Simultaneous detection of canine astrovirus and canine kobuvirus by duplex qPCR: validation and evidence of extraintestinal viral RNA in naturally infected dogs.

Canine astrovirus (mamastrovirus 5 [MAstV5], formerly CaAstV; family Astroviridae, taxon species Mamastrovirus canis) and canine kobuvirus (aichivirus A2 [AiV-A2], formerly CaKoV; Picornaviridae, Kobuvirus aichi) are emerging enteric viruses increasingly detected in both diarrheic and subclinical dogs. Although primarily associated with gastrointestinal illness, recent evidence suggests a potential for systemic dissemination, which remains insufficiently explored. To improve detection, we developed and validated a duplex quantitative real-time PCR (dqPCR) assay for simultaneous identification

Astrovirus infection alters gut microbial communities in a widespread neotropical bat across human-modified landscapes.

Astroviruses are becoming a growing concern in public and veterinary health. In humans, astrovirus infections can cause severe diarrhea and may lead to neuropathological encephalitis, whereas in wildlife, these enteropathogenic viral infections often lack overt symptoms and thus remain unnoticed. Yet their close interaction with the host's gastrointestinal microbiome might drive cascading effects with disadvantages for host health. Bats harbor many zoonotic viruses without showing signs of disease, and many species move freely along the gradient from pristine to agricultural landscapes. To bet

Astrovirus-Associated Polioencephalomyelitis in an Alpaca.

An 8-year-old alpaca was admitted to the emergency service of the Clinic for Ruminants in Bern due to a reduced general condition and progressive neurological signs. Despite supportive treatment, its condition deteriorated and the animal had to be euthanized. Histopathological analysis revealed a severe non-suppurative polioencephalomyelitis with neuronal necrosis, most likely of viral origin. We detected abundant neuronal labelling with antibodies directed against two different epitopes of Bovine Astrovirus CH13/NeuroS1 (BoAstV-CH13/NeuroS1), which is a common viral agent associated with non-

[Acute Viral Gastroenteritis: Viruses Other Than Norovirus].

Acute Viral Gastroenteritis: Viruses Other Than Norovirus Abstract. Norovirus is the leading cause of acute viral gastroenteritis. Norovirus is highly contagious, thus outbreaks of norovirus in hospitals and long-term care facilities are feared. Usually, stool samples of patients with a potentially viral gastroenteritis are first checked for the presence of norovirus. In recent years, sapovirus and astrovirus were increasingly reported as cause of acute gastroenteritis. Outbreaks of acute viral gastroenteritis caused by sapovirus or astrovirus are hardly distinguishable from those caused by no

Coinfection of parvovirus and astrovirus in gout-affected goslings.

Outbreaks of gosling gout have occurred in China since 2017 and caused a considerable economic impact on the poultry industry. While gosling astrovirus (GoAstV) is believed to be the main causal pathogen of gout, the full-blown disease of gout cannot be well reproduced by infecting the goslings with GoAstV, suggesting the possibility of other infectious agents being involved with the development of gosling gout. To assess other possible infectious agents, we collected tissues from gout-affected goslings in 12 goose farms in China, followed by PCR detection of GoAstV, goose reovirus (GRV), goos

Novel human astroviruses: Novel human diseases?

Astroviruses are small, non-enveloped, single-stranded positive RNA viruses that belong to the Astroviridae family. While classical human astroviruses (HAstV) are a well-recognized cause of acute non-bacterial diarrhea among young children worldwide, novel astroviruses, named HAstV-MLB and HAstV-VA/HMO, have been identified recently in humans by molecular assays. They are phylogenetically more related to animal astroviruses than to classical human astroviruses, thus suggesting cross-species transmission. Serological studies demonstrated a surprisingly high seroprevalence in certain populations

Indication of Cross-Species Transmission of Astrovirus Associated with Encephalitis in Sheep and Cattle.

We report the identification of a neurotropic astrovirus associated with encephalitis in a sheep. This virus is genetically almost identical to an astrovirus recently described in neurologically diseased cattle. The similarity indicates that astroviruses of the same genotype may cause encephalitis in different species.

Occurrence and Role of Selected RNA-Viruses as Potential Causative Agents of Watery Droppings in Pigeons.

The diseases with watery droppings (diarrhea and/or polyuria) can be considered some of the most severe health problems in domestic pigeons of various ages. Although they do not always lead to bird death, they can contribute to poor weight gains and hindered development of young pigeons and, potentially, to poor racing results in sports birds. The gastrointestinal tract disorders of pigeons may be of various etiology, but some of the causative agents are viral infections. This review article provides information collected from scientific reports on RNA-viruses belonging to the Astroviridae, Pi

Expression of the C-terminal ORF2 protein of duck astrovirus for application in a serological test.

Duck astrovirus (DAstV) is an important pathogen causing duck viral hepatitis (DVH), a highly contagious and fatal disease in young ducklings. To provide an antigen for a diagnostic serum test, the C-terminus of DAstV ORF2 protein was expressed in Escherichia coli. Four positive and 30 negative sera were used to validate the purified ORF2 protein by developing an indirect enzyme-linked immunosorbent assay (ELISA). No cross-reactions were found against other duck pathogens, including duck hepatitis A virus, duck plague herpesvirus, duck reovirus, Newcastle disease virus, and Riemerella anatipes

Isolation and genomic characterization of one novel goose astrovirus causing acute gosling gout in China.

