereby contributing to economic and environmental sustainability. Chitin from crayfish shells was isolated and converted into chitosan through a new protocol to retain astaxanthin, with a shorter procedure, reduced use of organic solvent and energy consumption. The resulting biopolymers were used to prepare chitosan-based films. Thermal, spectroscopic, barrier properties, a

Temperature affects predation of schistosome-competent snails by a novel invader, the marbled crayfish Procambarus virginalis.

The human burden of environmentally transmitted infectious diseases can depend strongly on ecological factors, including the presence or absence of natural enemies. The marbled crayfish (Procambarus virginalis) is a novel invasive species that can tolerate a wide range of ecological conditions and colonize diverse habitats. Marbled crayfish first appeared in Madagascar in 2005 and quickly spread across the country, overlapping with the distribution of freshwater snails that serve as the intermediate host of schistosomiasis-a parasitic disease of poverty with human prevalence ranging up to 94%

Differential biochemical and behavioral responses induced by cocaine and benzoylecgonine exposure to the red swamp crayfish Procambarus clarkii.

Cocaine (COC) and its main metabolite, the benzoylecgonine (BE), are the main illicit drugs measured in aquatic system worldwide, with concentrations up to hundreds of ng/L. Although their current environmental concentrations are low these molecules can induce adverse effects at sub-individual level in non-target organisms. In contrast, the information at individual and behavioral level are still scant. The present study aimed at investigating biochemical and behavioral effects induced by 14-days exposure to environmentally relevant concentrations (50 ng/L and 500 ng/L) of COC and BE towards P

[Haff disease - are we threatened with another epidemic?].

Haff disease is a group of symptoms caused by rhabdomyolysis following ingestion of some species of fish and crayfish. Pathophysiology remains unknown. Outbreaks of the Haff disease have been reported in many regions of the world. In this article we present the case of a 38-years-old patient, professional fisherman, suspected of suffering from Haff disease. He developed symptoms of diffuse myalgia, headache, chest pressure, brown-colored urine and elevated blood pressure. Predominant laboratory abnormalities were elevated serum creatine kinase and creatinine concentrations. During hospitalizat

DNA of crayfish spermatozoa as a target of waterborne pesticides - An ex vivo approach as a tool to short-term spermiotoxicity screening.

The spermiotoxic properties of aquatic contaminants might be the cause of low fertilization rate and decreased prolificacy, affecting the success of the impacted populations. The genotoxic potential of pesticides in spermatozoa as an undesirable effect on non-target organisms, namely aquatic invertebrates with external fertilization, emerges as a key question in ecogenotoxicological research. Thus, this study aimed to clarify if DNA integrity of red swamp crayfish (Procambarus clarkii) spermatozoa is affected by waterborne pesticides at environmentally relevant concentrations. By adopting an e

Ecological plasticity and commercial impact of invasive marbled crayfish populations in Madagascar.

BACKGROUND: The marbled crayfish (Procambarus virginalis) is a monoclonal, parthenogenetically reproducing freshwater crayfish species that has formed multiple stable populations worldwide. Madagascar hosts a particularly large and rapidly expanding colony of marbled crayfish in a unique environment characterized by a very high degree of ecological diversity. RESULTS: Here we provide a detailed characterization of five marbled crayfish populations in Madagascar and their habitats. Our data show that the animals can tolerate a wide range of ecological parameters, consistent with their invasive

Aphanomyces astaci genotypes involved in recent crayfish plague outbreaks in central Italy.

The oomycete Aphanomyces astaci is the causative agent of crayfish plague in native European freshwater crayfish. Molecular analyses showed that several distinct genotype groups of this pathogen, apparently associated with different original host taxa, are present in Europe. Tracking their distribution may contribute to understanding the introduction pathways of A. astaci. We used microsatellite markers to genotype the pathogen strains involved in 7 mass mortalities of the endangered indigenous crayfish Austropotamobius pallipes that occurred between 2009 and 2016 in the Abruzzi and Molise reg
