Chromosomal markerless integration of anthelmintic Cry proteins into the Bacillus thuringiensis genome.

Bacillus thuringiensis (Bt) is a Gram-positive bacterium that during sporulation produces insecticidal Crystal (Cry) proteins, which play a major role in insect control today. Some Bt Cry proteins, e.g., Cry5Ba, target nematodes and, when given orally, can cure animals of gastrointestinal nematode (GIN) parasites. To eliminate concerns about treating humans and animals with spores and live bacteria, we developed an asporogenous system for scalable and safe Cry protein delivery called IBaCC (Inactivated Bacteria with Cytosolic Crystal(s)), which results in production of a bioactive crystal and

Identification of the Extracytoplasmic Function σ Factor σP Regulon in Bacillus thuringiensis.

Bacillus thuringiensis and other members of the Bacillus cereus family are resistant to many β-lactams. Resistance is dependent upon the extracytoplasmic function sigma factor σP. We used label-free quantitative proteomics to identify proteins whose expression was dependent upon σP. We compared the protein profiles of strains which either lacked σP or overexpressed σP. We identified 8 members of the σP regulon which included four β-lactamases as well as three penicillin-binding proteins (PBPs). Using transcriptional reporters, we confirmed that these genes are induced by β-lactams in a σP-depe

Bacillus thuringiensis Cry14A family proteins as novel anthelmintics against gastrointestinal nematode parasites.

Bacillus thuringiensis crystal (Cry) proteins have been expressed in commercial transgenic crops for nearly 30 years, providing safe and effective control of insect pests and significantly reducing the application of hazardous chemical pesticides. B. thuringiensis crystal proteins have also been shown to target parasitic nematodes, including plant parasitic nematodes. Recently, transgenic soybean crops expressing Cry14Ab have been shown to provide control against the soybean cyst nematode Heterodera glycines, marking the first time a crystal protein is being commercialized in transgenic crops

Fluorescence assay based on aptamer-quantum dot binding to Bacillus thuringiensis spores.

A novel assay was developed for the detection of Bacillus thuringiensis (BT) spores. The assay is based on the fluorescence observed after binding an aptamer-quantum dot conjugate to BT spores. The in vitro selection and amplification technique called SELEX (Systematic Evolution of Ligands by EXponential enrichment) was used in order to identify the DNA aptamer sequence specific for BT. The 60 base aptamer was then coupled to fluorescent zinc sulfide-capped, cadmium selenide quantum dots (QD). The assay is semi-quantitative, specific and can detect BT at concentrations of about 1,000 colony fo

MclX modulates the process of mother cell lysis by SpoIIID in Bacillus thuringiensis.

UNLABELLED: Cell differentiation is essential for the survival and adaptation of organisms. In Bacillus species, sporulation is a tightly coordinated program that culminates in mother cell lysis, a critical step for spore release. Despite its importance, the mechanisms modulating the process remain poorly understood. Here, we show that MclX regulates mother cell lysis by inhibiting SpoIIID, a sporulation-specific transcription factor that represses cwlC, which encodes a key hydrolase required for lysis. SpoIIID directly binds to the cwlC promoter, and its overexpression during sporulation mark

A Sporulation-Independent Way of Life for Bacillus thuringiensis in the Late Stages of an Infection.

The formation of endospores has been considered the unique survival and transmission mode of sporulating Firmicutes due to the exceptional resistance and persistence of this bacterial form. However, nonsporulated bacteria (Spo-) were reported at the early stages following the death of a host infected with Bacillus thuringiensis, an entomopathogenic sporulating bacterium. Here, we investigated the characteristics of the bacterial population in the late stages of an infection in the B. thuringiensis/Galleria mellonella infection model. Using fluorescent reporters and molecular markers coupled to

Intraspecific Diversity and Pathogenicity of Bacillus thuringiensis Isolates from an Emetic Illness.