Novel goose astrovirus (NGAstV) is a member of the genus Avain Avastrovirus (AAstV) and the family Astroviridae. NGAstV-associated gout disease has caused huge economic losses to the goose industry worldwide. Since early 2020, NGAstV infections characterized by articular and visceral gout emerged continuously in China. Herein, we isolated a GAstV strain from goslings with fatal gout disease and sequenced its complete genome nucleotide sequence. Then we conducted systematic genetic diversity and evolutionary analysis. The results demonstrated that two genotypic species of GAstV (GAstV-I and GAs

Emerging fatal gout disease in Chinese goslings linked to acute kidney injury induced by novel goose astrovirus infection.

A novel goose astrovirus (GAstV) has broken out across China in recent years, causing widespread damage to the poultry industry. In goslings infected with GAstV, the leading cause of death is visceral gout. However, our understanding of the mechanism of gout formation in GAstV infection is largely inadequate. The aim of this study was to examine the pathogenicity of a GAstV strain and explore the molecular mechanisms of visceral gout caused by viral infection in goslings. The virulent GAstV strain HR2105/1 was effectively isolated from the visceral tissue of goslings in gout-affected areas. Th

Characterization of novel pesti- and astro-like viruses in hatchery-reared European seabass (Dicentrarchus labrax) associated with health disorders and low mortality.

European seabass is a major farmed species in Mediterranean aquaculture. Its early life stages are very sensitive to sanitary and environmental conditions and are therefore closely monitored. In this study, we characterized new viruses recently isolated from fingerlings in a hatchery showing whitish three-dimensional lesions on the fins and sides and distended abdomens and investigated their pathologic potential. A cytopathic effect was observed 17 days post-inoculation (dpi) of homogenized internal organs at 20 °C on a CHSE214 cell line. Viral presence was confirmed using the indirect fluores

Experimental Porcine Astrovirus 4-Associated Tracheitis and Bronchitis in Pigs.

Porcine astrovirus 4 (PoAstV4) is an emerging pathogen that has previously been associated with respiratory disease in piglets. The objective of this study was to inoculate naïve piglets with PoAstV4 and characterize the host-pathogen interaction and pathological lesions. Cesarean-derived colostrum-deprived (CDCD) piglets were inoculated intratracheally and intranasally with PoAstV4 PCR-positive tissue homogenate previously screened for the presence of other primary pathogens with next-generation sequencing (NGS). Nasal and fecal swabs for PCR identification of PoAstV4 were collected pre and p

Establishment and application of multiplex real-time qPCR detection for Tembusu virus and goose astrovirus genotypes 1 and 2.

The concurrent infection and epidemic of goose astrovirus type 1 (GoAstV-1), goose astrovirus type 2 (GoAstV-2), and Tembusu virus (TMUV) in geese have resulted in substantial economic losses to the goose industry. Therefore, establishing a rapid and accurate diagnostic method for these three viruses is imperative. In the present study, we designed specific primers targeting the Cap gene of GoAstV and the E gene of TMUV and developed a SYBR Green I-based multiplex real-time qPCR method for the simultaneous detection of GoAstV-1, GoAstV-2, and TMUV. The results revealed that the R2 values for t

Complete genome sequence of an astrovirus identified in a domestic rabbit (Oryctolagus cuniculus) with gastroenteritis.

A colony of domestic rabbits in Tennessee, USA, experienced a high-mortality (~90%) outbreak of enterocolitis. The clinical characteristics were one to six days of lethargy, bloating, and diarrhea, followed by death. Heavy intestinal coccidial load was a consistent finding as was mucoid enteropathy with cecal impaction. Preliminary analysis by electron microscopy revealed the presence of virus-like particles in the stool of one of the affected rabbits. Analysis using the Virochip, a viral detection microarray, suggested the presence of an astrovirus, and follow-up PCR and sequence determinatio

Viral metagenomics reveals diverse viruses in the fecal samples of children with diarrhea.

Diarrhea is the third leading cause of death in developing countries in children under the age of five. About half a million children die of diarrhea every year, most of which in developing countries. Viruses are the main pathogen of diarrhea. In China, the fecal virome of children with diarrhea has been rarely studied. Using an unbiased viral metagenomics approach, we analyzed the fecal virome in children with diarrhea. Many DNA or RNA viruses associated with diarrhea identified in those fecal samples were mainly from six families of Adenoviridae, Astroviridae, Caliciviridae, Parvoviridae, Pi

Human Astroviruses: A Tale of Two Strains.

Since the 1970s, eight closely related serotypes of classical human astroviruses (HAstV) have been associated with gastrointestinal illness worldwide. In the late 2000s, three genetically unique human astrovirus clades, VA1-VA3, VA2-VA4, and MLB, were described. While the exact disease associated with these clades remains to be defined, VA1 has been associated with central nervous system infections. The discovery that VA1 could be grown in cell culture, supports exciting new studies aimed at understanding viral pathogenesis. Given the association of VA1 with often lethal CNS infections, we tes

Natural selection dominates the codon preference and host adaptability of the novel goose astrovirus related to gout symptons, with non-structural protein playing a key role.

Since 2017, the primary cause of fatal gout in Chinese goslings has been identified as new goose astrovirus (N-GoAstV). Its infection can progress to severe liver and kidney damage, cause a mortality rate reaching up to 50 %, significantly impacting China's poultry industry. In this study, we conducted an assessment of codon usage bias (CUB) and host adaptation on the coding sequences of 179 N-GoAstVs derived from different hosts, including chickens, ducks, geese, and muscovy ducks. The results indicated that the complete coding sequences (CCDs) and open reading frames (ORFs) of N-GoAstV prefe
