Plasma-based fluorine ion implantation into dental materials for inhibition of bacterial adhesion.

The aims of this study were to evaluate the fluorine depth profiles of pure titanium (Ti), stainless steel (SUS), and polymethyl methacrylate (PMMA) modified by plasma-based fluorine ion implantation and the effects of fluorine ion implantation on contact angle, fluoride ion release, and S. mutans adhesion. Fluorine-based gases used were Ar+F2 and CF4. By means of SIMS, it was found that the peak count of PMMA was the lowest while that of Ti was the highest. Then, up to one minute after Ar sputtering, the presence of fluorine and chromic fluoride could be detected by XPS in the surface and sub

Hematogenous bacterial meningitis in an intercellular adhesion molecule-1-deficient infant mouse model.

Mice genetically deficient in the gene encoding for intercellular adhesion molecule-1 (ICAM-1) production were compared with wild-type mice after injection with Haemophilus influenzae type b (Hib) or Streptococcus pneumoniae. The incidence of Hib bacteremia was greater in the ICAM-1-deficient mice than wild-type mice (P = .007), but mortality was greater for wild-type mice at 24 h (P = .03). In contrast, the incidence of S. pneumoniae bacteremia was equivalent but mortality was greater in ICAM-1-deficient mice at 24 h (P = .0003). More ICAM-1-deficient mice had cerebrospinal fluid cultures (CS

Adsorption of enterobactin to metal oxides and the role of siderophores in bacterial adhesion to metals.

The potential contribution of chemical bonds formed between bacterial cells and metal surfaces during biofilm initiation has received little attention. Previous work has suggested that bacterial siderophores may play a role in bacterial adhesion to metals. It has now been shown using in situ ATR-IR spectroscopy that enterobactin, a catecholate siderophore secreted by Escherichia coli, forms covalent bonds with particle films of titanium dioxide, boehmite (AlOOH), and chromium oxide-hydroxide which model the surfaces of metals of significance in medical and industrial settings. Adsorption of en

[Inhibition of bacterial adherence by nitroxoline on cellular adhesion and on urinary catheter surfaces].

Nitroxolin or 5-nitro-8-hydroxyquinoline, used in the treatment of acute or recurrent uncomplicated urinary tract infection (UTI), has been investigated to demonstrate inhibitory effect on bacterial adherence to epithelial cells or solid surfaces. Nitroxolin in vitro and in urine inhibits bacterial adherence of E. coli 38 (MS/MS) on HeLa cells and epithelial cells from human bladder mucosa. In the same conditions, norfloxacin has no effect. Nitroxolin (MIC/8) decreases with a statistically significant difference (p less than 0.001) the bacterial attachment to a urinary catheter surface made in

[Bacterial adhesion to epithelial cells of the nasopharynx essential in the development of otitis media].

Otitis media develops when certain bacterial pathogens gain access to the middle ear cavity from the nasopharynx through the eustachian tube. Adhesion of bacteria, in particular Streptococcus pneumoniae and Haemophilus influenzae, to the non-ciliated epithelial cells of the nasopharynx, close to the opening of the eustachian tube, is significantly correlated to the otitis-prone condition in children. Otitis-prone children have significantly fewer bacteria in the nasopharynx coated with the immunoglobulin secretory IgA (SigA) then healthy children have. Adhesion and occurrence of middle ear pat

Niobium-Based Conditioning Layer to Reduce Bacterial Adhesion and Biofilm Formation on Titanium Surface.

Niobium metal has a wide range of applications; however, the development of Nb-coated surfaces with antimicrobial activity remains unexplored. This study investigates the antimicrobial and antibiofilm activities of ammoniacal niobium oxalate (ANO) and develops a methodology to deposit it on titanium-functionalized surfaces to prevent bacterial colonization and biofilm formation. ANO is dispersed in water and characterized for particle size, Fourier transform infrared spectroscopy, X-ray diffraction, ζ-potential, and in vitro cytotoxicity. Its antimicrobial activity is assessed by microdilution

Nanostructured Ti-6Al-4V Reduces Adhesion of Several Bacterial Species: An In Vitro Study.

Biomaterial-associated infections (BAIs) and insufficient early cellular response remain critical challenges for orthopedic implants. We introduce a comprehensive study that bridges current knowledge gaps by examining an early-stage antimicrobial effect on the clinically relevant alloy Ti-6Al-4V. It combines pathogenic strains with parallel osteoblast assays and tilted-view SEM analysis to obtain a qualitative understanding of the adhesion mechanisms. Detailed physicochemical characterization revealed a progressive increase in nanoscale roughness and oxide layer thickness, accompanied by selec

Hydrophobic 2-methacryloyloxyethyl phosphorylcholine polymer inhibits peri-implantitis-causing bacterial adhesion on titanium materials.

AIMS: To prevent peri-implantitis, we investigated the adhesion of periodontopathogenic bacteria to titanium surfaces using a hydrophobic 2-methacryloyloxyethyl phosphorylcholine (MPC) polymer to inhibit adhesion. METHOD AND RESULTS: We immersed titanium plates (TiPs) coated with a hydrophobic MPC polymer in a bacterial suspension for 30 min or 24 h and measured the number of adherent bacteria. Bacteria adhering to the TiPs were observed using scanning electron microscopy (SEM). Furthermore, mimicking an oral cavity, TiPs coated with MPC polymer and saliva, were immersed in bacterial suspensi

A main chain biodegradable polyurethane with anti-protein adsorption and anti-bacterial adhesion performances.