This study describes an emetic food-borne intoxication associated with a Bacillus cereus group species and the characterization of the bacterial isolates from the incident in aspects of molecular tying, genetic factors, cytotoxicity, and pathogenic mechanisms relating to emetic illness. Through the polyphasic identification approach, all seven isolates obtained from food and clinical samples were identified as Bacillus thuringiensis. According to multilocus sequence typing (MLST) analysis, intraspecific diversity was found within the B. thuringiensis isolates. Four allelic profiles were found,

Cellular metabolism network of Bacillus thuringiensis related to erythromycin stress and degradation.

Erythromycin is one of the most widely used macrolide antibiotics. To present a system-level understanding of erythromycin stress and degradation, proteome, phospholipids and membrane potentials were investigated after the erythromycin degradation. Bacillus thuringiensis could effectively remove 77% and degrade 53% of 1 µM erythromycin within 24 h. The 36 up-regulated and 22 down-regulated proteins were mainly involved in spore germination, chaperone and nucleic acid binding. Up-regulated ribose-phosphate pyrophosphokinase and ribosomal proteins confirmed that the synthesis of protein, DNA and

Characterisation of the binding properties of Bacillus thuringiensis 18 toxin on leukaemic cells.

BACKGROUND: Various strains of Bacillus thuringiensis (Bt) have been found to produce parasporal proteins that are cytotoxic to human cancer cells. This study aims to establish the binding affinity of purified Bt 18 toxin for CEM-SS (T lymphoblastic leukaemia cell line), to determine if competition exists between the toxin and commercial anticancer drugs for the binding site on CEM-SS and to localise the binding site of the toxin on CEM-SS. METHODS: In homologous competitive binding study, the purified toxin was labelled with biotin and allowed to compete with unlabelled toxin for binding sit

gyrB as a phylogenetic discriminator for members of the Bacillus anthracis-cereus-thuringiensis group.

Bacillus anthracis, the causative agent of the human disease anthrax, Bacillus cereus, a food-borne pathogen capable of causing human illness, and Bacillus thuringiensis, a well-characterized insecticidal toxin producer, all cluster together within a very tight clade (B. cereus group) phylogenetically and are indistinguishable from one another via 16S rDNA sequence analysis. As new pathogens are continually emerging, it is imperative to devise a system capable of rapidly and accurately differentiating closely related, yet phenotypically distinct species. Although the gyrB gene has proven usefu

Update on the detection of beta-exotoxin in Bacillus thuringiensis strains by HPLC analysis.

AIMS: The current work aimed to study the presence of beta-exotoxin by high-performance liquid chromatography (HPLC) in supernatant fluids from final whole cultures of the 69 type strains and 13 subtypes of Bacillus thuringiensis strains, as well as from some insecticidal strains. METHODS AND RESULTS: Results from HPLC and bioassays with Ephestia kuhniella (Lepidoptera Pyralidae) were compared. Type I beta-exotoxin was only detected in type strains representing serotypes H1, H9 and H10a,10b. Discrepancies between HPLC and bioassays were found in H8a,8b and some insecticidal strains, which sug

[Effect of mineral salts on exotoxin formation and productivity of a Bacillus thuringiensis culture].

The effect of ten mineral salts on the productivity and toxin synthesis was studied in Bacillus thuringiensis IPM-1140. Exotoxin synthesis was stimulated by Zn2+, Mn2+ and NH4+ ions as well as by potassium phosphates. The direct correlation between the number of viable spores and the exotoxin accumulation was disordered at extreme salt concentrations. Optimal salt concentrations in the yeast-polysaccharide medium were found using the method of a fractional factor experiment, which made it possible to increase the productivity of the culture to 5 X 10(9) spores/ml and the yield of the exotoxin

Characterization of monoclonal antibodies to a crystal protein of Bacillus thuringiensis subsp. kurstaki.

