Evolutionarily conserved protein sequences of influenza a viruses, avian and human, as vaccine targets.

BACKGROUND: Influenza A viruses generate an extreme genetic diversity through point mutation and gene segment exchange, resulting in many new strains that emerge from the animal reservoirs, among which was the recent highly pathogenic H5N1 virus. This genetic diversity also endows these viruses with a dynamic adaptability to their habitats, one result being the rapid selection of genomic variants that resist the immune responses of infected hosts. With the possibility of an influenza A pandemic, a critical need is a vaccine that will recognize and protect against any influenza A pathogen. One

Immunocytochemical study with an anti-transferrin binding protein serum: a marker for avian oligodendrocytes.

We have investigated immunocytochemically the localization of a transferrin binding protein (TfBP) in adult CNS of avian and mammalian species using a polyclonal antibody raised against the protein purified from hen oviduct membranes (alpha OV-TfBP). TfBP has recently been shown to be HSP108. An overall strong immunoreactivity was revealed in most parts of the avian brains, especially in the white matter. The main immunoreactivity originated in small, intensely reacting cells interpreted as oligodendrocytes. The density of TfBP-labeled oligodendrocytes of the avian brains was generally proport

Construction and immunogenicity evaluation of avian Escherichia coli-derived bacterial biomimetic vesicles displaying H9 subtype avian influenza virus HA1 protein.

Co-infection with H9N2 subtype avian influenza virus (AIV) and avian pathogenic Escherichia coli (APEC) is common in poultry, yet no vaccine is currently available that simultaneously prevents both infections. Biomimetic bacterial vesicles (BBVs) derived from avian E. coli are a platform for delivering heterologous subunit antigens. In this study, an antigen display strategy was adopted, which involved the genetic fusion of the H9 hemagglutinin subunit HA1 to the outer membrane-anchored protein cytolysin A (ClyA), followed by high-pressure homogenization to prepare BBV-HA1. Intranasal immuniza

Comparative Analysis of Epidermal Differentiation Genes of Crocodilians Suggests New Models for the Evolutionary Origin of Avian Feather Proteins.

The epidermis of amniotes forms a protective barrier against the environment and the differentiation program of keratinocytes, the main cell type in the epidermis, has undergone specific alterations in the course of adaptation of amniotes to a broad variety of environments and lifestyles. The epidermal differentiation complex (EDC) is a cluster of genes expressed at late stages of keratinocyte differentiation in both sauropsids and mammals. In the present study, we identified and analyzed the crocodilian equivalent of the EDC. The gene complement of the EDC of both the American alligator and t

Multiantigen epitope fusion recombinant proteins from capsids of serotype 4 fowl adenovirus induce chicken immunity against avian Angara disease.

Avian Angara disease caused by fowl adenovirus serotype 4 (FAdV-4) has spread widely and brought economic losses to the poultry industry in some countries. Effective vaccines for Angara disease control are currently lacking. In this study, four capsid proteins (hexon, penton, fiber1 and fiber2) from FAdV-4 were selected, and their optimal efficient antigenic epitopes predicted by bioinformatics software were tandemly linked with the flexible linker GGGGS. Based on their amino acid sequences, the DNA sequences for the genes encoding the multiantigen epitope tandem proteins (MAETPs) FAdV4:F1, FA

Wnt14 regulates the cellular inflammation induced by avian reovirus and interacts with the viral σB protein.

Avian reovirus (ARV) typically induces viral arthritis or tenosynovitis in chickens. The ARV structural protein σB plays a crucial role in viral replication and regulates cellular signaling pathways through interactions with host proteins. In this study, we demonstrate that both ARV infection and ARV σB protein can activate intracellular Wnt signaling pathways and induce inflammatory responses based on qPCR and Western blot analyses in HD11 cells. Interestingly, ARV infection can inhibit β-catenin ubiquitination and upregulate its protein expression levels. To further investigate the mechanism

Glycosylation of the viral attachment protein of avian coronavirus is essential for host cell and receptor binding.

Avian coronaviruses, including infectious bronchitis virus (IBV), are important respiratory pathogens of poultry. The heavily glycosylated IBV spike protein is responsible for binding to host tissues. Glycosylation sites in the spike protein are highly conserved across viral genotypes, suggesting an important role for this modification in the virus life cycle. Here, we analyzed the N-glycosylation of the receptor-binding domain (RBD) of IBV strain M41 spike protein and assessed the role of this modification in host receptor binding. Ten single Asn-to-Ala substitutions at the predicted N-glycos

The amino acid sequence of ovocleidin 17, a major protein of the avian eggshell calcified layer.