Biofilms are initially formed by substances such as proteins secreted by bacteria adhering to a surface. To achieve a durable antibacterial material, biodegradable dihydroxyl-terminated poly[(ethylene oxide)-co-(ethylene carbonate)] (PEOC(OH)2) with anti-protein adsorption properties was synthesized in this study. Further polycondensation of PEOC(OH)2 and isophorone diisocyanate (IPDI) led to biodegradable polyurethane (PEOC-PU) with PEOC as the soft segment. For comparison, polyurethanes with polyethylene glycol (PEG-PU) and polypropylene glycol (PPG-PU) as soft segments were also synthesized

Staphylococcus aureus endocarditis: distinct mechanisms of bacterial adhesion to damaged and inflamed heart valves.

AIMS: The pathogenesis of endocarditis is not well understood resulting in unsuccessful attempts at prevention. Clinical observations suggest that Staphylococcus aureus infects either damaged or inflamed heart valves. Using a newly developed endocarditis mouse model, we therefore studied the initial adhesion of S. aureus in both risk states. METHODS AND RESULTS: Using 3D confocal microscopy, we examined the adhesion of fluorescent S. aureus to murine aortic valves. To mimic different risk states we either damaged the valves with a surgically placed catheter or simulated valve inflammation by

Evaluation of bacterial adhesion in exposed orbital implants using electron microscopy and microbiological culture.

Three cases are presented of anophthalmic patients with exposed orbital implants. Although only one patient showed evident clinical signs of infection, all three implants were studied to determine the presence of microorganisms adhered to their surface using a scanning electron microscopy (SEM) and microbiological culture. The SEM allowed the visualisation of microorganisms adhered to the three implants, although only one of them showed growth in the microbiological cultures. In addition, the SEM technique used in case No.3 achieved a better orientation and appreciation of the microorganisms w

Surface immobilization of kanamycin-chitosan nanoparticles on polyurethane ureteral stents to prevent bacterial adhesion.

Bacterial adhesion is a major problem that can lead to the infection of implanted urological stents. In this study, kanamycin-chitosan nanoparticles (KMCSNPs) were immobilized on the surface of a polyurethane ureteral stent (PUS) to prevent urinary bacterial infection. KMCSNPs were synthesized using the ionic gelation method. The synthesized KMCSNPs appeared spherical with a ζ-average particle size of 225 nm. KMCSNPs were immobilized on the PUS surface by covalent immobilization techniques. The surface-modified PUS was characterized using attenuated total reflectance Fourier transform infrared

Prior contact lens wear reduces susceptibility of the superficially-injured cornea to bacterial adhesion.

Contact lens wear in humans and mice is consistently associated with asymptomatic corneal parainflammation. Here, we tested the hypothesis that the corneal response to lens wear alone can function to protect it against bacterial adhesion enabled by superficial injury. One eye only of mT/mG-LysMcre mice (cell membranes red; Lyz2+ cells green) wore a contact lens for 4-6 days. After lens removal, mice were anesthetized and both corneas superficially-injured before bacterial inoculation with either a mouse eyelid commensal (Macrococcus epidermidis) or a corneal pathogen (Pseudomonas aeruginosa).

ST6GALNAC5 promotes Glaesserella parasuis infection by enhancing bacterial adhesion and amplifying host inflammatory responses.

Glaesserella parasuis (G. parasuis), the causative agent of Glässer's disease in pigs, relies on adhesion to host epithelial cells and induction of inflammation for pathogenesis. However, the potential role of ST6GALNAC5, a host sialyltransferase involved in glycan modification, in G. parasuis infection remains unclear. Using the porcine kidney epithelial cell line LLC-PK1, we investigated the function of the host sialyltransferase gene ST6GALNAC5 during G. parasuis infection. Infection significantly upregulated ST6GALNAC5 mRNA expression. CRISPR/Cas9-mediated knockout of ST6GALNAC5 reduced ba

Long-Term Reduction of Bacterial Adhesion on Polyurethane by an Ultra-Thin Surface Modifier.

Indwelling urinary catheters are employed widely to relieve urinary retention in patients. A common side effect of the use of these catheters is the formation of urinary tract infections (UTIs), which can lead not only to severe medical complications, but even to death. A number of approaches have been used to attempt reduction in the rate of UTI development in catheterized patients, which include the application of antibiotics and modification of the device surface by coatings. Many of these coatings have not seen use on catheters in medical settings due to either the high cost of their imple

Human Enteric α-Defensin 5 Promotes Shigella Infection by Enhancing Bacterial Adhesion and Invasion.

Shigella is a Gram-negative bacterium that causes bacillary dysentery worldwide. It invades the intestinal epithelium to elicit intense inflammation and tissue damage, yet the underlying mechanisms of its host selectivity and low infectious inoculum remain perplexing. Here, we report that Shigella co-opts human α-defensin 5 (HD5), a host defense peptide important for intestinal homeostasis and innate immunity, to enhance its adhesion to and invasion of mucosal tissues. HD5 promoted Shigella infection in vitro in a structure-dependent manner. Shigella, commonly devoid of an effective host-adhes