Ten monoclonal antibodies were produced against a k-1-type crystal protein of Bacillus thuringiensis subsp. kurstaki. Eight of the antibodies belong to the immunoglobulin G1 (IgG1) subclass, with pI values ranging from 5.5 to 8.6, one could be assigned to the IgG2b subclass, and one could be assigned to the IgM class. Competitive antibody-binding assays and analysis of antibody specificity indicated that the 10 antibodies recognized at least nine distinct antigenic determinants. Eight antibodies bound to both protoxin and toxin, whereas the other two interacted with protoxin only. One antibody

[Toxicity of Bacillus thuringiensis var. israelensis for Simulium larvae, vectors of onchocerciasis].

On field trials in infested streams, the primary powder R 153-78 made of B. thuringiensis var. israelensis with a potency of 3,000 I.T.U. Aedes aegypti/milligramme is very toxic for Blackfly larvae, especially for Simulium damnosum s.l. larvae. The lethal concentration 100, in 24 h. is 0.2 X 10(-6) for a 10 min. treatment. This high toxicity, specific for Diptera larvae such as Mosquitoes and Blackflies, is related to the special characteristic of the bacterial crystal protein which has a clear serological and chemical individuality compared to the crystal proteins of the other B. thuringiensi

Immuno-physiological adaptations confer wax moth Galleria mellonella resistance to Bacillus thuringiensis.

Microevolutionary mechanisms of resistance to a bacterial pathogen were explored in a population of the Greater wax moth, Galleria mellonella, selected for an 8.8-fold increased resistance against the entomopathogenic bacterium Bacillus thuringiensis (Bt) compared with a non-selected (suspectible) line. Defense strategies of the resistant and susceptible insect lines were compared to uncover mechanisms underpinning resistance, and the possible cost of those survival strategies. In the uninfected state, resistant insects exhibited enhanced basal expression of genes related to regeneration and a

Urea-mercaptoethanol-soluble protein from spores of Bacillus thuringiensis and other species.

Treatment with urea-mercaptoethanol of purified spores of Bacillus thuringiensis, other Bacillus species, and Clostridium roseum solubilizes a protein fraction between 5 and 12% of the dry weight of the spores. This fraction behaves identically to the crystal protein of B. thuringiensis on acrylamide-gel electrophoresis. The protein from all of the Bacillus species shows partial homology with crystal protein, using the Ouchterlony immunodiffusion technique. A further fraction, similar in amount, can be removed from spores of B. thuringiensis by the addition of sodium lauryl sulfate to the urea

Cry1A toxins of Bacillus thuringiensis bind specifically to a region adjacent to the membrane-proximal extracellular domain of BT-R(1) in Manduca sexta: involvement of a cadherin in the entomopathogenicity of Bacillus thuringiensis.

Many subspecies of the soil bacterium Bacillus thuringiensis produce various parasporal crystal proteins, also known as Cry toxins, that exhibit insecticidal activity upon binding to specific receptors in the midgut of susceptible insects. One such receptor, BT-R(1) (210 kDa), is a cadherin located in the midgut epithelium of the tobacco hornworm, Manduca sexta. It has a high binding affinity (K(d) approximately 1nM) for the Cry1A toxins of B. thuringiensis. Truncation analysis of BT-R(1) revealed that the only fragment capable of binding the Cry1A toxins of B. thuringiensis was a contiguous 1

Discovery of a significant optical chromatographic difference between spores of Bacillus anthracis and its close relative, Bacillus thuringiensis.

A significant difference between two closely related Bacillus spores has been discovered using optical chromatography. This difference can be harnessed for the separation of microscopic particles using opposing laser and fluid flow forces. Particles of different size, composition, and shape experience different optical and fluid forces and come to rest at unique equilibrium positions where the two forces balance. Separations in excess of 600 mum have been observed between Bacillus anthracis Sterne strain and its genetic relative, Bacillus thuringiensis. These findings open new possibilities fo