The amino acid sequence of ovocleidin 17, a major protein of the chicken eggshell calcified layer, contains 142 amino acids including 2 phosphorylated serines. Data base searches show that ovocleidin belongs to a heterogeneous group of proteins consisting of a single C-type lectin domain (CTL). The most similar sequences with an average of 30% identical amino acids were those of pancreatic stone protein (lithostathine) and lectins and anticoagulant proteins from snake venom.

Copper binding to the N-terminal tandem repeat regions of mammalian and avian prion protein.

Mammalian prion protein (PrP) is a normal cellular protein (PrPc) which through post-translational modification produces the infectious prion protein (PrPsc). We have shown, using mass spectrometry, that synthetic peptides containing three or four copies of an octapeptide repeat sequence (PHGGGWGQ), found in a highly conserved N-terminal domain of PrP, preferentially bind copper over other metals. Peptides from the analogous region of chicken PrP, which contains an N-terminal repeat domain of the hexapeptide (NPGYPH), showed similar specificity for copper binding. In addition, gel filtration c

TPCK and quercetin act synergistically with vanadate to increase protein-tyrosine phosphorylation in avian cells.

We have found four compounds that act synergistically with the phosphotyrosine phosphatase inhibitor sodium orthovanadate (Na3VO4) to greatly increase the extent of protein-tyrosine phosphorylation in both uninfected chick embryo fibroblasts (CEF) and their Rous sarcoma virus-transformed counterparts (RSV-CEF). These four inhibitors fall into two categories: the chymotrypsin-specific protease inhibitors, tosyl-phenylalanine-chloromethyl ketone (TPCK) and N-carbobenzoxy-1-phenylalanine-chloromethyl ketone (ZPCK); and certain bioflavonoids which can competitively inhibit ATP-binding, quercetin a

The evolutionary landscape and serotypic dynamics of avian infectious bronchitis virus from spike protein.

Avian infectious bronchitis virus (IBV), a gammacoronavirus with substantial agricultural impact, offers a tractable model for dissecting coronavirus evolution. Here, we integrated 20 years of epidemiological surveillance with whole-genome sequence analysis of 624 IBV strains, including 136 newly isolated field samples, to investigate the evolutionary and structural dynamics of N-linked glycosylation at the spike protein. We identified three dominant glycosylation haplotypes defined by residues 51 and 77 of spike protein, which correlate with receptor-binding interfaces, clinical phenotypes, a

Generation and epitope mapping of monoclonal antibodies targeting the avian reovirus S1133 σC protein.

Avian reovirus (ARV) causes of multisystemic, immunosuppressive infectious diseases in poultry. Among them, the σC protein, a structural component of the outer capsid encoded by the S1 genome segment, plays a crucial role in viral attachment and cellular entry. In this study, we expressed the σC protein derived from the S1133 vaccine strain was expressed in a prokaryotic system and purified it. Then, using hybridoma technology, we generated two murine monoclonal antibodies (mAbs), designated 3H and 4B and shown to specifically recognize the ARV σC protein. In order to determine the linear B-ce

Mapping the mutational landscape of an avian retrovirus envelope protein across its evolutionary trajectory.

Many RNA viruses exhibit error-prone replication. Continuous generation of erroneous copies accelerates evolution. Avian leukosis virus subgroup J (ALV-J), an avian oncogenic virus, is a classical model virus for studying retroviruses. ALV-J's high mutation rates drive continuous evolution of its envelope and pathogenicity, posing significant challenges to the poultry industry. Here we employed deep mutational scanning to systematically assess envelope-wide mutation effects on ALV-J replication, integrating high-throughput sequencing with mutant libraries to identify critical envelope residues

Regulation of Agouti-Related Protein and Pro-Opiomelanocortin Gene Expression in the Avian Arcuate Nucleus.

The arcuate nucleus is generally conserved across vertebrate taxa in its neuroanatomy and neuropeptide expression. Gene expression of agouti-related protein (AGRP), neuropeptide Y (NPY), pro-opiomelanocortin (POMC), and cocaine- and amphetamine-regulated transcript (CART) has been established in the arcuate nucleus of several bird species and co-localization demonstrated for AGRP and NPY. The proteins encoded by these genes exert comparable effects on food intake in birds after central administration to those seen in other vertebrates, with AGRP and NPY being orexigenic and CART and α-melanocy

Mammalian adaptive mutations of the PA protein of highly pathogenic avian H5N1 influenza virus.

UNLABELLED: Highly pathogenic H5N1 influenza A viruses continue to circulate among avian species and cause sporadic cases of human infection. Therefore, the threat of a pandemic persists. However, the human cases of H5N1 infection have been limited mainly to individuals in close contact with infected poultry. These findings suggest that the H5N1 viruses need to acquire adaptive mutations to gain a replicative advantage in mammalian cells to break through the species barrier. Many amino acid mutations of the polymerase complex have been reported to enhance H5N1 virus growth in mammalian cells;

Influenza M2 envelope protein augments avian influenza hemagglutinin pseudotyping of lentiviral vectors.

Lentivirus-based gene transfer has the potential to efficiently deliver DNA-based therapies into non-dividing epithelial cells of the airway for the treatment of lung diseases such as cystic fibrosis. However, significant barriers both to lung-specific gene transfer and to production of lentivirus vectors must be overcome before these vectors can be routinely used for applications to the lung. In this study, we investigated whether the ability to produce lentiviral vectors pseudotyped with fowl plague virus hemagglutinin (HA) could be improved by co-expression of influenza virus M2 in vector-p

Avian and Human Seasonal Influenza Hemagglutinin Proteins Elicit CD4 T Cell Responses That Are Comparable in Epitope Abundance and Diversity.

Avian influenza viruses remain a significant concern due to their pandemic potential. Vaccine trials have suggested that humans respond poorly to avian influenza vaccines relative to seasonal vaccines. It is important to understand, first, if there is a general deficiency in the ability of avian hemagglutinin (HA) proteins to generate immune responses and, if so, what underlies this defect. This question is of particular interest because it has been suggested that in humans, the poor immunogenicity of H7 vaccines may be due to a paucity of CD4 T cell epitopes. Because of the generally high lev

Evaluation of recombinant Chinese avian hepatitis E virus (CaHEV) ORF2 and ORF3 proteins for protection of chickens against CaHEV infection.

Avian hepatitis E virus (HEV) is the etiologic agent of big liver and spleen disease in chickens. In 2010, the Chinese avian HEV (CaHEV) strain was isolated from chickens and demonstrated to cause the decreased egg production in layer hens. No avian HEV commercial vaccine has yet been developed to prevent virus infection in China. In this study, recombinant CaHEV truncated ORF2 and complete ORF3 proteins were evaluated separately for immunoprotection of chickens against CaHEV infection. First, truncated ORF2 and complete ORF3 proteins were expressed in Escherichia coli. Next, 48 specific-patho

Recombinant human adenovirus type 5-vectored vaccine expressing avian reovirus σC protein protects against prevalent genotype I avian reovirus in chickens.

Avian reovirus (ARV) is a globally important poultry pathogen causing substantial economic losses due to viral arthritis, enteritis and immunosuppression. Traditional vaccines based on the S1133 strain are less effective against circulating ARV variants due to genetic divergence. The σC protein is the primary antigen inducing neutralizing antibodies against ARV, making it a key target for vaccine development. Here, we constructed a recombinant human adenovirus type 5 (rHAdV5) vector vaccine expressing the σC protein of a prevalent Chinese ARV field strain (SDAU-1) and evaluated its safety, imm

Development of an indirect competitive ELISA based on σC protein for detection of antibodies against avian reovirus.

Avian reovirus (ARV) causes viral arthritis in poultry, and emerging variants in China demand improved serosurveillance. We developed an indirect competitive ELISA (ic-ELISA) using a monoclonal antibody against recombinant σC protein. After optimization (coating antigen 6.0 μg/mL, mAb 1:4000, blocking 90 min, serum 60 min, mAb 90 min), the cut-off percent inhibition was set at 44.5% based on 40 negative sera. The assay showed no cross-reactivity with six common avian pathogens, a detection limit of 1:400 serum dilution, and excellent reproducibility (intra-CV ≤0.70%, inter-CV ≤7.70%). Testing
